Mesenchymal stem cells (MSCs) are multipotent cells able to differentiate into several cell lineages, which has implications for cell therapy and reproductive biotechnologies. Although MSCs have been isolated from many species, including humans and animals, there is limited data on MSCs from large ruminants, such as bovines. In this study, we tried to isolate and characterize bovine tongue tissue-derived MSCs (boT-MSCs) by investigating phenotype morphology, performing proliferation properties, and determining cell surface marker expression patterns, self-renewal, and differentiation potentials. As a result, the boT-MSCs were successfully isolated by collagenase digestion and maintained proliferative capacity until 20 passages. Moreover, the boT-MSCs expressed pluripotency markers (OCT3/4, SOX2, and NANOG) and MSC-specific surface markers including CD44, CD90, and CD105, but not CD45 and MHC-II. The boT-MSCs could also differentiate into mesodermal (adipocyte, osteocyte, and chondrocyte) cell lineages. Our results suggest that the tongues of bovines could be used as a source of MSCs.
This study was conducted to Jindo dogsprovide whether Jindo dogs can distinguish cancer cells, early detection. Jindo dogs were trained for cancer detection based on obedience, smell recognition, and tracking. Cancer detection training was designed to enable detection of lung cells (A549), prostate cells (PC 3), or breast cells (MCF7) among 9 pure media. Four Jindo dogs were trained by the same trainer and our different results were obtained (true positive, true negative, false positive, false negative). The accuracy was 85%, 82%, 92%, and 81% for sensitivity and 95%, 89%, 95%, and 93% for specificity. In addition, 4 Jindo dogs can find what is the cancer cell cannot distinguish 3 kinds of (A549, PC3, MCF7). Jindo dogs can detect cancerous cells through human’s breath and urine.
Assays for detecting pregnancy-associated glycoproteins (PAGs) levels in the blood of pregnant cattle have been developed and commercialized to determine pregnancy status. This study was conducted to determine the accuracy of a PAG-based ELISA test for pregnancy detection in Hanwoo. The anticoagulated treated whole blood was sampled from 595 Hanwoo that were 28-71 d (mean 49) and 85-128 d (mean 106) after artificial insemination. At the 2nd blood collection time, a transrectal palpation test was used as the standard for pregnancy diagnosis. For PAGs and the transrectal palpation test (n=595), the sensitivity was 98.4% (pregnancy, 481/489), the specificity was 100% (open, 106/106), and the accuracy was 98.7% (pregnancy+open, 587/595). In conclusion, the rapid PAG-based ELISA pregnancy test was highly sensitive and specific for pregnancy detection in Hanwoo.
Mesenchymal stem cells (MSCs) have restricted life spans in vitro and can therefore only be expanded for a limited number of cell divisions before entering a senescent state and unequivocally stopping proliferation. Several types of cell culture systems have been used for large-scale expansion of MSCs. A recent trend in cell culture has been the change from serum-use to serum-supplement media. This study was conducted to investigate the proliferative effects of vegetable resources (VR) on equine adipose tissue-derived mesenchymal stem cells (eAD-MSCs) in the absence of serum and their possible mechanisms of action. Regulation of cell cycling is a key process involved in the fate of stem cells, including renewal and differentiation. In this study, we observed that the viability of eAD-MSCs was increased significantly when treated with VR under serum-free conditions. We also observed that expression levels of cell cycling-related proteins such as p53 and p21 were decreased, and proliferating cell nuclear antigen increased significantly in response to treatment with VR in eAD-MSCs under serum-free conditions. Furthermore, expression levels of cell survival-related proteins were increased in response to treatment with VR in eAD-MSCs under serum-free conditions. Therefore, our results suggest that VR promotes proliferation of eAD-MSCs under serum-free conditions.
The hepatitis E virus (HEV) is a leading causative agent of acute hepatitis in humans. Zoonotic HEV strains have been isolated from several animal species, including pigs. New HEV variants have been recently isolated from camels in the Middle East. In the present study, fecal samples from fallow deer, formosan deer, alpaca, and guanaco were analyzed for the detection of HEV. One HEV strain was detected from guanaco, a species of camelids. The nucleotide sequence of guanaco HEV was identical to those of deer HEV-3 strains, which implied the cross-species transmission of HEV-3 from deer to guanaco.
This study investigated the synergistic effects of a combination of cephalexin and gentamicin (CCG) against Streptococcus spp. isolated from bovine milk. Minimum inhibitory concentrations (MICs) and fractional inhibitory concentration (FIC) index were determined for 12 bacteria isolated from 12 bovine farms in Gyeongnam province. The MIC ranges of cephalexin, gentamicin and CCG were 0.25-0.5, 0.5-2.0 and 0.06-0.125 μg/mL, respectively. In addition, the FIC index range of CCG against the isolates was 0.32-0.34, showing a synergistic effect in the checkboard assay. In conclusion, CCG has a high antibacterial activity against Streptococcus isolates and may be utilized in the treatment of bovine streptococcal mastitis.
The current study identified risk factors associated with porcine circovirus type 2 (PCV2) infection on pig farms in the Republic of Korea using a multinomial logistic regression model to evaluate the PCV2 infection status of pigs at different growth stages. Compulsory disinfection of visitors (odds ratio [OR]: 0.019, 95% confidence interval [CI]: <0.001–0.378, p=0.0095), compulsory registration of visitors (OR: 0.002, 95% CI: <0.001–0.184, p=0.0070), regular blood testing (OR: 0.012, 95% CI: <0.001–0.157, p=0.0007), and running on-farm biosecurity learning programs for workers (OR: 0.156, 95% CI: 0.040–0.604, p=0.0072 and OR: 0.201, 95% CI: 0.055–0.737, p=0.0155, respectively) were identified as factors which could reduce the risk of PCV2 infection. However, visitation by a regular veterinarian (OR: 32.733, 95% CI: 3.768–284.327, p=0.0016) was associated with PCV2 infection.
This study examined the acute toxicity of Vital-Shoot containing reduced glutathione in male and female ICR mice. Mice were intraperitoneally injected the drug at dose levels of 0, 250, 500, 1,000 and 2,000 mg/kg body weight (BW) for single-dose toxicity test. There were no statistical differences in BW changes between the control and all treated groups. Based on hematological and blood biochemical analyses, the drug did not affect all parameters. In addition, markers for liver and kidney functions did not meaningfully change in all treated groups. Since there were no adverse effects from the drug in a single-dose intraperitoneal toxicity test, it was concluded that the lethal dose 50 (LD50) of Vital-Shoot is estimated to be >2,000 mg/kg BW.
