Canine mammary tumors account for ~30% of all tumors in the female dogs and approximately 50% of the tumors are malignant. Exosomes have been the focus of great interest, as they appear to be involved in numerous important cellular processes. In this study, we examined the anti-tumor effects of canine mesenchymal stem cells-derived exosomes (MSC-exosomes) in an experimental murine mammary tumor model using canine mammary carcinoma cells, REM134. The MSC-exosomes were injected tumor site and tail vein of REM134 xenografted mice. We found that tumor size of the MSC-exosomes-treated group decreased compared to those of the only tumor group in REM134-driven tumorigenic mouse model. In addition, the MSC-exosomes-treated tumor group showed meaningfully reduced expression levels of the MMP-3, IL-1β, IL-6, and TNF-α compared to those in the tumor group. Specifically, we confirmed that the expression level of the CD133, potent cancer stem cell (CSC) markers, decreased in the MSC-exosomes-treated tumor group compared to the tumor group. This study suggests that the MSC-exosomes exhibited anti-tumor effects through downregulating CSC-related markers in the canine mammary tumor murine model. Further study is needed in the future, and we are conducting research on the detailed anti-tumor mechanism of the MSC-exosomes.

Biosilica is a material extracted from the shell (cell wall) of Melosira nummuloides, a type of sea diatom, and is one of the widely distributed biominerals. Recently, some studies have revealed that biosilica has a characteristic bio-modulatory activity compared to synthetic silica. However, there has been little research on the effects and action mechanisms of biosilica in immune cells. In this study, we investigated the effect of biosilica water on mouse dendritic cells (DCs), the most potent antigen-presenting cells in immunity, and whether it may alter the function of DCs. Biosilica water decreased the metabolic activity of DCs at 20% concentration (v/v) and the production of IL-1 beta in a concentration-dependent manner. And also, more cells with fragmented nuclei were observed in the DCs treated with 20% biosilica water compared to other treatment groups. The mixed leukocyte response experiment showed the biosilica water-treated DCs significantly modulated the metabolic activity and proliferation of allogeneic spleen cells compared to control DCs. This result suggests that biosilica water may modulate the antigen-presenting capability of DCs. Taken together, this study shows the immunomodulatory activity of biosilica on DCs and may affect immune responses. Further research is needed to investigate the immunological activity of biosilica water.

The purpose of this study is to obtain data on the zones formed by the movement of livestock vehicles and to determine if such areas can be used to establish quarantine activities and quarantine policies for livestock epidemics. For this purpose, this study used mobile data on poultry-related livestock vehicles generated in 2019. InfoMap, a community detection method, was used for regional classification, and the results of the analysis were visualized on a map using GIS. The study results confirmed that the zone of the administrative unit can be classified based on the movement of livestock vehicles. In addition, the zones created by the vehicle movement could be seen to change depending on the purpose and timing of the operation of livestock vehicles. Some areas form relatively stable zones, such as Jeolla-do and Gyeongsang-do, while others change depending on the situation, such as Chungcheong-do, Gyeonggi-do, and Gangwon-do. Further, the zones derived for poultry differed from those derived for cattle and pigs in previous studies.

This study was to evaluate the skin sensitization of potassium hydroxide (KOH) by the guinea pig maximization test. In the preliminary test, the induction and challenge dose of KOH were determined as 1.0 and 0.2%, respectively. Guinea pigs were sensitized with intradermal injection of 0.1ml KOH for 24 h. After 1 week, 1.0% of KOH was treated on the site of injection and 0.2% of KOH was challenged 2 weeks later. The average skin reaction evaluated by erythema and edema on the challenge sites and sensitization rate in the KOH-treatment group at 24 h were 0.0 and 0%, respectively, which are substantially low compared with in the 2,4-dinitrochlorobenzene-treatment group (positive control group) (average skin reaction: 3.0, sensitization rate: 100%) and identical with in the distilled water-treatment group (negative control group), representing a weak sensitizing potential. The average skin reaction and sensitization rate observed at 48 h were 0.0 and 0% in the KOH-treatment group, respectively, and 2.0 and 100% in the positive control group, respectively. It was concluded that 1.0% of KOH classified to Grade I, induced no sensitization when tested in guinea pigs.

This study investigated the residue of tulathromycin (TLM) on the lung tissue of pigs intramuscularly injected with 2.5 mg/kg body weight as a single dose. Nine healthy cross swine were intramuscularly injected with the drug. Three treated animals were arbitrarily selected to be sacrificed at 7, 10 and 14 days after treatment. TLM residue concentrations in lung tissue were determined using liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). The drug was extracted from lung samples using acetonitrile followed by clean-up with n-hexane. The analyte was separated on a Hypersil GOLDTM liquid chromatography column using 0.1% formic acid in deionized distilled water and acetonitrile. The correlation coefficient (R2) of the calibration curve was 0.9981, and the limits of detection and quantification were 10 and 50 μg/kg, respectively. Recoveries at three spiking levels ranged between 95.0-98.2%, and relative standard deviations were less than 3.95%. In TLM-treated group, the concentration of TLM on 7, 10 and 14 days post-treatment was 627.3, 496.9, 259.1 μg/kg, respectively. The developed method is sensitive and reliable for detection of TLM in porcine lung tissues. In addition, it can detect below the maximum residue limits in animal-derived food products destined for human consumption.

Jellyfish envenomation is a world-wide health problem, which often seriously affect the fishery and bathing activities. To date, few individual jellyfish venoms proteins have been thoroughly characterized yet. In this work, four species of scyphozoan jellyfish (Nemopilema nomurai, Cyanea nozakii, Aurellia aurita and Rhopilema esculentum) are compared according to their, cytotoxicity, hemolytic potency, brine shrimp toxicity and protein components. Jellyfish venoms showed higher cytotoxicity in H9C2 heart myoblast than in C2C12 skeletal myoblast, with the exception of C. nozakii venom. This result suggests that the selective cytotoxic effects may be possibly related to their in vivo effects of cardiac tissue dysfunction. On the other hand, hemolytic activity could be also observed from all tested jellyfish venoms. N. nomurai jellyfish venom displaying the greatest hemolytic activity. As an alternative method of evaluating the toxicities of jellyfish venoms, the toxicity on brine shrimp was examined with the four jellyfish venoms. From this, the venom of N. nomurai showed higher toxicity against brine shrimp than the other jellyfish venoms, which is consistent with the results of cytotoxicity assay as well as hemolysis assay of the present study. SDS-PAGE analysis of four jellyfish venoms showed the similar pattern with molecular weight of around 40 kDa, and appeared to be the major protein components. These results provided that N. nomurai jellyfish venom was potently toxic than other scyphozoan jellyfish venoms and may explain to some extent the deleterious effects associated with human envenoming.

The current study explored the epidemiological associations between the 2016/18 highly pathogenic avian influenza (HPAI) epidemics and spatial factors, including the distance from a poultry farm to the closest groundwater source, migratory bird habitat, eco-natural area, and poultry farm altitude. We included 14 spatial factors as independent variables. The variables were used in the original continuous measurement format. In total, 288 poultry farms (144 HPAI-confirmed and 144 non-confirmed) were used as the dependent variable. In addition, the variables’ continuous measurement was converted to a categorical measurement format by using a general additive model. For risk factor analysis based on the continuous measurements of spatial factors, the non-graded eco-natural area distance (odds ratio [OR]: 1.00) and the grade one eco-natural area distance (OR: 0.99) were statistically significant independent variables. However, in the risk factor analysis based on the categorical measurement format of the spatial factors, the non-graded eco-natural area distance (OR: 0.08) and poultry farm altitude (OR: 0.44) were statistically significant independent variables in both a univariate and multiple logistic regression model. In other words, when a poultry farm was located far from the non-graded eco-natural area or in a highland area, the likelihood of an HPAI epidemic would decrease. From an HPAI control perspective, it is recommended that the government apply increased levels of biosecurity measures, such as bird-nets, fences, intensive disinfection of equipment, and regular bird health monitoring, for poultry farms located near non-graded eco-natural areas or in a lowland area.

The current study explored the movement characteristics of 14 migratory bird species that wintered in the Republic of Korea between 2014 and 2020. The migratory bird movement information was obtained via a global positioning system operated by the Korean government. The velocity of movement, number of clusters, and size of clusters of the migratory bird species during their movement from their departing country to the Republic of Korea were estimated by applying a method based on density-based spatial clustering of applications with noise. The average movement velocity of pintails (Anas acuta) that departed from China or Russia was 32.77 km/h, the highest velocity among those measured for the 14 migratory bird species. The average number of clusters for cinereous vultures (Aegypius monachus) was 43.00, which was the largest cluster number observed. However, herring gulls (Larus argentatus) had the largest cluster area with an average cluster radius of 27.43 km while wintering in the Republic of Korea. The findings of the current study could be useful in increasing the effectiveness of the Korean national highly pathogenic avian influenza (HPAI) surveillance program. The human and material resources of the HPAI surveillance could be allocated after considering the results of this study, revealing the movement characteristics of wintering migratory birds in Korea. The HPAI surveillance program should include fecal or swab sampling to detect the HPAI virus in both pintail and bean goose (Anser faballis) wintering sites. Sampling of those sites should have a higher priority than that for other migratory bird wintering sites since pintail and bean goose move faster and form larger clusters.

We investigated the cytotoxic potential of three different commercially available absorbent feminine hygiene products and one transdermal patch using direct contact and extract exposure methods. Two different cell lines were used – mouse fibroblast L929 and normal human skin fibroblast CCD-986sk cell lines. The test samples were extracted using three different methods in accordance to International Organization for Standardization (ISO). Viability of cells was analyzed using MTT assay and morphology of the cells were also observed using phase contrast microscopy. Overall, the direct contact method using L929 cells showed that all the test samples had no toxic effect when exposed to extracts for 1 h. For the exposure method, no toxic effect was observed in both L929 and CCD986sk cells incubated with all the test samples regardless of the extraction methods used.

Since the first detection of the African swine fever (ASF) virus in the Republic of Korea in 2019, the Korean government has applied interventions, including fencing, increasing the biosecurity level at domestic pig farms, and the capture-and-removal of wild boars. In particular, wild boars are an important risk factor for ASF control because they can spread disease among susceptible animals, such as wild boars or domestic pigs. A capture-and-removal method aims to reduce the likelihood of ASF transmission from wild boars to domestic boars or among wild boars by decreasing the number of susceptible wild boars. This study estimated the required number of wild boars captured and removed for ASF control using population viability analysis. Population factors, such as a life span, sex ratio, or an inbreeding depression with different capture-and-removal proportions of wild boars, were included in the analysis. Ten scenarios with different capture-and-removal proportions of wild boars and different periods of culling were considered. According to the results, a method in which 75% of wild boars are captured-and-removed for at least three years showed long-term effectiveness for more than ten years. The current ASF control method, in which 33% of wild boars are captured-and-removed, decreased the number of wild boars for three years, after which the wild boar population increased to more than its initial number. Given the limited human and material resources for controlling ASF in the Republic of Korea, it is recommended that resources be prioritized to increase the capture-and-removal proportion of wild boars to take full advantage of the ASF-control effectiveness.

The treatment of microbial infections requires a large use of antibiotics which is, partially, responsible for the appearance of resistant strains to antibiotics in dairy cows. However, many photochemical compounds, including the essential oils, are becoming interesting as potential source of natural bioactive molecules. The antibacterial activity of the studied essential oils was evaluated by aromatogram and microdilution in medium liquid. The results show that the essential oil of Thymus fontanesii has the biggest antibacterial action against all the bacterial strains comparing with the Eucalyptus oil. However, the aqueous extracts of Thymus fontanesii and Eucalyptus indicate a moderate antibacterial activity against the strains of Staphylococcus aureus. On the other hand there is no effect on Escherichia coli strains. The most strong activity inhibitory was get with the essential oil of Eucalyptus against Staphylococcus aureus strains with CMI of 0.39 μl/ml for Staphylococcus aureus ATCC and 1.562 μl/ml for Staphylococcus aureus pathogen in comparison with the essential oil of Thymus fontanesii which is more active against the Escherichia coli strains. The aqueous extract of Eucalyptus showed the best CMI and CMB against the Staphylococcus aureus strains in comparison with the aqueous extract of Thymus fontanesii. Comparative tests to the activity have been made with cefoxitin and gentamicin discs.

This study investigated the regenerative and healing effect of spirulina on corneal epithelial injury by acid burn in zebrafish. Corneal epithelium of adult zebrafish was evaluated at 0, 12, 24, 48 h after acid burn injury with or without immersion with 0.02 and 0.04 mg/mL of spirulina water extract. The pathological changes of corneal epithelium were assessed by light microscopy. Gene and protein expressions of proliferating cell nuclear antigen (PCNA), matrix metalloproteinase (MMP) 9 and MMP13 were determined by immunohistochemistry or real-time PCR. Corneal epithelium was denuded totally after acid burn and gradually regenerated. PCNA-positive cells significantly increased in spirulina treated group. MMP9 and MMP13 mRNA transcripts were significantly decreased in spirulina treated groups. In conclusion, spirulina has regenerative and healing properties by increased keratocyte proliferation, and inhibited extracellular matrix degradation during the regeneration process of corneal epithelial injury by acid burn in zebrafish.

The present study investigated the intra-vitam larval migration of Anisakis pegreffii, which is naturally infected, from the intestine to the viscera and the musculature of live spotty-bellied greenling, Hexagrammos otakii, under the different seawater temperatures. The results showed that the increase in the ratio of larval migration from the intestine to the viscera correlated with the increase in the water temperature in the aquarium. The intra-vitam larval migration ratios from the intestine to the viscera were 60% (63/105) at 15°C, 1.64% (3/183) at 12°C, 1.16% (2/172) at 9°C and 1.42% (2/141) in the control group (7.5°C). The free larvae showed no, slight and strong movement at 9°C, 12°C and 15°C, respectively, in saline water, and the larvae were present in a coiled state at 9°C and 12°C. The results suggest that temperature plays an important role in the intra-vitam larval migration of A. pegreffii. In addition, the free larval behavior of A. pegreffii in vitro increased with increase in temperature. Therefore, to prevent human anisakiasis caused by the consumption of raw fish, it is important that the temperature of the aquarium must be maintained below 12°C, which is the critical temperature zone.

Escherichia coli (E. coli) can easily contaminate the bovine mammary gland; hence, monitoring of bulk tank milk (BTM) samples helps detect mastitis. In this study, we investigated the characteristics of E. coli from BTM of a dairy company. A total of 58 BTM batches were collected from 29 dairy farms of a dairy company, and 40 E. coli were tested in this study. E. coli showed the highest resistance to cephalothin (52.4%), followed by colistin (28.6%), ampicillin (19.0%) and tetracycline (19.0%). However, E. coli showed low resistance from 0% to 9.5% against other 14 antimicrobials. In the distribution of antimicrobial resistant gene, β-lactamases genes blaOXA and tetracyclines-resistant gene tetB were detected in 15 (37.5%) and 2 (5.0) isolates, respectively, and blaTEM, aac (6')-Ib, aac (3)-II, tetA, sul1 and sul2 genes were only detected once in each (2.5%) isolate. The O127 (22.5%) serogroup was the most predominant, but all E. coli isolates were divided into 20 serogroups. The occurrence of antimicrobial-resistant bacterial infection can be problematic as it not only deteriorates the milk quality but also limits the therapeutic choices of antimicrobials in humans. Therefore, continuous monitoring is recommended to track antimicrobial-resistant E. coli in BTM and raw milk.
