        본 연구는 실내-외 우리가 있는 개방형 흑염소사에서 우리나라 5개월령 재래 흑염소를 대상으로 서열과 일주기 리듬이 하루 24시간 동안의 행동에 미치는 영향과 서열과 사료급여시간이 사료급여 후 30분 및 60분 동안의 행동에 미치는 영향을 조사하기 위해 수행하였다. 재래 흑염소의 행동은 CCTV를 사용해 녹화한 후 이들을 1분 단위로 측정하였고, 총 10개의 행동으로 분류하였다. 서열은 적대행동에 유의하게 영향을 미쳤으나 (p<0.05), 다른 행동에는 영향을 미치지 않았다. 일주기 리듬은 응립(p<0.05), 이동(p<0.01), 사료섭취(p<0.001), 적대행동(p<0.001), 비적대행동 (p<0.001), 사료탐색(p<0.05), 음수(p<0.001) 및 기타 행동(p<0.001)에 유의하게 영향을 미치는 반면 휴식(p>0.05)과 몸단장(p>0.05)에는 영향을 미치지 않았다. 서열과 일주기 리듬의 상호작용효과는 적대행동에만 유의하게 영향을 미쳤다(p<0.05). 서열은 사료급여 후 30분 동안 응립(p<0.05)과 사료섭취(p<0.05)에 유의하게 영향을 미쳤지만 다른 행동에는 영향을 미치지 않았다. 사료급여시간은 사료급여 후 60분 동안의 음수(p<0.05)에 유의하게 영향을 미쳤으나, 다른 행동에는 영향을 미치지 않았다. 서열과 사료급여시간의 상호작용효과는 모든 행동에 유의하게 영향을 미치지 않았다(p<0.05). 결론적으로, 서열, 일주기 리듬, 그리고 사료급여시간은 우리나라 5개월령 미성성숙한 재래 흑염소의 행동에 영향을 미치는 것으로 나타났다.

        The present study investigated effects of antifungal and carboxylesterase inoculant on rumen fermentation with different rumen pH. Corn silage was treated without inoculant (CON) and with a mixed Lactobacillus brevis 5M2 and L. buchneri 6M1 (MIX). Rumen fluid was collected from two cannulated Hanwoo heifers before morning feeding (high rumen pH at 6.70) and 3 h after feeding (low rumen pH at 6.20). Dried corn silage was incubated in the rumen buffer (rumen fluid + anaerobic culture medium at 1:2 ratio) for 48 h at 39oC. Eight replications for each treatment were used along with two blanks. Both in a high and a low rumen pH, MIX silages presented higher (p<0.05) the immediately degradable fraction, the potentially degradable fraction, total degradable fraction, and total volatile fatty acid (VFA) than those of CON silages. Incubated corn silages in a low rumen pH presented lower (p<0.05) total degradable fraction, ammonia-N, total VFA (p=0.061), and other VFA profiles except acetate and propionate, than those in a high rumen pH. The present study concluded that application of antifungal and carboxylesterase inoculant on corn silage could improve degradation kinetics and fermentation indices in the rumen with high and low pH conditions.

        To attenuate and control the spread of infectious disease, a body of research has been conducted to generate safe vaccines and to continue national-level surveillance. However, understanding on viability and persistence of avian influenza virus (AIV) in infected carcasses, and effective disposal approaches are still limited up to date. Here, using HA test and RT-PCR, we assessed active status of AIV and degradation of viral RNA in collected specimens at different sites and time points. First, AIV infectivity was recovered until day 2, and viral nucleic acids persisted to day 14 and 21 in inorganic and organic samples, respectively, in sealed vials incubated at room temperature. Second, AIV was totally inactivated in all examined specimens, and viral RNA was not detectable at all time points tested at least one month post-infection in AIV-inoculated carcasses buried directly in soil or fiber reinforced plastic (FRP) bin. Lastly, among different burial sites in South Korea, 6 out of 17 sampling sites in Jeonbuk province showed the presence of viral genetic materials, while the rest of the field samples displayed neither the presence of infective AIV nor detectable viral RNA. This study showed a linear relation between time and degradation degree of viral RNA in buried samples suggesting that burial disposal method is effective for the control or at least attenuation of spread of AI infection in infected animals although consistent monitoring is required to verify safety of disposal.

        This study evaluated the antimicrobial efficacy of different concentrations of ozonated water with organic matter, fetal bovine serum, at different concentrations and incubation times with bacteria. In the absence of organic matter, total eradication of up to 5 log of Escherichia (E.) coli was achieved, however, interference by organic matter led to inefficiency of ozonated water as a disinfecting agent. In addition, diminishing antimicrobial effects at higher temperatures, even in the absence of organic matter, were also demonstrated. These findings indicate that ozonated water will be a safe and effective disinfectant agent that could be useful in meat processing, especially an intestine processing, in Korean slaughter houses.

        The objective of this study was to determine the efficacy of ozone in sanitizing water experimentally inoculated with the gram-positive food-poisoning bacterium Staphylococcus aureus. The bactericidal effect was measured after experimentally inoculated solutions were exposed to 0, 0.5, and 1.0 ppm ozone at several time points and different temperatures, in the presence of varying concentrations of different organic matter, namely, fetal bovine serum (FBS) or cattle liver. Results revealed inhibition of the bactericidal effect in the presence of the lowest percentage of FBS, but a lower extent of the inhibition occurred when liver was used as the organic matter. It was also apparent that a higher temperature and shorter ozone exposure time had led to a more reduced bactericidal efficacy than that under a lower temperature and longer ozone exposure. This study provides insight into the potential use of ozonated water as an effective and safe disinfectant in an abattoir setting.

        Brucellosis is an important and re-emerging zoonotic disease worldwide. The prevention of human infection is achieved predominantly through the control of brucellosis in agricultural animals, which in turn depends on accurate diagnosis and vaccination. However, conventional serological diagnosis of brucellosis has several limitations, and currently available vaccines for animals have several drawbacks, including the ability to cause infection in humans. Phosphoglycerate kinase (Pgk) is one of the specific proteins reactive with mouse sera in the early stage of Brucella infection, and deletion of the pgk gene in B. abortus strain 2308 resulted in extreme attenuation of this strain in vitro and in vivo. Furthermore, the B. abortus pgk mutant has been used as a live vaccine, and in challenge experiments, it induced protection that was superior to that conferred by commercial strains. In this study, the pgk gene from Brucella abortus 544 was successfully amplified and cloned into a maltose binding protein fusion protein expression vector (pMAL). The recombinant protein was expressed in Escherichia coli DH5α and purified. The immunogenicity of purified recombinant B. abortus 544 Pgk (rPgk) was evaluated by western blot analysis using Brucella-positive mouse sera. rPgk could be used as an antigenic component for future serological tests and potential vaccine development.

        Brucellosis is a notorious zoonotic disease with global implications. Efforts to control the spread of the disease have been restricted to the agricultural livestock. Increasing incidences of accidental human infection have motivated researches to start working on alternative vaccines. At present, live attenuated vaccines are the only accepted type of vaccines used in developed countries for the prevention of brucellosis. Although serodiagnosis is occasionally unreliable, some countries have already claimed to have eradicated the disease, based on this testing. Live attenuated vaccines are not suitable for use in pregnant and immune-depressed animals. Moreover, these vaccines are not tolerated in humans. Therefore, many researches have been striving to discover alternative methods of vaccination. Most research has focused on the generation of subcellular, subunit, and DNA vaccines that are as efficient as the live attenuated vaccines. At present, none of the available vaccines has been able to replace the live attenuated vaccines. Therefore, additional research is necessary in order to discover a new brucellosis vaccine that is suitable for human use.

        Brucellosis is an important bacterial zoonotic infection worldwide responsible for economic losses in livestock industries which represents a considerable and increasing public health burden. The disease is caused by Brucella which possess the ability to invade and replicate within phagocytes and may result to chronic infection, leading to difficulty in medical therapy of the disease. The treatment for brucellosis employs conventional principles that have been applied for a long time and a combination of antibiotics is currently used due to low efficacy of monotherapies. However, vital effects with respect to health and safety are neglected. Nevertheless, the preventive methods and treatments for brucellosis using traditional medicine have not yet been thoroughly studied, hence, alternative therapies such as the use of natural plant extracts as traditional medicine that are safe, efficient and economical should be explored in order to identify candidates that eliminate complications due to brucellosis. Coptis chinensis Franch (Huanglian) is a traditional Chinese medicinal herb and its extract has been known to possess strong antibacterial activity. In this study, the antibacterial effects of C. chinensis Franch extracts, and the major components of the herb namely berberine and palmatine, were investigated on B. abortus. The C. chinensis Franch ethanol extracts (CCFE) showed bactericidal effects at 1,000 μg/ml concentration and berberine at 100 μg/ml concentration. However, C. chinensis Franch and its components did not affect invasion and intracellular growth of B. abortus in RAW 264.7 cells. In conclusion, C. chinensis Franch ethanol extracts, water extract and its major components such as berberine and palmatine would be a beneficial antimicrobial agent without affecting phagocytic pathway within macrophages, and further study for the precise mechanisms of antibacterial effect would be necessary.

        Avian influenza (AI) and Newcastle disease (ND) distress a variety of avian species, especially domestic poultry. Severe syndromes are caused by highly virulent specific virus strains termed highly pathogenic AI and velogenic ND viruses, which are potential agrobioterrorism agents. This outbreak emphasizes the need for continuing cooperation between public health and veterinary medical communities in controlling AI and ND when it has a zoonotic potential. Up to date, the stamping out and burying system were applied for controlling methods against these highly infectious diseases in the ordinary way, however these methods had many environmental problems, including leachat and effluvium. Thus, we assessed that sterilization effect of AI and ND virus dependent on several treatment conditions, such as autoclaving time and cutting types of chicken. As a result, we found that the cutting type of chicken meat revealed a reduced HA titer (20) against both of AI and ND virus after 10 min of autoclaving, while whole chicken showed same titer after 30 and 60 min. Therefore, we propose that the conditions of treatment on infected chicken should be developed for convenient, affordable, and effective prevention methods against for AI and ND.

        Salmonellosis is the commonest zoonosis worldwide that generally causes enterocolitis and foodborne poisoning which represents a considerable public health burden. Salmonella spp. are potential enteric pathogens and intracellularly replicates in host cells resulting in chronic infections. The medical treatments for salmonellosis have been difficult yet and had a serious problem including the increasing emergence of antibiotic resistance. The present report was designated to investigate the antibacterial effects of Saururus chinensis Baill ethanol extract (SCEE) on pure culture and infection with Salmonella enterica serovar Typhimurium (S. typhimurium) in murine derived macrophage RAW 264.7 cells. In determination of antibacterial activity of SCEE against S. typhimurium, bacterial viability was markedly decreased compared to the control. Also, SCEE significantly induced morphological change (p<0.05) of RAW 264.7 cells. In infection assay of S. typhimurium in RAW 264.7 cells pretreated with 100㎍/㎖ of SCEE, which is a non-cytotoxic concentration, bacterial uptake ability of macrophage was increased corresponding with morphological change, whereas bacterial survival rates within macrophage were markedly reduced compared with untreated control. Furthermore, nitric oxide (NO) production in SCEE-treated cells was slightly increased until 2 h but showed a tendency of decrease after 4 h until 24 h post infection compared with untreated control with S. typhimurium infection. Taken together, these findings demonstrated that SCEE has the antibacterial activity for S. typhimurium and the protective effects against S. typhimurium infection through activating murine macrophage independent on NO, suggesting that SCEE may be beneficial on the disease caused by intracellularly replicating pathogens as a safe alternatives of conventional chemotherapies.

        To improve the environmental management and resources, in this study, we aimed to investigate the effect of adding oyster shell powder to Hanwoo manure on its characteristics and microbial composition during the storage period. Additives were deposited on top of the manure surface at the rate of 0, 0.5, and 1% of oyster shell powder per 200 g of Hanwoo manure in a plastic container with three replicates; however, untreated manure litter served as the control. Manure characteristics (dry matter, organic matter and crude ash) and microbial composition (lactic acid bacteria, yeast, Bacillus subtilis, Salmonella, and E.coli) were evaluated at day 0, 2, 4, and 8. Manure characteristics exhibited an effect on dry matter, organic matter, and crude ash at day 2 and 8 (p<0.05), and not for day 0 and 4 (p>0.05). With the exception of yeast content at day 4 of storage, lactic acid bacteria, yeast, Bacillus subtilis, Salmonella, and E.coli exhibited no significant differences in all conditions during the storage period. Conclusively, addition of 1% oyster shell powder to Hanwoo manure resulted in slightly better manure characteristics; however, its microbial composition remained unchanged.
