        바퀴는 혐오감을 줄 뿐만 아니라 여러 종류의 병원체를 기계적으로 전파하는 주요한 위생곤충이며, 바퀴 방제를 위해 독먹이제를 가장 많이 사용한다. 본 연구에서는 4종류(chlorpyrifos, avermectin, hydramethylnon 및 fipronil)의 살충 주성분이 각각 함유된 국내 유통 독먹이제 3종 (Korea Combat Ultra Slim (K-CUS), Korea Combat Power (K-CP) 및 Korea Zaps Dual Bait (K-ZDB))과 미국 유통 독먹이제 3종(US Combat Source Kill Max (US-CSKM), US Hot Shot MaxAttrax Roach Bait (US-HSM) 및 US Raid Double Control Small Roach Bait (US-RDC))의 독일바퀴(Blattella germanica)에 대한 살충효력을 검증하였다. 6종의 독먹이제 중 fipronil을 살충 주성분으로 사용하는 세 가지의 독먹이제(K-CUS, K-CP 및 US-CSKM)가 속효성 및 높은 살충효과를 나타내었다. K-CUS과 K-CP의 살충효력의 지속성을 검정하기 위해 두 제품을 개봉 후 6개월 및 12개월 이후에 추가로 생물검정을 실시한 결과, 두 제품 모두 시간이 지남에 따라 제품 개봉 직후에 비해 살충효 과가 감소하는 경향을 보였지만, 40시간 이내 독일바퀴에 대한 사충률이 90%에 도달하는 것을 감안하면 적어도 이들 독먹이제의 옥내 설치 후 1 년동안은 살충효과가 유지되는 것으로 판단된다.

        The insecticidal activities of 13 Lamiaceae plant oils and their components against adult German cockroaches, Blattella germanica L. (Blattodea: Blattellidae), were evaluated using fumigant and contact bioassay. Among the tested oils, basil, pennyroyal, and spearmint showed the strongest insecticidal activities against adult B. germanica. Insecticidal activity of pennyroyal was 100% against male B. germinica at 1.25 mg concentration in fumigant bioassay. Basil and spearmint revealed 100% and 100% insecticidal activity against male B. germinica at 5 mg concentration, but their activities reduced to 80% and 25% at 2.5 mg concentration, respectively. In contact toxicity bioassy, basil, pennyroyal, and spearmint oils exhibited 100%, 100%, and 98% mortality against female B. germanica at 1 mg/♀, respectively. Among the constituents identified in basil, pennyroyal and spearmint oils, insecticidal activity of pulegone was the strongest against male and female B. germanica.

        The susceptibility values for field-collected (BG-BG) and laboratory (KS-BG) strains of German cockroaches, Blattella germanica (L.) to 5 synthetic pyrethroids and 3 organophosphates were assessed by topical application. Both KS-BG and BS-BG demonstrated the highest levels of susceptibility to deltamethrin. The KS-BG demonstrated the lowest susceptibility level to fenthion, while the BS-BG field population showed the lowest susceptibility to bifenthrin. The BS-BG field population showed resistance levels of 50 to 640 fold higher against the eight pesticides evaluated compared to the KS-BG. The resistance ratios (RR) of the eight pesticides were compared using populations of B. germanica that were collected from the same locality during 2007 and 2014. The field population of B. germanica collected during 2014 demonstrated significantly high resistance to an organophosphate and all pyrethroids when compared with those collected during 2007, but RR values were < 10 except for chlorpyrifos-methyl and chlorpyrifos with RR values of 74 and 104, respectively.

        We evaluated the repellent activity of 12 Apiaceae plant essential oils and their components against male and female adult German cockroaches, Blattella germanica, to find new natural repellents. Of the plant essential oils tested, ajowan (Trachyspermum ammi) and dill (Anethum graveolens) essential oils showed the most potent repellent activity against male and female adult German cockroaches. Of the compounds identified, carvacrol, thymol, and R-(-)-carvone showed >80% repellent activity against male and female adult German cockroaches at 0.2 mg/L air concentration. S-(+)-Carvone, (+)-dihydrocarvone, and terpinen-4-ol showed >70% repellent activity against male and female adult German cockroaches at 0.8 mg/L air concentration. Our results indicated that Apiaceae plant essential oils and their constituent have good potential as natural repellents against adult German cockroaches.

        The habitat of the cockroach varies by species. The German cockroach (Blattella germanica) lives in human dwellings, while the Japanese field roach (Blattella nipponica) lives in a mountainous region. Based on phylogenetic analysis of mtCO I, the two species are closely related to each other and B. germanica is divergent from wild species such as B. nipponica. Their habitats and walking speed differ even though the two species have similar morphology. We hypothesized that habitats might influence walking speed via changes to appendage morphology and enzyme-based physiological differences. We found that phenotype such as appendage length and esterase isozyme expression were clearly different between the two species. These differences might be responsible for the observed difference in walking speed.

        Lysozyme was the first humoral antibacterial factor to be studied in insects and it was considered the main immune factor in the haemolymph. Lysozymes are enzymes characterized by their ability to break down bacterial cell walls. We identified four lysozymes(c1, c2, c3, and I type)-encoding cDNAs from P. americana. It was deduced protein sequences are basic in nature, contain 138 amino acids(c1), 139 amino acids(c2), 141 amino acids(c3), and 143 amino acids(I) including conserved cysteine residues. Transcriptional profiles indicated that the predominant form is constitutively expressed and up-regulated upon immune challenge by heat stress. When injected lipoplysaccharide (LPS), lysozymes (I, c1, and c2) was up-regulated in the body. In addition, the expression levels of lysozymes(I, c1, and c2) in the P. americana significantly increased after two hour by injection of LPS. We were cloning and analysis of chitinase from B. germanica. The chitinase was deduced protein sequences are contain 539 amino acid residues long. The chitinase were up-regulated by injection of LPS in B. germanica. the expression levels of chitinase in the B. germanica significantly increased after four hour by injection of LPS. The cDNA encoding the chitinase of B. germanica was expressed as a 67-kDa band in the baculovirus-infected insect cells and the extracts of the recombinant baculovirus-infected cells showed anti-bacterial activity(0.0125㎍/㎖: 2.5±0.3, 0.00125㎍/㎖: 0.6±0.4mm) to C. albican on the plate.

        The habitat of the cockroach varies relative to species. The German cockroach (Blattella germanica) lives in human dwelling while B. nipponica lives in mountainous region. Even though, these two species have similar morphology, their habitats and walking speed were different. Underlying this fact, we hypothesized that habitats might influence on the walking speed via altering particularly nervous system including acetylcholinesterase (EC3.1.1.7, AChE). Full length ORFs were cloned from each species and sequenced. Sequence analyses showed that both genes in each species possess typical features of ace, and that Bgace1 and Bnace1. Those features are orthologous to the insect ace1 whereas Bgace2 and Bnace2 are to the insect ace2. Some SNPs were observed but Bgace1 and 2 showed high similarity (99%) with Bgace1 and 2, respectively. Multiple AChE bands were identified by native PAGE and IEF from three tissues (head, leg and other body) and their expression pattern was almost identical between two species. However, Esterase expression patterns were significantly different. Furthermore, length as well as detailed structure of antenna, leg and tail cerci was also significantly different. Taken together, various factors such as sensory organ detailed morphology and esterases are responsible for habitat and walking speed difference.

        The German cockroach, Blatella germanica, is one of the most important household pest that can cause disease on human. Conventional contact insecticides show less efficacy because it is hard to be contacted or digested by cockroach that lives inside small crevice. Ethyl formate(EF), which is a potential fumigant to control various insect pests such as stored grain insects and quarantine pests, can be penetrated wherever air exist and shows toxicity on respiratory system of insect. In this trials, we've demonstrated practical and commercial scale fumigations of EF on German cockroach. Efficacy of EF was described in terms of concentration × time (CT) product. At practical fumigation, EF gave 100% control of German cockroach nymph and adult when the CT product was 43.2g h m-3. When 600m3 scale warehouse fumigation for 15 hours with 5g m-3 of EF, nymph and adult stage of German cockroach were completely controlled. The ventilation time to reduce EF concentration under TLV(Threshold Limit Value, EF=100ppm) was less than 1 hour. These results show that EF can be applied at warehouse or mill house to control German cockroach effectively.

        Cockroaches are insects of the order Blattodea, sometimes also called Blattaria, of which about 30 species out of 4,600 total are associated with human habitats. About four species are well known as pests. Among the best-known pest species are the American cockroach, Periplaneta americana, which is about 30 mm long; the German cockroach, Blattella germanica, about 15 mm long. We researched growing condition of in-vitro(temperature and humidity) for P. americana and B. germanica. The cockroach is divided in three sections; the body is flattened and broadly oval, with a shield-like pronotum covering its head. A pronotum is a plate-like structure that covers all or part of the dorsal surface of the thorax of certain insects. They also have chewing mouth parts, long, segmented antennae, and leathery fore wings with delicate hind wings. The third section of the cockroach is the abdomen. We measured size of head, abdomen, and body length during differential stages. Also, we checked number of egg, size of egg, hatching rates, period of former laying eggs, and laying periods etc.

        Vitellogenins (Vgs) are precursors of the major egg storage protein, vitellin (Vn), in many oviparous animals. Insects Vgs are large molecules (200-kD) synthesized in the fat body in a process that involves substantial structural modifications (e.g., glycosylation, lipidation, phosphorylation, and proteolytic cleavage, etc.) of the nascent protein prior to its secretion and transport to the ovaries. However, the extent to which Vgs are processed in the fat body varies greatly among different insect groups. We were cloned Vgs partial genes PaVgs and BgVgs from Periplaneta americana and Blattella germanica. Real-time quantitative PCR shows that PaVgs and BgVgs were differential-regulated with aging. In insects, glutathione S-transferases (GSTs) are enzymes involved in detoxification of insecticides. We were cloned GST partial genes PaGST and BgGST from Periplaneta americana and Blattella germanica. Real-time quantitative PCR shows that PaGST and BgGST were up-regulated with aging, and the mRNA level of PaGST and BgGST was higher in 4℃ and 37℃ than room temperature. The expression level of PaGST and BgGST exposure to temperature stress suggests that PaGST and BgGST are up-regulated after exposure low and hige temperature treatments.

        Fumigant and contact toxicities of 11 Myrtaceae plant essential oils and their constituents against adult male and female Blattella germanica were evaluated. Of 11 Myrtaceae plant essential oils, Eucalyptus polybractea, E. smithii, E. radiata, E. dives, E. globulus, and Melaleuca uncinata, showed 100% fumigant toxicity against adult male German cockroaches at a concentration of 7.5 mg/liter air concentration. In contact toxicity tests, E. polybractea, E. smithii, E. radiata, E. dives, E. globulus, M. dissitiflora, and M. uncinata produced strong insecticidal activity against adult male and female German cockroaches. Of the essential oil constituents, terpinolene, α-terpinene, and terpinen-4-ol demonstrated strong fumigant toxicity against adult male and female B. germanica. Eugenol, isoeugenol, methyl eugenol, and terpinen-4-ol showed strong contact toxicity against adult male B. germanica. The toxicity of the constituent blends identified from M. dissitiflora essential oils indicated that terpinen-4-ol were major contributor to the fumigant activity or contact toxicity of the artificial blend. Only isoeugenol exhibited inhibition activity against male acetylcholinesterase. IC50values of isoeugenol were 0.22 mg/mL against male acetylcholinesterase.

        We evaluated insecticidal and acetylcholine esterase inhibition activity of 11 Apiaceae plant essential oils and their constituents against male and female adults of Blattella germanica. Of 11 Apiaceae plant essential oils tested, dill (Anethum graveolens), carvi (Carum carvi), and cumin (Cuminum cyminum) essential oils demonstrated > 90 % fumigant toxicity against male adults of German cockroach at 5 mg/filter paper. In contact toxicity test, dill (Anethum graveolens), carvi (Carum carvi), cumin (Cuminum cyminum), and ajowan (Trachyspermum ammi) essential oils produced strong insecticidal activity against male and female adults of German cockroach. Among test compounds, carvone, 1,8-cineole, trans-dihydrocarvone, cuminaldehyde, trans-anethole, p-cymene, and γ-terpinene demonstrated strong fumigant toxicity against male and female adults of B. germanica. In a contact toxicity test, carveol, cuminaldehyde, (S)-(+)-carvone, trans-anethole, thymol, and p-cymene showed the strong contact toxicity against B. germanica male and female adults. IC50 values of α-pinene, carvacrol, and dihydrocarvone against female acetylcholine esterase were 0.28, 0.17, and 0.78 mg/mL, respectively. The toxicity of blends of constituents identified in 4 active oils indicated that carvone, cuminaldehyde, and thymol were major contributors to the fumigant or contact toxicity of the artificial blend.

        Two acetylcholinesterases (AChEs; BgAChE1 and BgAChE2) from Blattella germanica were functionally expressed using the baculovirus system. Kinetic analysis demonstrated that BgAChE2 had higher catalytic efficiency but lower substrate specificity than BgAChE1. Except paraoxon, BgAChE1 was generally less sensitive to inhibitors than BgAChE2. Western blot analysis using anti-BgAChE antibodies revealed that BgAChE1 was far more abundant in all examined tissues compared to BgAChE2, which is only present in the central nervous system. Both BgAChEs existed in dimeric form, covalently connected via a disulfide bridge under native conditions. Most fractions of BgAChE1 had a glycophosphatidylinositol (GPI) anchor, but a small fraction comprised a collagenlike tail. BgAChE2 appeared to have a collagen-GPI-fused tail. Based on the kinetic and molecular properties, tissue distribution and abundance, BgAChE1 was confirmed to play a major role in postsynaptic transmission.

        The toxicity of Cyperus rotundus rhizome steam distillate constituents and their related compounds to adult females and males of the susceptible KS-BG strain and field-collected SR-BG and DR-BG colonies of Blattella germanica (L.) was examined using residual contact + fumigant and vapor-phase mortality bioassays. Results were compared with those of three organophosphorus (chlorpyrifos, dichlorvos, and fenitrothion), three carbamate (bendiocarb, dioxacarb, and propoxur), and three pyrethroid insecticides (bifenthrin, cypermethrin, and deltamethrin). In contact + fumigant mortality bioassay, p-cymene (LC50, 0.33 mg/cm2) was the most toxic terpenoid, followed by o-cymene, m-cymene, β-pinene, 1,8-cineole, and α-pinene, (LC50, 0.44–0.92 mg/cm2) against female B. germanica. These terpenoids were less toxic than the insecticides tested. Females were 3.1–3.6 times more tolerant than males to three cymene compounds. The toxicity of these monoterpenoids were almost identical against females from either of the three strains, even though the SR-BG and DR-BG females exhibited resistance to bifenthrin [resistance ratio (RR), 96 and 17], cypermethrin (RR, 67), deltamethrin (97 and 66), dioxacarb (19 and 22), and propoxur (24 and 32). In vapor-phase mortality bioassay, these monoterpenoids were effective in closed but not in open containers, indicating that the effect of these compounds was largely a result of vapor action. C. rotundus rhizome steam distillate and its constituents merit further study as potential insecticides for the control of insecticide-resistant B. germanica populations as fumigants with contact action.

        The susceptibility of the Blattella germcanica (L.) in the Republic of Korea (ROK) to insecticides was evaluated under laboratory conditions using 12 insecticides currently used by the local public health centers and/or pest control operators in the ROK. The insecticides included seven pyrethroids and five organophosphages. Based on their LD50values, the order of susceptibility of B. germanica adults to the insecticides was chlorpyrifos-methyl, profenofos, and chlorpyrifos with the values of 0.07, 0.29, and 0.88 ㎍/♀, respectively. The least susceptibility was obtained with tetramethrin at LD50 of 7.39 ㎍/♀. In the comparative resistance test, the resistance ratios (RR) of 12 insecticides were compared to each other using field collected B. germanica adults in Seoul between 1993 and 2007. Blattella germanica demonstrated higher RRs to pyrethroids such as λ-cyhalothrin, and low RRs among the organophosphates. Among the pyrethroids, λ-cyhalothrin had the highest RRs of 111 and 129 fold differences at LD50 and LD90 values, respectively. Among the organophosphates, profenofos was observed to have the highest RRs of 4 and 15 fold differences at LD50 and LD90 values, respectively. However, there were no significant differences in susceptibility to tetramethrin, chlorpyrifos, and fenitrothion. Blattella germanica was more susceptible to pyridafenthion showing a 0.7 fold difference in a resistance rate (RRLD50), respectively. Resistance ratio of tetramethrin was low, but susceptibility was also not high.

        바퀴벌레(Blattella germanica)에 대한 coriander oil (Coriandum sativum) 등 5종의 식물정유의 훈증독성을 조사하였다. 그들 중 spearmint oil이 20과 (air)의 약량처리에서 모두 의 높은 살충활성을 나타내었다. Coriander를 GC와 GC/MS로 분석한 결과, -pinene (), camphene (), -pinene (), -myrcene (), limonene (), -terpinene (), -cymene (), -terpinolene (), camphor (), linalool (), -tepineol (), geranyl acetate (), geraniol ()이 주요 구성성분으로 나타났으며, 그 중 -terpinene과 geranyl acetate가 (air) 약량에서 의 높은 훈증독성을 나타내었다.

        시판되고 있는 61종 살충제에 대한 식이법으로 바퀴(Blattella germanica) 수컷성충의 우수 살충제를 탐색하고, 활성이 있는 약제를 이용하여 처리부위와 검정법에 따른 감수성 차이와 발현속도를 비교하였다. 식이법에서 이상의 살충률을 나타낸 세 약제중에서 dichlorvos, fenitrothion, fipronil약제를 선발하여, 식이법과 여지접촉법으로 살충력을 비교한 결과, dichlorvos와 fenitrothion은 여지접촉법에서, fipronil은 식이법에서 더 높은 감수성을 나타내었다. 처리부위(머리, 가슴, 배, 다리)에 따라서는 dichlorvos와 fenitrothion은 머리처리에서, fipronil은 가슴처리에서 감수성이 가장 높았다. Fenitrothion과 fipronil의 (일)은 각각 1.05와 0.98로 속효성이었으며, boric acid와 hydramethylnon은 각각 3.92와 2.26으로 지효성이였다.
