Lonicera caerulea (Honey berry, HB) has been used in medical treatment in Russia, Japan, China and Korea. It has high level of vitamin C and polyphenolics. Polyphenolics can improve anti-inflammatory effect and prevent cancer, diabetes mellitus type 2. Also, Vitamin C is a representative anti-oxidant. however, it is still unknown what effect it will have on the oxidation stress of the reproductive system. In previous studies, ROS can be produced when it is exposed to heat stress and has negative effect on sperm’s maturation, capacitation, hyperactivation, acrosome reaction and fusion of egg and sperm. Therefore, the purpose of this study is to investigate the antioxidant effects of L. Caerulea on the sperm and mice. At first, it conducted using ICR mouse (n = 20) for 4 weeks. There are four groups of mice (n = 5 per group). Also, L. Caerulea was taken by oral gavage. Group I (control) kept at 23℃-27℃ and administer D.W (0.5 mL/day), Likewise, Group II (HB) kept at room temperature but gave HB (250 mg/kg, 0.5 mL/day), Group III (HB + HS) received heat stress (40℃) using hyperthermia induction chamber and gave HB at same dose. and Group IV (HS) exposed heat stress only. Mainly, we showed degree of gene expression using Western blot in SOD, HSP 70, 17β-HSD and Real-time PCR. It can find correlation between intracellular activity like steroid hormone, apoptosis under ROS and antioxidant activity of L. Caerulea.
Mitochondria is energy generating organelle. It synthesizes ATP, which is the essential energy source of many cellular processes. During producing energy, some redox centres leak electrons to oxygen and it is contributory to the reactive oxygen species. Besides, mitochondria have significant functions in metabolism, calcium homeostasis, and fatty acid oxidation.
Also mitochondria has importance to the breakdown of the ovarian follicles and could be factor determining oocyte of quality adversely. Increasing evidence shows that the number of mitochondria affect oocyte of developmental competence and maturation detrimentally during aging. Oocyte is the mitochondria-rich cell and enable the organelle to have competence for fertilization and early embryonic development. Occurrence of blastomere depends on distribution change of mitochondria which present in the egg.
Lonicera caerulea treatment inhibited ovarian mitochondrial oxidative damage by suppressing mitochondrial reactive oxygen species (mROS) generation, decreasing apoptosis, controlling disintegration of mitochondrial membrane potential and conserving respiratory chain complex activities.
The purpose of this study is to identify if mouse accepting treatment with L. caerulea could counter age-induced sterility and ovarian mitochondrial OS in a model organism of ovarian ageing.
Mammalian oocytes are sensitive to psychological stress at each period of follicular development. Especially, thermal stress interfere with reproductive condition by inducing formation of reactive oxygen species (ROS) and oxidative stress (OS). ROS lead to oocyte apoptosis, weakening oocyte quality and lowering the fertilization rate. As a result, the pregnancy rate is lowered, leading to infertility.
Thermal stress also seems to influence zygotes through physiological changes in the maternal environment surrounding them. Loss of developmental competence suggests hyperthermia-induced oxidative stress in embryos.
Interest in organic Lonicera caerulea berries has increased in recent years. They are abundant in various health-improving materials. Berries that found from natural products can be as free as possible from the bioactive toxicity of the active ingredient without side effects, and it can be a big advantage because it can work.
Mammalian oocytes are arrested at the first meiotic prophase stage and get their meiotic competence to produce offspring during the development of follicle. A series of nuclear and cytoplasmic maturations are involved in this process and these vary in temperature sensitivity.
Our study demonstrated that L. caerulea can relieve the negative effects of maternal hyperthermia by reducing ROS level at the developmental stage.

Previous studies have shown that Lonicera caerulea has a chemical protective effect. Phenolic and vitamin C contained in Lonicera caerulea prevent cancer, diabetes and cardiovascular disease, lower blood pressure and delay the aging process. However, the antioxidant mechanism of male reproductive system to heat stress is still unknown. Male reproductive system is very sensitive to heat. When scrotum temperature increase, oxidative stress can occur. Oxidative stress affects sperm motility and spermatogenesis, resulting in infertility. Therefore, we investigated the antioxidant effect of L. Caerulea in male genitalia by inducing oxidative stress by artificially exposing the testicles to heat at 42 ° C. The experiment was performed by dividing the ICR mouse into four groups. Each group is n = 5. Control group (C) and heat stress group (HS) were oral gavage administered D.W. Honey berry group (HB) and Honeyberry / heat stress group (HB + HS) were oral gavage administered honey berry (250mg / kg / day). HS groups (n=5, per n=5) received heat stress by exposing their lower bodies in the water bath at 42℃ for 30 minutes. We confirmed that there was a significant difference in the motility, morphology and the number of sperms using CASA(computer-assisted semen analysis). Lipid peroxidation assay results showed heat causes oxidative stress in serum. This study is conducting to investigate the antioxidant effect of L. Caerulea. Histologically analyzed the testicular form of each group, the activity level of heat shock protein and the level of reactive oxygen species were measured by Western blot and the level of catalase and HSP-90 was examined by RT-PCR analysis. Thus, studies of testicular morphology, sperm kinetics, hormone levels, heat shock protein expression and antioxidant enzymes under heat stress have shown that L. Caerulea ingestion has Anti-oxidant and thermal protective activity on the testis by heat damage.
본 연구는 붉은꽃인동덩굴의 적심높이(60, 120, 180 cm) 가 가지길이, 측지수 및 피복에 미치는 적심높이별 영 향을 알아 보기 위하여 수행하였다. 인동덩굴의 줄기길 이는 고절위(180 cm) 적심이 저절위(60 cm) 적심보다 길 었고, 측지수는 저절위 적심이 고절위 적심보다 많았다. 그러나 분지수는 적심처리간에 큰 차이가 없었다. 퍼골라 벽면의 피복도는 저절위 적심이 85%, 중절위 적심이 74%, 고절위 적심이 62%를 보였다. 개화는 5월 중순부터 10월 중순까지 계속되었고, 첫 개화는 60 cm 적심이 5월 중순으로 가장 빨랐다. 대체로 적심 높이가 낮고 적심시기가 빠를수록 개화가 빨랐고, 개화최성기는 8월 중순이었다. 붉은꽃인동덩굴을 이용한 퍼골라 벽면의 가로 3.0 m ×높이 2.2 m 녹화를 하기 위해서는 퍼골 라당 6주(1m당 2주)를 재식하고, 60 cm 벽면 높이에서 1회 적심하면 당년에 85%의 벽면스크린 및 녹화가 가 능하였다.
이상의 연구 결과로 미루어 볼 때, 댕댕이나무 잎과 가지 추추출물은 대장암 세포주 HCT116과 SW480세포의 생육을 억제 하였으나 열매추출물은 억제활성이 나타나지 않았다. 잎과 가지 추출물은 cell migration과 wound healing assay를 통해 비정상적인 세포증식 억제를 확인하였으며, β-catenin과 TCF4 의 단백질 수준을 감소시켜 비정상적인 Wnt 신호전달을 억제를 통해 대장암세포의 생육을 억제하는 것으로 판단된다. 따라서 댕댕이나무 잎과 가지는 항암을 위한 대체보완소재 및 천연 항암제 개발을 위한 소재로 활용이 가능할 것으로 판단된다.
본 연구는 각시괴불나무 종자의 휴면유형과 발아특성을 구 명하기 위하여 수행되었다. 수분흡수 실험결과, 각시괴불나무 종자는 수분흡수가 원활히 이루어졌다. 따라서, 물리적 휴면은 가지지 않는 것으로 판단된다. 각시괴불나무 종자는 탈리 시점에 미숙배를 가지고 있으며, 발아직전까지 배/종자 비율이 약 46% 증가하였다. 온도실험결과, 5℃에서는 발아가 전혀 이루어지지 않았다. 20℃에서는 파종 후 1주부터 발아가 시작되었으며 최종 발아율은 2주차에 86.7%였다. 15℃에서는 파종 후 2주부터 발아를 시작하였으며, 최종 발아율은 4주차에 75.0%였다. 25℃에서 파종 후 1주부터 발아가 시작되었으며 최종 발아율은 19주차에 48.3%였다. 결론적으로, 각시괴불나무 종자의 발아를 위한 최적온도는 가장 높은 최종 발아율과 가장 짧은 평균 발아 일수를 나타낸 20℃이다. 각시괴불나무 종자는 5℃를 제외 한 모든 온도에서 4주 이내에 발아하였기 때문에 생리적 휴면은 관찰되지 않았다. 따라서, 각시괴불나무 종자는 오직 형태적 휴면만을 가지는 것으로 판단되었다.
Background: Cynaroside is a flavone, a flavonoid-like compound, known by different names (luteoloside and cinaroside). It is commonly found in Lonicera japonica Thunb., Chrysanthemum moriflium, and Angelica keiskei. The process of cell death has been classified as necrosis and apoptosis. Necrosis refers to unregulated cell death induced by a chemotherapeutic agent. Doxorubicin is an anthracycline anti-cancer drug used to treat acute leukemia, cancer, and lymphoma. However, it induces nephrotoxicity including tubular damage. Therefore, we investigated the protective effect of cynaroside against doxorubicin-induced necrosis in HK-2 cells. Methods and Results: To confirm the beneficial effect of cynaroside on doxorubicin-induced necrosis, HK-2 cells, a human proximal tubule epithelial cell line were treated with 10 μM doxorubicin and 80 μM cynaroside. Doxorubicin treatment resulted in increased DNA fragmentation, caspase-3 activity and mitochondria hyperactivation during cell necrosis. However, pretreatment with 80 μM cynaroside attenuated DNA fragmentation, caspase-3 activity and mitochondria hyperactivation induced by 10 μM doxorubicin in HK-2 cells. Conclusions: These results indicated that pretreatment with cynaroside ameliorated doxorubicin-induced necrosis in HK-2 cells. Therefore, cynaroside be used as a therapeutic agent for improving doxorubicin-induced nephrotoxicity. However, further studies are required to evaluated the toxicity of cynaroside treatment in animals and to determine its protective effect against doxorubicininduced nephrotoxicity in an animal model.
Functional compounds including flavonoids, anthocyanins, polyphneols and antioxidants were extracted from blue honeysuckle (Lonicera caerulea L.) using highly efficient microwave-assisted extraction. And extraction process was modeled and optimized according to response surface methodology (RSM). The independent variables (Xn) were ethanol concentration (X1: 0, 25, 50, 75, 100%), irradiation time (X2: 1, 3, 5, 7, 9 min), and microwave power (X3: 60, 120, 180, 240, 300 W). Dependent variables (Yn) were total flavonoid contents (Y1), total anthocyanin contents (Y2), total polyphenol contents (Y3) and antioxidant activity (Y4). Four-dimensional response surface plots were generated based on the fitted second-order polynomial models to get optimal conditions. Estimated optimal conditions for 4 responses were ethanol concentration of 54-72%, irradiation time of 7.1-7.6 min, and microwave power of 243-251 W. Ridge analysis predicted the maximal responses of total flavonoid content, total anthocyanin content, total polyphenol content and antioxidant activity were 38.00 mg RE/g, 6.80 mg CGE/g, 14.90 mg GAE/g, 89.10%, respectively. Verification experiment was carried out at predicted optimal conditions and experimental values for total flavonoid content, total anthocyanin content, total polyphenol content and antioxidant activity were 38.10 mg RE/g, 6.72 mg CGE/g, 14.91 mg GAE/g and 89.13%, respectively. No significant difference was observed between predicted and experimental values, indicating good fitness of fitted model and successful application of RSM.
Background : Haskap berries commonly refer to fruits of Lonicera caerulea L., recognized by the Japanese aborigines as the “The elixir of life.”. Due to their recent arrival on the North American market, haskap berries have not yet been positioned among other berries and compared in terms of their phytochemical content. And haskap berries have higher ascorbic acid and anthocyanin content than other berries known for their health-promoting benefits, such as blueberries. However, no study has reported on the antioxidant and anti-cancer activity of Lonicera caerulea stem. The purpose of this study is to present the current research on the chemical content, antioxidant and anti-cancer activities of Lonicera caerulea stem. Methods and Results : The stem of Lonicera caerulea L. ware dried in the shade at room temperature and extracted with 100% methanol. The extract was suspended in deionized water and partitioned sequentially with n-hexane, chloroform, ethyl-acetate and butanol (water saturated BuOH) fractions. Antioxidant activities were measured by determination of antioxidants, DPPH (2,2-diphenyl-1-picrylhydrazyl). Cell viability was determined by the MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide] assay. All cell lines were purchased from the Korean Cell Line Bank (Seoul, Korea). All results were performed with three replications were processed statistically. By DPPH assay, the Lonicera caerulea L. the highest activity was obtained from the ethyl-acetate fraction (IC50=15.46 ㎍/㎖). By MTT assay, the chloroform fraction showed a significant growth inhibiting effect on MCF-7 (Human breast cancer, IC50=225.91 ㎍/㎖), COLO 205 (Human colon cancer, IC50=179.55 ㎍/㎖), but on AGS (Human stomach cancer) and other fractions it did not show effect. Conclusion : We demonstrated that Lonicera caerulea L. stem extract and fractions has antioxidant and antiproliferation activity in vitro. Further studies should identify the active constituents in Lonicera caerulea L stem to evaluate the potential in vitro antioxidant and antiproliferation activities of the extract.
Lonicera caerulea var. edulis is a rare species found in some alpine region of Korea. Genetic variation in L. caerulea var. edulis has been investigated by examining 161 individuals from six natural populations: Mt. Seorak 1, Mt. Seorak 2, Mt. Jeombong, Mt. Bangtae, Mt. Gyebang, Mt. Halla. The mean genetic diversity for all the six populations was 0.25 (S.I.). The highest genetic diversity was found in Mt. Seorak (S.I.=0.3158) and the lowest was in Mt. Gyebang (S.I.=0.1047). Comparatively low level of genetic diversity was observed (Ae=1.25, P= 64.6%, S.I.=0.25), which is a typical pattern for rare tree species. AMOVA showed exceptionally large proportion of genetic variations both for among populations (34.69%) and within populations (65.31%). Excluding Mt. Gyebang, the genetic variation among and within population was 18.71% and 81.29% respectively. The UPGMA dendrogram based on genetic distance is not suitable for geographic relationship. Genetic distance of Mt. Gyebang was most distant from the other populations. Excluding Mt. Gyebang, the genetic identities among the five populations were 0.95 to 0.97, which is very high similarity level of genetic identity. This low level of genetic variations and the lack of site in nature indicates that L. caerulea var. edulis demanded a serious conservation.
인동과에 속하는 인동덩굴(Lonicera japonica Thunb., LJ) 추출물이 활성산소의 일종인 glucose oxidase(GO)에 미치는 영향을 배양 C6 glioma 세포를 재료로 알아보았으며, 또한 LJ 추출물의 항산화 효과를 전자공여능(DPPH-radical scavenging activity)을 비롯하여 항산화 효소의 하나인 SOD의 활성과 비슷한 superoxide dismutase(SOD)-유사활성(SOD-like activity), 지질과산화 억제능(lipid peroxidation inhibitory activity) 및 lactate dehydrogenase(DH) 활성에 의하여 조사하였다. 본 연구에서 GO는 처리 농도 의존적으로 대조군에 비하여 배양 C6 glioma 세포의 생존율을 유의하게 감소시킴으로서 세포독성을 나타냈으며(P<0.01), 이 때 XTT50값은 35 mU·mL-1에서 나타났다. 또한, LJ 추출물은 GO에 의하여 감소된 세포생존율을 유의하게 증가시킴으로써 GO의 세포독성을 방어하였다. 한편, LJ 추출물은 전자공여능을 비롯한 SOD-유사활성, 지질과산화 억제능 및 LDH의 활성 감소를 보임으로서 항산화 효과를 나타냈다. 이상의 결과로부터 LJ 추출물은 항산화 효과에 의하여 GO의 세포독성을 유효하게 방어한 것으로 나타났다.
This study was conducted to evaluate the status of habitat distribution, environmental characteristics and plant species growing with Lonicera japonica in its habitats. In the distribution of plant coverage below 30%, it was 84.6% in the inland area and 80.7% in the seashore area. There was similar coverage in both inland and seashore areas. But the average relative coverage in the seashore area was 26.1%, which is a little higher than that of the inland area (22.5%). In the habitats, both inland and seashore areas showed a high distribution ratio below 10 degrees. But among them, inland areas showed a high distribution ratio (62.4%) below 10 degrees, which was about twice as much as the seashore areas (32.2%). Habitat distribution was most often found on the southern slopes in both inland and seashore areas. The average soil pH of Lonicera japonica habitat was 5.2 in the inland areas and 6.9 in the seashore areas. Also, the seashore areas showed little content of P2O5, but high content of K, Mg, Na on the other hand compared to the inland area. Miscanthus sinensis was the highest importance value (16.4) among plants growing with Lonicera japonica in their habitats in both inland and seashore areas. But in the inland areas, Artemisia princeps showed the highest importance value, and Miscanthus sinensis was the highest in the seashore areas.
