Interferon tau (IFNT) regulation, an anti-luteolytic factor produced by conceptuses of the ruminant ungulates, is essential for the maintenance of early pregnancy, but a definitive mechanism for its temporal transcription has not been elucidated. We and others have observed the T-box protein eomesodermin (EOMES) exhibited high mRNA expression in the ovine embryonic trophectoderm; thus, both caudal-relatedhomeobox-2 (CDX2) and EOMES coexist during the early stages of conceptus development. Objective of this study was to examine the effect of EOMES on ovine IFNT gene transcription when evaluated with CDX2, ETS2 and AP1 transcription factors implicated in the control of cell differentiation in the trophectoderm. In this study, quantitatively via reverse transcription-polymerase chain reaction (RT-PCR) analysis between ovine trophoblast cells was initially performed, finding that transcription factors CDX2 and ‘EOMES transcription factor mRNAs’ were specific to trophectoderm cells. These mRNAs were also found in days 15, 17, and 21 ovine conceptuses. Furthermore, human choriocarcinoma JEG3 cells (trophoblast cell line) were cotransfected with an ovine IFNT (-654bp)-luciferase reporter (-654-oIFNT-Luc) construct and several transcription factor expression plasmids. Cotransfection of the reporter construct with CDX2, ETS2 and AP1 increased transcription of -654-oIFNT-Luc by about 11-fold compared with transfection of the construct alone. When cells were initially transfected with EOMES followed by transfection with CDX2, ETS2 and/or AP1, the expression of -654-oIFNT-Luc was decreased. Also, EOMES factor inhibited the stimulatory activity of CDX2 alone. These results suggest that when conceptuses attach to the uterine epithelium, ovine IFNT gene transcription is down-regulated by an increase of EOMES factor expression in the attached ovine trophoblast cells.

We and others have observed that expression levels of bovine subtype IFNT genes in the tissues of ruminants; thus, bIFNT1 and other new type I (bIFNTc1/c2/c3) gene co-exist during the early stages of conceptus development and non-trophoblast cells. Its genes transcription could be regulated through CDX2 and ETS2 and JUN and/or cAMP-response element binding protein (CREB)-binding protein (CREBBP) expression, a transcription factor implicated in the control of cell differentiation in the trophectoderm. Bovine ear-derived fibroblast cells, were co-transfected with luciferase reporter constructs carrying upstream (positions -1000 to +51) regions of bIFNT1 and other new type I gene and various transcription factor expression plasmids. Compared to each - 1kb-bIFNT1/ c1/c2/c3-Luc increased when this constructs were co-transfected with CDX2, ETS2, JUN and/or CREBBP. Also, Its genes was had very effect on activity by CDX2, either alone or with the other transcription factors, markedly increased luciferase activity. However, the degree of transcriptional activation of the bIFNTc1 gene was not similar to that bIFNT1/c2/c3 gene by expression plasmid. Furthermore, Sequence analyses also revealed that the expression levels of bIFNT1/c2/c3 gene mRNAs expression were highest on day 17, 20 and 22 trophoblast and, Madin-Darby bovine kidney (MDBK), Bovine ear-derived fibroblast (EF), and endometrium (Endo) non-trophoblast cells. But, bIFNTc1 mRNA had not same expression level, bIFNTc1 lowest levels than those of IFNT1/c2/c3 gene in both trophoblast and non-trophoblast cells. These results demonstrate that bovine subtype bIFNT genes display differential, in the trophoblast and non-trophoblast cells.
Variance of conceptus interferon tau (IFNT), produced by the embryonic trophectoderm, is known as a major conceptus protein that signals the process of maternal recognition of pregnancy in ruminants, essential for the maintenance of early pregnancy. Similar to other IFN genes such as IFNA and IFNB, multiple IFNT genes are present. However, some kinds of IFNT genes actively transcribed and regulated in bovine conceptuses have not been well characterized. In this study, during the course of bovine IFNT gene transcription through the use of next generation sequencer SOLiD3, revealed that among 38 IFN genes registered, only two transcripts, IFNT1 and IFNTc1, were found in conceptuses during early pregnancy. Also, to identify a transcription factor(s) involved in the regulation of IFNT genes, mRNAs for various known transcription factors were investigated by real-time PCR in conceptus tissues, respectively. Furthermore, compared to the IFNT genes, IFNT1 and IFNTc1 had same active levels, which were previously shown to correlate with the appearance of effective antiviral activity. However, the expression levels of these Luc activities differed. Bovine ear fibroblast (EF) cells were cotransfected with luciferase reporter constructs carrying upstream (–631 to -51) promoter regions of IFNT1 or IFNTc1 and various transcription factor expression plasmids, CDX2, AP1(JUN), ETS2 and/or cAMP-response element binding protein (CREB)-binding protein (CREBBP). CDX2, either alone with the other 2 transcription factors, was found to increase luciferase activity approximately 14- and 11-folds, respectively. The degree of transcriptional activation of the IFNTc1 gene was not similar to that IFNT1 gene by AP1, ETS2 or/and CREBBP, expression plasmid. These results suggest that two isoforms of bovine conceptus IFNT genes are regulated differently in conceptuses during early pregnancy.
Interferon tau (IFNT), has known as a key signal molecule for a period of pregnancy in ruminants owing to the need on maternal recognition of pregnancy. It is generated in trophectoderm cells of the elongation bovine conceptus at day 13-21 and a peak output is at day 15-17 of pregnancy period. Moreover, other studies indicated that it can be effective in the embryonic development and quality. In previous study, there were 8 bovine IFNT, but only 2 forms of IFNTs, IFNT2 and IFN-tau-c1, were expressed by the conceptuses during the peri-implantation. In this study, we target the one between the two, IFN-tau-c1 and then the effect of IFNT knockout in donor cells to bovine cloned embryonic development by somatic cell nuclear transfer (SCNT) was investigated. In order to proceed this study, the immature oocytes from the ovaries at local slaughterhouse have been matured in vitro for 22 hours. For preparing the donor cell that have a mutation on IFNT gene, somatic cells were transiently transfected with Cas9 protein and single guide RNA targeting IFNT, and various single derived colonies with high proliferation were isolated and confirm the mutation by PCR. Finally, one colony had mono-allelic mutation (4bps deletion) was picked out and applied as the donor cell to SCNT. A donor cell was injected into an oocyte that nucleus was removed. Reconstructed oocytes with the donor cell were fused by electrical shock, activated by chemical stimulation and cultured for 7 days in chemically defined medium. In this study, control (n=199) and IFNT knockout-group (n=219) were compared with four replications. As results, there was no significant difference between control-and IFNT-knockout group not only in cleavage rate, but also blastocyst formation rate (Control: 12.3% ± 9.2, IFNT knockout-group: 20.1 ± 11%). In addition, the number of blastocyst cell was not different between control (91.7 ± 26.2) and IFNT knockout group (83.5 ± 21.3). Some IFNT mutated blastocysts from SCNT were randomly selected for confirmation of the deletion of IFNT and all samples were positive for mutation. In conclusion, these data indicated that the interruption of IFNT did not influence the embryonic development. In future study, we will transfer these mutated embryos toto test the effect of IFNT for pregnancy period. 과실파리류는 국내 60종의 검역 해충 가운데 42종을 차지하고 있다. 이들 대부분은 자연계 산물인 methyl eugenol (ME) 또는 raspberry ketone에 특이적으로 유인된다. 이들 유인물질을 파라핀 왁스를 이용하여 제형화하였고, 이를 대만에서 야외 유인 효과를 검증하였다. 이들 왁스 제형은 모두 델타트랩에 설치하고 8월 말경에 모니터링 한 결과 ME 왁스 제형은 오리엔탈과실파리(Bactrocera dorsalis)를 매일 60-80 마리 포획하 였다. 반면에 Cuelure 왁스제형은 오이과실파리(Bactrocera cucurbitae)와 B. tau를 유인하였다. 이러한 결과는 본 왁스제형이 이들 검역 해충에 대 한 국내 모니터링에 응용될 수 있다는 것을 제시하고 있다.

An antiluteolytic substance secreted by the bovine conceptus and primarily responsible for maternal recognition of pregnancy is bovine trophoblast protein 1 (bIFNT1), a new type I interferon tau (IFNT) genes. The objectives of this research were to investigate whether multiple, distinct gene encode bIFNT1 and other type I bIFNT gene in the bovine genome and to examine expression of bIFNT1 and other bIFNTc1 mRNAs during conceptus development. These transcrips could be regulated through caudalrelatedhomeobox-2 (CDX2) and ETS2 and/or AP1 (JUN) expression, a transcription factor implicated in the control of cell differentiation in the trophectoderm. The presence of mRNAs encoded by bIFNT1 and type I bIFNTc1 genes were examined quantitatively via reverse transcription-polymerase chain reaction (RT-PCR) analysis of total cellular RNA (tcRNA) extracted from on day 17, 20 and 22 bovine conceptuses. The expression level of bIFNT1 was higher on day 17 transcripts were gradually weakly detectable on day 20 and 22. However, the other bIFNTc1 gene examined transcripts was highly expressed on day 20 and transcripts were weakly detectable on day 17 and 22 bovine conceptuses. Furthermore, human choriocarcinoma JEG3 was co-transfected with an -1kb-bIFNT1/c1-Luc constructs and several transcription factor expression plasmids. Compared to each -1kb-bIFNT1/c1-Luc increased when this constructs were co-transfected with, ETS2, AP1(JUN), CREBBP and/or CDX2. Also, bIFNTc1 gene was had very effect on activity by alone ETS2, and AP1 (JUN) expression factors in choriocarcinoma JEG3 cell. However, bIFNT1 gene expression of the upstream region was not identified. We demonstrated that the activities of bIFN genes are regulated by differential, tissue-specific and developmental competence during pregnancy.
Interferon tau (IFNT), produced by the mononuclear trophectoderm, signals the process of maternal recognition of pregnancy in ruminants, but its expression in vivo is not well characterized. Objectives of this study were to determine IFNT gene isoforms expressed in the bovine uterus, and to identify differences in promoter sequences of IFNT genes that differ in their expression. Through the RNA-seq analysis of bovine conceptuses on days 17, 20 and 22 (day 0 = day of estrus), the expression of only two IFNT transcripts, IFNT1 and IFNTc1, were found, which were indeed classified into the IFNT gene clade. IFNT mRNAs were highest on day 17, and then decreased on days 20, and 22, which were also supported by the results of quantitative RT-PCR. Bovine ear-derived fibroblast (EF) cells were then cotransfected with luciferase reporter constructs carrying 5‘-upstream (positions -1000 to +51) regions of IFNT1 or IFNTc1 and various transcription factor expression plasmids. CDX2, either alone or with other Ap-1, ETS2 and/or CREBBP transcription factors, was found to increase luciferase activity approximately 10 and 18 fold more than twice of those cotransfected with bIFNT1, c1-Luc construct. Furthermore, The degree of transcriptional activation by a combination of the AP1, ETS2, CREBBP and/or CDX2 expression vectors was similar to that of CDX2 along plasmid. However, expression patterns of these Luc activity differented. Whereas bIFNTc1-Luc showed lowest antivity had than bIFNT1-Luc reports. Although, lowest antivity had of the bIFNTc1 –Luc report, cotransfected with the bIFNTc1-Luc construct and AP1(JUN) or/and ETS2 expression plasmid, Luc activity was enhanced approximately 2 and 4-fold more than the bIFNT1-Luc. Furthermore, along CDX2 expression factor had high effect on activity of bIFNT1-Luc reporter than the c1 gene in EF cells. These results suggest that two forms of IFNT genes are expressed in utero and their transcriptional regulations differ.

The results of spectral studies of the CTTS type young star RY Tau with spectrograms of the ultraviolet and the visual ranges are presented. We show the first detection of periodic variability of the emission line intensities in UV and visual ranges with a period of 23 days.
IWRAP 프로그램은 수로에서의 위험성을 평가하는데 유용한 프로그램이다. 그러나 이 프로그램의 기본 버전의 경우 AIS 데이터를 수집하는 기능이 포함되지 않아서, 더구나 베트남과 같은 개발도상국에서는 해상교통량 통계 데이터가 없는 실정이다. 사용자들은 수동으로 준비하여 입력하여야 한다. 이 연구는 IWRAP Mk2 프로그램 기본 버전을 사용하는데 있어 AIS 데이터를 전처리(pre-process) 할 수 있는 프로그램을 개발하고자 하였으며, 선박 형태, 선박 크기, 통과 시간 등으로 분류한 통항로에서의 선박 통항 척수 및 항로 배치와 같이 해역 내 해상교통에 관한 정보들을 사용자에게 제공하도록 고안되었다. 이렇게 개발된 통합 AIS 프로그램(Total AIS, TOAIS)은 베트남 Vung Tau 해역의 AIS 수집 데이터를 전처리할 수 있는지에 대하여 검증하였다. 그 결과, 통합 AIS 프로그램에서 전처리한 데이터를 이용한 IWRAP 프로그램은 베트남 해역의 위험성을 효율적으로 평가할 수 있었다.
This study is intended to examine the tDCS and Morris Water maze training in Alzheimer’s disease(AD) rats on Tau protein expression. Experiment groups were divided into four groups and assigned 16 rats to each group. Group Ⅰ was a control group(AD induced by scopolamine); Group Ⅱ was a experimental control group(AD injured by scopolamine and treatment tacrine); Group Ⅲ was a group of tDCS application after AD injured by scopolamine; Group Ⅳ was a group of morris water maze training after AD injured by scopolamine. In cognition test, the outcome of group Ⅱ was significantly lower than the groups(p<.001). and group Ⅲ, Ⅳ were significantly low result at 14 days(p<.05). In histological finding, the experimental groups were destroy of micro vessels and finding of cell atropy and swelling. Group Ⅲ, Ⅳ were decreased in degeneration of liver and kidney cells. In immuno- histochemistric response of BDNF and tau protein in hippocampus, BDNF expression of Group Ⅱ was more increase than the other groups. and increase of BDNF expression was Ⅲ, Ⅳ were higher than group Ⅰ at 21 days. Tau protein expression of Group Ⅱ was more decrease than the other groups. and decrease of Tau protein expression was Ⅲ, Ⅳ were lower than group Ⅰ at 21 days. These result suggest that improved tDCS and morris water maze training after scopolamine induced is associated with dynamically altered expression of BDNF and Tau protein in hippocampus and that is related with cognitive function.
Interferon-tau (IFNT) is regarded, generally, to be the conceptus protein that signals maternal recognition of pregnancy in ruminants. Although the discovery was made over two decades ago, the molecular mechanisms that regulate IFNT expression are not well understood. Previous studies demonstrated that transcription factors, caudal-related homeobox- 2 (CDX2), JUN, ETS2 and a transcriptional coactivator CREB binding protein (CREBBP) positively influenced IFNT gene expression, while OCT4 may exhibit negative regulation. We and others have observed that both CDX2 and OCT4 coexist during early stages of conceptus elongation but as development proceeds, OCT4 expression diminishes. The objective of this study was to evaluate the stimulatory and inhibitory effects of CDX2 and OCT4, respectively, on IFNT gene transcription when evaluated with other transcription factors. Human choriocarcinoma JEG3 cells were co-transfected with an ovine IFNT (-654 base pair)-luciferase reporter (-654-oIFNT-Luc) construct and several transcription factor expression plasmids. When the reporter construct was co-transfected with either CDX2, ETS2 or CJUN, transcription of -654-oIFNT-Luc increased about two-fold compared to -654-oIFNT-Luc alone. When -654-oIFNT-Luc was co-transfected with both c-jun and Ets-2, activity of -654-oIFNT-Luc was increased about four-fold; cotransfection with JUN, ETS2 and CDX2 increased -654-oIFNT-Luc expression 12X, indicating that the stimulatory activity of the transcription factors was additive. OCT4, when cotransfected with -654-oIFNT-Luc, reduced expression of the later about 40% when compared to -654-oIFNT-Luc alone. When co-transfected with JUN and/or ETS2, OCT4 abolished the stimulatory effect of these transcription factors. OCT4 also inhibited the stimulatory activity of CDX2 alone, but not when CDX2 was combined with JUN and/or ETS2. Therefore, when combined with the other transcription factors, CDX2 over the transcriptional inhibitory activity of OCT4. Conversely, when cells were transfected initially with OCT4 (0h) followed by transfection with CDX2, ETS2 and JUN 24 h later, -654-oIFNT-Luc expression was reduced to control (-654-oIFNT-Luc, alone) levels. Not surprisingly, 12S E1A, an inhibitor of transcriptional coactivator CREBBP, reduced stimulation of -654- oIFNT-Luc expression by CDX2, ETS2 and JUN, in combination, by about 40%, indicating that proper transcription complex formation is required for maximum expression. In conclusion, it is suggested that prior to conceptus elongation, pre-existing OCT4 may inhibit IFNT expression, but as elongation proceeds, IFNT expression increases, resulting from incremental increases in CDX2 expression, diminished OCT4 expression, and possibly proper transcription factor complex formation. Varroa destructor is an ecto-parasite mite and worldwide pest of the honey bee Apis mellifera L. The pyrethroid tau-fluvalinate (Apistan), an acaricide that is tolerated by honey bees, has been used for varroa mite control since the mid 1980s. Even though various resistances to tau-fluvalinate in varroa mites have been reported from Europe, Israel, and USA, the nature of tau-fluvalinate resistance in varroa mites in Korea has never been investigated. To investigate and understand tau-fluvalinate resistance in varroa mites in Korea, we conducted bioassay in several apiaries located different regions in Korea. To understand the molecular mechanisms underlying the difference of tau-fluvalinate resistances in varroa mites, partial genomic DNA fragments of a voltage-sensitive sodium channel gene from varroa mites were cloned and sequenced, since tau-fluvalinate is known to act on the sodium channels directly. Two novel mutations in sodium channels of varroa mites were present in eight apiaries. Two mutations might be a geographical polymorphism of sodium channel of varroa mites in Korea.
Sustainable development – including tourism development – is the general trend of all economies. With the orientation to build and develop the tourism industry into a key economic sector and become a tourist center of the Southeast region, Vietnam, Ba Ria-Vung Tau Province is demanding a proper, rational, development policy, while ensuring the development and exploitation of immediate natural advantages, and ensuring long-term sustainable development. The objective of this study is to examine the factors and their trends in sustainable tourism development in Ba Ria-Vung Tau Province, Vietnam. Methods used include descriptive statistical analysis, exploratory factor analysis and regression analysis. Data was collected from the results of a survey of 550 domestic and foreign tourists visiting Ba Ria-Vung Tau. With the help of SPSS statistical software, 12 groups of factors affecting sustainable development were identified including: economy, society, environment, humanistic tourism resources, natural tourism resources, tourism human resources, tourism products and service, tourism quality, infrastructure, technical facilities, governmental management and group of criteria to evaluate sustainable tourism development. The research results show that there are four groups of factors that significantly affect sustainable tourism development in Ba Ria-Vung Tau. These are groups of factors: Society, Environment, Tourism products and service, and Technical facilities.
This research examines the affect of destination image factors on revisit intention of domestic tourists at Ba Ria Vung-Tau (BRVT), by questioning directly 510 domestic tourists. The convenient sampling method is used in dividing the crowds into four groups; the four main surveyed areas in BRVT are: Vung Tau, Long Hai-Phuoc Hai, Xuyen Moc and Con Dao. A combination of qualitative and quantitative methodologies were utilized. A focus group of 10 domestic tourists was set up to review and explore the various factors as well as the conceptual model. An in-depth interview with 12 participants was developed to fine-tune measurement items. The questionnaire applied a 5-point Likert scale. Checking the reliability by Cronbach’s Alpha, exploratory factor analyzing and linear multiple regression were used with the SPSS program. The results show that there are eight main destination image factors affecting domestic tourists revisit intention to BRVT using linear regression and arranged by decreasing importance: Infrastructure, Variety Seeking, Accessibility, Local food, Atmosphere, Environment, Price Value, Leisure and Entertainment. From that, the research offers some suggestions and policy recommendations for the tourism business and provincial policy-makers to identify the main issues and develop better customer services to increase domestic tourists’ revisit intention.
