        Skeletal muscle is an organ that regulates biological metabolic energy. Its dysfunction causes decline of body functions and disability, thus deteriorating the overall quality of life. Various materials are being developed with an anti-sarcolytic effect. However, anti-sarcolytic effect of Sinomenium acutum rhizomes extract (SAE) remains unclear. Therefore, this study aimed to investigate anti-muscle atrophy effects of SAE and its alkaloids, including sinomenine (SIN), magnoflorine (MF), acutumine (ACU), and N-ferultyramine (NFT) isolated from SAE, on dexamethasone (Dex)-induced myotubules. C2C12 myogenic cells differentiated for 6 days were treated with 1 mM Dex for 24 hours. Induction of muscular atrophy was confirmed by a decrease in myogenin expression. We found that Dex increased expression levels of muscle-specific ubiquitin ligases MuRF1 and MAFbx/atrogin-1. However, mRNA and protein levels of these muscle-specific ubiquitin ligases were significantly reduced by cotreatment with SIN, MF, and NFT in myotubes. Glucose uptake reduced by Dex in myotubules were also restored by SIN, MF, and NFT treatments. These results suggest that SIN, MF, and NFT can reduce muscle wasting and enhance glucose uptake in Dex-treated myotubes, highlighting their potential as therapeutic agents to prevent muscle atrophy.

        우리나라의 해외 곡류 수입량과 수입국가의 다변화를 고려 해 볼 때, 호밀, 밀, 귀리 등 맥류에서 주로 발생하는 것으로 알려져 있는 맥각알칼로이드의 노출가능성도 배제 할 수 없다. 따라서 본 연구는 곡류 및 곡류가공품 중 맥 각알칼로이드에 대한 선제적 안전관리를 위해 호밀 및 가공품 (크래커, 맥주) 으로 부터 에르고메트린을 포함한 12 종 맥각알칼로이드의 공인 시험법을 개발하고자 하였다. 아세토니트릴이 함유된 2 mM 탄산암모늄용액 추출과 Mycocep catridge를 이용한 정제·농축이 전처리 방법으로 적합함이 확인되었고, 기기분석을 통해 표준물질 첨가 검량선의 직선성이 12개 알칼로이드 모두 R2 >0.99 이상이 었으며, 정량한계는 0.01 ~ 0.05 μg/kg수준이었다. 시험법 검증을 통해 대상 품목간의 차이는 있으나, 12개 맥각 알칼로이드 모두 회수율은 72~113% 수준이었으며, 반복성은 2~10% 수준으로 적합성이 확인되었다.

        빈카마이너 L.의 알칼로이드는 HPLC의 PDA 검출기를 이용하여 측정하고 이를 표준시료인 빈카민의 자외선/가시광선 흡수 스펙트럼과 비교하여 분석하였다. 플라보노이드류를 포함하여 추출물에서 9개 이상의 화합물에 대한 자외선/가시광선 흡수 스펙트럼을 얻었다. 추출방법에 따라 에탄올 추출물과 열수 추출물을 얻고 이를 DPPH 라디칼 소거법에 의해 항산화 효과를 조사하였다. 그 결과 에탄올 추출 물과 열수 추출물 모두 비교군인 케르세틴과 비타민 C 대비 약 14~15%의 항산화 효과를 보여 추출법에 상관없이 유사한 결과를 보였다.

        하동지역에서 생산되는 잎 녹차(우전, 세작, 중작, 대작)에 함유되어 있는 catechin류, alkaloid류 및 theanine를 HPLC를 이용하여 분석을 함과 동시에 녹차 추출물을 이용하여 총 페놀 물질과 항산화능을 측정하였다. Catechin류와 alkaloid류, theanine, 총 페놀 화합물의 함량은 물 추출물 보다 80% 알코올 추출물에서 더 높았다. 총 catechin과 alkaloid의 함량은 80% 에탄올로 추출한 우전 (172.33 mg/g, 30.80 mg/g)에서 가장 높았다. Theanine의 함량도 80% 에탄올 추출물에서 높았고 55.36에서 37.48 mg/g의 범위였다. 녹차의 총페놀 화합물은 우전에서 가장 높았고, DPPH법, FTC법 및 TBA법을 이용한 항산화 활성 측정에서도 우전에서 높은 결과를 나타내었다.

        Aza-indoles are important pharmacophores that have similar size and biological properties of indole. Here we have synthesized 4- and 7-azaindole tryptamines and showed that they are successfully incorporated in the biosynthesis of monoterepene indole alkaloids (MIAs) to form novel azaindole alkaloids by enzymatic reactions of strictosidine synthase(STR) and strictosidine glucosidase(SDG) monitored by UPLC/MS. By using HPLC equipped with a HPLC photo diode array(PDA) detector, each of the UV spectra of azaindole alkaloids was obtained and characterized. When hydrophilicity of azaindole alkaloids was compared, 4-azaindole alkaloids were more hydrophilic than 7-azaindole alkaloids.

        In this study, we examined the composition of catechins, theaflavins and alkaloids in leaves during processing to fermented black tea, which is produced by withering, roll breaking, and fermentation of Korean Yabukida tea plant. In addition, we determined the optimal conditions for the production of fermented black tea. The average moisture content in fresh leaves was 70.85%, which dropped to 3.07% in fermented black tea at the last stage of production. When the leaves were analyzed by HPLC, seven types of catechins, four types of theaflavins and three types of alkaloids were identified. The levels of catechins, theaflavins, and alkaloids were then evaluated after being processed into fermented tea. From these experiments, we found that the level of theaflavins, which determines the property of the tea, increased during fermentation. This effect resulted from the change in EGCG, ECG, EGC, EC during the process of fermentation. We also found that the maximal amount of theaflavins was created after 1-2 hours of fermentation. Thus, our results imply that the best condition for producing fermented black tea would be to ferment for 1-2 hours.

        This study was carried out to measure the contents of moisture, crude ash, crude fat, total amino acid, with amino acid composition, vitamin C, β-carotene, vitamin E, total catechins, EGCG, EGC, ECG, EC, GA, caffeine, theobromine and theophylline of the green tea I, II, III, oolong, and black tea. The content of crude fat of green tea I, II, III, oolong, and black teas was 1.1, 2.5, 4.9, 0.8 and 1.2% respectively, total amino acid content was 0.87, 0.78, 0.60, 0.63 and 1.05% respectively, and theanine content was 0.52, 0.48, 0.31, 0.41 and 0.61%, respectively. Total amino acid content of green tea increased in the order of green tea I $gt; green tea II $gt; green tea III, and among the teas, the content of theanine was the highest in the amino acids present. The content of vitamin C of green teal, II, III, oolong, and black tea was 101.6, 87.5, 95.9, 99.1 and 108.0 mg%, respectively, β-carotene content was 270, 268, 481, 80 and 181 ppm, respectively. Among the α-, β-, γ- and δ-tocopherol, the content of α-tocopherol was the highest in vitamin E present, and β- and δ-tocopherol were not detected in the samples of green teal, II, III, oolong, and black teas. The total catechins of green teal, II, III, oolong, and black teas was 10.5, 10.4, 7.2, 8.4 and 1.8% respectively, and among them, EGCG content was the highest. The content of EGC increased in the order of green tea I $gt; green tea III $gt; green tea II $gt; oolong tea $gt; black tea. The contents EGCG and ECG increased in the order of oolong tea $gt; green tea I $gt; green tea II $gt; green tea III $gt; black tea, and the highest contents of EGCG and ECG were observed in the samples of oolong tea. The content of GA was 0.01, 0.02, 0.05, 0.13 and 0.31%, respectively, and the highest contents of GA, caffeine and theobromine were observed in the sample of black tea. The highest content of theophylline, however, was observed in the sample of green tea I.

        Background : The 1,2-unsaturated PAs, reported to be widely present in medicinal plants belonging to Asteraceae, Boraginaceae, and Fabaceae, cause hepatotoxicity and genotoxicity in humans and animals. Hence, there is a need for an analytical method that allows these dangerous plant toxins to be determined. In this study, we developed a method that can be used for the rapid and accurate determination of nine toxic PAs in medicinal plants using ultra-pressure liquid chromatography–electrospray ionization–quadrupole–time-of-flight mass spectrometry (UPLC-ESI-Q-TOF). Methods and Results : The compounds were eluted onto a C18 column with 0.1% formic acid and acetonitrile, and separated with good resolution within 11 min. all analytes was characterized by its precursor ions generated by ESI-Q-TOF and fragment ions produced by collision-induced dissociation (CID), which were used as a reliable database. The proposed analytical method was verified with reference to the ICH guidelines. The proposed UPLC-ESI-Q-TOF method was applied to four medicinal plants, and lycopsamin, echimidine, senkirkine and senecionine were detected by matching with reference standard, and additional six PAs were tentatively identified though chemical profiling. In addition, the QuEchERS method was the most efficient in comparison with methods like hot water and methanol in extraction efficiency of pyrrolizidine alkaloids. Conclusion : The our proposed method can determine PAs rapidly and accurately in medicinal plants and will be utilize as an important data for other researchers who need analytical information of PAs.

        The quality control of natural products is principal key to guarantee the Good Manufacturing Practices (GMP) and Good Clinical Practices (GCP) for the functional food, pharmaceuticals and cosmeceuticals in the industry. In this study, we examined the quantitative analysis of berberine as marker substance of Coptidis Rhizoma by high performance liquid chromatography-photodiode array detector (HPLC-DAD). The HPLC method was validated and met all the requirements for the quality control analysis recommended by FDA and ICH. The berberine was separated on a Xterra C18 column (5 ㎛, 4.6 × 250 ㎜) using mobile phase consisting of distilled water and acetonitrile with KH2PO4 (3.4 g) and Na2SO4 (1.7 g). Calibration curve of berberine has been estimated (y = 42293.47x-41589 with the correlation coefficient 0.9999). The amount of berberine was calculated as 4.25%. And berberastine, palmatine, columbamine, jatrorrhizine, epiberberine, berberine and coptisine in the Coptidis Rhizoma were identified by high performance liquid chromatography - electrospray ionization-mass spectrometer (HPLC-ESI-MS) method.

        Scopolamine and hyoscyamine are important anticholinergic compounds obtained from Hyoscyamus niger. Adventitious roots induced from rhizome of H. niger and roots were cultured in SH medium supplemented with 3% (w/v) sucrose and 0.5 mg/L IBA. Roots were grown rapidly after 10 days of cultures. Scopolamine production was increased 7 times and hyoscyamine production was increased 12 times after 10 day of cultures. SH medium was best in root growth. But, highest scopolamine productivity was observed in WPM medium, followed White medium and best hyoscyamine productivity was resulted in MS medium. Sucrose was increased scopolamine and hyoscyamine production were increased the medium supplemented by sucrose comparing to than those by other carbon sources.

        Scopolia parviflora Nakai, a rare and endangered species, is the sole plant producing tropane alkaloids (TA) among the Korean native species. In order to enhance TA productivity the SP72 root line was selected by screening 100 of root line, and the optimal culture media for root growth and TA production were investigated with the SP72 roots. Based on the several media, SH and 2B5 medium were determined as growth medium and White and NN medium as production medium. Among the four combinations of two-stage culture, 2BN (2B5 as growth medium plus NN as production medium) showed more enhanced root growth and TA production as compared with production media of White and NN medium and growth media of SH and 2B5 medium, respectively. However, bubble column bioreactor (BCB) cultures applying two-stage culture did not reveal the effective results despite of the each successful operation of two-stage culture in conical flasks and BCB cultures.

        Hyoscyamine and scopolamine are two most common tropane alkaloids found in the Solanaceae. The pEB expression vector carrying Nspmt gene was transformed to Agrobacterium rhizogenes. The growth of transgenic hairy roots were approximately to 80% of the wild type root. Transgenic hairy roots are less developed on only their branch roots than those of the wild type root. The extracts from Nspmt transgenic hairy root lines, 3 and 5 contained between 3.52 and 4.23 mg/g dry weight as hyoscyamine and did between 5.23 to 6.40 mg/g dry weight as scopolamine. These results showed that the overexpression of the pmt gene enhanced tropane alkaloids production of S. parviflora transformed roots and this improvement affected both hyoscyamine and scopolamine production.

        미치광이풀의 모상근 뱌양에 의해 의학적으로 중요한 물질인 scopolamine과 hyoscyamine 생산을 증진시키기 위해 3종의 곰팡이와 1종의 효모 유래의 생물학적 elicitor를 supernatant와 homogenate의 형태로 처리하였다. Elicitor 중 Candida albicans는 supernatant와 homogenate 모두 scopolamine 생산성을 증가시켰다. Fusarium solani의 homogenate와 C. albicans의 supernatant 처리로 hyoscyamine 함량이 증가되었다. 그러나 나머지 fungus strain도 대조구에 비해 tropane alkaloids의 생산을 다소 증가시켰다. 본 연구에서는 C. albicans가 tropane alkaloids 생산에 적합한 elicitor임으로 구명되었다. 본 연구 결과는 미치광이풀 모상근 대량배양에 의한 tropane alklaoids 대량생산에 기여할 것이다.

        남부지방에 서식하는 식물체 중에서 꽃기린, 민들레, 박주가리, 후박나무, 팔손이 나무, 달맞이꽃, 동백나무 그리고 유동나무를 대상으로 약용성분의 일부분을 차지하고 있는 saponins 및 alkaloids의 검색을 실시한 결과를 요약하면, 1. Crude saponins 함량 중, crude 인삼 saponins 함량은 달맞이꽃이 가장 높았으며, crude saikosaponins 함량은 박주가리에서 가장 높게 나타났다. 2. Crude alkaloids 함량은 박주가리에서 가장 높게 나타났다. 3. Saponins 획분에 대해 HPLC분석을 실시한 결과, 꽃기린, 민들레 그리고 박주가리에는 인삼 saponin태 물질, 후박나무, 팔손이나무 그리고 달맞이꽃에는 인삼 saponin태 물질과 saikosaponin c태 물질, 동백나무와 유동나무에는 saikosaponin c태 물질의 존재가 추측되었다. 4. Alkaloids 획분에 대해 GC분석을 실시한 결과, 팔손이나무에서만 nicotine태 물질의 존재가 추측되었다.
