항생제는 과도한 사용으로 인해 폐수뿐만 아니라 다양한 수원에서 발견되는 새로운 오염 물질 중 하나입니다. 수 중 항생제 오염 물질을 처리하기 위한 고도 산화 공정, 생물학적 처리 등 다양한 기술이 있습니다. 이 두 가지 공정은 비효율 적이며, 부산물의 생성은 이 공정을 더욱 복잡하게 만듭니다. 오염 물질을 제거하기 위한 또 다른 대안으로 막 기술이 있습니 다. 항생제와 내성 유전자의 제거를 개선하기 위해 막 생물 반응기는 NaClO와 탄소 물질로 변형됩니다. 풍부한 반응성 종의 생성은 항생제의 내성 유전자에 대해 활성입니다.
우리는 길앞잡이(Cicindela chinensis)의 장에서 다양한 공생 미생물들을 분리하였다. 그중 다양한 곰팡이 성장 을 억제하는 세균을 동정하였고 “Ch-1”이라 명명하였다. 우리는 Ch-1 균주를 사용하여 10종의 식물 병원성 곰팡 이와 2종 곤충 병원성 곰팡이의 생장 억제를 확인하였다. 또한 8종의 항생제에 대한 저항성을 확인하였다. 동시 에, 본 균주의 genomic sequence를 수행하였고 유전적, 생화학적, 생리적 특성을 조사하였다. Ch-1균주는 특허등 록과 친환경 미생물제제로 등록하였고 향후 생물학적 방제제로써 활용될 수 있을 것으로 판단한다.
아미노글리코사이드계 항생제(Aminoglycosides, AGs) 는 그람음성균과 양성균에 광범위하게 작용하는 동물용 의약품으로, 최근 배양육에 사용된다고 알려져 있어, 안 전성 관리를 위한 분석법 마련이 반드시 필요하다. AGs 는 고극성 화합물로 성분 간의 분리를 위해 이온쌍 시 약(ion-pairing reagent, IPR)을 사용하고 있으나 IPR을 이동상에 첨가하는 기존 분석방법의 경우 용매가 흐르 는 동안 질량분석기로 주입되는 IPR로 인해 기기적인 문 제가 발생할 가능성이 높아, IPR를 바이알에 직접 첨가 하는 분석방법을 검토하였다. 본 연구에서 10종 AGs 성 분에 대한 분석방법을 확인하고 유효성을 검증하였다. 검출한계와 정량한계는 각각 0.0001-0.0038 mg/kg 와 0.004-0.011 mg/kg의 범위로 나타났으며, 0.01-0.5 mg/ kg 범위 내의 직선성(R2)은 0.99 이상이었다. AGs의 시 료 회수율을 확인하고자 소고기와 세포배양배지(cell culture medium) 매질에서 회수율과 상대표준편차로 나 타낸 정밀도를 확인한 결과 각각 70.7-120.6% 및 0.2 to 24.7%로 나타났다. 기존의 이동상에 IP 첨가 방법과 비 교하였을 때 유사한 수준으로 양호하였다. 검증된 AGs 분석법은 국내 유통되는 닭고기, 소고기, 돼지고기 15품 목과 배양육 배지 첨가제 6품목에 적용해보았다. 그 결 과 국내 유통되는 육류 15품목 모두 AGs 성분이 검출 되지 않았으나, 세포배양배지에서 streptomycin은 695.85- 1152.71 mg/kg, dehydrostreptomyci은 6.35-11.11 mg/kg 로 검출되었다. 따라서 IRR을 바이알에 직접 첨가하는 LC-MS/MS 방식은 육류, 세포배양배지, 배지첨가제 중 AGs 분석 및 안전성 평가를 위한 기초자료로 활용될 것 으로 기대된다.
The purpose of this study was to compare the efficiency of air and oxygen injected into the underwater non-thermal dielectric barrier discharge plasma (DBD plasma) device used to remove five types of antibiotics (tetracycline, doxycycline, oxytetracycline, clindamycin, and erythromycin) artificially contained in the fish farm discharge water. The voltage given to generate DBD plasma was 27.8 kV, and the measurement intervals were 0, 0.5, 1, 2, 4, 8, 16 and 32 minutes. Tetracycline antibiotics significantly decreased in 4 minutes when air was injected and were reduced in 30 seconds when oxygen was injected. After the introduction of air and oxygen at 32 minutes, 78.1% and 95.8% of tetracycline were removed, 77.1% and 96.3% of doxycycline were removed, and 77.1% and 95.5% of oxytetracycline were removed, respectively. In air and oxygen, 59.6% and 83.0% of clindamycin and 53.3% and 74.3% of erythromycin were removed, respectively. The two antibiotics showed lower removal efficiency than tetracyclines. In conclusion, fish farm discharge water contains five different types of antibiotics that can be reduced using underwater DBD plasma, and oxygen gas injection outperformed air in terms of removal efficiency.
활성 의약품 성분(APIs)의 존재가 수생 생태계와 인간의 건강에 위험하다는 증거가 있다. 물에 항생물질인 테트 라사이클린과 같은 API가 존재하면 미생물에 항균제 내성(AMR)이 발생해 개인과 사회에 막대한 비용이 발생한다. TiO2 또 는 비스무트 기반 촉매와 같은 촉매가 내장된 막은 유기 유출물을 분해하고 폐수로부터 분리한다. 촉매의 광촉매 활성은 귀 금속 도핑 및 탄소성 물질의 첨가 및 다른 반도체와의 헤테로 접합 형성으로 향상될 수 있다. 광촉매의 회수는 고분자 막에 서 광촉매의 고정화를 통해 가능하다. 이 검토에서는 물 속 항생제의 분해가 논의된다.
급성 췌장염은 담석, 음주 등의 원인으로 발생하는 급성 염증성 질환이다. 약 80%에서는 췌장에만 염증이 생기는 경증 췌장염으로 그치나, 약 20%의 환자에서는 증증 괴사성 췌장염으로 발전하고, 보고에 따라 사망률이 43%에 이른다. 이에 따라 초기의 집중적인 치료는 매우 중요하다. 본고에서는 급성 췌장염의 치료 중 영양과 항생제의 사용에 대해 살펴보고자 하였다. 전통적으로 급성 췌장염은 금식으로 췌장을 쉬게 한다는 개념으로 한 치료가 강조되었고, 이와 함께 대량 수액 공급, 통증 조절, 영양 공급, 적절한 항생제 치료가 주로 이루어졌다. 그러나 최근 조기에 경장 영양을 시작하는 것이 입원 기간 단축, 다발성 장기 부전, 전신 감염, 수술적 치료 필요성을 개선시키고 사망률을 개선시킨다는 것이 여러 연구들을 통해 입증되었다. 따라서 금기사항만 없다면 가급적 조기에 경장 영양을 시키는 것이 장려된다. 또한 무작위 대조군 연구들에 따르면 예방적 항생제가 감염을 예방하거나 사망률을 감소시키는 것에 도움이 되지 않고 오히려 항생제 내성을 유발하고 의료비 지출이 높아지므로 예방적 항생제 사용을 권장하지 않고 있다. 하지만 중증 급성 췌장염에서는 예방적 항생제를 쓰는 것에 대해 여전히 논란이 되고 있고 감염이 의심될 때에는 광범위 항생제를 조기에 쓰는 것이 필요하다. 결론적으로 급성 췌장염의 치료에 있어 전통적으로 행해 왔던 금식보다는 조기에 경장 영양을 시작하고 항생제는 적절히 사용하는 것이 중요하다.
Precise combinations of probiotics can be useful in dog nutrition, treatment and care. Also, host specificity must be considered in order to increase the effectiveness of probiotics. In this study, Lactobacillus acidophilus HY7032 and Lactobacillus reuteri HY7506 were used, which were isolated from feces of healthy dogs through the verification of pH, bile salt tolerance, and antibacterial activity. In addition, the selected strains were confirmed for activity in immune cells. Briefly, L. acidophilus HY7032 and L. reuteri HY7506 enhanced oxidative burst and phagocytosis of innate immune cell activities in peripheral blood. In addition, beagle were administered vancomycin 50 mg and polymyxin B 100 KU for 7 days, and then 107 CFU of L. acidophilus HY7032 and L. reuteri HY7506 were orally administered for 3 weeks to confirm the effect of improving hair quality. Also, compared with the placebo group, the health improvement effect including stool pattern were confirmed. These results imply that the microflora imbalance caused by antibiotics can be gradually improved through the intake of probiotics. Through this study, it was confirmed that L. acidophilus HY7032 and L. reuteri HY7506 are good probiotics that contribute to the welfare and health of companion animals and have the effect of improving hair quality.
Recurrent aphthous stomatitis (RAS) is a common oral mucosal disorder for which no curative treatment is available. We previously reported that decreased Streptococcus salivarius and increased Acinetobacter johnsonii on the oral mucosa are associated with RAS risk. The purpose of this study was to identify antibiotics that selectively inhibit A. johnsonii but minimally inhibit oral mucosal commensals. S. salivarius KCTC 5512, S. salivarius KCTC 3960, A. johnsonii KCTC 12405, Rothia mucilaginosa KCTC 19862, and Veillonella dispar KCOM 1864 were subjected to antibiotic susceptibility test using amoxicillin, cefotaxime, gentamicin, clindamycin, and metronidazole in liquid culture. The minimal inhibitory concentration (MIC) was defined as the concentration that inhibits 90% of growth. Only gentamicin presented a higher MIC for A. johnsonii than MICs for S. salivarius and several oral mucosal commensals. Interestingly, the growth of S. salivarius increased 10~200% in the presence of sub-MIC concentrations of gentamicin, which was independent of development of resistance to gentamicin. In conclusion, gentamicin may be useful to restore RAS associated imbalance in oral microbiota by selectively inhibiting the growth of A. johnsonii but enhancing the growth of S. salivarius.
Oxidation of erythromycin, sulfamethazine and sulfathiazole by ozone was experimentally investigated to see the effects of background water quality such as ultrapure water, humic acid and biologically treated wastewater and water temperature on the removal rate, consequently to provide design information when the ozone treatment process is adopted. Initial concentration of the antibiotics was spiked to 10 μg/l and ozone dose was 1, 2, 3, 5, 8 mg/l. While the removal rate of erythromycin under ultrapure water background by ozone oxidation was over 99%, that under humic acid and biologically treated wastewater background was markedly reduced to the range of 59.8%~99% and 17.0%~99%, respectively. When water temperature is decreased from 20℃ to 4℃, the removal rate is reduced from the range of 17.0%~99% to the range of 9.4%~97.4% under biologically treated wastewater background. The effects of background and temperature on the removal rate of sulfamethazine and sulfathiazole were similar to erythromycin, but the degree was different. Therefore, it is concluded that the background of water to be treated as well as water temperature should be taken into consideration when the design factor such as ozone dose is determined to meet the treatment objective in the ozone treatment process.
Aedes albopictus (Skuse) has been involved as the potential vector of imported zika virus disease and dengue fever throughout Republic of Korea. It has been reported that the successful introduction of a life-shortening strain of Wolbachia into Ae. albopictus halves adult lifespan and also directly inhibits the ability of a range of pathogens to infect this mosquito species. In our previous studies, Ae. albopictus collected from field was naturally infected by two strains of Wolbachia (wAlbA and wAlbB). However, virus regulation of two Wolbachia strains in Ae. albopictus have not been assessed, yet. In our studies, the two Wolbachia strains in about 20% among Ae. albopictus were removed for the virus regulation test using tetracycline. In this presentation, the detailed results and methods are introduced.
Agrobacterium tumefaciens-mediated transformation (AtMT) has been widely used for generation of fungal transformants and recently applied to Beauveria bassiana. In this study to comprehend how the AtMT promoter influences on the expression of selection marker (hygromycin B resistance gene; hph), two different Ti-Plasmids were constructed: pCeg (gpdA promoter-based) and pCambia-egfp (CaMV 35S promoter-based). Putative transformants were subjected to the PCR, RT-PCR and qRT-PCR to inspect the T-DNA insertion rate and gene expression level. In conclusion, more than 80% of the colonies succeeded in AtMT transformation and the hph expression level of AtMT/pCeg colonies was higher than that of AtMT/pCambia-egfp colonies. This result can provide useful information on the AtMT of B. bassiana, especially antibiotics susceptibility and promoter-dependant expression level.
A voltammetric analysis of doxycycline was developed using DNA immobilized onto a carbon nanotube paste electrode (PE). An anodic peak current was indicated at 0.2 V (versus Ag/AgCl) in a 0.1M NH4H2PO4 electrolyte solution. The linear working range of the cyclic and square wave stripping voltammetry was obtained to 1-27 ngL-1 with an accumulation time of 800 s. Final analytical parameters were optimized to be as follows: amplitude, 0.35 V; frequency, 500 Hz; and pH, 5.43. Here detection limit was found to be 0.45 ngL-1, this result can be applied in foods systems and in the biological diagnostics
Minimal inhibitory concentration (MIC) is the lowest antibiotic concentration that inhibits the visible growth of bacteria. Sub-minimal inhibitory concentration (Sub-MIC) is defined as the concentration of an antimicrobial agent that does not have an effect on bacterial growth but can alter bacterial biochemistry, thus reducing bacterial virulence. Many studies have confirmed that sub-MICs of antibiotics can inhibit bacterial virulence factors. However, most studies were focused on Gram-negative bacteria, while few studies on the effect of sub-MICs of antibiotics on Gram-positive bacteria. In this study, we examined the influence of sub-MICs of doxycycline, tetracycline, penicillin and amoxicillin on biofilm formation and coaggregation of Streptococcus gordonii, Streptococcus mutans, Actinomyces naeslundii, and Actinomyces odontolyticus. In this study, incubation with sub-MIC of antibiotics had no effect on the biofilm formation of S. gordonii and A. naeslundii. However, S. mutans showed increased biofilm formation after incubation with sub-MIC amoxicillin and penicillin. Also, the biofilm formation of A. odontolyticus was increased after incubating with sub-MIC penicillin. Coaggregation of A. naeslundii with S. gordonii and A. odontolyticus was diminished by sub-MIC amoxicillin. These observations indicated that sub-MICs of antibiotics could affect variable virulence properties such as biofilm formation and coaggregation in Gram-positive oral bacteria.
Antibiotics have been used to prevent disease, promote growth rate, and improve feed efficiency. However, the use of antibiotics in livestock has been restricted worldwide due to problems such as bacterial resistance. Therefore, probiotics among alternatives to antibiotics have gained attention in the livestock feed industry these days. This study was conducted to investigate the effects of dietary supplementation with probiotic 379D on safety, growth rate, and feed efficiency. In this study, bacterial strain 379D was isolated from soil and identified as a Bacillus sp. according to 16S rRNA sequence analysis. In an in vitro test, in-gel activity assay and antimicrobial susceptibility test were conducted to evaluate 379D. In an in vivo study, 379D was administered at concentrations of 0.1% and 1% to broiler chickens for 28 days. The results of in-gel activity assay and antimicrobial susceptibility test showed that strain 379D had broad spectrum antimicrobial activity. Furthermore, no adverse 379D-related effects were observed in 0.1% and 1% groups. Feed efficiency was higher in the 379D-treated groups than in the control group. In conclusion, 379D is expected to be used as a safe alternative to antibiotics in a feed supplement and will improve feed efficiency in broiler chickens.
