        본 연구는 만 65세 이상 85세 이하의 제2형 당뇨병 비만 여성 노인을 대상으로 12주간 복합운동 후 irisin, 신체조성 및 당 대사에 미치는 영향을 구명하기 위하여 복합운동군(n=20), 대조군(n=16)으로 구분하여 실시하였다. 복합운동 프로그램은 야외 걷기 운동과 탄성 밴드 운동을 12주간 주 3회, 1회 60분 간 실시하였으며, 야외 걷기 운동의 운동강도는 중강도(RPE 5~6)로 수행되었으며, 탄성 밴드 운동은 1~4주는 저강도(OMNI-RES 3~4), 5~8주는 중강도(OMNI-RES 5~6), 9~12주는 고강도(OMNI-RES 7~8)로 점진적으로 운동강도를 증가시켰다. 그 결과 복합운동군의 irisin이 유의하게 증가하였고(p<.001), 체지방률은 유의하게 감소하였으며(p<.001), 골격근량은 유의하게 증가하였다(p<.001). 당 대사 관련 인자 중 HbA1c(p=.020), 혈당(p<.001)은 유의하게 감소하였고, HOMA-β는 유의하게 증가하였다(p<.001). Irisin의 변화는 체지방률의 변화와 부적 상관이 있는 것으로 나타났고(r=-.423, p=.010), 유산소성 지구력 변화와 정적 상관이 있었다(r=.355, p=.034). 또한, HbA1c(r=-.351, p=.036)와 혈당(r=-.424, p=.010)의 변화는 부적 상관이 있었으며, HOMA-β는 정적 상관이 있는 것으로 나타났다(r=.411, p=.013). 결론적 으로 규칙적인 복합운동의 실천은 제2형 당뇨병 비만 여성노인의 혈중 irisin의 수준을 증가하고, 신체조성 변화에 긍정적인 영향을 주는 것으로 나타났다. 그리고 당화혈색소, 공복혈당 및 인슐린 분비능을 개선하여 당 대사 조절 능력에 도움을 주어 당뇨 예방 및 관리를 위해 효과적인 운동으로 권장될 것으로 사료된다.

        Streptozotocin(STZ)을 45mg/kg.b.w의 용량으로 흰쥐의 미정맥에 투여 한 후 유발된 당뇨 흰쥐에게 1일 1회 7일간 1,000mg/kg의 용량으로 도라지 뿌리 에탄올 추출물을 투여 후 glucose 함량과 당대사에 관여하는 효소인 glucose-6-phosphatase(G-6-Pase) glucose-6-phosphate dehydrogenase (G-6-PDH), glucokinase(GK)활성과 glycogen 함량, triglyceride(TG), total cholesterol등의 지질대사에 관여하는 물질들을 측정하였다 그 결과 도라지 뿌리 에탄올 추출물 투여군이 glucose, TG, total cholesterol등의 함량과 G-6-Pase 활성의 유의적인 감소(p<0.05)를 나타내었으며 glycogen 함량과 HDL-cholesterol, G-6-PDH, GK의 활성이 유의적인 증가(p<0.05)를 나타내었다. 이와 같이 도라지 뿌 리 에탄올 추출물이 항당뇨 개선효과를 갖는 유효성분을 함유하고 있음을 알 수 있었다.

        In the present study, we examined potential roles of glucose and pyruvate in nuclear and cytoplasmic maturation of porcine oocytes. In the presence and absence of 10% porcine follicular fluid (PFF), either 5.6 mM glucose or 2mM pyruvate effect on meiotic maturation and followed development ability. However, DOs doesn't take full advantage of the glucose in medium, only pyruvate can increase MII rate and follow early embryo development ability significance. COCs were matured with 200 uM pentose phosphate pathway (PPP) inhibitor (dehydroepiandrosterone, DHEA) or 2 μM glycolysis inhibitor (iodoacetate, IA), significantly lower levels of GHS in the DHEA an IA treated oocytes and the levels of ROS were higher significantly in the DHEA treated oocytes, treatment with DHEA significantly reduced the intra-oocyte ATP and NADPH level. Blastocysts from DHEA or IA treated group also presented higher apoptosis levels, meanwhile, the percentage of proliferating cells was dramatically lower than the non-treated group. In conclusion, our results suggest that 10% PFF promoted oocytes make full use of energy, glucose metabolism during in vitro maturation inseparable from the cumulus cells, PPP and glycolysis promoted porcine oocytes cytoplasmic maturation by supplying energy and reducing oxidative stress.

        The ethanol extraction yield of Meliae toosendan fructus(MT) was about 24.5% by extract apparatus. This study was done to investigate the carbohydrate metabolism related enzyme activities and antioxidative effects of MT in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) were significantly decreaed in MT treated group compared to the those of STZ-control group, also content of Total cholesterol was decreased. High density lipoprotein (HDL)-cholesterol was increased in MT treated group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in MT treated group. Also the activities of glucose-6-phosphate dehydrogenase(G-6-PDH) and glucokinase(Gk) were increaed in MT treated group. The content of hepatic glycogen was significantly increaed in MT treated group, in addition, content of malondialdehyde(MDA) was significanly decreased in MT treated group. Also, content of glutathione(GSH)was dereased in MT treated froup. whereas, activity of catalase(CAT) was significantly increaed in MT treated group compared to the those of STZ-control group. activity of glutathione peroxidase(GSH-Px) was inecreaed. In conclusion, these results indicated that ethanol extract of MT would have carbohydrate metabolism antioxidative effects in STZ-induced diabetic rats.

        The purpose of this study was to investigate the effects of dietary Indongcho(L. japonica Thunb) powder on blood glucose, serum lipid levels and antioxidative enzymes in normal and streptozotocin(STZ)-induced diabetic rats. Four groups of rats(3-week-old inbred Sprague-Dawley male rats) were normal rats fed control diet(NC), diabetic rats fed control diet(DC), normal rats fed Indongcho powder diet(NI), and diabetic rats fed Indongcho powder diet(DI). Diabetes was induced by single injection of streptozotocin(60mg/kg B.W., i.p.). The animals were fed ad libium each of the experimental diet for 5 weeks. Food and water intakes were determined everyday. Blood glucose and serum total cholesterol levels were determined every week. After 5 weeks the animals were sacrificed and activities of antioxidant enzymes and lipid peroxidation products were determined in their liver and kidney homogenates. We also determined serum concentrations of total lipid(TL), total cholesterol(TC), triglycerides(TG) and HDL-cholesterol(HDL-C). Blood sugar and water intake were higher in diabetic group(DC and DI group) than normal group(NC and NI group) and were not significantly decreased by dietary Indongcho intake. Body weight gain and FER(feed efficiency ratio) were reduced by STZ treatment. But, Final body weight was recovered by Indongcho-contained diet. LHR(LDL-cholesterol/HDL-cholesterol) of the DI g re up was significantly lower than the other experimental groups(NC, NI and DC groups). The hepatic glucose 6-phosphatase(G6Pase) activity of the groups fed Indongcho diet(NI and DI group) was lower than the groups fed control diet(NC and DC group) and the G6Pase activity of NI group was recovered to the normal levels(p<0.05). However, The glutathione peroxidase(GPx) and glutathione reductase(GR) activities in liver and G6Pase activity in kidney were not statistically different between the control and diabetic control groups. Renal GST activity of the DI group was recovered by Indongcho intake. In conclusion, these results confirm oxidative stress in the liver and kidney of rats with STZ diabetes and antioxidative effect of Indongcho.

        The purpose of this study was to investigate the improvement effect of 5% turmeric (Curcuma longa L.) on the blood glucose and lipid metabolism function of streptozotocin (STZ, 45 mg/kg body weight)-induced diabetic rats. Seven-week-old male rats were divided into four groups (n=6), and fed experimental diets containing turmeric meal [basal diet+5% turmeric (BT), basal diet+STZ+5% turmeric (ST)], and control (BD), BS groups (basal diet+STZ). The results of this study, turmeric diet groups (BT, ST) in lipid component as evidenced from the significantly reduction of serum total cholesterol, low density lipoprotein-cholesterol (LDL-cholesterol), atherosclerotic index (AI), cardiac risk factor (CRF), triglyceride (TG), phospholipid (PL), free cholesterol, cholesteryl ester, uric acid, blood glucose, non esterified fatty acid (NEFA), and elevation of high density lipoprotein-cholesterol (HDL-cholesterol). The serum albumin and globulin contents were increased in turmeric supplementation diet than STZ-induced diabetic rats (p<0.05). Concentrations of sodium (Na) and chlorine (Cl) in sera were lower in the turmeric diet than diabetic group. Total calcium (T-Ca), phosphorus (Pi) and potassium (K) concentrations in sera were higher in the BT, ST and BD groups than BS group. In vivo experiments with Sprague-Dawley rats showed that ingestion of turmeric (Curcuma longa L.) were effective in the blood glucose and lipid metabolism functional improvement.

        Menopause is characterized by a decrease in estrogen production by the ovaries. Furthermore, the risk of developing chronic diseases including cardiovascular disease, obesity, and diabetes increases in menopausal women. In this study, we evaluated the ability of Caragana sinica (CS) to affect lipid and glucose metabolism in an ovariectomized Sprague-Dawley rat model of induced menopause. The animals were divwided randomly into three groups: sham-operated rats (SHAM), ovariectomized rats (OVX), and ovariectomized rats treated with 1% water extract of CS (OVX-CS). The OVX-CS treated mice showed a significant decrease in body weight, adipose tissue, triglyceride, and total cholesterol. Improved lipid and glucose profiles were also observed, and were attributed to suppression of hepatic lipogenesis and adipokine production and regulation of glucose-regulating enzyme activity. Therefore, these findings indicate a potential use for CS as a functional food for menopause-induced hyperlipidemia and hyperglycemia.

        The ovariectomized Sprague-Dawley female rats were randomly assigned to Sham-Control, OVX-Control, OVX-Superjami (extract) groups. The results showed that the activity of glucokinase to keep the blood sugar constant is increased by increasing insulin secretion from pancreatic β- cells and the homeostatic regulation of glucose. Meanwhile the glyconeogenesis which is involved in the actions of the enzymes glucose-6-phosphatase and phosphoenolpyruvate carboxykinase showed that the glucose level is decreased. It was confirmed that these enzymes regulate the carbohydrate metabolism. On the other hand, results of the measurement of the lipid metabolism in the fat tissue and liver tissue, effect of β-oxidation enzymes and carnitine palmitoyl transferase which is involved in fatty acid oxidation for energy generation is increased. Moreover, the activity of fatty acid synthase, glucose-6-phosphate dehydrogenase and malic enzyme have been reduced, therefore, it was confirmed that these enzymes regulate the lipid metabolism.

        This study was designed in order to determine the influences of haw (Crataegus pinnatifida BUNGE) on the lipid metabolism syndrome. Sprague Dawley rats, 7 weeks old, were given four different types of diets for 5 weeks: ND group (noncholesterolemic diet), HE group (non cholesterolemic diet+haw extract), CD group (cholesterolemic diet), CH group (cholesterolemic diet+haw extract). Concentrations of LDL-cholesterol, triglyceride, phospholipid, free cholesterol, cholesteryl ester, blood glucose were remarkably higher in the CD group than the other groups. But by treatment of haw extract group with cholesterolemic diet (CH group) were significantly decreased compared with CD group. Cholesteryl ester ratio was no difference between CD group and CH group. These results indicate that Crataegus pinnatifida BUNGE would be effective in lipid metabolism syndrome.
