        Cotesia plutellae, an endoparasitoid wasp, parasitizes larvae of Plutella xylostella, and disrupt immune response of the host through parasitic factors. These immune disruption factors are maternal (venom proteins, polydnavirus, and ovary proteins) and embryonic (teratocytes) factors. In this study, we performed transcriptome analysis of venom glands of C. plutellae and identified neprilysin-1 (NEP1) known to be potential immunosuppression gene. Cp-NEP1 encoded 451 amino acid and belongs to hymeopteran NEP1 via phylogenetic analysis. Based on the structural comparison Cp-NEP1 lacks in conserved motifs such as substrate binding (NAYY/F), zinc-binding site (HExxH), zinc-binding site, protein folding and maturation (CxxW). To investigate function of Cp-NEP1, we constructed a recombinant Cp-NEP1 harboring N-terminally fused 6X His tag. Peptide sequencing revealed successful expression of the recombinant Cp-NEP1 in Escherichia coli. Pre-heated E. coli as antigen induced spike of nodule formation whereas co-injection of the recombinant Cp-NEP1 and pre-heated E. coli exhibited suppression of nodule formation in the host. Quantitative real-time PCR revealed that expression of phenoloxidase related to nodule formation was suppressed under co-injection of the recombinant Cp-NEP1 and E. coli. These results suggest that Cp-NEP1 contributes to immunosuppression of P. xylostella via phenoloxidase suppression and conserved motifs of neprilysin family are not required for host immune suppression.

        Human body and head lice are obligatory human ectoparasites. Although both body and head lice belong to a single species, Pediculus humanus, only body lice are known to be a vector of several bacterial diseases. The higher vector competence of body lice is assumed to be due to their weaker immune response than that of head lice. To test this hypothesis, immune reactions were compared between body and head lice following infections by two model bacteria, Staphylococcus aureus and Escherichia coli, and a human pathogen, Bartonella quintana. Following dermal or oral challenge, the number of these bacteria increased both in hemocoel and alimentary tract of body lice but not in head lice and the viability of the B. quintana was significantly higher in body louse feces, the major route of infection to human. In addition, body lice showed the lower basal/induced transcription level of major immune genes, cytotoxic reactive oxygen species and phagocytosis activity compared with head lice. These findings suggest that a reduced immune response may be responsible, in part, for the increased proliferation and excretion of viable bacteria which are associated with the high level of human infectivity seen in body versus head lice.

        Viral respiratory infections are common in horses, notably equine herpesvirus infection and equine influenza, which primarily initiate secondary bacterial respiratory infections such as strangles caused by Streptococcus equi equi. A decline in the production of stallions has been associated with these respiratory diseases leading to adverse financial implications. This study investigated the antibody responses against respiratory diseases in horses from Jeju Island a year after vaccination. A low level of equine herpesvirus type 1 (EHV-1) (11.36%) antibodies was detected from stallions, however a high level of EHV-4 (95.84%) antibodies was detected from horses without vaccination against this infection suggesting that EHV-4 is ubiquitous in this horse population. In case of equine influenza, ranch stallions showed low positive rate (12.06%) whereas stallions from Subtropical Livestock Research Institute displayed higher positive rate (81.32%). Antibody responses against equine influenza and strangles revealed positive rates of 26.32% and 55.12%, respectively. These findings may draw attention towards the importance of developing an improved disease prevention and/or immunization program that will effectively control respiratory diseases in horses.

        Sasa quelpaertensis Nakai is a type of edible bamboo grass distributed on Jeju Island, Korea. S. quelpaertensis has been used as afolk medicine for treatment of a variety of ailments. It has been reported to present biological effects, including anti-inflammatory and antioxidant effects. In this study, we demonstrate that S. quelpaertensis Nakai extract (SNE) rescues immunocytes from gamma radiation-induced apoptosis and oxidative DNA damage. We examined the cytotoxicity, cell proliferation, DNA damage, apoptosis, and generation of reactive oxygen species (ROS) in mice given SNE for 45 days in immune cells. To determine the splenocytes protection capability of SNE, gamma-ray was irradiated to the whole body of C57BL/6 mice. Our results suggest that SNE stimulated the proliferation of splenocytes without cytotoxic effects. In addition, SNE not only decreased DNA damage but also reduced apoptosis of splenocytes, and attenuated the production of ROS generation in hydrogen peroxide-induced splenocytes. Therefore, SNE can protect against gamma radiation-induced damage in mice.

        Macrophages play an important role in both the innate and adaptive immune responses. These include phagocytosis, killing of microorganisms, antigen presentation, and induction of immune cytokines and antimicrobial genes. Macrophage activity is reported to be controlled by diverse exogenous antigenic or endogenous metabolic molecules, and the underlying mechanisms are well documented in human and mouse macrophage cells. Bacterial lipopolysaccharide (LPS) is known to be one of the most potent stimuli activating macrophages through the toll like receptor 4 (TLR4) signaling pathway. There are other antigenic molecules, such as muramyl dipeptide (MDP) and outer membrane protein A (OmpA), that are also known to activate immune cells. On the other hand, short chain fatty acids (SCFAs) such as acetate and butyrate are produced by gut microbiota and control host energy metabolism and signal transduction through GPR receptors. However, there are few studies demonstrating the effects of these molecules in macrophages from domestic animals, including domestic pigs. In this study, we attempted to characterize gene expression regulation in porcine macrophages (PoM2, Pig Monocytes clone 2) following treatment with LPS, MDP, OmpA, and two short chain fatty acids using porcine genome microarray and RT-PCR techniques. A number of novel porcine genes, including anti-microbial peptides and others, appeared to be regulated at the transcriptional level. Our study reports novel biomarkers such as SLC37A2, TMEN184C, and LEAP2 that are involved in the porcine immune response to bacterial antigen LPS and two short chain fatty acids.

        Genetic polymorphisms within immunity-related candidate genes in pigs have been identified to control variations in immune functions and/or disease resistance. It has become necessary to evaluate the effects of other genetic markers of economically important traits prior to introducing them into marker-assisted selection programs. In this study, polymorphisms of porcine genes coding Interferon-induced Gunylate binding protein 1 (GBP1), GBP2, CD163, and CD169 were investigated for their association with growth and meat quality traits in a Korean native pig breed -Yorkshire inter-crossed F2 pig population (KY-F2). KY-F2 animals (n=346) have been successfully used for linkage mapping to identify quantitative loci that control meat quality, growth, and immunity traits. In our results, polymorphisms in genes GBP1 and GBP2 showed association with pig growth rate as well as meat quality traits such as crude fat, drip loss, and meat color (yellowness) in the KY-F2 population. The polymorphism in gene CD163 only showed association with crude fat, as a meat quality trait. CD169 gene was associated with pork tenderness. In conclusion, four immune-related genetic markers were validated for their association with growth and meat quality traits to gauge their potential use in a swine selection program. The results warrant further studies in other commercial pig populations.

        림프구는 면역기능의 중심적인 역할을 담당하는 세포이고, 운동에 의해 변화하고 노화에 의해 기능이 저하하는 것으로 잘 알려져 있지만, 고령자를 대상으로 운동에 의한 림프구의 변동에 대한 연구는 지금까지 미흡한 실정이다. 그래서 본 연구에서는 운동훈련이 고령흰쥐의 림프구에 미치는 영향에 대해 조사하였다. 8주간의 운동처리가 비장세포 중 그리고 장간막 림프절의 림프구에 미치는 영향을 분석했다. 비장세포중의 림프구에 대해서는 헬프-T 림프구, 세포상해성 T 림프구, 그리고 보조자극 신호 수용체 발현 T 림프구의 비율이 운동처리구에 있어 유의적으로 높은 수치를 나타냈다. 한편, 비장세포중의 B 림프구 및 장간막 림프절중의 림프구에 대해서는 두 처리구 간에는 유의적인 차이는 보이지 않았다. 이상의 결과들로부터, 운동처리구가 흰쥐 비장세포중의 T 림프구수에 영향을 미치는 것이 밝혀졌다. 그러나 면역기능을 평가하기 위해서는 림프구의 수 뿐만 아니라 기능에 대해서도 면밀히 검토해야 한다. 금후에는 운동처리구가 림프구의 기능에 미치는 영향을 해석할 필요성이 사료된다.

        Cows may suffer impaired ovarian function, often accompanied by reduced conception rates and increased embryonic loss. Cystic ovarian disease (COD) is one of the most frequently diagnosed gynecological findings in dairy cattle. It causes temporary infertility and is likely to affect reproduction as well as production parameters in cattle. Therefore, the purpose of this study was to determine the expression patterns of apoptosis (Bcl-2, Bax), implantation (E-cadherin) and immune related proteins (TNF-α, IL-10) in uterine endometrium of Hanwoo (Korean native cattle) with ovarian cyst and normal ovarian follicles. In the Western blot analysis, the expression of anti-apoptotic Bcl-2 protein was significantly higher in endometrium with normal ovarian follicles, whereas expression of pro-apoptotic Bax protein was significantly lower. Also, the expressions of E-cadherin and TNF-α proteins were significantly higher in uterine endometrium with normal ovarian follicles. On the other hand, the expression of IL-10 protein was significantly lower in uterine endometrium with normal ovarian follicles. Taken together, our results provided that the expressions of apoptosis, adhesion and immune related proteins in uterine endometrium with ovarian cyst were showed the aberrant patterns, and we suggest that different expression changes of these proteins may be affect to pregnancy ability of cattle.

        시스타틴(cystatin: CST)은 C1A류 시스테인 단백질분해효소에 대한 경쟁적 가역억제자로서 동식물류에서 파파인과 같은 캐셉신을 억제 대상으로 작용하게 된다. 바이러스 유래 CST (CpBV-CST1)이 폴리드나바이러스의 일종인 CpBV (Cotesia plutellae bracovirus)에서 동정되었 다. 기존 연구는 이 유전자의 과발현이 배추좀나방(Plutella xylostella) 유충의 면역 및 발육을 교란한다는 것을 보여 주었다. 본 연구는 이 유전자 의 단백질 기능을 분석하기 위해 세균발현시스템을 이용하여 재조합단백질(rCpBV-CST1)을 형성하여 단백질분해효소에 대한 활성억제효과를 결정하고, 곤충의 면역과 발육에 대한 생리적 억제효과를 분석했다. 이 유전자 번역부위는 138 개 아미노산으로 약 15 kDa 크기의 단백질로 추 정되었다. CpBV-CST1이 먼저 pGEX 발현벡터에 재조합되고, BL21 STAR (DE3) competent cells에 형질전환된 후 0.5 mM IPTG로 4 시 간동안 과발현되었다. 분리된 재조합단백질은 파파인에 대한 뚜렷한 억제효과를 나타냈다. 이 재조합단백질은 파밤나방(Spodoptera exigua)에 대 해서 혈구소낭형성의 세포성 면역반응을 억제하고, 경구로 처리할 때 배추좀나방의 유충발육을 처리 농도에 비례하여 제한시켰다. 이상의 결과 는 CpBV-CST1이 해충 밀도 억제에 응용될 수 있음을 제시하고 있다.
