        포도의 레스베라트롤 함량은 품종에 따라 매우 다양하나, 대체적으로 자옥, 피오네, 머스캇함부르크 등의 흑색계 품종에서 가식부인 과실의 레스베라트롤 함량이 높은 것으로 조사되었으며 머스카텔루소, 골드핑거, 뉴나이 등의 일부 청색계 품종에서도 레스베라트롤 함량이 비교적 높았다. 그러나 과경과 과축을 포함하면 자옥, 머스캇오브알렉산드리아, 베이비핑어 등의 품종에서 레스베라트롤 함량이 높았으며 이러한 품종은 친환경 재배에 유리할 것으로 생각된다. 레스베

        A resveratrol synthase (RS) gene was isolated from peanut (Arachis hypogaea, L. cv. Jinpoong) plants. This gene was placed under the control of the cauliflower mosaic virus 35S promoter (CaMV35S) and introduced into two Korean varieties of potato (Solanum tuberosum L. cvs. Jasim and Jowon) plants by Agrobacterium-mediated gene transfer. Putative transformants were screened by PCR with primers designed from CaMV 35S promoter, NOS terminator and RS gene. Most of selected transgenic potato plants showed the amplification of expected fragments by PCR of genomic DNA with gene-specific primers, while they were absent in untransformed control plants. Expression of the resveratrol synthase gene was also examined by northern blot analysis. The transformants showed a band which was lacking in the control plant, confirming that the introduced gene is transcribed into mRNA in the transformants. The strength of the band, which reflected the level of mRNA expression, differed among the individual transformants. Among the transformants obtained, the highest trans-resveratrol content in the transgenic young leaves of purple-fleshed "Jashim" was 2.11 μgg-1 fresh weight and that in the microtubers in vitro of purple fleshed "Jashim" was 8.31 μgg-1 fresh weight. This amount of resveratrol may have a positive biological effect on human health.

        The high-performance liquid chromatography(HPLC) method for the determination of trans-resveratrol in 34 germplasms and processing methods of peanut seeds has been modified. Peanut germplasms contained trans-resveratrol contents of 0.14~4.96μg/g, but findings for the testa color were not significant. However, two germplasms, 'KIGAN' and 'CS1', contained more trans-resveratrol contents than the other germplasms. The contents of their were 2.26μg/g and 4.96μg/g. The tested processing methods caused no significant changes in trans-resveratrol contents. The contents of fresh, boiled, and roasted peanuts were 0.36, 0.32, and 0.40μg/g, respectively in cv. Palkwang, and 0.22, 0.22, and 0.26μg/g, respectively, in cv. Jakwang. Differences were not significant among fresh, boiled, and roasted peanuts. The grains of 'Palkwang' and 'Jakwang' contained trans-resveratrol contents of 0.34μg/g and 0.24μg/g, and testa contained 1.12μg/g and 1.00μg/g, respectively. However, when comparing absolute quantity, the trans-resveratrol contents appears to be approximately 3~4 times higher in the testa than in the grain of the peanut, although the total contents were not different because the ratio of testa was low in peanut seeds.

        포도, 와인에 함유되어 있는 레스베라트롤을 정량적으로 분석할 수 있는 HPLC 정량 분석법을 확립하였으며, 국내에서 생산된 13개의 포도 재배 품종의 부위별과 13종류의 와인에 대한 레스베라트를 함량 분석 결과는 다음과 같다. 1. 13 품종의 평균 레스베라트를 함량은 143.5ug/g 이었으며, 과피, 과육, 종자, 송이가지는 각각 0.9~104.8ug/g,0.1~0.5ug/g1.7~173.9ug/g2.9~247.4ug/g 의 함량 분포를 나타냈다. 2. 과피, 과육, 종자, 송이가지 부위별 평균 레스베라트롤 함량은 각각 35.0, 0.2, 25.8, 82.5ug/g 이었으며, 과피의 경우 용보 품종이 104.8ug/g ,종자의 경우 청수 품종이 173.9ug/g , 송이가지의 경우 델라웨어 품종이 247.4ug/g 의 가장 높은 레스베라트를 함량을 나타냈다. 3. 13종류의 와인에 대한 레스베라트롤은 253.3~11,271.6ug/L 의 함량 분포를 나타냈다.

        국내에서 재배되고 있는 뽕나무의 resveratrol 및 oxyresveratrol 함유량을 측정한 결과 부위로는 뿌리껍질, 품종으로는 신일 및 수원품종이 가장 많은 resveratrol 및 oxyresveratrol를 함유하는 것으로 나타났다. 따라서 뽕나무로부터 resveratrol 및 oxyresveratrol을 유효성분으로 얻고자 할 때는 신일 및 수원품종의 근피를 사용하는 것이 가장 효과적일 것으로 판단된다.

        Resveratrol, which is both a phytoalexin with antifungal activity and a phytochemical associated with reduced cancer risk and reduced cardiovascular disease, is synthesized in a limited number of plant species including peanut. Resveratrol synthesis is catalyzed by the enzyme stilbene synthase including resveratrol synthase (RS). Resveratrol synthase gene (RS3) obtained from peanut, Arachis hypogaea, Fabaceae has been transferred into chinese foxglove, Rehmannia glutinosa by using Agrobacterium mediated transformation. RS t-DNA introduced to chinese foxglove (R. glutinosa L) by transformation and its reaction product, resveratrol-3-O-β-D-glucoside was isolated and characterized using HPLC. Also its biological effects was tested in inhibition of the lipid peroxidation of mouse LDL by glycosylated stilbenes derivatives obtained from transgenic plants. Resveratrol-3-O-β-D-glucoside isolated from transgenic R. glutinosa L. showed antimicrobial activity of the growth inhibition zone against Escherichia coli and Salmonella typhimurium. Therefore, this compound can be contributed to be useful as a phytoalexin for plant health as well as a phytochemical for human health.

        The objectives of this study were to establish the genetic transformation system of stilbene synthase in Rehmannia glutinosa. Resveratrol, which is both a phytoalexin with antifungal activity and a phytochemical associated with reduced cancer risk and reduced cardiovascular disease, is synthesized in a limited number of plant species including peanut. Resveratrol synthesis is catalyzed by the enzyme stilbene synthase including resveratrol synthase (RS). Stilbene synthase gene (RS3) obtained from peanut, Arachis hypogaea, Fabaceae has been transferred into chinese foxglove, Rehmannia glutinosa by using Agrobacterium mediated transformation. PCR analysis with RS3 primer confirmed that the targeted gene was introduced into the plant genome, 904 bp in size. Further analyses of identification of transformation using developed other molecular techniques and transgenic plants that RS t-DNA introduced to chinese foxglove (R. glutinosa L) and its reaction product, stilbene such as resveratrol will be isolate and characterize using NMR, MS, and HPLC.

        Secondary phenolic metabolites play an important role in plant defense mechanisms, and increasing evidence indicates that many phenolic compounds are important in human health. To date, few studies have investigated the impact of various medicinal plants on levels of secondary plant metabolites. To address this issue, 82 species of Korean medicinal plants were screened to determine their resveratrol contents. Among 82 medicinal plants, 5 species such as Gardenia jasmonoides, Phlomis umbrosa, Rheum palmatum L., Polygala tenuifolia, Rubus chingii HU contained relatively high concentrations of resveratrol (179.75~42.71 μg/g). But, 40 species including Adenophora triphylla var. japonica HARA were only observed low concentrations or trace of resveratrol, and 20 species including Alpinia officinarum HANCE did not contain a resveratrol.

        본 연구에서는 땅콩의 resveratrol에 대한 체계적인 연구를 위하여 고속액체크로마토그래피HPLC)를 이용한 땅콩의 trans-resveratrol 분석체계 확립과 품종 및 생육시기별 trans-resveratrol 함량 변화를 구명하여 금후 고부가가치 고함유 기능성 품종개발의 기초 자료로 활용하고자 수행한 결과를 요약하면 다음과 같다. 1. HPLC를 이용한 trans-resveratrol 분석조건은 컬럼은 Xterra C18, 이동상 용매조성은 (A) glacial acetic acid DCW(52.6 : 900)과 (B) acetonitril solution A(80:20), 용매흐름속도는 1.1 ml/min, 검출기는 UV-Vis detector 308 nm 이었다. 2. 땅콩에서 trans-resveratrol 추출시 추출용매의 경우 80%EtOH과 80%MeOH이 유의차가 없었고, 추출온도의 경우에도 온도간 유의성이 인정되지 않았으며, ICH guideline에 따른 안전성과 경제성을 고려한 trans-resveratrol의 최적추출 조건은 80% EtOH, 25℃ , 45분이었다. 3. 땅콩 품종별 trans-resveratrol 함량 분포범위는 0.018-1.125 ug/g으로 품종별 유의차가 있었으나, 초형과 립 크기에 따른 trans-resveratrol 함량은 일정한 경향을 보이지 않았다. 4. 종실과 종피를 포함한 종실의 trans-resverarol함량은 종실(0.152-0.305 ug/g) 보다 종피를 포함한 종실(0.188-0.345 ug/g)에서 더 높았다. 5. 생육시기별 trans-resveratrol 함량은 신남광땅콩(0.227ug/g 대광땅콩(0.418 ug/g)은 파종 후 110일에 팔광땅콩(0.486 ug/g)과 밀양16호(0.441 ug/g)는 파종 후 130일에 가장 높았다.
