        CD63, a member of tetraspanin membrane protein family, plays pivotal role in cell growth, motility, signal transduction, host-pathogen interactions and cancer. In this work, the cDNA encoding CD63 homologue (TmCD63) was cloned from larvae of coleopteran beetle, Tenebrio molitor. The cDNA is comprised of an open reading frame of 705 bp, encoding putative protein of 235 amino acid residues. In silico analysis shows that the protein has four putative transmembrane domains and one large extracellular loop. The characteristic ‘Cys-Cys-Gly’ motif and ‘Cys188’ residues are highly conserved in the large extracellular loop. Phylogenetic analysis of TmCD63 revealed that they belong to the insect cluster with 50-56% identity. Analysis of spatial expression patterns demonstrated that TmCD63 mRNA is mainly expressed in gut and Malphigian tubules of larvae and the testis of the adult. Developmental expression patterns of CD63 mRNA showed that TmCD63 transcripts are detected in late larval, pupal and adult stages. Interestingly, TmCD63 transcript was upregulated the maximum 4.5 fold in response to DAP-type peptidoglycan during the first 6 h, although other immune elicitors also made significant increase in the transcript level at later time-points. These results suggest that CD63 might contribute to T. molitor immune response against various microbial pathogens.

        We have identified novel ricin-type (R-type) lectin by sequencing of random clones from cDNA library of the coleopteran beetle, T.molitor. The cDNA sequence is comprised of 495 bp encoding a protein of 164 amino acid residues and shows 49% identity with galectin of Tribolium castaneum. Bioinformatics analysis shows that the amino acid residues from 35 to 162 belong to ricin-type β-trefoil structure. The transcript was significantly upregulated after early hours of injection with peptidoglycans derived from Gram (+) and Gram (-) bacteria, beta-1, 3 glucan from fungi and an intracellular pathogen, L. monocytogenes suggesting putative function in innate immunity.

        The centipede Scolopendra subspinipes mutilans has been a medically important arthropod species by using it as a traditional medicine for the treatment of various diseases. In this study, we derived a novel lactoferricin B like peptide (LBLP) from the whole bodies of adult centipedes, S. s. mutilans, and investigated the antifungal effect of LBLP. LBLP exerted an antifungal and fungicidal activity without hemolysis. To investigate the antifungal mechanism of LBLP, a membrane study with propidium iodide was first conducted against Candida albicans. The result showed that LBLP caused fungal membrane permeabilization. The assays of the three dimensional flow cytometric contour plot and membrane potential further showed cell shrinkage and membrane depolarization by the membrane damage. Finally, we confirmed the membrane-active mechanism of LBLP by synthesizing model membranes, calcein and FITC-dextran loaded large unilamellar vesicles. These results showed that the antifungal effect of LBLP on membrane was due to the formation of pores with radii between 0.74 nm and 1.4 nm. In conclusion, this study suggests that LBLP exerts a potent antifungal activity by pore formation in the membrane, eventually leading to fungal cell death.

        Entomopathogenic fungi are natural pathogens of insects and contribute to the regulation of host insect populations in the environment. Several these fungi produce a wide range of secreted enzymes, secreted protein toxins and secondary metabolites to overcome host defenses and ultimately kill the host, and to defend host resources against competing pathogens and saprophytes. Therefore, this study was performed to select the antimicrobial activity of entomopathogenic fungi form Korea soils against plant pathogenic bacteria Ralstonia solanacearum and plant pathogenic fungi Botrytis cinerea using dual culture technique on SDYA. In addition, we also performed to screening of 2,2-diphenyl-1-picrylhydrazyl (DPPH) radicals scavenging activity compounds from liquid culture filtrates of entomopathogenic fungi and investigate to it’s anticancer activity. As results, 12 isolates, 6 isolates and 25 isolates showing of these fungal metabolites produced antibacterial, antifungal and radicals scavenging activity compounds, respectively. The preferential antimicrobial and radical scavenging activities give evidence that these entomopathogenic fungal metabolites might be useful as a source for plant pathogen control and pharmaceutical interests.

        In South Korea, ants are predicted to shift their distributions northwards and upwards. It was predicted that ant fauna will be changed dramatically in highlands due to the range shifts. The Mymica ants which are most abundant in high altitudinal areas in South Korea will be nearly disappeared there in 2050s, and replaced by Aphaenogaster japonica which is abundant in lowlands. It was recently found that A. japonica shifted upwards in Mt. Hanla in Jeju island, South Korea. Interestingly, A. japonica is similar in shape and size with the Myrmica species, which indicate niche overlap and resulting intensive competition. To find elevational change of competitions between two specie, we investigated ants during two ant foraging periods (2010 and 2011) in a high mountain (Mt. Gaebang) using pitfall traps and bait traps along elevational gradients. Ten ant species were collected from a.s.l. 800 m to a.s.l. 1577 m. Myrmica sp. 1 was most abundant (35% of total ants) and collected at all elevations. Myrmica sp. 2 (19.7%) occurred from 1000 m to 1577 m. Meanwhile, A. japonica occurred up to 1200 m. Fights between ants were observed 22 times; fights between these two species were most frequently found. Although, competitive hierarchy was not recognized due to a low frequency of interspecific encounters, A. japonica is likely to be superior over Myrmica species in food competition when considering the slower and more timid behavior of Myrmica species compared with A. japonica. Therefore, it is likely that A. japonica would replace easily Myrmica species in Korean highlands when thermal barrier (i.e., cold climate in high elevations) will be removed due to climate warming.

        Black fungus gnats play an important role as the decomposers in the forest ecosystem. Their larvae live on various organic matters, and the subfamily Sciarinae, in particular, have restricted habitats that feed on dead woods in the deep forest. We compared the biodiversity of family Sciaridae from two different habitats, the clear-cutting area (CA) and the pitch pine forest (PF) by the emergence traps (ET) and the pitfall traps (PT) from May to October, 2010. A total of 12 species were reliably identified to the species among 19 unique taxa belonging to 9 genera 4 subfamilies from 716 collected black fungus gnats. Taxonomic studies show that Corynoptera bicuspidata (Lengersdorf, 1926) (200 individuals, 27.9 % of total) and Cory. sinedens Hippa, Vilkamaa & Heller, 2010 (74 individuals, 10.3 % of total) dominate in the PF and the CA, respectively. Species richness and abundance of CA is lower than those of PF. Any species of subfamily Sciarinae were not collected in the CA. These findings indicate that the biodiversity of Sciaridae was influenced by forest management such as clear cutting, which leads to the changes of canopy cover, dead woods and leaf litter layer.

        신선편의 샐러드와 유기농 채소에 오염되어 있는 식중독균인 B. cereus, S. aureus 그리고 E. coli를 대상으로 살균 소독제와 유기산에 대한 감수성을 평가하여 신선편의 식품의 미생물학적 안전성의 확보를 위한 올바른 세척제와 사용량을 제시하고자 하였다. S. aureus는 2% acetic acid, malic acid, tartaric acid와 1% CaO에 대하여 고농도의 염소계, 과산화수소계 보다 높은 살균 효과를 보였고 E. coli의 경우에는 1% acetic acid 와 CaO처리에 의해 80%의 분리주가 감소하여 50%에탄올과 400 ppm 차아염소산나트륨과 대등한 감소율을 보였다. B. cereus 분리주에 대한 1% CaO, 0.5% acetic acid, 2% malic acid와 tartaric acid 처리는 200 ppm의 차아염소산나트륨보다 높은 감소율을 나타내었고, 1%의 과산화수소와 대등한 감소율을 나타내었다. 본 연구에 따라 천연 물질 유래 살균 소독제인 CaO와 유기산은 기존의 화학적 살균제를 대체할 수 있는 천연 살균 소독제로 활용 될 수 있을 것이고 특히 환경 친화적이고 안전한 유기산은 고품질의 신선편의 채소를 안전성으로 공급하는데 효과적인 것으로 판단되었다.

        The high quality contact between graphene and the metal electrode is a crucial factor in achieving the high performance of graphene transistors. However, there is not sufficient research about contact resistance reduction methods to improve the junction of metal-graphene. In this paper, we propose a new method to decrease the contact resistance between graphene and metal using directly grown graphene over a metal surface. The study found that the grown graphene over copper, as an intermediate layer between the copper and the transferred graphene, reduces contact resistance, and that the adhesion strength between graphene and metal becomes stronger. The results confirmed the contact resistance of the metal-graphene of the proposed structure is nearly half that of the conventional contact structure.

        The performance of graphene-based electronic devices is critically affected by the quality of the graphene-metal contact. The understanding of graphene-metal is therefore critical for the successful development of graphene-based electronic devices, especially field-effect-transistors. Here, we provide a review of the peculiar properties of graphene-metal contacts, including work function pinning, the charge transport mechanism, the impact of the process on the contract resistance, and other factors.

        Most patients with chronic low back pain experience functional disability of trunk muscle, and limitations in physical activity. While there are many types of exercise programs available, in recent years sling exercise has been emerging as the exercise program for spinal stabilization. It has been supported by a great amount of research with positive findings on its effectiveness. This research studies the effects of bridging exercise, conducted on a sling, on pain level and trunk muscle activation in supine, sidelying, and prone positions during a 4 weeks period. 10 healthy people(normal group, n=10) and 28 patients with low back pain participated in this study. 28 patients were divided into two groups; one group participated in exercise with the sling(experimental group, n=14) and the other group exercised without the sling(control group, n=14). They were asked to use the Numerical Rating Scale(NRS) to answer to the level of their pain they felt (no pain: 0 point, severe pain: 10 points). During sling bridging exercises, the muscle activity level in each muscle measured in each position was standardized as three seconds of EMG signals during five seconds MVIC. In conclusion, the experimental group with four weeks of sling bridging exercise experienced a statistically significant reduction in the pain level(p<.05) and increase in the muscle activities of erector spinae when in supine position, internal oblique when in sidelying position, and rectus abdominis in prone position(p<.05). Regular sling bridging exercise reduces the low back pain and enhances other trunk muscle activation, thereby positively affect spinal stabilization.

        The purpose of this study was to investigate spirometric lung pattern, respiratory function and degree of fatigue by lung function tests and fatigue tests of 39 elderly people in a care facility aged 65 and over. The respiratory function tests were used to the Spirovit SP-1 and fatigue tests were used modified Piper fatigue scale(mPFS). Regarding the respiratory function, the FVC was 1.41±0.36ℓ, the FVC % predicted was 69.10±14.98%, the FEV1 was 1.02±0.31ℓ, the FEV1 % predicted was 63.27±16.05%, the FEV1/FVC was 72.77±13.40%, and the fatigue score was 5.83±1.09. As for the spirometric lung pattern, 19 patients had a restrictive pattern(48.7%), followed by 11 with a mixed pattern(28.2%), 5 with an obstructive pattern( 12.8%), and 4 with a normal pattern(10.3%). Regarding the respiratory function and fatigue by spirometric lung pattern, the FVC and the FVC % predicted of patients with a normal pattern or an obstructive pattern were greater than other groups at a statistically significant level. As for the FEV1, that of patients with a normal pattern was significantly higher than others, and for the FEV1 % predicted, that of patients with a normal pattern or a restrictive pattern was significantly higher(p<.001). Fatigue score by patients with a normal pattern was significantly less than patients of other patterns(p<.001). Therefore, pulmonary physical therapy is considered necessary to improve respiratory function and fatigue degradation of elderly in a facility.

        Lepidoptera is one of the largest insect orders, but the phylogenetic relationships within this order, have yet to be completely described. One of the unresolved relationships includes the monophyly of Papilionoidea in relationship with the monotypic superfamily Hesperioidea. We newly sequenced five hesperid mitochondrial genomes (mitogenomes), representing four subfamilies: Pyrginae (Daimio tethys and Lobocla bifasciatus), Coeliadinae (Choaspes benjaminii), and Hesperiinae (Potanthus flavus), and Heteropterinae (Carterocephalus silvicola). Along with these newly sequenced hesperid genomes phylogenetic analysis was conducted with all available lepidopteran mitogenomes including three reported species of Hesperiidae that consisted of ~70 species in ten lepidopteran superfamilies. The test for the effect of optimization schemes, such as exclusion and inclusion of third codon position of 13 PCGs, other genes (22 tRNAs and two rRNAs), and with and without partitions also was performed. Majority of datasets consistently placed the monophyletic Hesperiidae the sister to ((Pieridae + Lycaenidae) + Nymphalidae), placing another true butterfly family Papilionidae as the basal lineage of this group, presenting the relationships (Papilionidae + (Hesperiidae + ((Pieridae + Lycaenidae) + Nymphalidae))). Consistent to previous result, Pyraloidea was placed as the sister to ((Bombycoidea + Geometroidea) + Noctuoidea), placing the Macrolepidoptera as non-monophyletic group.

        The larch hawk moth, Sphinx morio, belongs to the lepidopteran family Sphingidae that has long been studied as a family of model insects in a diverse field. In this study, we describe the complete mitochondrial genome (mitogenome) sequences of the species in terms of general genomic features and characteristic short repetitive sequences found in the A+T-rich region. The 15,299-bp long genome consisted of a typical set of genes (13 protein-coding genes, two rRNA genes and 22 tRNA genes) and one major non-coding A+T-rich region, with the typical arrangement found in Lepidoptera. The 316-bp long A+T-rich region located between srRNA and tRNAMet harbored the conserved sequence blocks that are typically found in lepidopteran insects. Additionally, the A+T-rich region of S. morio contained three characteristic repeat sequences that are rarely found in Lepidoptera: two identical 12-bp repeat, three identical 5-bp long tandem repeat, and six nearly identical 5~6 bp long repeat sequences.

        Inhibition of acetylcholinesterase (AChE) is one of the important modes of action available for the control of insects. An assessment was made of the electric eel AChE inhibitory activity of 55 endophytic fungi isolated from the leaves of Huperzia serrata collected in Eunshi, Hubei Province, China. Of 55 mycelial extracts and 55 ethyl acetate-soluble fractions from liquid culture of the fungi, the ethyl acetate-soluble fraction of strain ES056 exhibited strong AChE inhibitory activity (88%) at a concentration of 100 μg/mL. Based on morphological characteristics and nuclear ribosomal DNA ITS sequence analysis, this fungus showed greatest similarity to members of the order Eurotiales and was shown to be most closely related to members of the Aspergillus aculeatus group. It was designated Aspergillus sp. MY056. This strain merits further study as a potential anti-AChE product.

        The present study is aimed to evaluate forest health in a fragmented urban forest using insects which are rarely used for evaluation of forest health. It is supposed that forest would be health if insect community in an urban forest is similar to that in healthy forests. Ants (Formicidae) and ground beetles (Carabidae) surveyed by pitfall trap method in the Hongneung forest located in the urban area of Seoul were compared with those in the Gwangneung forest. The Gwangneung forest is supposed to be health due to its high biodiversity and well-conserved nature. Ants of the Hongneung forest was much more abundant compared with those of the Gwangneung forest. However, ground beetles showed the opposite patterns; much more abundant in the Gwangneung forest than in the Hongneung forest. Species richness was higher in the Gwangneung forest than in the Hongneung forest, but species diversity was higher in the Hongneung forest. This dominance decreased species diversity in the Gwangneung forest. Thus, characteristics of insect community in the Hongneung forest were greatly different with that in the Gwangneung forest. This great difference of insect communities between the target and reference sites may indicate whether the Hongneung forest would not be health according to the working hypothesis, or may indicate simply the characteristic of the fragmented urban forest. In order to determine the significant decision, the repetitive studies are needed on this regard.

        Distributions of ants are mainly dependent on temperature. Hence, global warming would lead to shifts of distribution and abundance in ants. We surveyed ants at 366 forest sampling sites in South Korea using pitfall traps to predict change of ant abundance. Abundances of ant species were projected on the projected temperature based on A1B climate scenario. Common species (> 10 % occurrence) were analyzed on the relationship between abundance and temperature, and 16 species of the common species were selected for projection of abundance owing to future climate warming. Abundance of the selected species were projected on 900 m2 grid cells in 2011 and 2060 using GIS tool. In the projection, 11 species are predicted to decrease in 2060, whereas 5 species are predicted to increase. Myrmica kotokui, a most dominant highland species is expected to decrease up to 2.5% of current abundance in 2060, whereas Pachycondyla chinensis is expected to increase up to 264%. This abrupt change of ants would lead to cascade effects on other organisms such as insects, plants, mushrooms, and birds, which would disturb various functions and services of forest ecosystems.

        This study was carried out to clarify the response of butterfly communities on forest degradation in the Gwangneung Forest, Korea. We monitored butterfly communities with varying degrees of human activities by conducting line transect twice a month in 2011. A total of 70 species and 4,676 individuals butterflies were observed in four sites: natural forest, plantation forests, and Korean National Arboretum. Species richness increased with increasing open land. The result on niche breadth and habitat type of butterfly was consistent with our predictions; specialist species and forest interior species were abundant in natural forest, whereas generalist species and grassland species were abundant in Korea National Arboretum, the most modified area. Also, habitat breadth of butterflies clearly indicated the difference on the degree of forest degradation. Butterfly diversity associated with landscape patterns based on aerial photographs supported mosaic concept which indicates that species diversity increases as habitat heterogeneity and variability increase. Forest management plan that maintains various habitats and ensures grasslands is necessary to increase butterfly diversity in forest.
