The complete mitochondrial genome of Ostrinia palustralis memnialis Walker, 1859 (Lepidoptera: Crambidae) was determined to be 15,246 bp with a typical set of genes (13 protein-coding genes [PCGs], 2 rRNA genes, and 22 tRNA genes) and one non-coding region, with an arrangement identical to that observed in most lepidopteran genomes. The A+T content of the whole genome, PCGs, srRNA, lrRNA, tRNAs, and the A + T-rich region all are well within the range found in other Pyraloidea. Phylogenetic analyses of the concatenated sequences of the 13 PCGs using Bayesian inference and Maximum-likelihood methods placed O. palustralis as a sister group to O. furnacalis and O. nubilalis, with the highest nodal support. The subfamilies within Crambidae, such as Nymphulinae, Spilomelinae, and Pyraustinae, all formed monophyletic groups with the highest nodal support.
Insect killing fungus Beauveria bassiana has been widely studied as a biological control agent. However, many studies have been focused on lab or field-based management. Herein this work, comparison of three B. bassiana strains was investigated under a molecular level. The whole genome sequences of ERL836, JEF-007 were analyzed by PacBio (35.5 Mb of ERL836 and 36.5 Mb of JEF-007) and ARSEF2860 referenced from GenBank (33.7 Mb). To compare the three strains, virulence, thermotolerance and chemical resistance were assayed. The transcriptomes of non-infecting B. bassiana and infecting B. bassiana against western flower thrips were analyzed using RNA-seq. This work can provide that genome features, functions, morphology and gene expression could be different under the molecular level, even if in the same species.
Japanese pine sawyer, Monochamus alternatus, is the main pest that mediates pine wilt nematode, Bursaphelenchus xylophilus, that causes serious damage to pine forests. In this study, we studied the strategy to control M. alternatus using entomopathogenic fungi. The fungi were collected from soil by an insect-baiting method and two fungal isolates (Metarhizium anisopliae JEF-197 and JEF-279) showed high virulence against M. alternatus. The Metarhizium isolates were evaluated for insecticidal activity against M. alternatus by spray treatment on live pine trees and wintering trees, and the M. anisopliae JEF-197 showed high insecticidal activity. In addition, the interaction of fungi and M. alternatus were analyzed by RNA-seq. This result can contribute to the development of insect control agents using entomopathogenic fungi.
The serious emergence of chemical-mediated residual toxicity and insect resistance have been enforced the regulation of synthetic pesticides. Future decisions to select more realistic control options probably depend on the speed of technological development in chemical and biological pesticides. Now, a strategic collaboration between synthetic pesticides and biopesticides has been progressed, such as distribution and R&D in collaboration and M&A for obtaining microbial resources. Recently registered microbial pesticides are entomopathogenic fungi in pest management. A concept of e-biopesticide could be properly combined with digital agriculture and accelerate the use of biological control agents in the future farming.
Drosophila suzukii Matsumura (Diptera: Drosophilidae) is known as a pest that has a wide host range. This study was carried out to investigate the occurrence of D. suzukii by trapping D. suzukii in Korean strawberry greenhouse and its surrounding woodland from November, 2017 to April, 2019. As a result of trap monitoring, the greatest capture was recorded in fall and secondary peak was confirmed in summer in a lower capture. It was confirmed that there was a statistically significant difference between the occurrence among inside and outside of the greenhouse and the woodland. The strawberry greenhouse cultivation area of Korea is mainly located in the plain region, whereas most of the other host plants for D. suzukii are usually grown at the ridge of the mountain or hillside. Therefore, if considering the living environment of host plant, monitoring result that showed more capture in woodland than strawberry greenhouse would be explained.
In order to accumulate the ecological information of Drosophila suzukii for export negotiations of domestic strawberry, we weekly monitored occurrence of D. suzukii at three strawberry fields located in Gok-seong, Dam-yang and Hwa-sun using the best combination of trap and attractant from November 2017 to May 2018, the period of strawberry production. Monitoring was carried out at three sites (a hill located 150 meter distant to greenhouse, outside greenhouse and inside greenhouse) in Gok-seong. Drosophil suzukii was trapped at three short periods from November to the beginning of December, from the end of December to the beginning of January, and from April in a hill. At outside greenhouse, D. suzukii was trapped at two short periods from November to the early December and the beginning of May. In inside greenhouse, it occurred once after May 24th. Gok-seong data can be summarized as more occurrence for longer period as the distance to greenhouse increases. Dam-yang occurred once in a short period inside greenhouse and hill, respectively on May. In case of Hwa-sun, D. suzukii did not trapped in whole trapping sites during trapping period, except for the surrounding grape and peach cultivation areas at the end of November. Summarized, no occurrence periods are from November 20th, 2017 to May 14th, 2018 inside greenhouse; from December 11th, 2017 to May 3rd, 2018 outside greenhouse; and from January 8th, 2018 to April 12th, 2018 on a hill. In connection with strawberry culture, the D. suzukii tends to occur at the beginning and the end of the strawberry production period. We expect this result can serve as a helpful data for export negotiation for quarantine.
2018년 현재까지 농림축산검역본부 호남지역본부로 수입된 식물은 220종 17,030건으로 우드펠렛(18%), 주정박(8%), 페스큐 건초(8%), 알파파건초(8%), 귀리건초(6%), 혼합건초(5%), 티모시건초(5%), 라이그래스건초(5%), 목화씨(사료용)(4%), 소나무원목(3%) 등의 순이다. 이와 같이 호남지역은 비재식 사료류(건초 등) 수입비중이 높으며 이는 전국 수입 건수의 약 67%에 달한다. 검역해충은 795건 검출되었으며 이 중 관리해충 294건, 잠정규제해충 146건, 비검역해충 307건 등이다. 관리해충 중 Carpophilus obsoletus가 210건(72%), 잠정규제해충 중 Cryptoletes ferrugineus가 56건(39%)으로 높은 검출율을 보였다. 또한 호남지역으로 수입 된 재식용 구근류는 생강 및 생강구근, 마늘구근이며 전국 수입 건수의 약 42%를 차지한다. 수입 된 생강 및 생강구근 91건 중 37건에서 해충(선충)을 발견하였고 Meloidogyne sp.(잠정규제해충) 검출로 9건이 폐기·반송되었다. 수입된 마늘구근 34건 중 29건에서 해충을 발견하였고 Carpophilus obsoletus(관리해충) 18건, Cryptoletes ferrugineus(잠정규제해충) 11건이 검출되어 소독하였다.
Japanese pine sawyer beetle, Monochamus alternatus Hope (Coleoptera: Cerambycidae) is considered as a serious pest in pine trees. To develop an eco-friendly strategy to manage this forest insect, we collected entomopathogenic fungi from Korean soil and assessed their virulence against the adults of the insect in laboratory conditions. As a result, two isolates with conidial suspension (1.0×107conidia/ml), showed 87% and 90% mortality 12 days after fungal treatment, respectively. We assessed the potential of the fungi-derived destruxin and protease as additives to the fungal isolates, and they showed insecticidal activity via feeding and spraying treatments. Finally, we produced fungal conidia in massive solid cultures and formulated wettable powders, and now studying optimal conditions of oil-based formulation with two isolates. Based on these results, we are evaluating the control efficacy of the fungal agents against M. alternatus in field conditions.
본 연구에서는 국내 유통 중인 농산물 9품목(n = 578)에 대한 납과 카드뮴 함량을 조사하고 이들의 섭취로 인한 위해성을 평가하고자 하였다. 납과 카드뮴의 함량은 마이크로웨이브 분해 후 ICP-MS로 분석하였다. 조사대상 농산물의 납 평균 함량은 각각 보리 0.014 mg/kg, 완두콩 0.010 mg/kg, 강낭콩 0.008 mg/kg, 녹두 0.006 mg/kg, 파인애플 0.008 mg/kg, 살구 0.016 mg/kg, 매실 0.015 mg/kg, 자두 0.021 mg/kg, 대추 0.019 mg/kg이었고, 카드뮴 평균함량은 보리 0.017 mg/kg, 완두콩 0.004 mg/kg, 강낭콩 0.007 mg/kg, 녹두 0.005 mg/kg, 파인애플 0.001 mg/kg, 살구 0.002 mg/kg, 매실 0.002 mg/kg, 자두 0.002 mg/kg, 대추 0.003 mg/kg이었다. 모든 시료의 납, 카드뮴 함량은 EU, CODEX 및 국내 기준보다 낮은 수준이었다. 조사 대상 농산물에 대한 납, 카드뮴의 인체노출량을 산출한 결과, 납은 잠정주간섭취허용량(PTWI, 25 μg/kg b.w./week)의 0.067%, 카드뮴은 월간잠정섭취허용량(PTMI, 25 μg/kg b.w./month)의 0.28%이었다. 이상의 결과는 조사 대상 농산물의 납, 카드뮴 오염도와 이들의 섭취에 의한 위해성이 모두 낮은 수준이라는 것을 보여준다.
본 연구는 기존 노인용 기능성 게임이 갖는 장점과 한계점을 분석하고 체감형 게임이 갖는 매체적 가능성을 접목하여 노인용 체감형 기능성 게임의 스토리텔링 전략을 탐색하는 것을 목적으로 한다. 따라서 본 연구는 노인용 기능성 게임에 대한 이론적 탐색과 경험적인 접근을 바탕으로, 노인이라는 대상의 특수성과 체감 인터페이스라는 기술적 속성, 그리고 재미와 기능이라는 이질적인 두 속성의 결합이 만들어내는 혼재성을, 게임의 형식으로 구조화하기 위해 스토리텔링 전략을 수립하고 게임을 기획하였다.
<The four seasons>는 동작인식센서 키넥트를 활용한 재활치료용 체감형 기능성 게임이다. 지루하고 힘든 재활치료를 재미있게 훈련할 수 있는 체감형 기능성 게임으로 편마비 재활 치료를 위한 동작을 반복할수록 스크린 너머 가상세계가 풍요로워진다. 게임 플레이 과정은 뇌졸중으로 인해 편마비가 있는 중, 장년층이나 신체의 움직임이 원활하지 못한 노인들에게 정신적 위안과 신체적 건강을 얻게 한다. <The four seasons>게임은 감정 유희와 동작 유희가 인과적으로 연결 된 게임으로 동기부여, 동작반복, 피드백의 순환이 게임 내적 매커니즘을 통해 흥미롭게 전개되도록 하기 위해 특정 감정을 의도적으로 이끌어내고 그에 따른 결과로 행동을 유도한다. 본 연구 에서는 <The Four Seasons>의 스토리텔링 기획 과정을 체계적으로 기술함으로써 노인용 체감형 기능성 게임의 실제적 개선을 위한 스토리텔링의 구체적 방법론과 새로운 방향성을 모색하고자 하였다.
벼메뚜기의 인공사료 개발을 위하여 일본의 Konno식 먹이를 대조구로 하고, 28개의 배합비율별 인공사료를 제작하여 급이시험을 하였다. 시험결과, Konno식 보다 6주 후 1마리당 체중은 22개 처리에서 높았고 우화성공율은 23개 처리에서 높았다. 폐사율은 부화입식 후 45일까지 Konno식 대비 24개 처리에서 높았다. 1마리당 체중, 우화성공율, 폐사율을 종합해 볼 때 탄수화물원으로는 옥수수 건조잎 분말 또는 통밀가루와 단백질원으로는 2~5%의 탈지대두분 또는 어분의 배합비율이 벼메뚜기의 인공사료로서 가능성이 높았다.
벗초파리는 우리나라에서는 심각한 해충으로 인식되지 않아 발생 기간 및 패턴, 기주 범위 등의 생태적인 연구뿐만 아니라 모니터링을 위한 우수 유인제 및 트랩의 연구도 제한적으로 수행된 실정이다. 본 연구는 기존 연구 결과를 참조하여 사과식초 함유 유인제 2종과 화학적 루어 2종 그리고 트랩 2종에 대한 선발 시험을 수행하였다. 선발 시험은 사육중인 벗초파리를 이용한 choice test와 함께 수목원, 딸기 농장, 블루베리 농장에서의 6반복 유인력 검증 실험을 수행하였다. 유인력 비교 결과 ACV + wine의 포획량이 높았으며, 블루베리 농장 인근 결과값은 전체 조사 항목에서 통계적 유의성을 보였다 (P < 0.05). 트랩 선발 실험 결과 모든 실험 장소에서 Dreves 트랩이 높은 포획량을 보였다. 이상의 결과를 통해 ACV + wine과 Dreves 트랩 조합을 선발하였으며, 이를 이용하여 향후 지속적인 모니터링, 월동 발생 조사 등의 연구에 활용할 수 있을 것으로 생각된다.
Previously, a partial mitochondrial COI and CytB gene sequences have been used to infer genetic diversity and gene flow of the species. In this study, we additionally collected C. tripartitus from one previous and two new localities, sequenced the COI and CytB genes. Sequence divergence of current samples showed slightly lower values, but nearly equivalent to previous study. Haplotype diversity was still high and nucleotide diversity indicating that Seogwi-dong showed the highest estimates in both gene sequences. Gene flow among populations is high, but a significance difference was detected between Gulupdo and Anmado; and between Gulupdo and Seogwi-dong only in COI sequences (P < 0.05). Considering the high genetic diversity and gene flow in C. tripartitus populations, one of the major issues regarding conservation seems not to be the recovery of genetic diversity as has been suggested before.
지난 18년간(2000~2018년 3월) 검역현장에서 검출된 해충의 종류는 1,980종이다. 이중 딱정벌레목은 전체의 42.6% (곤충은 1,707종으로 86.2%)로 검역적으로 매우 중요하며, 여기에서 가는납작벌레과(Silvanidae)는 딱정벌레목 머리대 장상과에 포함되고, 이들의 검출 종은 21종, 횟수는 15,516회로 가장 많은 비율을 차지한다. 최근 18년간 검출률 결과 Ahasverus sdvana(쌀머리대장)로 6,404회로 가장 많이 검출되었으며, Slivanus didentatus(두니가는납작벌레)는 6,225회, Oryzaephilus surinamensis(머리대장가는납작벌레)는 1,123회, Oryzaephilus mercator(곡물가는납작벌레)는 415 회의 검출률을 보여준다. 가는납작벌레과는 딱정벌레목중 가장많은 검출률을 보이는 과로 검역현장에서 매우 중요한 과에 속한다. 본 연구는 검역과정에서 검출되는 가는납작벌레과 곤충 중 검역현장에서 최초로 발견된 Monanus 속의 한 종을 발견하여 보고 한다.
This study was designed to optimize ethanol extraction process of unfertilized corn silk (UCS) to maximize phytochemical contents and bioactivities. The response surface methodology (RSM) with central composite design (CCD) was employed to obtain the optimal extraction conditions. The influence of ethanol concentration, extraction temperature and extraction time on total polyphenol contents, total flavonoid contents, maysin contents, 2,2-diphenyl-1- picrylhydrazyl(DPPH) radical scavenging activities and tyrosinase inhibition were analyzed. For all dependable variables, the most significant factor was ethanol concentration followed by extraction temperature and extraction time. The following optimum conditions were determined by simultaneous optimization of several responses with the Derringer’s desirability function using the numerical optimization function of the Design-Expert program: ethanol concentration 80.45%, extraction temperature 53.49°C, and extraction time 4.95 h. Under these conditions, the predicted values of total polyphenol contents, total flavonoid contents, maysin contents, DPPH radical scavenging activity and tyrosinase inhibition were 2758.74 μg GAE/g dried sample, 1520.81 μg QUE/g dried sample, 810.26 mg/100g dried sample, 56.86% and 43.49%, respectively, and the overall desirability (D) was 0.74.
This study was aimed to evaluate the dose-response the effects of nano-encapsulated conjugated linoleic acids(CLAs) on in vitro ruminal fermentation profiles. A fistulated Holstein cow was used as a donor of rumen fluid. Nano-encapsulated CLAs(LF, 5% of nano-encapsulated CLA-FFA; HF, 10% of nano-encapsulated CLA-FFA; LT, 5% of nano-encapsulated CLA-TG; HT, 10% of nano-encapsulated CLA-TG) were added to the in vitro ruminal fermentation experiment. In the in vitro ruminal incubation test, the total gas production on incubation with nano-encapsulated CLAs was increased significantly according to the incubation time, compared with the control(p<0.05). The tVFA concentrations on addition of LF and HT were significantly higher than that of the control(p<0.05). Thus, nano-encapsulated CLAs might improve the ruminal fermentation characteristics without adverse effects on the incubation process. In addition, the population of Butyrivibrio fibrisolvens which is closely related to ruminal biohydrogenation was increased by adding HT, while decreased by adding LF at 12 h incubation. These results showed that nano-encapsulated CLA-FFA could be applied to enhance CLA levels in ruminants by maintaining the stability of CLA without causing adverse effects on ruminal fermentation profiles considering the optimal dosage.
Anthriscus sylvestris (L.) Hoffm. is a perennial herb found widely distributed in various regions of Korea, Europe, and New Zealand. The root of A. sylvestris have been extensively used in the treatment for antitussive, antipyretic, cough remedy in Oriental medicine, but the physiologically active function of the leaf of A. sylvestris is as yet unknown. In this study, we investigated the anti-cancer activity and the mechanism of cell death of water extracts of leaf of Anthriscus sylvestris (WELAS), on human FaDu hypopharyngeal squamous carcinoma cells. Our data showed that WELAS treatment inhibited cell viability in a concentration- and time-dependent manner. In addition, the treatment of WELAS markedly induced apoptosis in FaDu cells, as determined by the viability assay, DAPI stain and FACS analysis. WELAS also increased the proteolytic cleavage of procaspase-3, -9 and PARP (poly(ADP-ribose) polymerase). In addition, exposure to WELAS decreased the expression of Bcl-2 (an anti-apoptotic factor), but increased the expression of Bax (a pro-apoptotic factor), suggesting that mitochondria-dependent apoptotic pathways are mediated in WELAS-induced apoptosis. Taken together, these results indicate that water extracts of leaf of A. sylvestris inhibits cell growth and induces apoptosis via the mitochondrial-dependent apoptotic pathway in FaDu human hypopharyngeal squamous carcinoma cells. Therefore, we propose that the water extracts of leaf of A. sylvestris is a novel chemotherapeutic drug, having growth inhibitory properties and induction of apoptosis in human oral cancer cells.
