        The Stockholm Convention was adopted in Sweden in 2001 to protect human health and the environment, including Persistent Organic Pollutants Rotors, such as toxic and bioaccumulative. Currently, there are 28 kinds of materials. This prohibits and limits the production, use, and manufacture of the product. Korea is a party to the Convention and it is necessary to prepare management and treatment plan to cope with POPs trends. In the text, we have discussed HCBD materials. HCBD belongs to halogenated aliphatic unsaturated hydrocarbons. It is a toxic, organic mixture of bioaccumulation. A study on the treatment of waste containing HCBD substance, We decided to treat the waste containing HCBD thermally. So six samples were selected. Waste water treatment sludge, rubber plate, insecticide, tarpaulin, tire rubber, mixed sample. The tire rubber injected HCBD as a technical sample. HCBD analysis showed that 59.345 ~ 18,238.355 ug/kg was detected. For the thermal treatment, we analyzed element. As a result of thermogravimetric analysis, the weight change due to the decomposition of the material started at 200℃. The material decomposition was completed within 800℃. The thermal treatment was performed on a Lab-scale (1kg/hr). After exhaust gas analysis result, HCBD was detected at 0.01 to 0.09 ug/kg. The decomposition rate is estimated to be 99.848 ~ 99.999%. As a result of dioxin analysis in the exhaust gas, the highest concentration was found in the tarpaulins and the emission limit was exceeded. The concentrations of Cd, Pb, Cr, Cu, Ni and Zn in the residues were very low. Considering the decomposition rate of HCBD containing wastes, incineration treatment at 2 ton/hr or more is considered to be possible. And unintentional persistent organic pollutants such as dioxins in the exhaust gas. Therefore, it is considered safe to operate the incineration temperature at more than 1100℃.

        To achieve energy efficiency improvement is used to lower temperature for emission gas at catalyst inlet, or to reduce/stop using steam to reheat emission gas. Saved energy from this process can be used as power source in order to increase generation efficiency. Dry emission gas treatment, on the other hand, is the technology to increase generation efficiency by using highly efficient desalination materials including highly-responsive slaked lime and sodium type chemicals in order to comply with air pollution standards and reduce used steam volume for reheating emission gas. If dry emission gas is available, reheating is possible only with the temperature of 45℃ in order to expect generation efficiency by reducing steam volume for reheating. Retention energy of emission gas from combustion is calculated by emission gas multiplied by specific heat and temperature. In order to obtain more heat recovery from combustion emission gas, it is necessary to reduce not only exothermic loss from boiler facilities but emission calorie of emission gas coming out of boiler facilities. In order to reduce emission calorie of emission gas, it is efficient to realize temperature lowering for the emission gas temperature from the exit of heat recovery facility and reduce emission gas volume. When applying low temperature catalysts, the energy saving features from 0.03% to 2.52% (average 1.28%). When increasing the excess air ratio to 2.0, generation efficiency decreases by 0.41%. When the inlet temperature of the catalyst bed was changed from 210℃ to 180℃, greenhouse gas reduction results were 47.4, 94.8, 118.5, 142.2 thousand tons-CO2/y, CH4 was calculated to be 550.0, 1100.1, 1375.1, 1650.1 kg-CH4/y, and N2O was 275.0, 550.0, 687.6, 825.1 kg-N2O/y. In the case of high efficiency dry flue gas treatment, reduction of greenhouse gases by the change of temperature 120~160℃ and exhaust gas 5,000 ~ 6,500 ㎥/ton is possible with a minimum of 355,461 ton/y of CO2 and minimum 4,125 tons of CH4/y to a maximum of 6,325 ton/y and N2O to a minimum of 2,045 kg/y to a maximum of 3,135 kg/y.

        Shoot-fresh-weight (SFW) is one of the parameters, used to estimate the total plant biomass yield in soybean. Understanding the genetic and molecular basis of SFW could help increase the total biomass production. In this particular study, we identified QTLs associated with SFW in a Recombinant Inbred Line (RIL) population derived from interspecific cross of PI483463 and Hutcheson. A total of 551 (535 SNP and 16 SSR) markers, were found to be polymorphic between the parental lines and were used to screen the RILs to develop the genetic map. Linkage analysis and QTL mapping were performed using with the software QTL IciMapping version 4.0, with the minimum LOD score of 3.0 and estimating the likelihood of a QTL and its corresponding effects at every 1cM. QTLs with LOD value > threshold LOD, as determined by 1000 permutation tests at p > 0.05 were considered as significant QTLs. The analysis identified a total of 5 QTLs associated with shoot fresh weight over two environments, with the phenotypic variation (PV) ranging from 6.34 to 21.32%, and the additive effect from -0.54 to 0.33. Among these QTLs, qFW1314_19_1 had the largest LOD scores, with PV of 21.32%. Interestingly, three QTLs, qFW2013_19_1, qFW2014_19_1, and qFW1314_19_1 identified on chromosome 19(L), showed negative additive effects, indicating the contribution from the wild parent PI483463. The QTLs identified in this study can be the targets to identify the candidate genes for the SFW and can help in developing cultivars with increased biomass potential.

        Mungbean (Vigna radiata) is a fast-growing, warm-season legume crop that is primarily cultivated in developing countries of Asia. We constructed a draft genome sequence of mungbean to facilitate genome research into the subgenus Ceratotropis and to enable a better understanding of the evolution of leguminous species. The draft genome sequence covers 80% of the estimated genome, of which 50.1% consists of repetitive sequences. In total, 22,427 high confidence protein-coding genes were predicted. Based on the de novo assembly of additional wild mungbean species, the divergence of what was eventually domesticated and the sampled wild mungbean species appears to have predated domestication. Moreover, the de novo assembly of a tetraploid Vigna species (Vigna reflexo-pilosa var. glabra) provided genomic evidence of a recent allopolyploid event. To further study speciation, we compared de novo RNA-seq assemblies of 22 accessions of 18 Vigna species and protein sets of Glycine max and Cajanus cajan. The species tree was constructed by a Bayesian Markov chain Monte Carlo method using highly confident orthologs shared by all 24 accessions. The present assembly of V. radiata var. radiata will facilitate genome research and accelerate molecular breeding of the subgenus Ceratotropis.

        Soybeans have been the favored livestock forage for centuries. However, little studies have been succeed in estimating forage quality of soybean by near-infrared reflectance spectroscopy (NIRS). To establish NIR equations for soybean forage quality, 353 forage soybean samples, including an 181 recombinant inbred line population derived from PI 483463 (G. soja) ´ Hutcheson (G. max), 104 cultivated soybeans (G. max) and 68 wild soybeans (G. soja) were used to develop NIR for four quality parameters: crude protein (CP), crude fat (CF), neutral detergent fiber (NDF), and acid detergent fiber (ADF). Two NIR spectroscopy equations developed for CP and CF (2,5,5,1; multiple scatter correction [MSC]) and for NDF and ADF (1,4,4,1; MSC) were the best prediction equations for estimating these parameters. The coefficients of determination in external validation set (r2) were 0.934 for CF, 0.909 for CP, 0.767 for NDF, and 0.748 for ADF. The relative predictive determinant (RPD) ratios for MSC (2,5,5,1) calibration indicate that the CP (3.34) and CF (3.45) equations were acceptable for quantitative prediction of soybean forage quality, whereas the NDF (2.34) and ADF (1.97) equations were useful for screening purposes.         Soluble-NSF attachment protein receptor (SNARE) proteins play a role in vesicle fusion, exocytosis, and intracellular trafficking in neuronal cells as well as in fertilization and embryogenesis. We investigated the expression patterns of two SNARE proteins, SNAP-25 and synaptotagmin VII (SytVII), and their regulation by pregnant mare serum gonadotropin (PMSG) during mouse ovarian follicular development. Ovaries were obtained at 0, 12, 24, 36, and 48 h post-PMSG injection of immature mice. SNAP-25 and SytVII mRNA expression levels increased gradually in a time-dependant manner. However, protein levels revealed different patterns of expression, suggesting different translational regulation following PMSG stimulation. SNAP-25 and SytVII expression was closely associated with thickening of the granulosa cell (GC) layer and follicle morphological changes from a flattened to a cuboidal shape. To explore follicle stimulating hormone receptor (FSHR)-mediated regulation of their expression, GCs from preantral follicles were cultured to examine the effects of FSHR siRNA knockdown. FSHR siRNA abolished upregulation of the SNAREs in both PMSG and FSH-stimulated GCs. This abolished gene expression was rescued by adding dibutyryl cyclic AMP to the cultures. These results suggest that SNAP-25 and SytVII expression is regulated via the FSHR-cAMP pathway during follicular development.

        Scientific studies have shown that essential fatty acidintake can have a dramatic impact on human health. Soybean [Glycine max(L.) Merr.] oil from current commercial cultivars typically containsaround 8%linolenic acid (18:3) known as omega-3 fatty acid. Omega-3 fatty acid plays an important role to prevent cardiovascular disease and cancer. Relatively high 18:3 content in seed oil is a trait of the wild soybean (Glycine soja Sieb. and Zucc.) ancestor of modern soybean cultivars. Wild soybean is native to Korean peninsula and recently thousands of wild soybeans collected by soybean researchers in Korea. The objective of this study were to determine the linolenic acid content for wild soybean collection and to determine the stability of linolenic acid content derived from wild soybean over environments. Fatty acid profile for 1,806 wild soybean accessions collected from South Korea was determined by GC. The range of linolenic acid was 7.3 to 23.7% with an average 15.6%. We developed a recombinant inbred population from a cross PI483463 (wild soybean with 15% 18:3) and Hutcheson (cultivar with 8% 18:3). Three RILs, RIL156, RIL159 and RIL166, with high linolenic acid content (over 14%), parents and Williams 82 as checks were grown in nine environments over 2008-2011. Results showed that the content of linolenic acid for the PI483463, Hutcheson, and Williams 82 ranged from 14.8 to 17.1, 8.5 to 9.7, and 6.9 to 8.4 % and averaged 15.4, 9.2 and 8.0%, respectively. However selected RILs 156, 159, and 166 ranged from 10.7 to 15.7, 14 to 15.8, and 14.8 to 15.8, and averaged 13.9, 14.9, and 15.2, respectively. Among the tested accessions, RIL166 was the most stable with the lowest range and CV, and had a relatively lower stability coefficient value than other genotypes. Genes related to high linolenic acid from wild soybean may be useful in developing higher linolenic acid soybean genotypes and would broaden the use of soybean in food applications to improve human nutrition and health.

        Soybean is desirable as a forage crop because of it has high protein and oil concentration. Wild soybean, a progenitor of cultivated soybean, has a softer stem and higher protein content in seed than cultivated soybean. There is little information on yield and forage quality for wild soybean and its derivatives. The objective of this study was to determine the forage yield and quality of wild soybeans and selected soybeans derived from a cross G. max ×G. soja. Forage yield and quality were assessed for three grain soybean cultivars, three wild soybeans and three selected lines from G. max×G. soja. Forage quality attributes such as crude protein (CP), crude fat (CF), neutral detergent fiber (NDF), acid detergent fiber (ADF), digestible dry matter (DDM), dry matter intake (DMI) and relative feed value (RFV) were determined at the R2, R4 and R6 developmental stages. Forage yield and CF were highest at stage R6 in G. max, G. soja and selected G. max×G. soja lines. CP content was similar between R2 and R4 but increased sharply after R4 and peaked at R6 in G. max and selected lines from G. soja×G. max. On the other hand, CP content was similar between R4 and R6 stage in wild soybeans. Generally, NDF and ADF were highest at stage R4 but decreased at stage R6. DDM, DMI, and RFV increased between R4 and R6. These results suggest that R6 was the optimal harvest stage to provide forage of highest quality and yield. A study was conducted in 2011 to evaluate forage yield and quality at stage R6 in 25 lines from PI483463 (G. soja)×Hutcheson (G. max) and four cultivated grain soybeans. Hutcheson had the highest forage yield with 24.7t/ha infresh weight (FW) among grain soybeans. Line W11 had the highest forage yield(25.7t/ha,FW) among G. soja×G. max selections and four other lines had similar forage yield compared to Hutcheson. Generally the 25 lines from this G. max×G. soja cross had thinner main stems and branches than cultivated soybeans. When the 25 lines were evaluated for their feed quality as per forage grade by AFGC, nine lines rated prime grade and all 25 lines were classified as forage Grade 1. Results of this study indicate crosses between wild and cultivated soybean show promise for improving soybean as a forage crop.

        Controllable transgenic expression systems in transgenic animal model are valuable to the development of therapeutic approaches in human medical fields. The aim of this study was to 1) produce a transgenic cloned dog using inducible tetracycline vector system, and 2) investigate whether the transgenic cloned dog could be induced the transgene expression using doxycycline (Doxy). Canine fetal fibroblasts were infected with retroviral vectors designed to express the enhanced green fluorescent protein (eGFP) gene under the control of tetracycline-inducible promoter. For somatic cell nuclear transfer (SCNT), nucleus of an in vivo matured oocyte was removed and an eGFP expressed cell cultured with 1 ㎍/㎖ of Doxy was injected. After electrical fusion and chemical activation, the reconstructed embryos were transferred to a recipient and pregnancy diagnosis was performed by ultrasonography. Experiment I evaluated the mean fluorescence intensity (MFI) of infected cells while the cells were cultured in the presence of 1 ㎍/㎖ of Doxy for 5 days, and then in the absence of Doxy for 7 days using fluorescence-activated cell sorter. Experiment II was designed to produce an eGFP controllable transgenic cloned dog via SCNT. For verification of transgenic dog, experiment III was performed Southern Blot analysis and observation in vivo regulation of eGFP expression in the cloned dog treated with 100 ㎎/㎏ of Doxy every 2 days for 2 weeks under ultraviolet light. In experiment IV, western blot was used to detect eGFP increase and decrease in skin tissues of transgenic dog under the presence or absence of Doxy. In the results of experiment I, the MFI for infected cells was rapidly increased to approximately 42.3 times after 3 day-treatment compared to pre-treatment and quickly decreased 3 days after ceasing the treatment. In experiment II, a total of 203 embryos were transferred to nine recipients and three pregnant delivered three pups (Tet-on eGFP 0, Tet-on eGFP 1, and Tet-on eGFP 2) by C-sec and Tet-on eGFP 2 among them is still alive. All cloned pups were genetically identical to the donor cell. Tet-on eGFP 2 showed an apparent in vivo eGFP expression on her body after Doxy administration in experiment III. The result of Sothern blotting showed that the transgene insertion was detected from the three cloned dogs and all organs of Tet-on eGFP 1. Experiment IV indicated that a robust eGFP expression in skin tissue of Tet-on eGFP 2 rapidly increased after Doxy treatment and gradually decreased to basal level on 9 weeks after ceasing the treatment. In conclusion, we report here for the first time an inducible transgenic system in canine species and it can stably induce the transgene expression at intended time. This study has demonstrated the capacity to generate transgenic model dog which could regulate the transgene and it would contribute to human medical research fields.

        "Early Valley", is an early maturing potato cultivar with high yield potential. "Early Valley" is a clonal selection resulting from the cross between 'Suncrisp' and 'A87109-10'. It has medium plant height and light green foliage. "Early Valley" has medium flowering habit and white flowers. Tubers are smooth, yellow skin, light yellow flesh, round tuber shape, medium eye depth, and medium dormancy and good keeping quality. It has stable yield under wide range of climatic conditions. "Early Valley" is resistance to late blight, but moderately susceptible to common scab and hollow heart. This cultivar is also resistant to potato rotting at harvesting during the raining season. "Early Valley" has high level of antioxidant activity (about three times higher) and vitamin C (higher by 40%) than the 'Superior'. This cultivar has high level of tuber uniformity and capable of yielding 36.56 t/ha which is 17.07% higher than the control potato cultivar 'Superior' under optimum agronomical practices.

        Bovine embryos produced by in vitro maturation, feretilization and development was examined for presevation and transfer. The fertilization medium used BO medium with 5 mM/ caffeine and 10/ heparin and adjusted to a pH of 7.2 to 7.4. The final concentration of spermatozoa was adjusted to 1 cells/ motile sperm during fertilization in vitro. At 8~10 hrs after insemination, the oocytes were transferred into CR1aa medium and cultured for 7 days. Embryos were preserved by vitrification method for transfer. When the embryos of early, blastocyst and expanded blastocyst stages were frozen-thawed, the proportions of embryos with normal morphology 83.6, 88.1 and 85.2%. (중략)

        DNA methylation is involved in epigenetic processes such as X-chromosome inactivation, imprinting and silencing of transposons. DNA methylation is a highly plastic and critical component of mammalian development The DNA methyltransferases (Dnmts) are responsible for the generation of genomic methylation patterns, which lead to transcriptional silencing. The maintenance DNA methyltransferase enzyme, Dnmt 1, and the de novo methyltransferase, Dnmt3a and Dnmt3b, are indispensable for development because mice homozygous for the targeted disruption of any of these genes are not viable. The occurrence of DNA methylation is not random, and it can result in gene silencing The mechanisms underlying these processes are poorly understood. It is well established that DNA methylation and histone deacetylation operate along a common mechanistic pathway to repress transcription through the action of methyl-binding domain proteins (MBDs), which are components of, or recruit, histone deacetylase (HDAC) complexes to methylated DNA. As a basis for future studies on the role of the DNA-methyl-transferase in porcine development, we have isolated and characterized a partial cDNA coding for the porcine Dnmt1. Total RNA of testis, lung and ovary was isolated with TRlzol according to the manufacture's specifications. 5 ug of total RNA was reverse transcribed with Super Script II in the presence of porcine Dnmt 1 specific primers. Standard PCRs were performed in a total volume of 50 ul with cDNA as template. Two DNA fragmenets in different position were produced about 700bp, 1500bp and were cloned into pCR II-TOPO according to the manufacture's specification. Assembly of all sequences resulted in a cDNA from 158bp of 5'to 4861bp of 3'compare with the known human maintenance methyltransferase. Now, we are cloning the unknown Dnmt 1 region by 5'-RACE method and expression of Dnmt 1 in tissues from adult porcine animals.
