        미이라병은 Diaporthe/Phomopsis complex에 의해 유발되는병으로 콩 재배기간 중 따뜻하고 습한 환경에서 종자가 성숙되면 감염률이 높아지며 감염된 콩 종자는 외관상 품질뿐만아니라 종자 활력이 저하된다. 미이라병에 대한 연구를 수행하기 위하여 대만에 위치한 아시아채소개발연구센터(AVRDC)의 콩 시험포장에서 미이라병 병징을 보이는 콩 줄기를 채집하고 이로부터 3개의 곰팡이 균주(isolate)를 분리하였다. 배지위에서의 곰팡이 균사의 생육특성, 현미경하에서 관찰된 알파,베타 분생자(conidia)의 모양 그리고 PCR-RFLP 분석으로, 3개의 균주는 Diaporthe phaseolorum var. sojae 으로 확인되었다. 한편, 미이라병 저항성 육종을 위해서는 유전자원과 계통의 검정이 선행되어야 하는데, 인공접종을 위해서 분생자의최적 배양조건을 탐색하였다. 그 결과 배지는 PDA, 온도는24oC에서 잘 배양되었으며, 일장은 암조건에서는 균사체만 유도되고 분생자는 유도되지 않았으며, 24시간과 15시간의 일장에서는 균사체 유도 및 분생자의 유도 정도에 차이가 없었다.또한 잎-줄기와 꼬투리, 두 개의 접종 부위에 따른 미이라병감염률을 조사하였는데, 두 접종 부위에 따른 미이라병 감염정도는 통계적인 유의차는 나타나지 않았으나 잎-줄기에 접종한 개체 보다 꼬투리에 접종한 개체의 종자 감염률이 높은 경향을 보였다.

        Background : Some of invasive plants, which were introduced from foreign countries, have caused problems in Korea. Invasion of these invasive plants in the ecosystem threatens the habitat of endemic species, reducing biodiversity, and causing a disturbance in the ecological system. Hypochaeris radicata L. (Asteraceae), the most invasive plants in Korea, particularly in Jeju Island, invade farmland, and autochthonous forest, establishing monocultures and modifying the ecosystem structure. This invasive species has become a serious environmental problem because they displace the indigenous plant species. This study was conducted to evaluate the antioxidantive effects of ethanolic extracts from different parts (root, stem, seed and leaf) of the invasive exotic species Hypochaeris radicata L. Methods and Results : The aim of present study was to estimate the total phenolic and flavonoid contents and to investigate in vitro antioxidant potential of ethanolic leaf, root, seed, and stem extracts of the Hypochaeris radicata. Antioxidant activity was assessed by using 2,2-diphenyl-1-picryl-hydrazyl (DPPH) assay, reducing power activity, [2,2-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid)] ABTS+ assay and ferrous ion chelating activity. The total phenolic and flavonoid contents were also determined and expressed in gallic acid and quercetin equivalent respectively. The results of the study indicate that the ethanolic extracts of the leaf, root, seed, and stem of H. radicata posses significant scavenging activity against DPPH (21.25% for leaf, 34.98% for root, 60.76% for seed and 45.25% for stem at 250 μg/ml each) and ABTS+ radical scavenging activity (14.85% for leaf, 17.40% for root, 35.91% for seed and 24.70% for stem at 250 μg/ml each), reducing power activity (0.178 absorbance at 300 μg/ml for leaf, 0.211 absorbance at 300 μg/ml for root, 0.447 absorbance at 300 μg/ml for seed, 0.276 absorbance at 300 μg/ml for stem). The free radical scavenging and antioxidant activities may be attributed to the presence of adequate phenolic (gallic acid content is 361.92.98 μg/g in leaf, 356.59μg/g in root, 719.72 μg/g in seed and 512.08 μg/g stem) and flavonoid compounds (219.52 μg/g in leaf, 75.67μg/g in root, 281.39 μg/g in seed and 215.66 μg/g stem). This study revealed that the ethanolic extracts of both leaf, root, seed and stem of H. radicata has demonstrated significant antioxidant activity. Conclusion : In conclusion, the present study has demonstrated that Hypochaeris radicata seed ethanol extracts are rich in phenolics and have a strong antioxidant activity and a radical-scavenging action in all of the tested methods. This suggests that Hypochaeris radicata is a good source of natural antioxidants.

        Background : More than 1250 bamboo species, belonging to 75 genera, are distributed all over the world. Sasa quelpaertensis Nakai is a type of bamboo grass widely distributed in Halla mountain, Jeju Island, which has been used as antidiabetic, anti-cancer and anti-inflammatory drugs. In this study, we investigated the antioxidant and antimicrobial activities of Sasa quelpaertensis Nakai leaf extracted with different ethanol concentration and demonstrated the potent bioactivities of the extracts suitable to be used as natural antioxidant compounds or pharmaceutical supplements. Methods and Results : Antioxidant and anti-microbial activities of Sasa quelpaertensis Nakai extracts were studied. At first, different ethanol concentrations (0, 20, 40, 60 and 80%) were compared for determining of the best solvent for extraction of phenolic compounds from Sasa quelpaertensis Nakai. Forty percent Ethanol extract with 990.01±28.9 (mg of gallic acid equivalents/g sample) were the best solvent in the extraction of phenolic compounds. But, 60% ethanolic extracts were highest antioxidant activity appeared such as DPPH radical scavenging (IC50 21.20±0.42 μg/ml), ABTS radical scavenging (IC50 49.85±1.27 μg/ml) and reducing power. However, 80% ethanol extracts showed the strongest SOD like activity. The anti-microbial capacity was screened against Gram positive and Gram negative bacteria, and yeast. Sixty percent and 80% ethanol extracts inhibited the growth of Gram positive bacteria; Bacillus cereus was the most susceptible one with MIC of 125 μ g/ml and 250 μg/ml for the 60% and 80% extracts, respectively. Conclusion : The results of this study show that the extract of Sasa quelpaertensis Nakai can be used as easily accessible source of natural antioxidants and as a possible food supplement or in pharmaceutical industry. However, the components responsible for the antioxidant and antimicrobial activities of both extracts of Sasa quelpaertensis Nakai are currently unclear. Therefore, it is suggested that further works should be performed on the isolation and identification of the antioxidant components in Sasa quelpaertensis Nakai.

        Soybean (Glycine max (L.) Merr) is a short day plant and has been adapted to various climates and environments during cultivation. However, the cultivation area is restricted to a very narrow range of latitudes. To date, nine major genes (E1 to E8 and J) have been reported to control the flowering time and maturity. Here, we evaluated the role of E2, E3, E4, and their paralogue genes in late flowering soybean cultivars under long day (LD) conditions using Soybean yellow common mosaic virus (SYCMV)-based virus-induced gene silencing (VIGS) system. A total of nine VIGS constructs were infiltrated into two fully expanded cotyledons and primary leaves. After inoculation with these VIGS constructs on Jangyeobkong, which is a late-flowering cultivar, phenotypic traits were evaluated for the first flowering dates (FFDs) and pod maturities under LD conditions. The FFDs of the silenced plants occurred 50-56 days after sowing (das), while the non-silenced plants bloomed on 60-61 days. We found that the E3 paralogue-silenced plants flowered the fastest and responsive genes were identified to be associated with the promotion of flowering time. As the knock-down of E3 paralogue, expression of E1 was up-regulated, E2 was no difference, E3 and E4 genes were down-regulated in the silenced plants. Expression of GmFT2a and GmFT5a is known to be controlled by E3 and E4. Interestingly, GmFT5a were highly expressed in SYCMV:E3 paralogue-silenced plants, whereas the expression of GmFT2a was not significant. These results support that GmFT5a is able to independently promote flowering under LD conditions.

        Mungbean (Vigna radiata) is a fast-growing, warm-season legume crop that is primarily cultivated in developing countries of Asia. We constructed a draft genome sequence of mungbean to facilitate genome research into the subgenus Ceratotropis and to enable a better understanding of the evolution of leguminous species. The draft genome sequence covers 80% of the estimated genome, of which 50.1% consists of repetitive sequences. In total, 22,427 high confidence protein-coding genes were predicted. Based on the de novo assembly of additional wild mungbean species, the divergence of what was eventually domesticated and the sampled wild mungbean species appears to have predated domestication. Moreover, the de novo assembly of a tetraploid Vigna species (Vigna reflexo-pilosa var. glabra) provided genomic evidence of a recent allopolyploid event. To further study speciation, we compared de novo RNA-seq assemblies of 22 accessions of 18 Vigna species and protein sets of Glycine max and Cajanus cajan. The species tree was constructed by a Bayesian Markov chain Monte Carlo method using highly confident orthologs shared by all 24 accessions. The present assembly of V. radiata var. radiata will facilitate genome research and accelerate molecular breeding of the subgenus Ceratotropis.

        Undomesticated soybeans (Glycine soja) are an important source of genetic variation for introducing useful traits to domesticated soybeans (Glycine max). Although Korea is known as the origin of the soybean, a little is known about genetic diversity and structure analysis of G.soja. The objectives of this study were to investigate the genetic diversity and the structure analysis of wild soybeans, and to construct a core collection of G. soja accessions in Korea. To evaluate the genetic diversity and structure analysis of G.soja, we analyzed allelic profiles at 21 SSR loci of 1028 accessions using POWERMARKER V3.25. These markers generated a total of 581 alleles over all loci. The number of alleles per locus ranged from 21 to 40, with a mean of 28 alleles per locus and a mean gene diversity of 0.886 in this accessions tested. Polymorphic information content value ranged from 0.737 to 0.946, with an average of 0.877. Using STRUCTURE V2.34, wild soybean originated from Korea was divided into two distinct populations, largely corresponding to two geographic regions. Population 1 consisted of eight sub-groups corresponds to mountains; population 2 to entire regions in Korea. Based on theses 21 SSR markers, a core collection development was performed by POWERCORE V1.0. A G. soja core collection consisted of 148 accessions which were established from 1028 accessions in Korea. Most accessions of the core collection were belonged to population 2 and only four were belonged to population 1. These results of this study would provide valuable information for future breeding programs using the G. soja core collection.

        R genes are a key component of genetic interactions between plants and biotrophic bacteria and are known to regulate resistance against bacterial invasion. The most common R proteins contain a nucleotide-binding site and a leucine-rich repeat (NBS-LRR) domain. Some soybean NBS-LRR genes have also been reported to function in disease resistance. A total of 319 genes were determined to be putative NBS-LRR genes in the soybean genome. The number of NBS-LRR genes on each chromosome was highly correlated with the number of disease resistance QTL in the 2-Mb flanking regions of NBS-LRR genes. In addition, the recently duplicated regions contained duplicated NBS-LRR genes and duplicated disease resistance QTL, and possessed either an uneven or even number of NBS-LRR genes on each side. The significant difference in NBS-LRR gene expression between a resistant near-isogenic line (NIL) and a susceptible NIL after inoculation of Xanthomonas axonopodis pv. glycines supports the conjecture that NBS-LRR genes have disease resistance functions in the soybean genome. The number of NBS-LRR genes and disease resistance QTL in the 2-Mb flanking regions of each chromosome was significantly correlated, and several recently duplicated regions that contain NBS-LRR genes harbored disease resistance QTL for both sides. In addition, NBS-LRR gene expression was significantly different between the BLP-resistant NIL and the BLP-susceptible NIL in response to bacterial infection. From these observations, NBS-LRR genes are suggested to contribute to disease resistance in soybean. Moreover, we propose models for how NBS-LRR genes were duplicated, and apply Ks values for each NBS-LRR gene cluster.
