The green peach aphid, Myzus persicae, is one of the most important pests of vegetable crops worldwide. This species cause direct damage by feeding on plant nutrients and indirect damage as transmits many virus vectors. The control of aphid has relied on the use of chemical insecticides and their intensive use over many years has led to the development of resistance. To surmount or avoid these problems, we screened the entomopathogenic fungi against green peach aphid and evaluated their virulences. Several entomopathogenic fungal isolates were selected with high virulence to green peach aphid. Additionally, the antimicrobial activity of the selected fungal isolates was tested and analyzed to phytopathogens. Consequently, these entomopathogenic fungi would be used effectively for dual control agents for the green peach aphid and phytopathogen.

To investigate unknown molecualr and cellular mechanisms underlying polyphenisms in the migratory locusts, we performed whole transcriptome analysis from 12 different groups. Significantly differentially expressed coding and long non-coding (LNC) RNAs were present according to habitats, phases, and genders. Those differentially expressed coding and LNC RNAs were confirmed by quantitative-realtime-reverse transcriptase PCR. Those identified coding and LNC RNAs could be a target for developing environmental friendly management tools in the near future.

Polyphenism is one of the most intriguing phenomenon observed from various insect species. Polyphenism is changing morphological, behavioral, and physiological phenotypes without changing genome informations. Therefore, polyphenism is thought to be controlled by epigenetic methods. Epigenetic controls are including DNA methylation, histone acetylation, small RNAs, long non-coding RNAs, and protein phosphorylation. To investigate the mechanism involved in polyphenism, various genomic researches have been completed. However, metabolite changes in different phases have not been reported yet. Thus, we studied the metabolite changes and found the changed metabolites. These studies will make an important contribution to reveal the difference between gregrious and solitary phases and reveal eco - friendly control techniques.

Earthworms, especially Eisenia andrei, are cultivated for a variety of purposes including waste disposal and compost production. In this study, bacteria from earthworm gut were cultured and the dominant species identified. Subsequently, we isolated bacteriophages able to lyse the isolated gut bacteria. Two dominant genera of gut bacteria, Aeromonas sp. and Citrobacter sp. were identified by using MALDI-TOF MS analysis and a library was constructed to find lytic phages. Phage EF1 showed lytic activity for C. freundii and two Citrobacter isolates, C. braakii and C. murliniae. These 3 species have similar sensitivities to EF1. Several aspects of the life cycle of EF1 were investigated by using C. freundii under optimal growth conditions. EF1 infects C. freundii with a moderate latent period, approximately 25 min, and a large burst size averaging 5 × 109 per infected cell. Moreover, the antimicrobial activity of EF1 was well maintained under diverse conditions including a broad temperature range of 40°C to 50°C and a wide pH range of 4 to 11. In conclusion, the results indicate that earthworm casting contain a wide range of bacteria species, for which there are various corresponding bacteriophages.

This study was conducted in order to investigate the antioxidant capabilities and quality characteristics of instant porridge made of foxtail millet (Setaria italica Beauv.) from different varieties with different endosperm characteristics and processing using different methods. The two foxtail millet varieties used in this study were waxy foxtail millet, ‘Samdachal’, and non-waxy foxtail millet, ‘Samdame’. The means of processing food to create instant porridge were dry cereal frying, wet cereal frying, freeze drying after steaming, popping, and no treatment. The WAI (water absorption index) of instant porridge made of waxy foxtail millet was higher than that of non-waxy foxtail millet, and popping created the highest WAI among the treatments tested. Whean we analyzed RVA characteristics of non-waxy foxtail millet instant porridge, peak viscosity and setback values were decreased in all processed foxtail millet instant porridge compared to non-waxy foxtail millet instant porridge. In waxy foxtail instant porridge, the peak and final viscosities of processed treated instant porridge increased. The viscosity just after pouring hot water into the instant porridge was higher in waxy foxtail millet porridge treated by freeze drying after steaming, popping, and wet cereal frying; the viscosity of non-waxy foxtail porridge was increased after popping as compared with the other treatments. The polyphenol contents and antioxidant activities were increased in all processed, treated-instant porridge except for freeze dried porridge after steamingnt. The popping treatment showed the highest polyphenol contents (1.5 fold), and ABTS antioxidant activity (1.2 fold), compared to no treatment. Non-waxy foxtail millet porridge produced by popping earned the highest sensory evaluation scores.

The cabbage whitefly Aleyrodes proletella L. (Hemiptera: Aleyrodidae) is a specialized insect that uses its mouthparts(stylets) to feed from the phloem of its host plants, which are found among the Brassicaceae and Asteraceae. Female whiteflies lay their eggs in circular patterns embedded in wax on the underside of the leaves. These were collected from the leaf of triangular lettuce, Crepidiastrum sonchifolium throughout the whole Chungnam province, 2013. We reared the cabbage whitefly on the egg-plant. The elongate-oval eggs are laid upright in a semicircle on the underside of Brassica leaves but didn’t in lab. Initially pale and translucent, the eggs become darker. Nymphs are scale-like and covered with wax; their color is white with two yellow spots on the abdomen. On the dorsal surface of the last abdomial segment is the vasiform orifice charactersitic of the group. The fourth instar is called the “pupa”. The pupa is thicker, immobile, and pale in color with red eyes. The adults are tiny, about 1.5 mm long, and moth-like. The head and thorax are dark. The abdomen is yellow and covered by a conspicuous white waxy layer. The forewings have a faint, dark bar. If disturbed, the adults fly readily. The duration of development of immature stages of Aleyrodes proletella decreased with increase in temperature, the mortality was higher below 16 ℃ and above 28 ℃.

Even though plant protections using chemical pesticides have several advantages, non-specific toxicities to other beneficial insects and humans and rapid development of tolerance and/or resistance of target pests to chemicals are major disadvantages. Recent researches suggested that using double stranded RNA (dsRNA) could be species specific and environmental friendly pest management protocols. However, efficiency of dsRNA treatments are known to be variable according to its application methods. For example, injections of dsRNAs to pests were known to be effective in all species. However, efficiencies of oral application of dsRNAs were known to be dependent on species. Thus, development of tools that could enhance the efficacy of orally treated dsRNAs are utmost important for widening usages of dsRNAs in plant protection. Recently, we found that the efficacy of oral treated dsRNAs to target pests could be enhanced by nano-technologies. I will show how applying nano-technology to dsRNAs enhance the efficiency of dsRNAs. Porcine epidemic diarrhea virus (PEDV) causes acute diarrhea and dehydration in pigs with high mortality. PEDV is belong to Coronavirus, enveloped, single-stranded, positive-sense RNA virus. PEDV particles were composed of four structure proteins such as a glycosylated peplomer (spike, S) protein, envelope (E), glycosylated membrane (M) protein, and unglycosylated RNA-binding nucleocapsid (N) protein. Many of previous studies talk about this four structure proteins have a great potential to diagnosis and prevent PEDV. In this study we investigated expression of these structure proteins using the bacterial and baculovirus expression system. In bacterial expression system, our results showed that structure proteins fused polyhedrin and intein gene were expressed higher than non-fusion structure proteins. The expressed fusion proteins were used to immune mice for generating a polyclonal antibodies. In baculovirus expression system, co-infection of insect cells with these four recombinant baculoviruses led to self-assembly of virus-like particles as demonstrated by Transmission electron microscopy. They were confirmed by western blot analysis using pre-made polyclonal antibodies. Finding in this study may provide important information for vaccine and diagnostic development.
