Dietary fiber plays a important role in improving gut micriobiome and reproductive performance in gestating sows. This study aimed to evaluate effects of dietary supplementation with domestically produced alfalfa, provided in pelleted or mashed form, on body weight, body condition, litter performance, fecal score and fecal microbiome in gestating sows. A total of nine crossbred F1 gilts (Yorkshire × Landrace) with an average body weight of 230.33 ± 12.63 kg at 50 days of gestation were randomly assigned to three dietary treatments (n = 3 per treatment): (1) control (CON, basal diet), (2) Pellet (basal diet + 10% pelleted alfalfa), and (3) Mash (basal diet + 10% mashed alfalfa). Results indicated that alfalfa supplementation had no significant effect on weight gain during gestation (p>0.05), although sows in both alfalfa-supplemented groups showed numerically reduced weight loss during lactation compared to the control. The mashed alfalfa improved average daily weight gain of piglets during lactation (p<0.05). Moreover, the pelleted alfalfa alleviated constipation and numerically increased the relative abundance of positive microbes such as Firmicutes (phylum level), Lactobacillaceae and Ruminococcaceae (family level), and Clostridium_sensu_stricto_1, Lactobacillus, and Ruminococcaceae_UCG_002 (genus level) (p<0.05). On the other hands, it numerically decreased the relative abundance of negative microbes such as Proteobacteria in phylum level and Escherichia_Shigella in genus level. These findings suggest that domestically grown alfalfa, particularly in pellet or mash form, may be a valuable dietary fiber source for gestating sows to support piglet performance, gut health, and welfare. However, further studies with larger sample sizes are warranted to confirm these effects.
This study was conducted to secure basic data for developing technologies to reduce the generation of odor substances by investigating the effects of environmental temperature on growth performance and the generation of odor substances from feces in growing pigs. A total of 16 pigs (Landrace × Yorkshire × Duroc, average body weight 56.49±0.47kg) were randomly assigned to two treatments: thermal-neutral (TN) and heat stress (HS) conditions. The experiments were conducted for two weeks, with average temperature-humidity indices of 68.91±0.09 for TN and 85.98±0.08 for HS. The results showed that HS significantly decreased average daily feed intake (ADFI, 33.3%) and average daily gain (ADG, 25.8%) compared with TN (p<0.05). Non-esterified fatty acid in serum was lower (36.2%) in HS compared with TN (p<0.05). However, protein, blood urea nitrogen, cholesterol, triglyceride, glucose, and IgG in serum showed no difference between HS and TN. Phenol (350.0%) and skatole (416.3%) were significantly higher in HS than in TN (p<0.05). The decrease in growth performance is attributed to reduction in ADFI. The increase in phenol and skatole in HS is presumed to be due to the effect of HS on the metabolism of intestinal microbial composition. Digestion rate, intestinal microbial composition, and urine emissions are known to affect odor substances. Further research on the content of odor substances in urine, nutrient digestion rate, and intestinal microbial composition is considered necessary to determine the exact associations.
This study aimed to develop the in vitro method using domestic commercial diets to estimate nutrient digestibility in dogs. The existing in vitro method were tested and compared with literature data to develop new in vitro method. The development of in vitro method progressed as follows: modification of pepsin solution to an activated form and supplementation with 1% lipase. All the in vitro method progressed to 4 hours of stomach simulation and 2 hours of small intestine simulation. In vivo digestibility was measured using the same diets as beagle dogs. The supplementation of lipase methods showed significantly improved (p < 0.05) DM, OM, and EE than the existing and modified pepsin solution methods. The correlation between in vitro and in vivo data in DM, OM, and EE digestibility was high (r2 = 0.889, 0.907, and 0.721, respectively), and the correlation between in vitro and in vivo data in CP and GE digestibility was medium (r2 = 0.681 and 0.536, respectively). The current in vitro method is similar to in vivo digestibility and helps potentially predict digestibility for dogs. In conclusion, this developed in vitro method suggests that it can help estimate the nutrient digestibility of dogs' diets without in vivo experiments.
본 연구는 소양강댐 하류에서 서식하는 생태계교란 생물 종인 브라운송어와 그 먹이원으로 이용되는 저서성 대형무척추동물에 대한 파악을 위해 2022년부터 2023년까지 총 8회에 걸쳐 소양강댐 하류(St.1~St.3)와 지류 (St.4)에 대해 브라운송어와 공서종, 브라운송어의 위 내용물, 저서성 대형무척추동물의 종조성 및 기능군 분석을 실시하였다. 저서성 대형무척추동물의 경우, 하루살이목에서 가장 많은 분류군이 확인되었으며(27.1%), 그 중 붙는 무리(CL)와 헤엄치는 무리(SW)가 높은 비율을 차지하는 것으로 확인되었다. 브라운송어 채집 결과, 전장은 26∼246mm까지 총 105개체가 채집되었으며, 전장-체중 관계의 매개변수 b값이 3을 초과하여 안정적인 성장이 이루어지는 것으로 확인되었다. 브라운송어의 위 내용물에 대한 먹이원 분석 결과, 빙어(0.2%, TL: 246mm)와 육상곤충(2.7%, TL: 154mm, 183∼185mm)을 섭식한 개체는 매우 적었으며 상대적으로 전장이 큰 개체에서 확인 되었다. 대부분 수서곤충(73.8%)과 물 속에서 서식하는 비곤충류(23.3%)를 섭식하는 것으로 나타났다. 브라운 송어의 전장에 따른 먹이 섭식 패턴을 파악하기 위해 위 내용물에서 확인된 종들과의 상관분석을 실시한 결과, 브라운송어의 먹이원 중 유수성 환경 선호 종들의 경우 전장과 양의 상관관계(p<0.05)를 나타낸 반면, 모래 기질 이하의 흐름이 적은 서식처를 선호하는 종들의 경우 전장과 음의 상관관계(p<0.05)를 나타냈다.
The facile production of high-purity mesophase pitch has been a long-standing desire in various carbon industries. Recently, polymer additives for mesophase production have attracted much attention because of their convenience and efficiency. We propose polyvinylidene fluoride (PVDF) as a strong candidate as an effective additive for mesophase production. The mesophase content and structural, chemical, and thermal properties of pitches obtained with different amounts of added PVDF are discussed. The influence of PVDF decomposition on mesophase formation is also discussed. We believe that this work provides an effective option for mesophase pitch production.
A pregnancy diagnosis is an important standard for control of livestock’s reproduction in paricular dairy cattle. High reproductive performance in dairy animals is a essential condition to realize of high life-time production. Pregnancy diagnosis is crucial to shortening the calving interval by enabling the farmer to identify open animals so as to treat or re-breed them at the earliest opportunity. MicroRNAs are short RNA molecules which are critically involved in regulating gene expression during both health and disease. This study is sought to establish the feasible of circulating miRNAs as biomarkers of early pregnancy in cattle. We applied Illumina small-RNA sequencing to profile miRNAs in plasma samples collected from 12 non-pregnant cows (“open” cows: samples were collected before insemination (non-pregnant state) and after pregnancy check at the indicated time points) on weeks 0, 4, 8, 12 and 16. Using small RNA sequencing we identified a total of 115 miRNAs that were differentially expressed weeks 16 relative to non-pregnancy (“open” cows). Weeks 8, 12 and 16 of pregnancy commonly showed a distinct increase in circulating levels of miR-221 and miR-320a. Through genome-wide analyses we have successfully profiled plasma miRNA populations associated with pregnancy in cattle. Their application in the field of reproductive biology has opened up opportunities for research communities to look for pregnancy biomarker molecules in dairy cattle.
In this work, α-Fe2O3 nanocrystals are synthesized by co-precipitation method and used as adsorbent to remove Cr6+, Cd2+, and Pb2+ from wastewater at room temperature. The prepared sample is evaluated by XRD, BET surface area, and FESEM for structural and morphological characteristics. XRD patterns confirm the formation of a pure hematite structure of average particle size of ~ 40 nm, which is further supported by the FESEM images of the nanocrystals. The nanocrystals are found to have BET specific surface area of ~ 39.18 m2 g−1. Adsorption experiments are carried out for the different values of pH of the solutions, contact time, and initial concentration of metal ions. High efficiency Cr6+, Cd2+, and Pb2+ removal occur at pH 3, 7, and 5.5, respectively. Equilibrium study reveals that the heavy metal ion adsorption of the α-Fe2O3 nanocrystals followed Langmuir and Freundlich isotherm models. The Cr6+, Cd2+, and Pb2+ adsorption equilibrium data are best fitted to the Langmuir model. The maximum adsorption capacities of α-Fe2O3 nanocrystals related to Cr6+, Cd2+, and Pb2+ are found to be 15.15, 11.63, and 20 mg g−1, respectively. These results clearly suggest that the synthesized α-Fe2O3 nanocrystals can be considered as potential nano-adsorbents for future environmental and health related applications.
Interferon tau (IFNT) regulation, an anti-luteolytic factor produced by conceptuses of the ruminant ungulates, is essential for the maintenance of early pregnancy, but a definitive mechanism for its temporal transcription has not been elucidated. We and others have observed the T-box protein eomesodermin (EOMES) exhibited high mRNA expression in the ovine embryonic trophectoderm; thus, both caudal-relatedhomeobox-2 (CDX2) and EOMES coexist during the early stages of conceptus development. Objective of this study was to examine the effect of EOMES on ovine IFNT gene transcription when evaluated with CDX2, ETS2 and AP1 transcription factors implicated in the control of cell differentiation in the trophectoderm. In this study, quantitatively via reverse transcription-polymerase chain reaction (RT-PCR) analysis between ovine trophoblast cells was initially performed, finding that transcription factors CDX2 and ‘EOMES transcription factor mRNAs’ were specific to trophectoderm cells. These mRNAs were also found in days 15, 17, and 21 ovine conceptuses. Furthermore, human choriocarcinoma JEG3 cells (trophoblast cell line) were cotransfected with an ovine IFNT (-654bp)-luciferase reporter (-654-oIFNT-Luc) construct and several transcription factor expression plasmids. Cotransfection of the reporter construct with CDX2, ETS2 and AP1 increased transcription of -654-oIFNT-Luc by about 11-fold compared with transfection of the construct alone. When cells were initially transfected with EOMES followed by transfection with CDX2, ETS2 and/or AP1, the expression of -654-oIFNT-Luc was decreased. Also, EOMES factor inhibited the stimulatory activity of CDX2 alone. These results suggest that when conceptuses attach to the uterine epithelium, ovine IFNT gene transcription is down-regulated by an increase of EOMES factor expression in the attached ovine trophoblast cells.
This study was performed to investigate the effect of heat-stressed environment on rumen microbial diversity in Holstein cows. Rectal temperature and respiration rate were measured and rumen fluid was collected under normal environment (NE; Temperature humidity index (THI)=64.6) and heat-stressed environment (HE; THI=87.2) from 10 Holstein cows (60±17.7 months, 717±64.4 kg) fed on the basis of dairy feeding management in National Institute of Animal Science. The rumen bacteria diversity was analyzed by using the Illumina HiSeq™ 4000 platform. The rectal temperature and respiratory rate were increased by 1.5゚C and 53 breaths/min in HE compared to that in NE, respectively. In this study, HE exposure induced significant changes of ruminal microbe. At phylum level, Fibrobacteres were increased in HE. At genus level, Ruminococcaceae bacterium P7 and YAD3003, Butyrivibrio sp. AE2032, Erysipelotrichaceae bacterium NK3D112, Bifidobacterium pseudolongum, Lachnospiraceae bacterium FE2018, XBB2008, and AC2029, Eubacterium celulosolvens, Clostridium hathewayi, and Butyrivibrio hungatei were decreased in HE, while Choristoneura murinana nucleopolyhedrovirus, Calothrix parasitica, Nostoc sp. KVJ20, Anabaena sp. ATCC 33047, Fibrobacter sp. UWB13 and sp. UWB5, Lachnospiraceae bacterium G41, and Xanthomonas arboricola were increased in HE. In conclusion, HE might have an effect to change the rumen microbial community in Holstein cows.
Interferon-tau (IFNT) is known as a major conceptus protein that signals the process of maternal recognition of pregnancy in ruminants. Also, multiple interferon genes exist in cattle, However, molecular mechanisms of these bovine IFNT (bIFNT) genes whose expressions are limited have not been characterized. We and others have observed that expression levels of bovine subtype IFNT genes in the tissues of ruminants; thus, bIFNT1 and other new type I (bIFNTc1/c2/c3) gene co-exist during the early stages of conceptus development and non-trophoblast cells. Its genes transcription could be regulated through CDX2 and ETS2 and JUN and/or cAMP-response element binding protein (CREB)-binding protein (CREBBP) expression, a transcription factor implicated in the control of cell differentiation in the trophectoderm. Bovine ear-derived fibroblast cells, were co-transfected with luciferase reporter constructs carrying upstream (positions -1000 to +51) regions of bIFNT1 and other new type I gene and various transcription factor expression plasmids. Compared to each - 1kb-bIFNT1/ c1/c2/c3-Luc increased when this constructs were co-transfected with CDX2, ETS2, JUN and/or CREBBP. Also, Its genes was had very effect on activity by CDX2, either alone or with the other transcription factors, markedly increased luciferase activity. However, the degree of transcriptional activation of the bIFNTc1 gene was not similar to that bIFNT1/c2/c3 gene by expression plasmid. Furthermore, Sequence analyses also revealed that the expression levels of bIFNT1/c2/c3 gene mRNAs expression were highest on day 17, 20 and 22 trophoblast and, Madin-Darby bovine kidney (MDBK), Bovine ear-derived fibroblast (EF), and endometrium (Endo) non-trophoblast cells. But, bIFNTc1 mRNA had not same expression level, bIFNTc1 lowest levels than those of IFNT1/c2/c3 gene in both trophoblast and non-trophoblast cells. These results demonstrate that bovine subtype bIFNT genes display differential, in the trophoblast and non-trophoblast cells.
Variance of conceptus interferon tau (IFNT), produced by the embryonic trophectoderm, is known as a major conceptus protein that signals the process of maternal recognition of pregnancy in ruminants, essential for the maintenance of early pregnancy. Similar to other IFN genes such as IFNA and IFNB, multiple IFNT genes are present. However, some kinds of IFNT genes actively transcribed and regulated in bovine conceptuses have not been well characterized. In this study, during the course of bovine IFNT gene transcription through the use of next generation sequencer SOLiD3, revealed that among 38 IFN genes registered, only two transcripts, IFNT1 and IFNTc1, were found in conceptuses during early pregnancy. Also, to identify a transcription factor(s) involved in the regulation of IFNT genes, mRNAs for various known transcription factors were investigated by real-time PCR in conceptus tissues, respectively. Furthermore, compared to the IFNT genes, IFNT1 and IFNTc1 had same active levels, which were previously shown to correlate with the appearance of effective antiviral activity. However, the expression levels of these Luc activities differed. Bovine ear fibroblast (EF) cells were cotransfected with luciferase reporter constructs carrying upstream (–631 to -51) promoter regions of IFNT1 or IFNTc1 and various transcription factor expression plasmids, CDX2, AP1(JUN), ETS2 and/or cAMP-response element binding protein (CREB)-binding protein (CREBBP). CDX2, either alone with the other 2 transcription factors, was found to increase luciferase activity approximately 14- and 11-folds, respectively. The degree of transcriptional activation of the IFNTc1 gene was not similar to that IFNT1 gene by AP1, ETS2 or/and CREBBP, expression plasmid. These results suggest that two isoforms of bovine conceptus IFNT genes are regulated differently in conceptuses during early pregnancy.
The objective of this study was to evaluate the effect of treating dairy cattle with exogenous human chorionic gonadotrophin (hCG), five (5) days post artificial insemination (AI) on serum progesterone (P4) concentration and pregnancy rate. In this experiment, five days after AI, cows were assigned randomly to two groups namely: a) treated group (67) which were administrered with 1500 IU hCG (Chorulon) and b) control group (61), which received no treatment. On day 5, 10, 15 and 20 after the artificial insemination, blood samples from a total of 8 cows (4 from each group) were collected and were analyzed for serum P4 concentration. Cows were detected for estrus according to standing heat by visual observation. Cows that were detected still in estrus after days 18-24 were re-inseminated and recorded as not pregnant (open). Pregnancy diagnosis was conducted by ultrasonographic examination and transrectal palpation of the uterus on approximately 60 days in cows that observed to be not in estrus. The conception rate in hCG treated and control groups were 52.5 and 36.1%, respectively. The results proved that there were no significant differences in conception rate between two groups (p=0.0568). However, pregnancy rates were reduced by hCG treatment. Average serum P4 concentrations did not differ between Hcg-treated and control groups on day 5 (0.377 versus 0.375 ng/ml). On day 20 serum P4 concentrations were greater in the treated group compared with the control group (3.085 versus 2.010 ng/ml). The treatment with hCG seemed to increase P4 level compared with the control. In conclusion, the results of this study showed that 1500 IU of hCG administered on 5 day post AI increased conception rate in dairy cows. This was supported by the results on serum P4 concentration which was greater in hCG treated group.
Some of the deicer ingredients are the same as fertilizer ingredients from agricultural activities. The goal of this study is to distinguish the components of highway deicer from the components used for agriculture. Among the deicer ingredients, Ca and Cl are substances which can be supplied by fertilizer or livestock manure. However, fertilizer and livestock manure contain plenty of nitrogen, organic matter, phosphoric acid and magnesium, including deicer ingredients. The soil physico-chemical characteristics were analyzed according to the distance from the highway. The physico-chemical analysis items of the soil include electrical conductivity, total nitrogen (TN), available phosphate (Av.P), organic matter (OM), ammonia nitrogen (NH4-N), Cation Exchange Capacity (CEC). In the case of effective phosphoric acid, leaching into the subsoil is very low due to various binding mechanisms in the soil. However, it is considered that the compost (fertilizer) applied to the topsoil is mixed with the soil by the tillage or flattening operation. In addition to the organic matter content of the subsoil appear to 10% or more is that in some cases exhibit a similar level as the topsoil. The average soil organic matter content of about 2.6%. This results shows that an excess of organic compost in the study area flows into subsoil and was mixed with the subsoil. The high EC value at the points outside the snow removal effect range level is not considered to be affected by the deicer. The results of the survey showed that the values were differently detected by agricultural activities, and that they should be approached in a complex way in their interpretation.
A study on indigenous diatoms was carried out at 10 sites from May 2014 to December 2016 in marine and freshwater in Korea. Seventeen species of diatoms are new to Korea and they are divided into 3 classes, 4 subclasses, 10 orders, 14 families, and 16 genera. The nomenclatures, references, dimensions, specimens examined, local habitat, distribution in Korea, and photograph are reported here. Seventeen species found in marine, freshwater, and brackish water showed species-specific habitats.
We performed a survey for flavivirus infection and distribution of Aedes albopictus that known as Zika and Dengue virus vector using black–light trap and BG-sentinel trap around urban area in Korea. Mosquitoes were collected in 27 cities during March to November (twice a month) year 2016. Total numbers of mosquitoes collected 102,102 including 19 species 8 genera during collecting period. Total 21,467 Ae. albopictus was collected that 20,961(24.3%) by BG-sentinel trap and 506 (3.2%) by Black-light trap in urban area. Trap index(trap/night) of Ae. albopictus was showed highest in Hamyang (TI:992.3) and lowest in Taebaek (TI:0.3) there was only collected by Black-light trap. A total of 894 pools from all collecting Ae. albopictus were performed a Flavivirus detection. Flavivirus was not detected during study period. This study may provide basic information for surveillance of imported diseases (include Zika virus) and vectors in Korea.
The tobacco cutworm, Spodoptera litura(Fabricius), is a major pest of tomato and frequently demands control measures. The timing of insecticide application is a key factor in determining its efficiency, so an experiment was designed to investigate this. Application of insecticide was based on three criteria: (i) the number of trap-caught moths in a Delta-type trap with a commercial sex pheromone lure placed in the center of the target area, soon after plant emergence; (ii) the percentage of plants exhibiting pinhole-type damage (10% or 20%) and (iii) the percentage of plants exhibiting shot hole-type damage (10% or 20%) compared to a check plot without any control measures. We found that the number of trap-caught moths was, compared to the other methods, the best means of deciding on insecticide application in tomato plant to control the tobacco cutworm. Using pheromone traps, we obtained the best performance of the insecticide Shinnago, causing > 90% larval mortality. Without insecticide application, tomato yield reduction due to the tobacco cutworm larva damage was 27%.
Brachymystax lenok tsinlingensis (family Salmonidae), cold freshwater fish, is endemic to Asia. This species is currently distributed throughout Russia, Mongolia, China and the Korean Peninsula. B. lenok tsinlingensis in South Korea was severely affected by anthropogenic activities such as habitat destruction, agricultural run-off and water pollution, and hence this fish has recently been dramatically decreased in its population sizes and become now critically endangered. To recover the number of individuals of B. lenok tsinlingensis, stocking or translocation programs have been conducted continuously by local governments since 1970s. However, these programs made little effort to clarify populations that may have originated from stocked, translocated or introduced fish. An understanding of genetic characteristics of endangered populations is critical to develop effective conservation and restoration plans especially because genetic diversity ensues their future fate. Therefore, we assessed the “conservation status” of this species by estimating the level of genetic diversity and genetic structure among ten geographic populations including restored populations via reinforcement and supplementation. Also, we aimed to trace the genetic origins of the newly translocated population (Chiak) through a restoration practice program. Moreover, we inferred the phylogenetic relationships among Korean lenok populations as well as across the Northeast Asia. Two hundred eighteen individuals of B. lenok tsinlingensis were sampled from ten localities (Yanggu, Injae, Seorak, Bangtae and Hongcheon: North Han River basin; Pyeongchang, Chiak and Jeongseon: South Han River basin; Taebaek and Bonghwa: Nakdong River basin in South Korea). Based on mitochondrial DNA (mtDNA) control region and eight nuclear microsatellite loci, we found extremely low levels of within-population genetic diversity, which suggests small effective population sizes (Ne) within populations. For mtDNA control region, each population housed one, or at most, two haplotypes that are restricted to the respective localities, meaning that these ‘genetically unique’ lineages will be lost permanently if the local populations undergo extinction. The overall values of haplotype diversity (h) and nucleotide diversity (π) for the entire Korean population were 0.703 ± 0.024 and 0.021 ± 0.010, respectively. In the case of microsatellites, average number of alleles across the eight loci for the entire population was 9.1 and allelic richness (AR) per population ranged from 2.375 to 4.144 (mean = 3.104). The values of observed heterozygosity (HO) and expected heterozygosity (HE) were similar to each other [HO: 0.400 ~ 0.590 (mean = 0.518); HE: 0.407 ~ 0.608 (mean = 0.504)]. The inbreeding coefficient (FIS) values were generally low, ranging from 0.048 to 0.279. Consequently, the majority of the populations (except Yanggu and Pyeongchang) were not significantly deviated from Hardy-Weinberg equilibrium (HWE), suggesting random mating at these loci tested. In addition, we found that Korean lenok populations were significantly genetically isolated from each other, with private mtDNA haplotypes and microsatellite alleles, indicating limited gene flow among populations, strong effects of genetic drift due to small Ne, or a combination of both. The Mantel test of microsatellites revealed a significant correlation (r = 0.414, P = 0.04) between genetic and geographic distances for pairwise comparisons among the ten populations, while that of mtDNA showed a lack of correlation. Given the shared identical mtDNA haplotype and similar microsatellite allelic distributions between Chiak and Hongcheon populations, we suggest that the restored (introduced) Chiak population would be inferred to be genetically originated from Hongcheon population. Phylogenetic relationships among Northeast Asian populations showed that South Korean lineages have more recently diverged from China (Yellow River), than between North Korea and Russia. Although the phylogenetic relationship would be expected to be associated with geography, South-North Korea and China populations with a similar latitude was more phylogenetically closely related. These findings may suggest a possible scenario for the historical movements of B. lenok tsinlingensis in Northeast Asia during Last Glacial Maximum (LGM). It would be supported by the line of evidence that most lenok populations migrated to southward from Northern Asia such as Russia and Mongolia during LGM because the Korean Peninsula was landlocked as inland epoch and functioned as a southern shelter with Yellow River. For this reason, the Korean Peninsula is suggested to be an important geographical region for better understanding phylogenetic relationships and evolutionary histories of B. lenok tsinlingensis across the Northeast Asia. Despite large efforts made to develop several restoration programs in South Korea for B. lenok tsinlingensis, it is still unknown whether these past restoration efforts were successful or fruitless, mainly because of little attention paid to post-restoration monitoring research. Hence, there was a lack of their published official records. In the future, conservation and restoration projects of the Korean lenok populations should consider the genetic data for a better understanding of their ecological and evolutionary trajectories. And finally, we hope that our findings here can help inform on the future effective conservation and restoration plans for B. lenok tsinlingensis populatio ns in South Korea.
