        The black-veined white, Aporia crataegi (Lepidoptera: Pieridae), which is distributed mainly in Eastern Asia is presumed to be extinct in South Korea, only with some numbers of dried specimens left, whereas the species is found casually in circumferential countries. One of the common conservation practices for such species is to launch introduction program, but prior population genetic analysis between donor and donee populations might be essential for long-term conservation. In this study, we developed 11 microsatellite markers specific to A. crataegi using Illumina paired-end sequencing to investigate the genetic relationships of A. crataegi populations from South Korea and circumferential Asian countries (China, Russia, Mongolia, and Japan). Further, two mitochondrial DNA (mtDNA) gene segments (COI and CytB) were sequenced from the samples. The population- and individual-based Principal Coordinates and STRUCTURE analyses collectively suggested that the South Korean population of A. crataegi is most differentiated from the Japanese population, whereas it was closer to Mongolian and Chinese populations. The STRUCTURE analysis based on two concatenated mtDNA gene sequences also supported different genetic composition of Japanese population from the remaining populations including that of South Korea and rather similar genetic composition between the populations of South Korea and Mongolia. These results collectively suggest that northern populations, in particular, Mongolian populations can be considered as the most genetically compatible one as doner population, when reintroduction program is launched.

        Bumblebee, Bombus ardens ardens (Apidae: Hymenopera), is an important resource for pollination that is most widely distributed in Korea. This study utilized microsatellite markers for investigation of genetic diversity and geographic relevance of the B. a. ardens populations in Korea. Through Next Generation Sequencing analysis, we identified 10 microsatellite markers and genotyped for 107 individuals of B. ardens collected from 10 populations. At each locus the number of alleles ranged from 10 to 23; the observed and expected heterozygosities ranged from 0.8909 to 1.0000 and 0.6641 to 0.8422, respectively; and inbreeding coefficient(FIS) ranged from –0.5053 to –0.0891. Significant deviation from the Hardy–Weinberg equilibrium was not observed at any locus. Population structure analysis indicated that there are three genetic groups in Korea with each Jeongseon and Ulleung-do composed of different gene pool from the remaining other populations. Similarly, Principal coordinates analysis also showed the same pattern. FST and RST analyses showed that each Jeongseon and Ulleung-do population had a significant genetic distance from other populations. Considering these results, genetic isolation of Ulleung-do may be explained by “Oceanic island” status and Jeongseon, which showed the positive FIS (0.069) and genetic isolation may be caused by its location on the east side of Baekdudaegan and by on-going inbreeding with a small population size.

        The nerippe fritillary butterfly, Argynnis nerippe , is listed as an endangered species in Korea. Establishment of effective conservation strategies can be aided by the development and application of molecular markers that can be used to investigate the population genetics of the butterfly. Therefore, in this study, we identified ten microsatellite markers specific to A. nerippe using the Next-Seq 500 platform, and applied these markers to investigate the characteristics of five South Korean butterfly populations. Genotyping of 48 A. nerippe individuals from five localities showed that at each locus the number of alleles ranged from 4 to 14, and that the observed and expected heterozygosities were 0.324–0.863 and 0.138–0.985, respectively. Significant deviation from the Hardy–Weinberg equilibrium was not observed at any locus. Population structure analysis indicated that there are two genetic groups in Korea, but no population-based gene pool assignments were found. Analysis of FST, RST, and a principal coordinates analysis suggested that the Gureopdo and Yaecheon populations were isolated from other populations. Genetic isolation of the Gureopdo population may be a consequence of unequal population change between Gureopdo and inland populations and to the offshore habitat of Gureopdo. Genetic isolation of the Yaecheon population may be a consequence either of the southernmost location of the population or of the limited sample size available. Further studies with increased sample sizes will be necessary to draw robust conclusions on population isolation and to devise conservation strategies.

        In this study, we developed 12 microsatellite markers specific to N. pygmaea using Illumina paired-end sequencing. Forty individuals of N. pygmaea collected from three currently known localities in South Korea were genotyped to validate these markers and to preliminarily assess population genetic characteristics. No locus showed significant deviation from the Hardy–Weinberg equilibrium (HWE). Our preliminary data indicate an absence of inbreeding in all populations and an absence of obvious genetic difference. The microsatellite markers developed in this study will be useful for studying the population genetics of N. pygmaea collected from other regions, including additional sites in South Korea.

        Aphis gossypii is considered as one of the most polyphagous aphid species and found throughout the world. This pestinduce plant deformation and transmit viruses by feeding the host plant. In Korea, A. gossypii has been occurred in manycrops, however, the population genetic structure of this pest has not been revealed. In this study, we used eight microsatellitemarkers to analysis the population genetic structure of A. gossypii in Korea. We collected A. gossypii specimens frompepper greenhouse in 2016 (18 sites) and 2017(15 sites). As a result, two genetic cluster (△K=2) showed in structureanalysis. As we compared the cluster result between 2016 and 2017, several populations appeared opposite clustering groupeven the samples were collected in same site and crop. Therefore, more samples are required from different greenhouse(same host) to identify the accurate genetic cluster of province in Korea. From this study, the genetic variation revealedfrom A. gossypii and this information may develop sustainable management of A. gossypii.

        쇠검은머리쑥새(Emberiza yessoensis)는 국내 및 중국, 일 본 등에서 월동하는 겨울철새로, 이들의 번식지는 국내를 제외한 러시아, 몽골, 중국, 일본의 일부지역에 분포하는 것으 로 알려져 왔다. 그러나 최근 국내 번식 사례가 최초로 관찰되 어, 이후 이들 개체군을 보전하기 위한 기초연구 필요성이 대두되었다. 쇠검은머리쑥새는 국제자연보전연맹(IUCN)에 서 준위협(NT)종으로 취급되고 있으며, 국내의 경우 멸종위 기야생생물 II급으로 지정되어있다. 이들의 국내 서식지는 현재까지 지속적으로 감소하고 있어, 월동 및 번식개체군 모두 위협받고 있는 것으로 판단된다. 그럼에도 불구하고 개체군 크기, 서식지 면적 등 이들의 정확한 국내 개체군 기초정보는 현재까지 알려지지 않아 이들의 멸종위기 위험정 도를 객관적으로 파악하는데 어려움이 있다. 우리의 연구목 적은 쇠검은머리쑥새의 국내 멸종위기 등급을 판정하는데 도움을 줄 수 있는 유전정보를 제공하는 것이다. 우리는 이를 위해 10개 이상의 microsatellite 분자표지자를 개발하고 있 다. 현재까지 29개의 분자표지자를 디자인 하였으며, 10개의 다형성을 나타내는 microsatellite 유전자좌를 관찰하였다. 이 중 4개 유전자좌의 유전다양성을 경기 안산지역에 서식중 인 32개체의 쇠검은머리쑥새 번식개체군을 대상으로 분석한 결과, 대립유전자수(A)는 2~16개, 관찰이형접합빈도(HO)와 기대이형접합빈도(HE)는 각각 0.06~0.78, 0.12~0.90의 범위 를 나타냈다. 향후 10개 이상의 microsatellite 분자표지자를 개발하여 쇠검은머리쑥새 번식개체군의 유전다양성 및 유전 구조를 파악 할 것이며, 이를 통해 개체군 기초정보가 알려져 있지 않은 국내 쇠검은머리쑥새 번식개체군 보전방안 수립을 위한 객관적인 기준을 제시할 것이다. 이 연구에서 개발한 microsatellite 분자표지자는 국내뿐만 아니라 국외에 서식중 인 쇠검은머리쑥새 개체군 보전 연구에 유용하게 활용될 것이다.

        The Japanese oak silkmoth, Antheraea yamamai Guérin-Méneville 1861 (Lepidoptera: Saturniidae), is one of the important natural resources possessing industrial value for silk fiber production. In this study, ten microsatellite markers and two mitochondrial DNA (mtDNA) gene sequences (COI and ND4) were used to investigate the genetic variation and geographic structure of A. yamamai populations in South Korea. Two mtDNA gene sequences revealed very low total genetic variation and resultant low geographic variation, validating to use further variable molecular markers. Population-based FIS, FST, RST, and global Mantel test consistently support that A. yamamai populations are overall well interconnected with a relatively high gene flow. Nevertheless, STRUCTURE analysis using microsatellite data and mtDNA sequences coincidently indicate the presence of two genetic pools in many populations.

        The red-spotted apollo butterfly, Parnassius bremeri Bremer, 1864 (Lepidoptera: Papilionidae), has been listed as an endangered species in Korea. We developed microsatellite markers by nest-generation sequencing (NGS), selected 12 markers, and applied the markers to available South Korean populations to understand population genetic characteristics. The genotyping of 40 P. bremeri individuals from three localities in South Korea showed that at each locus, the observed number of alleles ranged from 17 to 43, the observed and expected heterozygosities were 0.84722-0.90556 and 0.76045-0.79208, respectively, and FIS was –0.155 to –0.121. STRUCTURE analysis supported the presence of two genetic pools in all three populations, although an immediate reason for this subdivision is not known. The population based FST, RST, and front wing length collectively suggest that at least Samchuk population in Gangwon Province has a significant distance. Further scrutinized analysis is undergoing.

        The tiny dragonfly, Nannophya pygmaea (Odonata: Libellulidae) is one the smallest dragonflies in the world and listed as a second-degree endangered wild animal in Korea. We developed microsatellite markers and applied selected markers to South Korean populations to understand population genetic characteristics, along with two mitochondrial DNA (mtDNA) gene sequences (COI and ND5). Two mtDNA-based population genetic analysis indicates substantially reduced genetic diversity in an island population (Muuido) compared to others. On the other hand, population-based FST and RST consistently support that N. pygmaea populations are overall well interconnected with a relatively high gene flow. These results may collectively indicate that N. pygmaea populations in South Korea may have rather larger population size than we previously acknowledged based on a single-locus mtDNA sequence and field observation.

        이 연구의 목적은 성염색체 특이적 초위성체마커의 대립 유 전자 다양성과 빈도를 조사하여 한국 재래소 3품종의 (칡소, 한우 그리고 제주흑우) 유전적 근연관계 및 특징을 조사하여 우리 고유 유전자원으로서의 가치를 구명하고자 하였다. 한국 재래소 3품종은 30개의 초위성체 마커에서 선별된 4개의 초 위성체 마커(INRA30, TGLA325, UMN0905 및 UMN0929) 를 이용하여 식별하였다. 대립 유전자의 다양성, 대립 유전자 빈도, 이형접합도 그리고 다형 정보량(PIC)을 산출 하였다. 칡 소, 제주 흑우와 한우에 대한 관찰 이형접합도의 평균은 각각 0.541, 0.406 및 0.61이었고 PIC의 평균은 각각 0.542, 0.377 그리고 0.6이었다. 제주흑우의 경우 칡소나 한우에 비해 낮은 이형접합도와 PIC를 보여준다. 이들 4개의 초위성체 마커를 이용하여 칡소, 제주흑우 그리고 한우의 품종을 구별에 이용 할 수 있다. 이러한 결과들로 볼 때 한국 재래소 3품종은 가 축유전자원으로서 중요한 가치를 지니고 있으며 이들 품종의 보존, 관리 및 활용에 중요한 기초자료로 이용될 것으로 사료 된다.

        최근 가축의 유전적 다양성 유지 및 식량 안보에 있어서 재래품종의 중요성은 점차 증대되고 있다. 제주흑우는 멸종위험에 처한 품종이며, 2013년 7월 문화재청에 의해 천연기념물 제546호로 지정되었다. 본 연구의 목적은 제주흑우(124두)의 유전적 다양성, 유연관계 및 유전적 구조의 평가이며, 그 비교 대상으로 한우(128두) 및 외래품종 홀스타인(73두)을 공시하였다. 분자유전학적 특성을 평가하기 위해 11개 초위성체 마커(BM1824, BM2113, ETH10, ETH225, ETH3, INRA23, SPS115, TGLA122, TGLA126, TGLA227, TGLA53)의 대립유전자형을 분석하였으며, 그 결과를 토대로 유전적 다양성 지수들을 산출하였다. 품종별 평균 기대이형접합도(HExp)는 0.605-0.738, 관측이형접합도(HOsb)는 0.667-0.747 그리고 다형정보지수(PIC)는 0.644-0.773의 범위를 보였다. 특히, 제주흑우의 유전적 다양성 지수는 가장 낮은 결과를 보였다. STRUCTURE를 이용한 군락 분석 결과 유전적으로 3개의 군락으로 구분되었으며, 주성성분분석(PCA) 결과 또한 3개의 군집으로 분류됨을 확인하였다. 따라서 본 연구의 결과는 제주흑우의 유전적 고유성 및 유전자원으로써 가치 판단을 위한 과학적 근거가 될 것으로 사료된다.

        초위성체 표지인자는 가축에서 유전자 다양성, 개체식별 연구를 위한 유전자 마커로 활용되고 있지만 가축의 가금류에서는 이러한 연구가 미비한 실정이다. 이러한 문제를 해결하기 위해 닭에서의 초위성체 마커에 대한 유전정보를 확보하고 보다 정확하고 신속한 유전자 분석 방법의 개발이 필요하다. 본 연구의 목적은 12개의 초위성체 표지인자(MS Marker)를 1세트로 구성된 다중중합효소연쇄반응(Multiplex PCR)을 이용하여 닭의 대립유전자와 대립유전자 빈도 및 이형접합도을 결정하는 마커를 개발하는 것이다. 닭 96수를 이용하여 12개 MS marker를 분석한 결과, MS marker에 대한 대립유전자수는 평균 3.08개로 관찰되었다. 12개 초위성체 마커의 이형접합도은 평균 0.563이고 다형정보도(Polymrphism information content : PIC)는 0.482로 계산되었다. 이 결과는 전국적으로 닭의 유전자를 이용한 개체식별 및 친자감별에 이용하기 위한 기초자료 뿐만 아니라 닭 이력제를 정착시킬 수 있는 중요한 기술로 활용 될 수 있을 것이라 사료된다.

        White-backed planthopper, Sogatella furcifera (Horvath) (Hemiptera: Delphacidae), has been a serious migratory pest in Korea. It is important to figure out the migration route and gene flow of S. furcifera. Microsatellite marker (SSR) shows high efficiency as molecular markers. Unfortunately, various microsatellite marker of S. furcifera has not been developed to see genetic diversity. S. furcifera samples were collected from Vietnam, Laos and three different sites of Bangladesh in 2012. We extracted DNA by using QIAamp DNA Mini Kit and ran next generation DNA sequencer (NGS) Roche 454 to develop a new microsatellite marker. Roughly, about 18 singleton primers and 14 contigs primers were found. We will test these primers with S. fucifera DNA samples, and figure out the accurate new microstatellite marker.

        The swine is one of the most widespread mammalian throughout the whole world. Presently, many studies concer-ning microsatellites in swine, especially domestic pigs, have been carried out in order to investigate general diversity patterns among either populations or breeds. Until now, a lot of time and effort spend into a single PCR method. But simple and more rapid multiplex PCR methods have been developed. The purpose of this study is to develop a robust set of microsatellites markers (MS marker) for traceability and individual identification. Using multiplex-PCR method with 23 MS marker divided 2 set, various alleles occurring to 5 swine breed (Berkshire, Landrace, Yorkshire, Duroc and Korea native pig) used markers to determine allele frequency and heterozygosity. MS marker found 4 alle-les at SW403, S0227, SWR414, SW1041 and SW1377. The most were found 10 alleles at SW1920. Heterozygosity represented the lowest value of 0.102 at SWR414 and highest value of 0.861 at SW1920. So, it was recognized appro-priate allele frequency for individual identification in swine. Using multiplex-PCR method, MS markers used to determine individual identification biomarker and breed-specific marker for faster, more accurate and lower analysis cost. Based on this result, a scientific basis was established to the existing pedigree data by applying genetics additio-nally. Swine traceability is expected to be very useful system and be conducted nationwide in future.

        The aim of this study was to identify allele variability and frequencies of six microsatellite markers (BM861, INRA124, INRA189, UMN0103, UMN0307 and UMN0504) specific to the Y-chromosome. DNA samples from 147 males of three Korean native and seven exotic cattle breeds were tested. Three (Chikso, Heugu, and Jeju black cattle) Korean native cattle (KNC) breeds showed Bos taurus genotype. In the neighbor-joining tree, based on Nei’s DA genetic distance, ten breeds represented main group. In addition, Bayesian clustering result showed that 3 main clusters are for individual allele variability and frequencies. Moreover, KNC breeds showed differences in genotype with B. taurus type when compared to previous studies. The molecular information of paternal lineage in this study would be useful for the conservation and utilization of three KNC breeds as genetic resources.

        This study was carried out to examine a molecular marker system for parentage test in Jeju Black cattle (JBC). Based on the preliminarily studies, we finally selected for construction of a novel genetic marker system for molecular traceability, identity test, breed certification, and parentage test in JBC and its related industrial populations. The genetic marker system had eight MS markers, five indel markers, and two single nucleotide polymorphisms (SNPs; g.G299T and g.del310G) within MC1R gene which is critical to verify the breed specific genotypes for coat color of JBC differing from those of exotic black cattle breeds such as Holstein and Angus. The results showed lower level of a combined non-exclusion probability for second parent (NE-P2) of 4.1202×10-4 than those previously recommended by International Society of Animal Genetics (ISAG) of 5.000×10-4 for parentage, and a combined non-exclusion probability for sib identity (NE-SI) of 2.679×10-5. Parentage analysis has been successfully identified the JBC offspring in the indigenous population and cattle farms used the certified AI semens for production using the JBC-derived offspring for commercial beef. This combined molecular marker system will be helpful to supply genetic information for parentage test and traceability and to develop the molecular breeding system for improvement of animal productivity in JBC population.

        This study was conducted to find a useful marker for gene polymorphism analysis using Microsatellite marker (MS marker) in Gyeongju Donggyeong dog. Twenty three MS marker analyzed the genetic features of DNA using 100 Gyeongju Donggyeong dogs in Gyeongju area. It was performed multiplex PCR with 3 set primer divided 9, 10 and 4 by analysis of conditions among MS markers. The results were calculated heterozygosity, polymorphic information content (PIC), allele frequency and number of allele at each locus using Microsatellite Toolkit software and Cervus 3.0 program. Total 148 alleles were genotyped to determine and average 6.43 alleles was detected. FH3381 had the highest of 15 alleles and FH2834 had the lowest of 2 alleles. Expected heterozygosity had a wide range from 0.282 to 0.876 and had average value of 0.6496. Also, Observed heterozygosity had a more wide range from 0.200 to 0.950 and had average value of 0.6404. PIC had range from 0.262 to 0.859 and average PIC was calculated 0.606. Especially, FH2998 represented the highest rate of observed heterozygosity of 0.950 and FH3381 represented the highest rate of expected heterozygosity of 0.876 and PIC of 0.859. The use of these markers was considered to be useful to study genetic traits of Gyeongju Donggyeong dog.
