        The oxidative stability of palm oil with moisture addition during heationg was investigated and the antioxidant effects of tocopherol, citric acid and sodium polyphosphate were evaluated. The addition of natural tocopherol up to 200 ppm level did not improve the oxidative stability of palm oil with moisture (1m1 H2O additive to 200g of oil at 180℃). The maximum antioxidant effect was observed at the Concentration of 400 ppm. Addition of citirc acid did not influence the effective concentration of tocopherol. Whereas addition of sodium polyphosphate and citric acid increased the antioxidant effect of tocopherol. The maximum synergistice effect was observed at the concentrations of 50 ppm citric acid and 50 ppm of sodium polyphosphate. Addition of 50 ppm citric acid and 50 ppm sodium polyphosphate to 100 ppm of tocopherol exceeded the antioxidant effect of 400 ppm tocopherol alone.

        정상사료내의 지질에 철분이 어떤 영향을 미치는가를 알아보기 위하여 정상사료(Common feed)와 물을 혼합한 비정상 사료내에 Vitamin E와 유리 불포화 지방산 함량을 측정한 결과 다음과 같은 성적을 얻었다. 1. 정상사료(P<.05)와 비정상사료(P<.01)내의 Vitamin E 함량은 1.75mg/g(실험 1일째) 로부터 0.25mg/g(실험 11일째)로 지속적으로 감소되었다. 2. 정상사료내의 유리 불포화 지방산 함량은 23.8mg/g(대조군)로부터 17.6mg/g(20% 철분 혼합군)로 역시 지속적으로 감소되었다(P<.01). 3. 비정상사료내의 유리 불포화 지방산 함량은 23.9mg/g(대조군)로부터 17.4mg/g(20% 철분 혼합군)로 지속적으로 감소되었다(P<.01). 4. 정상사료내의 Vitamin E와 유리 불포화 지방산 함량은 비정상사료내의 함량보다 다소 높았으나 유의차는 인정되지 않았다. 5. 정상사료와 비정상사료내의 Vitamin E와 유리불포화 지방산에 있어서 날짜 경과별 및 철분 혼합 수준별 간의 상호작용이 인정되었다.

        ‘Tocomi-1’, a new japonica rice cultivar derived from a 200 Gy gamma ray irradiation with high tocopherol content and red pericarp. The local adaptability test of MRXII-1001-1 was carried out from 2012 to 2014 and it was named as ‘Tocomi-1’ in 2014. This variety is medium matured with heading date of August 12 in honam plain area of Korea. This variety is about 80 cm tall culm length and 106 spikelets per panicle. Its 1,000 grain-weight of rice seeds is 25.4 g. The yield potential of this variety is about 5.15 MT/ha in local adaptability test for three years. This variety exhibited greater seed longevity than the Donganbyeo, indicating a crucial role for tocopherols in maintaining viability during quiescence, and displayed faster seedling growth during the early growth stage. Tocopherol contents was 50% higher than the Donganbyeo. To study the molecular mechanism underlying vitamin E biosynthesis, we examined the expression patterns of seven rice genes encoding vitamin E biosynthetic enzymes. Accumulation levels of the OsVTE2 transcript and OsVTE2 protein in the ‘Tocomi-1’ were significantly higher than in the Donganbyeo. Sequence analysis revealed that the ‘Tocomi-1’ harbored a point mutation in the OsVTE2 promoter region, which resulted in the generation of MYB transcription factor—binding cis-element. These results help identify the promoter regions that regulate OsVTE2 transcription, and offer insights into the regulation of tocopherol content in ‘Tocomi-1’.

        Tocopherols (α-, β-, γ- and δ-tocopherols) represent a group of lipophilic antioxidants which are synthesized only by photosynthetic organisms. It is widely believed that protection of pigments and proteins of photosynthetic system and polyunsaturated fatty acids from oxidative damage caused by reactive oxygen species (ROS) is the main function of tocopherols. In the present study, NtTC, which encodes a tobacco tocopherol cyclase ortholog, was cloned and characterized. Compared with control plants, NtTC transgenic rice showed higher tolerance to drought stress, and total tocopherol content increased by 52 % in leaf. Additionally, total antioxitant activity of NtTC transgenic lines was increased significantly by 19%. These results demonstrate that over-expressing NtTC could improve the tolerance to abiotic stress in rice, and tocopherols play a crucial role in the protection of oxidative stress.

        This study investigated the functional substances, including phenolics, tocopherol compounds in rice (Onnuri, Nampyeong, Dongjin, Deuraechan, Hopum, Iksan-515, Iksan-526) that were cultivated in Iksan region. Specially, Iksan-515 and Iksan-526 were produced resveratrol rice. Resveratrol is health-beneficial compound with strong antioxidant activity. Iksan-515 and Iksan-526 were contained resveratrol compound while, other rice varieties did not showed resveratrol. The total phenolic compounds showed similar concentrations in 7 rice varieties. Especially, Iksan-526 revealed 273.3 ㎍ g-1 and Iksan-515 was 264.8 ㎍ g-1. In this study, we analyzed vitamin E contents of 7 varieties of rice. The total tocopherols revealed 418.3 ㎍ g-1 in Iksan-526, 401.2 ㎍ g-1 in Iksan-515 and 413.3 ㎍ g-1 in Hopum. We determined 4 tocopherols (α-, β-, γ-, δ-) and 4 tocotrienols (α-, β-, γ-, δ-). Among the all samples, β-tocotrienol showed higher average concentrations (101.3 ㎍ g-1) than other compounds and, γ-tocotrienol revealed second higher concentration (94.5 ㎍ g-1). On the other hand, δ-tocopherol was not detected in all rice samples.

        In order to get information on tocopherol content and composition in fifty-six soybean germplasms were evaluated by HPLC. From the principle component analysis, the first three components accounted for 71.6% of the total variance of tocopherol content in the germplasms. Principal component 1 showed significant correlations with all the morphological markers except 100-seed weight. Soybean germplasms were divided into three groups by the first two principal components. The highest content of α -tocopherol was 38.6~mug per g in Tanner, while that of ~gamma -tocopherol was 195.6~mug in PI 91073. In case of δ -tocopherol, IT 105622 showed the highest value as 29.8~mug . The contents of tocopherol were gradually increased from the late August to late September, which was 10 days before maturity, in Alchankong and PI 96322. The tocopherol content was higher in seeds from the plants sown early than those sown late. Total tocopherol contents of PI 96188, Geomjeongkong 2, and Suwon 183 grown in Gimje were higher than those grown in Iksan, but the difference was not observed in PI 96322. These results suggest that the contents of tocopherol in soybean were affected not only by the genotypes but also by environment.

        품종 및 재배지역에 따른 생리활성성분(isoflavone, anthocyanin 및 tocopherol 함량, 항산화 활성)변이를 구명하여, 생리활성성분 고함유 품종선발과 생리활성성분 축적에 유리한 재배지역을 발굴하여 지역특화작목으로 선정하기 위한 기초자료로 활용하고자 연구한 결과는 다음과 같다. 100립중은 품종간에는 차이 없었으나, 지역간에는 2004년에는 뚜렷한 차이를 보였으나, 2005년에는 화성이나 진주보다 나주에서 현저하게 가벼웠다. 총 isoflavone 함량은 2004년에는 일품검정콩이 1,064.9ug/g 으로 청자콩의 801.3ug/g 에 비해 높았으나, 2005년에는 오히려 일품검정콩에 비해 청자콩이 높았다. 재배지역간에는 2004년에는 두 품종 모두 화성지역에서 뚜렷하게 높았으나, 2005년에는 진주지역이 화성이나 나주보다 현저하게 높았다. 총 anthocyanin 함량은 품종 간에는 일품검정콩이 7.22 mg/g으로 청자콩의 6.83 mg/g에 비해 높았으며, 재배지역간에는 진주지역에서 9.16 mg/g으로 나주, 화성지역에 비해 가장 높았다. 총 tocopherol 함량은 품종 간에는 큰 차이가 없었으며, 지역 간에는 화성, 진주지역이 각각 217.2ug/g , 216.3ug/g 으로 나주지역 189.7ug/g 에 비해 높았다. tocopherol 성분비율은 ~gamma -tocopherol이 56.2~59.9% 로 가장 높았다. 전자공여능은 품종간에는 일품검정콩이 청자콩보다, 지역간에는 화성이나 진주보다 나주에서 EDA 값이 55.6%로 가장 높았다.

        The purpose of this study is to establish an extraction and analysis method for α-tocopherol, and to then distinguish among varieties. The α-tocopherol contents of 22 varieties of peanut seeds were analyzed by HPLC. Peanut seeds of cv. Palkwang were processed in two ways, by boiling and roasting. The α-tocopherol contents of the two types of peanuts were compared with fresh seeds without any processing. α-Tocopherol was detected at a retention time of 2.95 minutes. Five standards of α-tocopherol covering a range of 20~100μg/ml were made up in 2% isopropyl alcohol/n-hexane and analyzed in duplicate. The α-tocopherol contents differed according to extraction temperature. The contents were 85μg/g or less at 10℃ and 20℃ and 94μg/g at 30℃, but they were decreased at 40℃ or higher. The α-tocopherol contents in 22 peanut varieties were 61.36~96.80μg/g according to variety. Fresh peanuts contained 106.7μg/g of α-tocopherol, while boiled peanuts contained 108.8μg/g of α-tocopherol, and roasted peanuts contained 109.2μg/g of α-tocopherol.

        Tocopherol belongs to the Vitamin E class of lipid soluble antioxidants that are essential for human nutrition. Tocopherols consist of a chromanol ring and a 15-carbon tail derived from homogentisate (HGA) and phytyl diphosphate, respectively. Condensation of HGA and phytyl diphosphate, the committed step in tocopherol biosynthesis, is catalyzed by HGA phytyltransferase (HPT). To increase tocopherol level in cucumber fruit, we performed Agrobacterium-mediated transformation with mdhpt gene, encoding HPT, isolated from apple (Malus domestica). We obtained two independent T0 cucumber plants from transforming 2,142 explants. Southern blot analysis of T0 showed that mdhpt was inserted in the genome. In order to test the tocopherol level, T1 cucumber fruits were screened by GC analysis. From 39 T1 fruits tested, some fruits showed 1.5-2 fold in α- and β-tocopherol content comparing to control. However, most of fruits contained lower levels of γ- and δ- tocopherol indicating a partition of metabolites in tocopherol biosynthesis pathway. We selected these T1 line and made a self-cross to obtain T2 seeds.
