        Background: Gastrodia elata Blume is a saprophytic perennial plant in the Orchidaceae family, because of its agricultural and medicinal effectiveness, researchers focus on its genome and chemical components. However, cytogenetic information based on the chromosome structure and composition to construct chromosomal backbone for genome sequencing research and for the development and breeding of plants is very limited. Methods and Results: We determined the metaphase chromosome composition of the G. elata genome by fluorescence in situ hybridization (FISH) using 5S and 45S rDNAs and telomeric repeat probes. The nuclear genome of G. elata was organized into 2 n = 36, with relatively small (2.71 - 5.50㎛)chromosomes that showed gradual decrease in size. Conglutination phenomenon was observed among the metaphase chromosomes, and it was distinguished from that in other plant metaphase chromosome spreads. One pair of signal was detected for each 5S and 45S rDNA in the pericentromeric region and interstitial region on the short arm of chromosomes 10 and 4, respectively, and telomeric DNA signals were detected in the terminal region of most chromosomes. Conclusions: To our knowledge, this is the first FISH chromosome composition result in G. elata and could be useful in more comprehensive molecular cytogenetic and genomic analyses as well as breeding programs of the medicinal plant G. elata.

        Dual-color fluorescence in situ hybridization karyotype analysis was created using repetitive sequences including two types of rDNA repeats (45S and 5S rDNAs) and Arabidopsis-type telomere sequence repeats. The somatic metaphase cells of Carthamus tinctorius were observed as diploids (2n=2x=24). A symmetrical or slightly asymmetrical karyotype with seven pairs of metacentric and five pairs of submetacentric chromosomes was observed. The lengths of the somatic metaphase chromosomes ranged from 4.18 to 6.53 ㎛, with a total length of 60.71 ㎛. One locus of 45S rDNA was located on the pericentromeric regions of three pairs of chromosomes and the other pair was situated on the terminal regions of the short arms of a single pair of chromosomes. One locus of 5S rDNA was detected on the interstitial regions of the short arms of two pairs of chromosomes. Arabidopsis-type telomeric repeats were detected on the terminal regions of all pairs of chromosomes. Co-localization of loci between telomeric repeats and 45S rDNA was observed in a single pair of chromosomes. The results provide additional information for the existing physical mapping project of C. tinctorius and will also serve as a benchmark to a more intricate cytogenetic investigation of C. tinctorius and its related species.

        본 연구는 V. nashicola에 의해 발생하는 배 검은별무늬병 에 저항성을 가지는 유전자를 선발하고자 고도 저항성 ‘93-3-98’과 고도 감수성 ‘스위트스킨’에 병 접종후 24시간, 48시간후 엽을 채취하여 suppression subtractive hybridization 을 수행하였다. ESTs 분석결과 ‘93-3-98’에서만 발현된 유전 자는 24H(tester, 24시간 ’93-3-98; driver, 24시간 ‘스위트스킨’)과 48H(tester, 48시간 ’93-3-98; driver, 48시간 ‘스위트 스킨’)에서 각각 9개, 14개였으며 방어 또는 스트레스에 관련 된 유전자의 비율은 각각 40%와 42%였다. 식물의 방어반응에 관련된 PR protein family 유전자로써는 24H에서 pathogenesisrelated protein 1a, major allergen Pyr c 1과 allergen mal d 1, 48H에서는 major allergen Mal d 1.03B 유전자가 특이 적으로 발현되었고 다수의 방어반응에 관련된 유전자들도 확 인되었다. Major allergen Pyr c 1, F-Box/kelch-repeat protein, flavoprotein wrbA, hypothetical protein POPTRDRAFT_ 783792은 고도저항성 ‘Bartlett’과 ‘93-3-98’에서만 높게 발 현 되었으며 중도저항성 ‘감천배’와 감수성 ‘원황’, 고도 감수 성 ‘신고’와 ‘스위트스킨’에서는 낮게 발현되어 검은별무늬병 저항성 연관 유전자로 확인되었다.

        Genome sequencing researches for considerable numbers of crops and wild plants are being developed. Cytogenetic researches according to chromosome number and size are essential to confirm and comprehend ploidy level and genome size before genome sequencing project is actually conducted. Cytogenetic researches on six food crop plants were carried out by DAPI staining and fluorescence in situ hybridization (FISH) method. Fagopyrum esculentum Moench showed 2n=2x=16, each chromosome length of 1.42㎛ to 1.77㎛, total chromosome length of 13.31㎛, and karyotypic formula of 2n=8m; Phaseolus angularis W.F. Wight, 2n=2x=22, 2.01㎛ to 3.84㎛, total 28.03㎛, 2n=9m+2sm, Perilla frutescens var. japonica Hara, 2n=2x=40, 1.73㎛ to 2.76㎛, total 44.36㎛, 2n=5m+13sm+2st. Chromosome sizes of the other three species such as, Panicum miliaceum L., 2n=2x=36, total chromosome length of 30.83㎛, Sesamum indicum L., 2n=2x=26, 27.39㎛, lpomoea batatas L., 2n=2x=30, total 33.51㎛ were too small for each chromosome type to be identified and analyzed. The result of FISH analysis using 5S and 45S rDNA probe showed species-specific chromosome locations in the genome. These preliminary analyses were carried out to decide which food crop to prioritize for genome sequencing.         Tongil (IR667-98-1-2) rice, developed in 1972, is a high-yielding rice variety derived from a three-way cross between indica and japonica. Tongil contributed to staple food self-sufficiency of Korea, an achievement that was termed the ‘Korean Green Revolution’. In this study, we analyzed the nucleotide-level genome structure of Tongil rice and compared it to those of the parental varieties. A total of 17.3 billion Illumina Hiseq reads, 47× genome coverage, were generated from Tongil rice. Three parental accessions, two indica and one japonica types, of Tongil rice were also sequenced for approximately 30x genome coverage. A total of 2,149,991 SNPs were detected between Tongil and Nipponbare; the average SNP frequency of Tongil was 5.77 per kb. Genome composition based on the SNP data by comparing with the three parental genome sequences on sliding window of Nipponbare genome sequence revealed that 91.8% of the Tongil genome originated from the indica parents and 7.9% from the japonica parent, different from the theoretical expectation in a three-way cross, i.e., 75% indica and 25% japonica parental origins on average. Copy number of SSR motifs, ORF gene distribution throughout the whole genome, gene ontology (GO) annotation, yield-related QTLs or gene locations, and polymorphic transposon insertions were also comparatively analyzed between Tongil and parents using sequence-based tools. The results indicated that each genetic factor was transferred from parents into Tongil in proportion to the whole-genome composition. The Tongil rice is the first successful superior cultivar derived from indica × japonica hybridization in Korea. Defining of genome structure demonstrates that the Tongil genome is composed mostly of the indica genome with a small proportion of japonica genome introgression.         This paper deals with an experimental study on the tensile performance of recently developed “FRP Hybrid Bars” by authors. The objective of this study is to investigate tensile strength of FRP Hybrid Bars. FRP Hybrid Bar uses the concept of material hybridization to increase elastic modulus to be used in concrete structures, especially for marine and port concrete structures. The effect of hybridization on tensile properties of FRP Hybrid Bars was evaluated by comparing the results of tensile test with those of non-hybrid FRP bars. The results of this study indicated that the elastic modulus of the hybrid GFRP bar was increased by up to approximately 5 to 204 percent by the material hybridization.

        Copy number variations (CNVs) are considered major sources of genetic variation, and CNVs may influence phenotypic variation and gene expression. To detect CNVs, rice seeds were exposed with 100~400 Gy of gamma-ray (GA, 60Co), cosmic-ray (CR) by Russia ISS, and 30 and 40 Gy of ion beam (IB, 220 Mev carbon ion). After the exposed rice seeds were cultured in 1/2 MS medium for 14 days, they were used for array-based Comparative genomic hybridization (CGH) analysis using Agilent’s RICE CGH array. As a results, the highest number of CNVs (Gain 808 and Loss 24,080) were detected in the CR treatment, whereas GA100 (100 Gy of GA) was identified the least CNVs. Compared individual chromosome, the chromosome 8 and 11 were identified the highest CNVs, the chromosome 3 had the least CNVs. Most of identified CNVs existed in the range of 10~500kb. In particular, the same CNV locations among different types of ionizing radiation were observed in chromosome 12, and these CNVs contained the commonly 5 amplified genes, containing retrotransposon protein, NADH-ubiquinone oxidoreductase chain 3, heavy metal transport/detoxification protein domain containing protein, and 2 unknown proteins. Other studies were reported that Ty1 (Long Terminal Repeat-retrotransposon family 1) transcription and retrotransposition were induced by different environmental stresses such as ionizing radiation, UV-light exposure, DNA damage and nutrient starvation in Saccharomyces cerevisiae. Our results also show that retrotransposon protein (LOC_Os12g34016) was specifically amplified by different types of ionizing radiation.

        Breeding the whole crop siliage rice cultivar basically requires priority in high biomass and grain yield and tolerance biotic and abiotic stresses. In this context, AA genome wild rices are potential resources to broaden genetic variability for whole crop silage cultivar. We have developed several introgression populations from crosses with three AA genome wild rices, O.glaberrima, O.rufipogon, and O.longistamina. Among these populations, the three BC4F8 near isogenic lines (NILs) derived from a cross of Milyang 23 and O.glaberrima(acc. 101154) could produce the high-yielding lines surpassing Milyang 23 by 20% in grain yield. Introgression lines from crosses with 4 accessions of O.rufipogon with japonica rices, Hwaseongbyeo and Ilpumbyeo had huge agronomic variability and very tall and vigorous lines possessing culm length of 125 ∼ 187cm were selected as promising potential parents to improve biomass of existing cultivars. These selections with high yield potential and biomass will be crossed to create another gene pool to combine high yield and biomass and anther culture breeding could be useful to develop genetically fixed F2 lines with both traits.

        The Brown planthopper (BPH) is one of the most serious pests of rice affecting rice yield and quality throughout the country. AA genome wild rice, O.rufipogon is a useful source of insect resistance to BPH. To broaden the BPH resistance in the existing quality japonica cultivars, crossess were made between a highly susceptible quality rice cultivar, Ilpumbyeo, and five accessions (acc.106109, 106286, 106424,105908,80671) of O.rufipogon. Among 126 F8 RIL lines evaluated for Bph resistance at NICS, the highly resistant 22 introgression lines were identified and Ilpumbyeo*2/O.rufipogon (acc.105908) showed different genotype at the marker loci of Bph1, Bph18 and the resistance reaction to BPH in the BC1F2 population fitted to 3:1 segregation mode, indicating the possible new dominant resistance gene might be involved. Polymorphism between Ilpumbyeo and resistant line was low with 36 (27%) SSR markers among 130 markers detected. Currently mapping population of BC2F2 is established and SSR marker analysis will be carried out to find out the resistance loci.

        Conventional staining and fluorescence in situ hybridization (FISH) karyotypes of the non-genetically modified (GM) parental rice line, 'Nakdong' (Oryza sativa L. japonica), and its four GM rice lines, LS28 (event LS30-32-20-1), Cry1Ac1 (event C7-1-9-1), and LS28 × Cry1Ac1 (events L/C1-1-3-1 and L/C1-3-1-1) were analyzed using 5S and 45S rDNAs as probes. Both parental and transgenic lines were diploids (2n=24) with one satellite chromosome pair. The lengths of the prometaphase chromosomes ranged from 1.50 to 6.30 μm. Four submetacentric and eight metacentric pairs comprised the karyotype of 'Nakdong' and its four GM lines. One pair of 5S rDNA signals was detected near the centromeric region of chromosome g in both the parental and transgenic lines. The 45S rDNA signals were detected on the secondary constrictions of the satellite chromosome pair in both the parental and transgenic lines. There was no significant difference in chromosome size, length, and composition between 'Nakdong' and its four GM lines. This research was conducted as a preliminary study for chromosomal detection of transgenes in GM rice lines and would be useful for their breeding programs.

        ABSTRACT: Wide hybridization was used to broaden the gene pool of japonica rice. Different approaches involving direct crosses between four japonica cultivars and eight wild species in combination with anther culture, embryo rescue, and molecular markers were used to produce interspecific hybrids, advanced alien introgression lines, dihaploids, and characterization of alien introgression. Interspecific hybrids were produced between Jinmibyeo, Ilpumbyeo, and Hwaseongbyeo cultivars of rice (O. sativa, 2n=24 AA) and wild species, O. rufipogon (2n=24, AA), O. longistaminata (2n=24, AA), O. punctata (2n=24, BB), O. minuta (2n=48, BBCC), O. alta (2n=24, CCDD), including African rice species, O. glaberrima (2n=24, AA). Crosses involving species other than A genome were produced through embryo rescue and were sterile. Following backcrossing with the recurrent japonica parents, advanced progenies have been produced for transfer of alien genes into japonica rice. As results of yield trials, three elite lines were generated from this cross; they are "Suweon 487" with resistance to black streak dwarf virus, "Suweon 497" with blast and bacterial blight resistance and "Suweon 506" with blast resistance, emonstrating that wild species genes have now become important component of japonica rice breeding.
