Background: Because oxidative stress can induce decreased quality of caprine semen during the storage, there has been limitation for the use of stored semen in the assisted reproductive technologies. The present study, therefore, assesses the potential of Annona muricata (A. muricata ) to reduce semen storage associateddamages. Methods: Semen was collected by electro-ejaculation from ten bucks, and extended with Tris-egg yolk (TEY) supplemented with A. muricata leaf aqueous extract (SAE) at 20 (SAE20), 40 (SAE40), and 80 (SAE80) μg/mL. Sperm variables including motility, motion characteristics, viability, membrane functionality, and DNA integrity were assessed at different storage periods (6, 24, 48, and 72 hr). In addition, oxidative stress indicators in the extender supplemted with SAE were also assessed for each group. Results: By adding SAE at 80 μg/mL in TEY, the storage of goat buck semen was improved, resulting in reduced loss of sperm motility, viability, DNA fragmentation, and membrane integrity during chilled storage at 4℃ for up to 72 hr. In addition, enrichment of TEY extender with SAE significantly (p < 0.05) reduced malondialdehyde, an indicator of oxidative stress, compared to the negative control. Conclusions: Supplementation of SAE in TEY extender can reduce buck spermatozoa liquid storage associated damages due to oxidative stress.

The normal feeding approach of goats might be due to their precise anatomical and physiological characteristics of entity, which permit them to be highly selective, to eat legume silages and wild green grass. This review has been designed to consider the grazing behavior, fodder selection, and feed composition of goats. Various herbs and corns consumed by goats have numerous nutritive resources. Based on the general herbaceous intake activities and behavior of goats, they prefer wild grass such as grass grown in the steep hills than soft grass. Because the digestion capacity of cellulose feed has higher digestion level compared to other non-ruminants within rumen and it is advantageous to use wild forest or mountain grass which comprises high proportion of cellulose feed for goat. In South Korea, there are abundant feed resources for goats because of occupying large areas of mountains. Thus, goat production and feeding costs could be reduced if plants are used from the wild forest as a feed for goats relative to grassland grazing. Also, it is expected to contribute in improvement of goat farming with harmonious relationship between the grassland and wild forest while satisfying animal welfare and physiological desires of livestock.

A case of caseous lymphadenitis (CL) was detected in a native Korean goat without manifestation of superficial nodules but with pulmonary abscesses that were characterized microscopically by granulomatous inflammation consisting of a central necrotic region, peripheral fibrous capsule, and numerous cellular infiltrates with epithelioid macrophages, giant cells, and lymphocytes. Presence of Corynebacterium pseudotuberculosis was identified by using bacterial isolation and multiplex PCR. In addition, a 43% seropositivity to C. pseudotuberculosis was observed in goats from the same farm. This case highlights the risk of exposure of farmers to the causative agent and the urgent need for national monitoring for CL, a potential zoonotic.

This study was conducted to determine grazing intensity of growing Korean native goats(Capra hircus coreanae) on mountainous pasture. It was carried out to obtain basic information for improvement of mountainous pasture management and establishing feeding system of Korean native goat. A total of 20 goats were grouped by feeding systems [A mountainous pasture grazing group (Concentrated feed of 1.5% body weight, treatment 1, T1, n=10) and a barn feeding group (TMR, treatment 2, T2), n=10] to conduct study from April to September. The average forage productivity of the mountain pasture was 500.9 ± 61.41 kg/ha. The average dry matter intake in T1 was 0.64 and the calculated grazing intensity was 21 head/ha. In productivity, when the two treatments(T1, T2) were compared, the dry matter intake was about two to three times the difference. The average daily gain per day during the experiment was 63.3 in the mountain pasture and 120 g in barn feeding. When grazing, considering mountainous pasture productivity it is necessary to increase the productivity through proper feeding. The feed costs of black goats raised by grazing on the grassland in the same period showed an average 75% reduction compared to barn feeding. As a result of this study, it can be expected that a considerable reduction of feed costs can be expected in the breeding of Korean native black goat using the mountain pasture.

The study was conducted to determine effects on forage productivity, feed value, grazing intensity and livestock productivity in growing Korean native female goat grazing in native pasture. Its with average initial body weight of 14.10±3.6kg and an average age of 4 months were used in this study. Dry matter content of native pasture was the highest at 33.48 ± 2.56% in June, and the content was significantly increased from spring to autumn (p<0.05). Crude protein was maintained between 11% and 12% on average. Nutrient content was maintained at a certain level in native pasture, but there were differences due to the different types of wild grasses produced in each season. The productivity of forage crops increased from June, but decreased after August and showed a characteristic of grassland where productivity decreased rapidly in spring and autumn. The average grazing intensity are 39 head/ha. Black goat average daily gain was 80.2g/d. The stable weight gain in grazing is that the nutrient requirement of the black goat was met by supplementing the concentrated feed during grazing. study, can be expected that the productivity of livestock can be increased through the proper feeding of supplementary feed and maintenance of grazing intensity

The objective of this study was to determine the effect of various roughage sources on nutrient digestibility and enteric methane (CH4), and carbon dioxide (CO2) production in goats. Four castrated black goats (48.5 ± 0.6 kg) were individually housed in environmentally controlled respiration-metabolism chambers. The experiment design was a 4 × 4 balanced Latin square design with 4 roughage types and 4 periods. Alfalfa, tall fescue, rice straw, and corn silage was used as representative of legume, grass, straw, and silage, respectively. Dry matter digestibility was higher (p < 0.001) in corn silage than in alfalfa hay. Dry matter digestibility of alfalfa hay was higher than those of tall fescue or rice straw (p < 0.001). Neutral detergent fiber digestibility of tall fescue was lower (p < 0.001) than those of alfalfa, rice straw, or corn silage. Daily enteric CH4 production and the daily enteric CH4 production per kilogram of BW0.75, dry matter intake (DMI), organic matter intake (OMI), digested DMI, and digested OMI of rice straw did not differ from those of tall fescue but were higher (p < 0.001) than those of alfalfa or corn silage. Roughage type had no effect on enteric CO2 emission in goats. Straw appeared to generate more enteric CH4 production than legume or silage, but similar to grass.

This study was conducted to evaluate the protein requirement for maintenance of fattening Korean black goat (Capra hircus coreanae). Six male goats with average initial body weight (BW) of 31.78±4.54kg and an average age of 8 months were used in this study. The experiment had a replicated duplicated 3×3 Latin square design for balancing carryover effects. In the course of the experiment, each of Black goats were fed three diets that were formulated to contain T1 (13%), T2 (16%) and T3 (19%) levels of crude protein (CP). A 14-day diet adjustment period was followed by a 5-day collection period. Dry matter intake (DMI) of groups fed diets with T2 was 966.67g/d which was higher than group fed diets with T1 and T3 were 925.14g/d and 936.08g/d each. Average daily gains (ADG) of black goats were the highest in T2(167.13g/d) But, there was no significant difference. Dietary protein levels affected the apparent digestibility of CP (p<0.05). A significant difference was found in CP intake among treatments and goats receiving T3, T2, and T1 recorded 181.23, 154.57, and 128.78g CP/d, respectively. This was excepted because CP intake is proportional to CP content of diet, which from highest to lowest was as follows: T3 (19%) > T2 (16%) > T1 (13%). Intercept of the regression equation between CP intake and CP balance indicated that maintenance CP requirement was 1.63g/BW0.75.

Neuron-specific enolase (NSE), a brain specific isoenzyme of the glycolytic enzyme, is characterized by its consistent occurrence in the cytoplasm of mature neurons. The purpose of the present study was to investigate the expression of NSE in the developing tongue of Korean native goats. The tongues of the fetuses were removed from 2- to 4-year-old female Korean native goats by caesarean section performed under general anesthesia. The expression of NSE in the developing tongue of goat fetuses (60, 90, 120, and 150 days) was studied using immunohistochemistry. In 60-day-old fetuses, NSE-immunoreactivity (IR) exhibited weak appearance in lamina propria of the basal portion and the apical epithelia of the tongue. In 90-day-old fetuses, NSE-nerve fibers were extended in the core part of connective tissue, and primordia of the taste buds was moderately positive. In 120-day-old fetuses, NSE-IR was strongly expressed in taste buds and gustatory nerve fibers. In neonates, the taste buds of vallate papillae were strongly positive for NSE, and development of nerve fibers was synapsed with connective tissue of well innervated taste buds. These results indicate that NSE expressions were associated with the sign of nerve innervation in prenatal development of goat tongues and NSE may be a useful neuronal marker to understand the development of gustatory nerve innervations.

This study was conducted to determine grazing intensity of growing Korean native goats (Capra hircus coreanae) on mountainous pasture. It was carried out to obtain basic information for improvement of mountainous pasture management and establishing feeding system of Korean native goats. Castrated, male goats (n=10) with average initial body weight (BW) of 23.33±2.15kg and an average age of 4 months were used in this study. Grazing goats were supplemented by concentrates with 1.0% of BW. The crude protein content of forage was the highest in October (22.71±0.25%) and there were significantly differences (p<0.05) in monthly comparison. The forage productivity of pasture was the highest from May to June (1718.7±207.5~ 1672.0±422.8 kg/ha) but it was decreased in July (1356.0±103.8 kg/ha) because of drought and summer depression. Average daily gains (ADG) of goats were the highest in June (99.5±6.4 d/g). Grazing intensity was calculated by forage productivity and dry matter intake (DMI) and was the highest in May (65 head/ha). As shown in the results of this research, grazing intensity was suggested to average 39 head/ha from May to October. It is desirable that adequate grazing intensity was maintained by adjusting supplemental feed.

Cytokeratin (CK) comprises the intermediate filament cytoskeleton of epithelial cells. Patterns of CK expression can be regarded as a specific marker for epithelial differentiation status. The aim of this study was to identify CK expression on tongues of Korean native goats ranging from 60-day-old fetuses to newborns during prenatal development using immunohistochemistry. The tongues of fetuses were removed from 2- to 4-year-old female Korean native goats by caesarean section performed under general anesthesia. Immunohistochemistry was performed to assess CK expression patterns on developing goat tongues using serial paraffin-embedded sections. Light zones signifying CK immunoreactivity in dorsal lingual epithelia were weakly positive in 60-day-old fetuses. In 90-day-old fetuses, deep areas in dorsal lingual epithelia were strongly positive for CK expression and superficial areas were moderately positive. In 120-day-old fetuses, light zones of lingual epithelia in the vallate papilla were strongly positive for CK expression, whereas ducts of von Ebner’s glands were moderately positive. In neonates, taste buds were positive for CK expression, whereas non-taste epithelial cells and von Ebner’s glands were negative. These findings indicate that goat tongues have different patterns of CK expression during development and provide a morphological basis for studies on the biological mechanism of epithelial differentiation.

The purpose of this study was to identify the composition and organization of lingual tissues underlying the histo-structural change of developing tongue in Korean native goats by light microscopy (LM). Tongues of the fetuses on days 60, 90, 120 and neonate were examined for the morphological development. In the 60-day-old fetuses, the tongue tissues were differentiated into epithelium, lamina propria and muscle layer. Primordia of filiform, conical, lentiform, fungiform and vallate papillae appeared and rudiments of taste bud were observed in the epithelia of the primordia of the gustatory papillae. The dorsal surface of the lingual epithelia showed a weak PAS positive reaction. Collagenous fibers and small blood vessels were shown in the connective tissues. In the 90-day-old fetuses, Von Ebner’s glands were moderately PAS positive while the muscle fibers and connective tissue were strongly positive for PAS. The collagenous fibers increased and came to have a more complex arrangement in the tongue. The muscle fibers were spread out at various directions and developed in striated muscle bundles. In the 120-day-old fetuses, taste buds were observed in the epithelia of the gustatory papillae, and several well-developed tissues visible such as blood vessels, collagenous fibers, muscle fiber bundles and Von Ebner’s glands. In the neonates, many taste buds were found in a transverse section of the vallate papilla. The muscle layers, Von Ebner’s glands, collagenous fibers and blood vessels were more developed than those of the 120-day old fetuses. These findings indicate that goat tongues have a variety of different shapes during prenatal development.

The purpose of this study was to explore the morphologi- cal characteristics of developing lentiform papilla (LP) in Korean native goats by scanning electron microscopy (SAM). Tongues were removed from fetuses on days 90, 120, neonates, and juveniles on days 30, 60, 90, 120, 150, and 180. In prenatal development, the primordia of LP in 90-day-old fetuses were round and spotted on the inner most part of the torus linguae of the tongue. Primordia of LP in 120-day-old fetuses also had a lens-like shape. In neonates, LP displayed similar features as the adult one. In postnatal juveniles on days 30 and 60, LP continually increased in size without much difference in structure compared to that of neonates. By postnatal day 90, detached pieces of keratinized super- ficial epithelia were observed. Microridges and microplicae were well developed on the epithelial surface of LP in 60- to 120-day-old goats. The lengths of LP were 476~514 μm in neonates, 687~962 μm in the weaning period of 60-day- old goats, and 1,068~1,567 μm in the maturing period of 180-day-old goats. These findings indicate that goat LP has different sizes and shapes from those of other species during development.

The aim of this study was to investigate the morphological development pattern of conical papilla (CP) during prenatal and postnatal periods in native Korean goats by scanning electron microscopy. Tongues were removed from fetuses on days 90, 120, and neonates and from juveniles on days 30, 50, 60, 75, 90, 120, 150, and 180 after birth. The lengths of CP were 194~240 μm in neonates, 335~485 μm in the weaning period of 60-day-old goats, and 526~662 μm in the maturing period of 180-day-old goats. The primordia of CP in 90-day-old fetuses, shaped like mountain berry, were sprouted. The CP of 120-day-old fetuses was a cylinder shape with an apex whose center was slightly concave and the margin was sticking up like a low fence. The CP of neonates was empty inside and the secondary papillae were irregularly arranged fence-like structures on it. In 60-day-old goats, CP was an obliquely sectioned cylinder shape compacted in inner surface; the shapes of CP varied in 120-day-old goats, and had a smooth surface. A low elevation of CP was observed in 150- and 180-day-old goats. The microridges, microplicae, and micropits were well developed on the epithelial surface of lingual papillae in 60- to 120-day-old goats. These findings indicate that CP of goats has a variety of sizes and shapes during development.

The aim of this study was to investigate the morphological development of fungiform papillae in native Korean goats by scanning electron microscopy (SEM). Tongues were removed from fetuses on days 120 and 150 of gestation and from juveniles on days 30, 60, 90, 120, 150, and 180 after birth. The sizes of fungiform papillae in the newborn were 225.0-250.0 μm, the weaning period of 60-day-old goats was 271-294 μm and the maturing period of 180-day-old goats was 385-398 μm, respectively. The primordia of fungiform papillae had the typical feature of a mushroom shape in a 120-day-old fetus. In a 150-day-old fetus (neonate), fungiform papillae had features similar to those of the adult form and showed a gradual increase in size with slight morphological changes until 150-day-old goats. Microridges, microplicae, and micropits were well developed on the epithelial surface of lingual papillae from 60 to 150-day-old goats. The results obtained in this study might provide some basic data on the mechanisms that control the morphogenesis and development of the lingual papillae in ruminants.

The experiment was divided into two phases. In phase-I fresh embryos were transferred and in Phase-II frozen embryos were transferred. Embryos were collected by using Dulbecco's phosphate buffered saline. In phase-I total of 65 ova were collected out of 107 ovulation in 18 goats. Recovery of ova was 60.74%, of which 51 (78.46%) was fertilized. Sixteen embryos were transferred to 10 recipient goats and kidding was observed in 6 goats, that produced 10 kids. Thus, 62.50% embryo survival and 60% kidding were achieved in phase-I. In phase-II of the experiment, 17 regular cyclic Black Bengal goats were used. The main purpose was to study the viability of caprine embryos after cryopreservation. In this phase the embryos were collected and frozen using Bio-cool freezers. A two step addition of cryoprotectants (5% glycerol and 10% glycerol) and three-step dilution of cryoprotectants with 1mole (M) sucrose was used. Embryos were preserved for 10 to 45 days. Out of 27 embryos preserved, 18 were recovered after freezing and thawing (37 water bath) with 33.33% embryonic loss. Seventeen frozen and thawed embryos were transferred in 9 recipient goats, out of which kidding was observed in 6 goats and 7 kids were produced, giving a 66.66% kidding and embryo survival of 41.17%. The technique utilized for fresh and frozen embryo transfer can be successfully utilized to produce goats of superior genetic merits. The protocol used for addition of cryoprotectant, freezing, thawing and dilution was found suitable for caprine embryo freezing.
