        This study was carried out to obtain useful information on the cost-effective and labor-saving methods in the on-farm pigmented rice cultivation trials that were performed in Gangjn, Jindo and Gimje provinces. Two different pigmented rice cultivars were tested at each site from 2014 to 2015. From the common rice transplant technologies, Wet Hill Direct Seeding Technology (WHDST) was applied and was compared to Transplanting Machine (TM) Technology, which was mainly used for rice transplanting. The results of the study showed that during the trial period, the average C3G contents of pigmented rice ‘Heugkwang’ in Gangjin was 145.5 mg/100g using the WHDST. This was 23.5 mg/100g higher than using TM in Gangjin area. On the other hand, the C3G contents of functional pigmented rice variety ‘Seonhyangheukmi’ was 163 mg/100g under the WHDST while 189.3 mg/100g under the TM. Therefore, it was observed that the C3G contents of functional pigmented rice changed depending on the variety and according to the annual environmental variation. In Jindo area, two years average C3G contents of functional pigmented rice variety ‘Josaengheugchal’ was resulted to 268.1 mg/100g using TM. This C3G contents was 97.8 mg/100g more than the amount obtained using WHDST. For functional pigmented rice variety ‘Shinnongheugchal’, the C3G contents was 247.3 mg/100g in the WHDS, while 249.1 mg/100g in the TM. This indicated that C3G contents tended to be similar based on cultivation method. The C3G contents of the rice variety ‘Heugkwang’ in Gimje area were 34.7 mg/100 g and 20.2 mg/100g using WHDST and TM, respectively. Meanwhile, the C3G contents of functional pigmented rice variety ‘Heugsujeong’ was 85.6 mg/100g in the WHDST and 43.62 mg/100g in the TM. In conclusion, the study showed that the C3G contents of the pigmented rice are affected by the yearly environmental variation, the regions, and the cultivated variety.

        Cycloamyloses (CA) from high-amylose rice starch (Dodamssal, 50.94% amylose) was produced to form a complex with anthocyanin extracted from black rice (sihtoheukmi). The effects of CA concentration (1, 5, 10 and 30%, w/v), pH (2, 4, 6, and 8), UVB irradiation time (~24h) and thermal treatment time (~24h) on the oxidant capacity of the complex were investigated. Anthocyanin consisting of 95% cyanidin-3-glucoside (C3G) was extracted from the black rice by suspending in 60%(v/v) ethanol containing 0.2%(v/v) hydrochloric acid for 90 min at 50°C. Cycloamyloses (CA) produced by 4-α-glucanotransferase (4αGTase) formed complexes with C3G by shaking the mixture for 48 hours at 30°C. Antioxidant activities (radical scavenging capacity) were determined by using ABTS [2,2'-azinobis-(3-ethylbenzothiazoline- 6-sulfonic acid)]. The antioxidant activities of the complexes increased with pH increasing. Upon UVB irradiation and heat treatment, the antioxidant activities were more retained for the complexes incorporating 10% CA concentration or above compared to the C3G control. Degradation rate constant (Kd) and half-life (T1/2) were obtained from degradation data of CA complex with cyanidin-3-glucoside. The effect of CA complex formation with C3G is significantly observed at pH4. The results suggest that the CA complex formed with C3G indeed has an enhanced antioxidant potential toward UVB and thermal degradation.

        ‘Neugeunsuperjami’, a late maturing and blackish purple pigmented rice cultivar, was developed from a cross between Gumjeongbyeo and Hwaseonchalbyeo. During selected by pedigree breeding method until F8 generation, a promising line, SR28757-22-2-4-2-1-1-2-B, was selected and advanced by late maturing and content of Cyanidin 3-glucoside. Advance line designated as the name of ‘KNOU 13’. This variety headed on August 28, 140 days to heading after sowing. The culm length and the panicle length of ‘Neugeunsuperjami’ was 91.0 cm and 20.0 cm, respectively. The number of panicle per hill was 10.5 and the number of grain per panicle was 88.6. The dehulling recovery of ‘Neugeunsuperjami’ was about 80.5% and 1,000-grain weight was about 19.7 g, heavier than that of ‘Heugjinju’. ‘Neugeunsuperjami’ has 3~5 times higher Cyanidin-3-glucoside(C3G) content compared with ‘Heugjinju’.

        ‘Pareunsuperjami’, a early maturing and blackish purple pigmented rice cultivar, was developed from a cross between Heugjinju and Suwon 425. During selected by pedigree breeding method until F8 generation, a promising line, C3GM(CG2-2-70-1-1-3-2-B), was selected and advanced by early maturing and contents of cyanidin 3-glucoside. Advance line designated as the name of 'KNOU 3'. This variety headed on July. 30, 115 days to heading after sowing. The culm length and the panicle length of 'Pareunsuperjami' was 63.5cm and 21.3cm, respectively. The number of panicle per hill was 9.4 and the number of grain per panicles was 98.0. The ratio of fertility of ' Pareunsuperjami’ was about 80.5% and 1,000-grain weight was about 18.8 g, heavier than that of ‘Heugjinju’. ' Pareunsuperjami ' has 3~4 times higher Cyanidine-3-glucoside(C3G) contents compared with ‘Heugjinju’.

        전통적인 교배육종을 통하여 천연색소(C3G) 함량을 높인 ‘슈퍼자미2호’(흑진주벼/수원 425호//대립벼 1호) 품종의 작물학적 특성과 품질특성을 조사 분석한 중부평야지에서 평균 출수기는 8월 30일로 만생종이며, 간장은 106 cm 정도이며 임실률은 82.0%였다. 또한 현미천립중은 30.1 g 정도로 ‘슈퍼자미’(26.2g)보다 무거운 품종이다. ‘슈퍼자미2호’ 품종의 현미 장폭비는 2.08의 장원형으로 흑진주벼에 비해 길이와 폭이 12%, 26%로 증가된 대립종이다. 미량원소 중 K, Ca 함량은 ‘흑진주벼’에 비해 낮았고, 단백질, 회분 함량도 낮게, 열량, 지방, 탄수화물 함량은 비슷한 수준으로 나타났다. ‘슈퍼자미2호’ 품종의 C3G 색소함량은 2013년에는 1,782 mg(100 g 종자), 2014년에는 1,980 mg(100 g 종자)으로서 ‘슈퍼자미’ 보다는 다소 낮았으나 ‘흑진주벼’ 보다는 9배 이상 높았다.

        Brown rice grain pigments of black rice have a higher content of bioactive substances such as anti-mutagenic substance than the non-pigmented rice grain. The major anthocyanin pigment contained in black rice was cyanidin-3-glucoside. This study was conducted to establish a rapid analysis method for determining cyanidin-3-glucoside contents in flour and whole rice seeds of black rice using VIS/NIRS technique. A total of 60 black rice samples were used for VIS/NIRS equation model development and validation. The value of coefficient of determination of external validation (r 2 ) and standard error of performance (SEP) in whole rice seed sample were 0.653 and 97.2, respectively. Therefore, the value of it seemed to be difficult to analyze cyanidin-3-glucoside content in whole rice seed samples using VIS/NIRS. However, in rice flour sample, the best accurate equation model was obtained from the partial least square regression (PLS) method. The value of r 2 , SEP and bias were 22.5, 0.922 and -1.45 in the calibration transformed to the N-point smooth of log 1/R signal, 5 factors, respectively. Therefore, the results of our study clearly demonstrate that the VIS/NIRS method would be applicable only for rapid determination of cyanidin-3-glucose content in black rice flour samples.

        ‘대립자미벼’는 한국방송통신대학교 농학과에서 2009년도에 육성한 종실이 크고 C3G 함량이 높은 복합기능성 유색미 품종이다. 주요 특징으로 중부평야지 평균 출수기는 8월 18일로 중만생종이며, 간장은 80.6 cm 정도이고, 종피색이 흑자색인 유색메벼 품종이다. 현미 장폭비는 1.88로 단원형이고, 현미천립중은 27.9 g으로 무거운 대립 품종이다. 종피의 안토시아닌 주색소인 C3G (Cyanidin 3-glucoside) 함량이 ‘흑진주벼’ 보다 3.6배 높은 품종으로 건강기능성 소재 및 가공용으로 활용가치가 높은 것으로 기대된다. 중부평야지역 보통기 보비재배 2개소에서 평균 450 kg/10a 수준으로 ‘흑진주벼’ 대비 16% 증수 되었다.

        ‘큰눈자미벼’는 한국방송통신대학교 농학과에서 2009년도에 육성한 쌀눈이 크고, C3G 함량이 높은 복합기능성 유색미 품종을 개발하였다. ‘큰눈자미’의 중부평야지 평균 출수기는 8월 13일로 중생종이며, 간장은 87 cm 정도이고, 종피색이 흑자색인 유색메벼 품종이다. 현미천립중은 18.9 g 정도이고, 장폭비는 2.14로 중원형이며, 현미에서 쌀눈의 비율이 8.2% 수준으로 ‘흑진주벼’의 2.5배 정도이며, 현미 1립 기준 쌀눈의 무게가 ‘흑진주벼’의 2.8배 수준인 거대배아미 품종이다. 현미의 지방함량과 Lysine 함량이 ‘흑진주벼’ 보다 높고, 종피의 안토시아닌 주색소인 C3G 함량이 ‘흑진주벼’에 비해 2배 정도 높은 품종으로 건강 기능성 소재로서 활용가치가 높을 것으로 기대된다. 중부평야지역 보통기 보비재배 2개소에서 평균 410 kg/10a 수준으로 ‘흑진주벼’ 대비 5% 증수 되었다.

        슈퍼자미'는 한국방송통신대학교 농학과에서 2009년도에 육성한 C3G 고함유 흑자색 품종으로 주요특성과 수량성을 요약하면 다음과 같다. 1. 중부평야지 평균 출수기는 8월 25일로 만생종이며, 간장은 75 cm 정도이고, 종피색이 흑자색인 유색메벼 품종이다. 2. 현미천립중은 26.2 g 정도이고, 장폭비는 1.87로 단원형이며, 종피는 흑자색으로 안토시아닌 주색소인 C3G 함량이 '흑진주벼'의 10배 수준으로 색택이 균일한 품종이다. 3. 중부평야지역

        뽕나무 오디함유 천연색소 C3G의 식용색소화 및 안정생산을 위한 최적 전처리 조건은 0.1 % citric acid-70% EtOH로서 오디로부터 제조한 순수 오디색소 분말의 수율은 8%이었다. 실제 농가에서 오디 색소 추출시 추출 시간 및 제조 비용을 대폭 절감할 수 있는 추출 방법을 개발하였다. 오디색소 추출시 품종의 선택이 중요하므로 C3G 함량이 높은 품종을 선택하도록 해야 할 것이다. 오디 색소 분말은 알코올, 물 및 밀가루 등 식품 재료나 가공방법에 상관없이 적색소의 특성이 유지되므로 다양한 식품에 적용하여 고부가가치를 향상시킬 수 있을 것으로 판단된다.

        Cyanidin-3-glucoside(C3G) 고함유 흑자색미 계통을 육성하기 위해 흑진주, 수원425호, 상해향혈나, 용금1호, C3GHi 품종 및 계통들 간에 완전이면교배에서 얻은 20개 조합 F1 의 현미 천립중과 C3G함량에 대한 diallel analysis 및 조합 능력 분석 결과는 다음과 같다. 1. 교배모본의 현미 천립중은 16.6 g(흑진주) - 30.4 g(상해향혈나)이고, 현미 100 g당 C3G 함량은 C3GHi가 흑진주의 5배 정도 높은 1043.5 mg으로 나타났다. 2. 완전이면교배 결과 현미 천립중과 C3G 함량은 비대립 유전자 효과는 나타나지 않았고, 유전자의 상가적 및 우성적 유전효과만 인정되었다. 3. 현미 천립중과 C3G 함량의 유전 양식은 두 형질 모두 상가적 및 우성효과에 의한 부분우성을 보였으며, 평균 우성정도는 각각 0.80 및 0.69였고, 교배모본에서 정의대립 유전자 비율이 높았으며, 우성친의 빈도가 높고, 후대에서는 감소하는 방향으로 작용하였다. 4. 두형질의 상가적 유전효과에 의한 협의의 유전력은 각 각 0.66, 0.70이고, 광의의 유전력은 각각 0.98, 0.98로 높게 나타났다. 5. 현미 천립중에 대한 일반조합능력 효과는 상해향혈나가 가장 높았고, 흑진주가 가장 낮았으며, 흑진주/수원425 호, 수원425호/C3GHi 조합의 특정조합능력 효과가 가장 높았다. 6. C3G함량에 대한 일반조합능력 효과는 C3GHi가 가장 높았고, 상해향혈나가 가장 낮았으며, 흑진주/C3GHi 조합에서 특정조합능력 효과가 가장 낮게 나타났다.

        Anthocyanins are water-soluble glycosides and acylglycosides of anthocyanidins, having different color variations due to its substitution patterns. Anthocyanins, present in various fruits, vegetables and crops as natural colorant, have been well characterized for its bioactive properties, anti-oxidant, anti-cancer, anti-proliferative and anti-inflammatory properties. During extraction and purification, the factors, such as pH, temperature, oxygen, light, enzymes, nucleophilic agents, sugar derivatives and co-pigments, have affected on anthocyanin stability. For this reason, the extraction method should be thoroughly checked for the qualitative/quantitative analysis of anthocyanin in particular plant material. To identify the optimum extraction method of cyanidin-3-glucoside, major anthocyanin of dark purple-colored grains, Oryza sativa cv. Heugjinjubyeo, Phaselous vulgaris, Phynchosia gngularis, Sesamum indium, Rhynchosia nulubilis and Lablab purpureus, reversed-phase HPLC analysis using solvent system of acetonitrile, methanol and water were accomplished.
