본 연구는 닭 진드기 구제를 목적으로 개발된 살비제인 와구잡이 II® (WGJB, 편백정유 : 계피정유 = 20 : 56)에 대하여 토끼와 기니픽을 이용하여 피부 자극성 및 감작성 평가를 각각 수행하였다. 일차피부자극시험에서 토끼의 피부에 WGJB를 24시간 동안 처리한 후 피부 자극성을 확인한 결과, WGJB는 비찰과 부위에서 홍반과 부종과 같은 어떠한 부작용도 일으키지 않았으나, 몇몇 토끼의 찰과 부위에서 매우 약한 홍반과 부종을 나타내어 WGJB의 1차 피부자극 지수는 0.625이었다. 따라서 WGJB는 약한 자극성이 있는 물질로 분류되었다. 피부감작성 시험에서 기니픽에 0.1 mL의 WGJB을 피내주사한 후 24시간 동안 감작시켰다. 감작 1주일 후 WGJB를 함유한 패취를 주사 부위에 부착하여 48시간 동안 처리한 다음, 2주 후에 WGJB 를 함유한 패취를 부착하여 감작을 야기시켰다. WGJB는 어떠한 알러지 반응도 나타내지 않았다. 따라서, 본 연구를 통하여 WGJB는 약한 피부 자극성을 가지며 감작성을 야기하지 않는 물질로 평가되었다.
The poultry red mite (PRM), Dermanyssus gallinae, is one of the most important external parasites in the poultry industry. PRM adheres to birds’ bodies, and its hematophagy causes anemia and itching, and reducing egg production (~20%). Fipronil is a toxic pesticide used to control PRM. Thus, an environmentally friendly alternative control agent is needed. In this study, the effectiveness of various environmentally friendly PRM control measures including an acaricide consisting of synthetic silica, clove extracts, garlic extracts, and a probiotic solution were evaluated, comparing them to a combined treatment using the chemical acaricides. Each agent was tested in vitro using two approaches, in which PRMs were either added to environments containing acaricide, or sprayed in situ. Within 10 hours of inoculation, all the environmentally friendly acaricides except for the probiotic solution killed or rendered immotile significantly more mites than the control, normal saline (p < 0.05). The performance of the environmentally friendly acaricides, except for synthetic silica, was significantly weaker than that of commercial chemical acaricides. (p < 0.05). Further tests to determine the optimal concentration of amorphous synthetic silica agent for complete PRM control found this to be 400 g/L. PRM motility following treatment showed the same pattern: the synthetic silica agent and chemical acaricide killed mites, while application of other environmentally friendly agents resulted in living, but immotile, insects, or those with reduced motility. As a result, these environmentally friendly acaricides, especially the synthetic silica agent, could be used as alternatives to chemical acaricides for PRM control.
The present study evaluated the repellent activity and the improvements in egg production and mortality rate on laying hens infested with poultry red mites (PRM) administered with a mixture of Korean traditional herbal methanolic extracts (CHE, Foeniculum vulgare : Sophora flavescens : Cinnamomum cassia : Glycyrrhiza glabra = 1:1:1:1, w/w/w/w). PRM numbers per laying hens in CHE-0.5 administered with CHE 0.5 kg/ton feed were significantly decreased compared to those in Non-CHE (control) during overall experimental periods (1st week post-treatment, p<0.05; 2nd week post-treatment, p<0.01; 3rd-4th week post-treatment, p<0.001). In CHE-1.0 administered with CHE 1.0 kg/ton feed, PRM numbers were significantly decreased compared to those in Non-CHE during the whole experimental periods (1st week post-treatment, p<0.01; 2nd-4th week post-treatment, p<0.001). After administration of CHE for 4 weeks, egg production and mortality rate in CHE-0.5 and CHE-1.0 were significantly improved compared to those in Non-CHE (p<0.05). In addition, hematological and blood biochemical parameters in all CHE-treated groups were insignificantly different compared to those in Non-CHE. This study showed that CHE (0.5 and 1.0 kg/ton feed) had a repellent activity on PRM in laying hens and effects on the improvement in egg production and mortality rate of laying hen. CHE could be an effective and safe candidate for the control of PRM and the improvement in egg production and mortality rate of laying hens.
Red mite, Dermanyssus gallinae is one of the serious pests, damaging to the egg production in chickens. Moreover, the overuse of chemical insecticides caused pest resistance and environmental residual toxicity. Therefore, this work provides a screening method to select entomopathogenic fungi as a candidate of environmentally safe control agent, having high miticidal activity against D. Gallinae. The virulence test was conducted using a spraying method. Of the several species, Beauveria species showed high virulence and mycosis were observed. Some isolates were produced on eight cereal grains in Petri dish conditions. Millet, perilla seed and barley showed the high conidia production. Conidia of the isolates produced on millet and rice showed high thermal stability, when exposed to 45℃. Based on these results, this work suggests that entomopathogenic fungi could be used to control the D. Gallinae.
The susceptibility of Dermanyssus gallinae adults, collected from poultry farms in 5 regions, to 11 acaricides was investigated. When bifenthrin and formic acid with pyridaben were diluted 100 times, bifenthrin showed 100% acaricidal activity; however, formic acid with pyridaben showed less than 20% acaricidal activity for poultry farms in 3 regions (Gyeongju, Chilgok, and Geumsan) except Yeoncheon and Anseong. Clothianidin, thiamethoxam, fenitrothion, and formic acid with pyridaben showed differences in acaricidal activity among regions. Therefore, farmers should concentrate during the selection of these acaricides. However, carbaryl, cartap hydrochloride, dichlorvos, and bifenthrin showed high activity against D. gallinae collected from poultry farms in five regions. To control D. gallinae effectively, the alternation of acaricides is necessary, and indiscriminate pesticide use should be avoided. Therefore, this study can serve as a basis for controlling D. gallinae.
5개 지역 양계장에서 채집한 닭진드기(Dermanyssus gallinae) 성충에 대한 11종 살비제의 감수성을 조사하였다. Bifenthrin과 formic acid + pyridaben은 100배의 희석배수로 처리한 결과, bifenthrin은 5가지 지역에서 모두 100%의 살비율을 보였으나 formic acid + pyridaben 합제 는 연천과 안성을 제외한 나머지 3지역(경주, 칠곡, 금산)에서 20% 이하의 낮은 살비율을 보였다. 카바메이트계 약제인 carbaryl은 경주(45.0%) 를 제외한 4지역에서 100%의 살비효과를 보였다. 유기인계 약제인 dichlorvos는 연천을 제외한 4지역 모두 100%의 살비효과를 보였지만 연천 은 66.7%의 살비율을 보였다. 연천과 안성지역의 양계장이 살비제에 대한 감수성이 높게 나타났으나 다른 3지역(경주, 칠곡, 금산)의 경우에는 약제에 따라 활성의 차이를 보였다. Clothianidin, thiamethoxam, fenitrothion, formic acid + pyridaben은 지역에 따라 살비활성의 효과에 차이를 보여 사용 시 주의를 기울여야 할 것이나, carbaryl과 cartap hydrochloride, dichlorvos, bifenthrin은 실험에 사용된 모든 지역에서 활 성이 높아 모든 지역에서 사용가능할 것으로 생각된다. 양계장별 살비제 활성이 차이를 보이는바 효과적인 닭진드기의 방제를 위해서는 살비제의 활성평가와 더불어 약제의 교호살포와 무분별한 사용을 지양해야할 것으로 보이며 본 연구가 닭진드기 방제를 위한 기초 자료로 활용될 수 있을 것으로 생각된다.
Survival rate and development of the two spotted spider mite, Tetranychus urticae are decreased by UV-B irradiation (Otsuka and Osakabe, submitted). This is considered to be a reason why this mite prefers lower leaf surfaces host plants. On the contrary, the citrus red mite, Panonychus citri is known to remain on upper leaf surfaces of host plants. Such difference in the distribution was assumed to depend on the difference in UV resistance. Hatchability of P. citri eggs was decreased by UV-B irradiation, yet their eggs were more protected from the UV-B irradiation in company with T. urticae.
Eggs of P. citri are reddish, while those of T. urticae are white. To test whether the reddish color invest UV resistance in eggs, pale white eggs of albino strain and reddish eggs of wild type of P. citri were simultaneously irradiated by UV-B. Reddish eggs of wild type showed higher resistant to the UV-B irradiation than the pale-white eggs of albino strain. For the next step, the wild type and the albino eggs of P. citri under UV opaque film (-UV) or under UV transparent film (+UV) were exposed to solar irradiation. Hatchability of albino eggs was signifycantly lower than that of wild type eggs under the + UV condition. Under UV –, both eggs showed higher hatchability than under UV+ conditions. From these results, the wild type eggs of P. citri were likely to have some mechanism protecting itself from ambient UV irradiation, and the red pigment possibly act as a shield againt solar UV radiation.
F1 female proceeded by reciprocal crosses between the wild type and the albino strain exhibited reddish body color in their adult stage, indicating that the albino was recessive. Notwithstanding, F1 eggs from the crosses between albino female and wild type male were pale white. After hatching, the F1 larvae also exhibit pale white body color, then the females turned to reddish at their first quiescent stage. F1 progeny from the crosses between wild type female and albino male were all reddish during egg to adult stage. These revealed that the color of eggs and also larvae were conferred as a maternal effect (: eggs of P. citri might be protected by the red pigments as the first gift of their mother.)
In Jeju cirtrus orchards, Panonychus citri, citrus red mite is the most important pest requiring 3 times acaricide sprays. In open field conventional orchards, P. citri usually shows three population peaks; from end of Jun to July, from end of Aug. to Sep., from end of Oct. to Nov. However, natural enemy complex and its function regulating P.citri are poorly understood.
From the survey of P.citri natural enemy in citrus orchard in Jeju from 2004 to 2006, predatory beetle, Oligota spp. was most abundant. Three predatory mite, N. californicus, Amblyseius eharai, and N. barkeri, were identified. Among them, the predatory mite, Neoseiulus californicus (McGregor) (Acari: Phytoseiidae), was first found in Korea. Even though it was first found, N. californicus was the dominant species occupying 84% of phytoseiid mites. These predatory mites mostly occurred in Jun and peaked at July, which was accorded with the high humidity season of the year. From the survey, the density relationship with P. citri was unclear.
From the study conducted in 2005-2007, N. californicus was more abundant in greenhouse citrus (var. Shirahuhi) than in open field orchards; conventional or organic mandarine citrus. In greenhouse citrus, phytoseiid mites showed suppressing P. citri population. As a next step, the inundative biological control study was conducted using commercial strain of N. californicus, which was originated from Jeju, 2005, in greenhouse citrus. One thousand N. californicus per 1a were released 2 times at 10 day interval on citrus leaves when the initial density of P. citri was about 0.2 per leaf. The release effects were variable depending on the field condition. However, N. californicus did successfully reduce P. citri in greenhouse citrus orchards.
귤응애 성충의 표본추출법을 개발하기 위하여 2개년(2001-2002년)동안 8월부터 11월까지 잎과 열매에 대해 각 조사일에 평균밀도를 조사하였다. 잎과 열매에서 귤응애 성충 밀도의 관계를 시기별로 비교하였으며, Taylor's power law (TPL)와 Iwao's patchiness regression (IPR)을 이용하여 분산지수를 비교하였다. 잎(X)과 열매(Y)에서 귤응애 성충 밀도의 관계는 ln(Y+l)=1.029 ln(X+l)(=0.80)의 직선적인 관계가 있었으며, 열매가 성숙될수록 잎보다 열매에서 귤응애 밀도가 높아지는 경향이었다. 잎과 열매의 표본조사에서 TPL이 IPR보다 평균-분산 관계를 더 잘 나타내었으며, TPL의 기울기와 절편은 두 표본단위간, 연도간에 차이가 없었다. 정착도 0.25일 때 요방제밀도 2.0, 2.5와 3.0에서의 의사결정을 위한 표본추출조사법을 개발하였으며, 조사에 필요한 최대조사나무수(고정표본크기에서 필요 나무수)는 각각 19, 16과 15주였으며, 이 때의 의사결정임 계값 은 각각 554,609와 659였다.,609와 659였다.
국내 자생종인 긴털이리응애(Amblyseius womersleyi Schica)와 도입종인 Amblyseius fallacis Garman, Typhlodromus occidentalis Nesbit 등 3종 이리응애의 감귤원 귤응애에 대한 포식능력과 피식자에 대한 밀도억제효과를 사육실(, RH 65%, 16L : 8D)에서 조사한 결과는 다음과 같다 귤응애 알에 대한 포식량은 A. fallacis 20: 1개, 긴털이리응애와 T. occidentalis는 각각 1.1개와 9.1개였다. 귤응애 유충과 제1약충 그리고 제2약충에 대해 긴털이리응애는 각 23.8, 16.5, 9.0마리, A. fallacis는 각 26.1, 18.2, 7.4마리 그리고 T. occidentalis는 각 7.8, 4.2, 4.2마리를 포식하였다. 20:1의 비율에서 귤응애 밀도억제효과는 긴털이리응애와 A. fallacis는 효과가 좋았으나 T. occidentalis는 효과가 매우 낮았다. 에서 알에서 우화까지의 발육기간은 긴털이리응애는 6.1일, A. fallacis는 5.9일로 두 종간에 유의차는 없었다
제주지역의 온주밀감 과수원에서 귤응애(Panonychus citri (McGregor)) 밀도에 대해 잎당 응애수를 2년에 걸쳐 조사하였다. 이항표본조사법은 잎당 귤응애의 밀도와(m)와 귤응애가 T마리보다 많이 존재하는 잎의 비율()과의 관례를 기본으로 하며, T는 경험적 이항분포모형 []에서의 tally threshold로서 본 연구에서는 1, 3, 5, 7을 사용하였다. 표본단위 수의 증가는 T와 관계없이 이항분포 모형의 정확도에 영향이 거의 없었던 반면에 T는 값이 증가함에 따라 표본수를 증가시켜도 정확도가 낮아졌다. 이항분포모형의 정확도는 T=1일 때 가장 높았으며, 최적의 tally threshold인 것으로 나타났다. 또한, 이항표본조사의 유효성을 조사하기 위하여 독립된 표본을 추출, 조사하였으며, 그 결과 온주밀감원에서 귤응애 밀도추정에는 T=1인 경우가 적합한 것으로 판단되었다. 또한, 귤응애 밀도를 분류하기 위한 이항표본조사과정을 개발하여 action threshold가 귤응애 밀도가 잎당 2마리일 때의 이항표본조사 프로그램을 작성하였다.
귤응애의 예찰방법을 개발하기 위하여 제주지역의 온주밀감원에서 귤응애 분산형태에 대해 2개년(1999~2000년)에 걸쳐 잎 표본에 대하여 각 조사일에 평균밀도를 조사하였다. Taylor's power law와 Iwao's patchiness regression을 이용하여 분산지수를 비교하였으며, 잎 표본 조사에서는 일반적으로 Taylor's power law가 Iwao's patchiness regression보다 평균-분산 관계를 더 잘 나타내었다. Taylor's power law의 기울기와 절편은 조사한 포장 간에 차이가 없었으며, 여기에서 얻어진 상수값을 이용하여 잎 표본 조사에 의한 귤응애 약 .성충에 대한 고정정확도수준에서의 표본조사법을 개발하였다. 이 조사법에 대해 resampling 기법을 이용하여 독립된 4개의 조사자료를 이용하여 분석한 결과 실질 고정정확도(D)값이 요구되는 D값보다 항상 낮았으며, 나무당 귤응애 밀도가 8마리 이상에서 필요한 조사 나무수는 18주보다 작았다.
