        This paper presents a study of the port logistics activities at the port of Santos (Brazil). The study follows a qualitative approach and it is based on in-depth interviews with some key actors from Santos port logistics chain. Based on these interviews, the main dysfunctions and improvement opportunities associated to the container port logistics processes at Santos were identified. The results show that the main dysfunctions are related to the existing information flows. Due to this, a new information flow related to the studied port is proposed and some probable results of the implementation are identified. The findings contribute both to the studied port and to the academic community, as the number of studies addressing port logistics activities is still limited.

        To date, most serodiagnostic methods for brucellosis screening are based on antibodies against lipopolysaccharides of Brucella spp. However, this approach has the drawback of yielding false-positive results due to cross-reactivity with lipopolysaccharides of other related pathogens, especially Yersinia enterocolitica O:9. In this study, Brucella abortus AspC was cloned and expressed by PCR amplification into a pCold TF expression system to obtain recombinant AspC (rAspC). The immunogenicity of rAspC was confirmed by western blotting of Brucella-positive bovine serum. rAspC-based ELISA was performed to determine whether rAspC could be used in the serodiagnosis of bovine brucellosis. rAspC reacted strongly with anti-Brucella antibodies in positive sera in the tube agglutination test (TAT), but did not show strong reaction with most negative samples. In particular, the average OD492 value at the highest TAT titer showed a 1.4-fold increase with respect to the cutoff value. The accuracy, specificity, and sensitivity of rAspC were 71.88%, 78.33%, and 68%, respectively. These findings suggest that rAspC might be valuable for the serological diagnosis of bovine brucellosis.

        Dow AgroSciences has a long history of proactive insecticide resistance management efforts. In our experience, the key to managing resistance is to reduce selection pressure on any one mode-of-action by convincing farmers to rotate among effective insecticide products with different modes of action and to use only the number of insecticide applications required for effective Integrated Pest Management (IPM). To accomplish this, farmers may need to use non-chemical control methods and to rotate to insecticide products that are effective but may not provide the highest levels of control. Integrating rotation of effective insecticides with other IPM techniques can provide high quality and quantity of the harvested crop. We will share our experience managing spinosyn resistance with a series of best management practices for western flower thrips (Frankliniella occidentalis Pergande) and our resistance management recommendations for ISOCLASTTM (sulfoxaflor), our new insecticide for control of sap-feeding insect pests. Dow AgroSciences is an active member of the IRAC (Insecticide Resistance Action Committee) and strongly supports the placement of IRAC mode-of-action group numbers on insecticide product labels. This simple approach greatly facilitates effective product rotation. Resistance management is critical to maintaining the effectiveness of the current arsenal of conventional insecticides and transgenic insect-resistant crops for as long as possible. Responsibility for resistance management is shared between manufacturers, formulator-distributors, retailers, influencers (universities, government), and farmers.         Brucellosis is an important and re-emerging zoonotic disease worldwide. The prevention of human infection is achieved predominantly through the control of brucellosis in agricultural animals, which in turn depends on accurate diagnosis and vaccination. However, conventional serological diagnosis of brucellosis has several limitations, and currently available vaccines for animals have several drawbacks, including the ability to cause infection in humans. Phosphoglycerate kinase (Pgk) is one of the specific proteins reactive with mouse sera in the early stage of Brucella infection, and deletion of the pgk gene in B. abortus strain 2308 resulted in extreme attenuation of this strain in vitro and in vivo. Furthermore, the B. abortus pgk mutant has been used as a live vaccine, and in challenge experiments, it induced protection that was superior to that conferred by commercial strains. In this study, the pgk gene from Brucella abortus 544 was successfully amplified and cloned into a maltose binding protein fusion protein expression vector (pMAL). The recombinant protein was expressed in Escherichia coli DH5α and purified. The immunogenicity of purified recombinant B. abortus 544 Pgk (rPgk) was evaluated by western blot analysis using Brucella-positive mouse sera. rPgk could be used as an antigenic component for future serological tests and potential vaccine development.

        Although ex situ target strength (TS) measurements using dual- and split-beam systems have become the primary approach of estimating fish abundance, theoretical model estimation is a powerful tool for verifying the measurements, as well as for providing values when making direct measurements is difficult. TS values for 20 samples of live bambooleaf wrasse (Pseudolabrus japonicus) whose target length (TL) ranged between 13.7 and 21.3 cm were estimated theoretically using the Kirchhoff-ray mode model, and the TS values for 18 live fish samples were additionally measured at ~0° tilt angle to the swimming aspect using a tethered method at a frequency of 120 kHz to verify the theoretical values. The digitizing intervals used to extract the fish body and swim bladder morphology in the X-ray photographs significantly affected the calculated TS patterns, but variations based on the speed of sound and density ratio values for the general range of fish flesh were relatively small (within 1 dB). Close agreement was observed between the measured and theoretical TS values, and the correlation between the average TS and body length of the fish could be calculated accurately as <TS120kHz>= 20logTL (cm) –71.6 using the theoretical method.

        Brucellosis is a notorious zoonotic disease with global implications. Efforts to control the spread of the disease have been restricted to the agricultural livestock. Increasing incidences of accidental human infection have motivated researches to start working on alternative vaccines. At present, live attenuated vaccines are the only accepted type of vaccines used in developed countries for the prevention of brucellosis. Although serodiagnosis is occasionally unreliable, some countries have already claimed to have eradicated the disease, based on this testing. Live attenuated vaccines are not suitable for use in pregnant and immune-depressed animals. Moreover, these vaccines are not tolerated in humans. Therefore, many researches have been striving to discover alternative methods of vaccination. Most research has focused on the generation of subcellular, subunit, and DNA vaccines that are as efficient as the live attenuated vaccines. At present, none of the available vaccines has been able to replace the live attenuated vaccines. Therefore, additional research is necessary in order to discover a new brucellosis vaccine that is suitable for human use.

        Brucellosis is an important bacterial zoonotic infection worldwide responsible for economic losses in livestock industries which represents a considerable and increasing public health burden. The disease is caused by Brucella which possess the ability to invade and replicate within phagocytes and may result to chronic infection, leading to difficulty in medical therapy of the disease. The treatment for brucellosis employs conventional principles that have been applied for a long time and a combination of antibiotics is currently used due to low efficacy of monotherapies. However, vital effects with respect to health and safety are neglected. Nevertheless, the preventive methods and treatments for brucellosis using traditional medicine have not yet been thoroughly studied, hence, alternative therapies such as the use of natural plant extracts as traditional medicine that are safe, efficient and economical should be explored in order to identify candidates that eliminate complications due to brucellosis. Coptis chinensis Franch (Huanglian) is a traditional Chinese medicinal herb and its extract has been known to possess strong antibacterial activity. In this study, the antibacterial effects of C. chinensis Franch extracts, and the major components of the herb namely berberine and palmatine, were investigated on B. abortus. The C. chinensis Franch ethanol extracts (CCFE) showed bactericidal effects at 1,000 μg/ml concentration and berberine at 100 μg/ml concentration. However, C. chinensis Franch and its components did not affect invasion and intracellular growth of B. abortus in RAW 264.7 cells. In conclusion, C. chinensis Franch ethanol extracts, water extract and its major components such as berberine and palmatine would be a beneficial antimicrobial agent without affecting phagocytic pathway within macrophages, and further study for the precise mechanisms of antibacterial effect would be necessary.

        Brucella spp. are facultative intracellular pathogens that have the ability to survive and multiply in professional and nonprofessional phagocytes and cause abortion in domestic animals and undulant fever in humans. Brucella species can survive in a variety of cells, including macrophages and their virulence and chronic infections are thought to be due to their ability to avoid the killing mechanisms within macrophages. Inhibition of phagosome-lysosome fusion has been proposed as a mechanism for intracellular survival of Brucella in professional and nonprofessional phagocytes. Toll-like receptors (TLRs) are part of a skillful system for detection of invasion by microbial pathogens. Recognition of microbial components by TLRs triggers signaling pathways that promote expression of genes and regulate innate immune responses. Recent studies for the interaction between TLRs-Brucella have indicated the importance of control of Brucella infection. Here, we review selected aspects of TLRs-Brucella interaction, which may be helpful to understanding the mechanism of Brucella pathogenesis.

        Brucellosis is an important bacterial zoonosis in humans and domestic animals. Brucella spp. are taken up, and survive within non-professional and professional phagocytes. In common belief, diabetes mellitus increases susceptibility to pathogenic infection. In this study, Brucella (B.) abortus was inoculated into a diabetic animal model, db/db mice, in order to show the course of brucellosis in diabetic state. The liver proliferation, bacterial burden of the liver, level of cytokines in serum and macrophage migration into liver, were investigated at 14 days post-infection. In comparison with the uninfected control mice, the results revealed that the weight of the liver of infected db/db mice was higher but with lower bacterial load in this organ. The level of MCP-1 mRNA expression in the liver was lower, the levels of IL-12p70, IL-10, TNF-α and IFN-γ in serum was significantly higher and the macrophages migration was significantly lower in infected mice than in the control group. In conclusion, this present study suggested that MCP-1 suppression by B. abortus infection may inhibit the macrophages migration, and consequently may induce to abrogate the bacterial survival in db/db mouse liver. Furthermore, the increased inflammatory cytokines may contribute to inhibition of B. abortus proliferation in diabetic mice.

        Feedback from accreting BH (AGN) is thought to be responsible for the co-evolution of BHs and galaxies. It is likely to be prominent in the most luminous dust-obscured quasars, particularly those containing radio sources too luminous to be powered by starbursts. In order to investigate the feedback mechanism in detail, we select a unique sample containing ~ 200 of the most luminous obscured QSOs by cross-matching the WISE catalog with the FIRST and NVSS radio surveys. We present overall statistics for the observed range of colors and radio/mid-IR flux density ratio. We also present our efforts to understand the physical and evolutionary nature of these extreme feedback candidates using various telescopes such as Magellan, SOAR, Herschel, and ALMA.

        This article reports a case study on the impacts of an interactive, culturally-sensitive computer-based learning environment on at-risk teenage students' motivation and learning of American history in an alternative school in the United States of America. Hip-hop culture combined with interactive software and digital audio workstations was applied to motivate at-risk teenagers and to promote active and effective learning. Data were collected through interviews, archived multimedia data, learning artifacts, teacher’s notes and reflections. Multiple positive impacts were found on both learners’ motivation and learning outcomes. Implications on practice and research were discussed, together with suggestions for educators, researchers and practitioners.

        Interferon-tau (IFNT) is regarded, generally, to be the conceptus protein that signals maternal recognition of pregnancy in ruminants. Although the discovery was made over two decades ago, the molecular mechanisms that regulate IFNT expression are not well understood. Previous studies demonstrated that transcription factors, caudal-related homeobox- 2 (CDX2), JUN, ETS2 and a transcriptional coactivator CREB binding protein (CREBBP) positively influenced IFNT gene expression, while OCT4 may exhibit negative regulation. We and others have observed that both CDX2 and OCT4 coexist during early stages of conceptus elongation but as development proceeds, OCT4 expression diminishes. The objective of this study was to evaluate the stimulatory and inhibitory effects of CDX2 and OCT4, respectively, on IFNT gene transcription when evaluated with other transcription factors. Human choriocarcinoma JEG3 cells were co-transfected with an ovine IFNT (-654 base pair)-luciferase reporter (-654-oIFNT-Luc) construct and several transcription factor expression plasmids. When the reporter construct was co-transfected with either CDX2, ETS2 or CJUN, transcription of -654-oIFNT-Luc increased about two-fold compared to -654-oIFNT-Luc alone. When -654-oIFNT-Luc was co-transfected with both c-jun and Ets-2, activity of -654-oIFNT-Luc was increased about four-fold; cotransfection with JUN, ETS2 and CDX2 increased -654-oIFNT-Luc expression 12X, indicating that the stimulatory activity of the transcription factors was additive. OCT4, when cotransfected with -654-oIFNT-Luc, reduced expression of the later about 40% when compared to -654-oIFNT-Luc alone. When co-transfected with JUN and/or ETS2, OCT4 abolished the stimulatory effect of these transcription factors. OCT4 also inhibited the stimulatory activity of CDX2 alone, but not when CDX2 was combined with JUN and/or ETS2. Therefore, when combined with the other transcription factors, CDX2 over the transcriptional inhibitory activity of OCT4. Conversely, when cells were transfected initially with OCT4 (0h) followed by transfection with CDX2, ETS2 and JUN 24 h later, -654-oIFNT-Luc expression was reduced to control (-654-oIFNT-Luc, alone) levels. Not surprisingly, 12S E1A, an inhibitor of transcriptional coactivator CREBBP, reduced stimulation of -654- oIFNT-Luc expression by CDX2, ETS2 and JUN, in combination, by about 40%, indicating that proper transcription complex formation is required for maximum expression. In conclusion, it is suggested that prior to conceptus elongation, pre-existing OCT4 may inhibit IFNT expression, but as elongation proceeds, IFNT expression increases, resulting from incremental increases in CDX2 expression, diminished OCT4 expression, and possibly proper transcription factor complex formation.         The necessity of conditional gene expression in pigs for transgenic models is raised. Thus, in this study, Cre-loxP conditional expression in porcine fetal fibroblasts was investigated and the transformed fibroblasts were reprogrammed in enucleated oocytes for further early embryonic development. Fetal fibroblasts from miniature pigs were used for transfection with pCALNL-DsRed including floxed neomycin resistant gene and selected with 750 ug/mL neomycin for two weeks. The transfected cells did not express DsRed under fluorescence microscope. After transient transfection of plasmid DNA expressing Cre, the fibroblasts began to express DsRed. The cells expressing Ds- Red were employed into somatic cell nuclear transfer (SCNT). A total of 121 oocytes were used for SCNT and 76 cloned embryos (62.8%)were cleaved. Six blastocysts were grown up after SCNT and expressed DsRed. Deletion of floxed neomycin resistant gene was confirmed by RT-PCR in cloned blastocysts. Taken together, this study demonstrated that Cre-loxP recombination in miniature pig fibroblasts were successfully worked and those sequential transformed cells were developed into pre-implantation stage via SCNT.
