        본 연구는 우리나라에 자생하고 있는 팥배나무 11개 집단으로부터 잎의 형태적 특성과 변이를 조사하여 조경수로서의 팥배나무 선발 및 육종에 대한 기초자료를 제공하고자 수행하였다. 잎의 형태적 특성 12개를 조사하고 집단 간 차이를 비교한 결과 11개 특성에서 유의적인 차이가 인정되었다. 특히 잎의 길이와 너비, 엽면적 등 잎 크기와 관련된 인자들은 마니산 집단이 모두 상위그룹에 속하여 비교적 큰 경향을 나타냈으나, 백운산과 두륜산은 하위 그룹으로 분류되어 비교적 작은 경향을 나타냈다. 주성분 분석을 실시한 결과 제1 주성분은 29.9%의 설명력이 있으며 엽면적, 잎의 너비, 잎의 길이 순으로 높은 상관을 나타내어 잎의 형태적 차이를 설명하는 주요 요인으로 나타났고, 제5 주성분까지 72.9%의 누적 설명력을 나타냈다. 제5 주성분까지의 득점치를 새로운 변량으로 하여 유집분석을 실시한 결과 Ⅰ그룹인 광교산과 안면도 집단, Ⅱ그룹인 덕유산 등 3집단, Ⅲ그룹인 발왕산 등 3집단, Ⅳ그룹인 가지산과 축령산 집단, Ⅴ그룹인 마니산 집단 등 5개 그룹으로 분류되었고, 지리적으로 인접 집단 간의 구분은 명확하게 이루어지지 않았다.

        Background : Kalopanax pictus is one of the most popular wild vegetables in Korea. In recent years Kalopanax pictus has been developed which has no thorns that can be efficiently harvested. Kalopanax pictus, now without thorns, breeds only by root cutting. This makes the price of ineffective and invisible Kalopanax pictus high. Therefore, tissue culture is necessary for efficient mass proliferation of visibly absent Kalopanax pictus. This study was conducted to investigate the effect of anti-browning treatment on tissue culture of thornless Kalopanax pictus. Methods and Results : Thornless Kalopanax pictus young leaves were cut into 5 ㎜ in diameter and cultured for 1 month at 2,4-D 1 ppm + TDZ 0.1 ppm + 3% sucrose. Add 0.05%, 0.1%, 0.15% and 0.2 ㎎/ℓ of browning inhibitor such as ascorbic acid, citric acid and activated charcoal to the MS + 2,4-D 0.2 ppm + 5% sucrose medium. And cultured at 25℃ for 24 hours Light condition for 1 month. In order to determine the optimum concentration of browning agent, characteristics such as callus size, adventitious shoot formation rate and browning degree were investigated. As a result, the callus size of ascorbic acid 0.05 ㎎/ℓ was the largest at 2.26 ± 0.4 ㎝ and the adventitious shoot formation rate was the highest at 16.4%. The degree of browning was 40.30 ± 4.92 in L value and 5.00 ± 2.23 in b value. Conclusion : When ascorbic acid 0.05 ㎎/ℓ is added to the secondary culture medium, the size of the callus increases and the degree of browning decreases. Using the above-mentioned results, we can establish a mass proliferation system through tissue culture of Thornless Kalopanax pictus.

        Background : For the green approach of nanoparticles synthesizing, plant based technology has been considered as cost-effective and eco-friendly mass production. The oriental medicinal crop, Kalopanax septemlobus (Thunb.) Koidz. (Korean name: 음나무), the deciduous tree and a family of Araliaceae. Endemic tree of Asian countries, K. septemlobus being used for the treatment of various diseases. Phytochemicals of K. septemlobus such as polyphenols has highly probability of reducing agent for biosynthesizing nanoparticles. Methods and Results : In this study, we applied K. septemlobus ZnO nanoparticles (Ks-ZnO NPs) with procedures including green approach one-pot synthesis. For the characterization of nanoparticles, UV–Vis, FTIR, XRD, SEM and TEM were used. The formation of ZnO nanoparticles, the aurface plasmon resonance were observed at 372 ㎚ in UV-Vis spectroscopy. The presence of functional groups which as a capping agent and formation of ZnO nanoparticles were confirmed in FTIR result. The crystallization and morphology showed by XRD, TEM and SEM respectively. The photocatalytic activity of ZnO nanoparticles, was determined using Methylene blue (MB) dye degradation under UV irradiation (365 ㎚) which resulted rate constant is (−k) 0.1215 with 97.5% of degradation in 30 min. Conclusion : The result shows that phytochemicals in K. septemlobus extract have a potential as a reducing agent to form ZnO nanoparticles. The ZnO NPs are capable to degrade MB with in brief time.

        Background : Oplopanax elatus Nakai. is distributed in Korea and China. In this study, we have used high performance liquid chromatography (HPLC) to compare the internal standards contents [uracil, adenosine, protocatechuic acid, syringin (eleutheroside B) and scoparone (6,7-dimethoxycoumarin)], and compared the antioxidant activity. Methods and Results : Samples were prepared two different temperature conditions (90℃ and 100℃). Total phenolic contents and total flavonoid contents were analyzed while gallic acid and quercetin were used as standard. Anti-oxidant activities were measured by determination of DPPH and reducing power assay. HPLC was reported as five standard compounds equivalent using the following linear equation based on the calibration curve. According to the results, the anti-oxidant effects of Korean O. elatus Nakai. stem extracts in 90℃ water showed more activity than that of Chinese in DPPH assay. However, the amount of internal compounds was higher in Chinese O. elatus Nakai.. The anti-oxidant effects of Korean O. elatus Nakai. stem extracts in 90℃ water showed more activity than Korean O. elatus Nakai. stem extracts in 100℃ water in DPPH assay. In this study, we had found that, at over the 100℃ temperature all the anti-oxidant effects of O. elatus Nakai. extracts were reduced. However, all five standard compounds were detected at similar value. Conclusion : These results suggests that Korean O. elatus Nakai. has higher anti-oxidant activities which can be use for bioactivity assay.

        Background : Hippophae rhamnoides L. are known for antioxidant, immunodeficiency, skin protection, influenza infection and prevention of heart disease. This study was carried out to confirm the possibility of functional food by changing the antioxidant effect using H. rhamnoides L. leaf extracts to the Gamju (sweet rice drink). Methods and Results : A total of 12 samples were made of different processes. Briefly, the H. rhamnoides L. leaf were extracted at 60℃ in two different conditions (EtOH 100%, water 100%). Gamju was fermented into three different koji (Aspergillus oryzae – red, yellow, black). In addition, The addition of H. rhamnoides L. leaf extracts were mixed in two ways (simultaneous saccharification, mixed after saccharification). Antioxidant activities were estimated by 2,2-diphenyl-1-picryl-hydrazil (DPPH) and reducing power assay. Total phenolic content (TPC) was determined by Folin-Ciocalteu method. In this study, we found that Gamju mixed with H. rhamnoides L. leaf increased antioxidant effects and TPC than the control (original Gamju). Moreover, the anti-oxidant effects of the mixed H. rhamnoides L. leaf with Gamju after saccharification exhibited more activity than simultaneous saccharification in DPPH assay. Conclusion : These results demonstrated that samples of added to the H. rhamnoides L. leaf could be use as functional food.

        Background : Cornus kousa fruit has been used as a medicine for a long time. Therefore, for the purpose of utilizing C. kousa fruit as a health functional food, antioxidant activity was tested by extracting ethanol concentrations to identify optimal extraction conditions. Methods and Results : C. kousa fruits used in this study were washed, dried and pulverized. Then, the samples and solvents by ethanol concentration (0, 20, 40, 60 80 and 100%) were extracted at 80℃ for 90 minutes at 1 : 20 ratio, respectively. Afterwards, it was filtered, concentrated, frozen and dried. Total phenol and flavonoid content, DPPH and ABTS radical scavenging activity, ferric reducing antioxidant power and oxygen radical absorbance capacity were measured. The total phenol and flavonoid content is the highest in 60% ethanol extract at 20.08 ㎎·GAE/g, 0.86 ㎎·QE/g. In addition to all antioxidant studies showed the highest activitiy in 60% ethanol extract. Also, when HepG2 cells were treated with ethanol extract at 200 ㎍/㎖ concentrations, the survival rate was more than 90% and cytotoxicity was not observed. Then, HepG2 cells were treated with tBHP to induce oxidative stress, and the cytoprotective effect and inhibitory activity of ROS production were measured. As a result, 60% ethanol extract showed the best activity. Conclusion : When extracting C. kousa fruits with ethanol as solvent, It is expected best to use 60% ethanol to maximize potential as a material for health functional foods.

        Background : In this study, we investigated the formation of Abeliophyllum distichum callus which has not been reported until now and the culture optimization. The potential of callus cultures was examined by comparing the components and antioxidant activities of callus and wild planting. Furthermore, it was intended to provide data on the possibility of substitution for the active ingredient production of Abeliophyllum distichum callus extraction. Methods and Results : We tried to induce callus by introducing it in vitro culture by using leaves and stem of Abeliophyllum distichum grown in Goesan Chungbuk. In order to investigate the effect of growth regulators on the callus, TDZ, NAA, 2,4-D, IBA and BAP were treated with 0.25 to 5 ㎎/ℓ of single hormone, and the combination hormones were treated with two concentrations of BAP (0.1, 1.0 ㎎/ℓ) and 2,4-D, NAA, IBA at 0.1 ㎎/ℓ and 1.0 ㎎/ℓ. And investigate the effect of inorganic salt concentration on somatic embryogenesis, the incidence of callus was examined by culturing 1X, 1/2X MS medium for 4 weeks. As a result, the effect of growth regulator and treatment concentration on the callus formation of Abeliophyllum distichum, callus induction was the fastest in the 2,4-D condition, but the callus indection was slow. The highest amount was produced ine the NAA condition. The axillary bud was best grown in TDZ condition, but no root was formed. LC chromatogram was able to compare the contents of leaf, stem extracts and callus extracts. Antioxidant ativity showed excellent antioxidative activity in the extracts of the wild planted stem. Conclusion : The optimum culture conditions for callus were 1/2X MS, 1 ㎎/ℓ BAP and 0.1 ㎎/ℓ of 2,4-D. As a result of LC chromatogram, it was confirmed that callus extract contained a large amount of the same components as the stem and leaf extracts grown on the wild planting. Currently, Studies on mass culture using bioreactor to utilize this are underway, We could confirm the potential possibility of Abeliophyllum distichum cultured products.

        Background : Oplopanax elatus has many compounds such as essential oils, saponin, flavonoids, anthraquinones, and polyacetylenes etc. in all part of stems, roots, and leaves. In previous study, we isolated five compounds (uracil, adenosine, protocatechuic acid, syringin, and scoparone) from the water extract of in stems of O. elatus. In this study, we confirmed the variation of chemical constituents and antioxidant activity in leaves of O. elatus by different cultivation environment. Methods and Results : We analyzed three types of O. elatus in different cultivation environment (in vitro plant, in vivo plant and wild plant). We detected five compounds (uracil, adenosine, protocatechuic acid, syringin, and scoparone) in three types of plants by using HPLC. The contents of five compounds varied depending on the different cultivation environment. Syringin and adenosine were detected on all plants and showed different contents, respectively. We compared antioxidant activities such as total phenol contents (TPC), total flavonoid contents (TFC), DPPH and reducing power assay. The values of antioxidant activities (DPPH and reducing power) in leaves of in vitro plants were higher than other plants. Also TPC and TFC in leaves of in vitro plants showed the highest contents. Conclusion : These results could be basic data for cultivation methods about enhancement of syringin and adenosine compounds contents in leaves of O. elatus.

        Background : We examined the inorganic compounds, compressive strength, yield, and functional substances of five steam-treated deciduous tree species to address the problem of the low degradation activity of conventional oak trees in the cultivation of Gastrodia elata in indoor facilities. Methods and Results : With the exception of Ca in cherry blossom tree (CBT), the content of the major inorganic elements (N, P, Mg, K, and Ca) in alternative trees were 1.5 - 3.6, 1.3 - 2.6, 3.3 - 5.3, 4.4 - 7.5, and 0.9 - 3.9 times those in oak (control group), respectively. The content of trace elements (Zn, Cu, Fe, and Mn) were equivalent to or slightly lower than those in oak, depending on tree species. The compressive strength of alternative trees was 9.3%–34.9% lower than that of oak. The compressive strength after steaming was lower in the order CBT (8.0%), mulberry tree (MBT: 7.0%), chestnut tree (CNT: 7.2%), Siebold's crab (SBC: 6.9%), black locust (BLL: 6.1%), and oak (5.4%). The total weight of oak was 893 g in comparison with the 1,107, 902, 952, 813, and 813 g of MBT, CNT, CBT, SBC, and BLL, respectively. The mature tuber and seed tuber ratios were 31%, 34%, 32%, 32%, 34%, and 30%, and 67%, 75%, 68%, 70%, 73%, and 65%, respectively. The yield index increased overall by 1% - 14%. Total weight after steaming increased by 10%, 18%, 14%, 16%, 20%, and 12% in oak, MBT, CNT, CBT, SCB, and BLL, respectively. Mature tuber and seed tuber ratios after steaming increased by 1%, 1%, 0%, 1%, and 2%, and 5%, 3%, 7%, 2%, 5%, and 5%, respectively. There were no significant difference among tree species in drying ratio, hardness, chromaticity for quality comparisons, and steaming. However, the results of functional substance analysis revealed significant differences in gastrodin, gastrodigenin, p-hydroxybenzaldehyde, and ergothioneine according to tree species. Conclusion : From the above results, we may believed that five tree species including MBT might have useful as alternative to oak when the cultivation of G. elata in Indoor Facilities.

        Background : Among medicinal plant sources, Abeliophyllum distichum is widely used in traditional Korean medicine. we report on the synthesis of nanostructured zinc oxide particles by both chemical and biological method. Highly stable and spherical zinc oxide nanoparticles are produced by using zinc nitrate and Abeliophyllum distichum leaf extract. Methods and Results : Zinc oxide (Ad-ZnONPs) nanoparticles synthesized from Abeliophyllum distichum at room temperature by aqueous extract of dried leaf and stem. The plant endemic in Korea alone and it is a monotypic flowering plant genus of olive family, Oleaceae. Catalytic and toxicity effect against human keratinocyte and adenocarcninomic human alveoloar. Ultra violet visible (UV-Vis) spectroscopy, field emission transmission electron microscopy (FE-TEM), energy-dispersive X-ray (EDX) spectroscopy, elemental mapping, X-Ray powder diffraction (XRD), selected area electron diffraction (SAED) and Fourier transform infrared (FTIR) spectroscopy were engaged to illustrate the biosynthesized nanoparticles. The Zn-AdNPs has the ability in catalytic action and the cytotoxicity agent against different cell lines as investigated by 3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium bromide (MTT) assay Conclusion : The present studies reveals that facile approaching the biological synthesis of zinc oxide nanoparticles by using the A. distichum leaf and stem extract, which is revealed that recyclable method. The method is well suited for the green synthesis and dual function molecule as reducing agent and stabilizing agent for synthesis of nanoparticles. The nanoparticles also showing promising biological activities.

        This study was carried out to investigate the environment factors including community structure and soil characteristics in the wild habitats of Cephalotaxus koreana, and offers the basic information for habitats conservation and restoration. Most of the wild habitats were located at altitudes between 148~835 m with inclinations ranged as 12~32°. The average soil pH was 4.7~5.9, soil organic matter was 5.72~15.99%, cation exchange capacity was 14.1~19.9 cmolc/㎏-1 and exchangeable K+, Ca2+, Mg2+ was 0.25~0.48 cmolc/㎏, 0.79~6.68 cmolc/㎏, 0.31~1.73 cmolc/㎏, respectively. The dominant species of tree layer were found to be dominated by Quercus dentata in Jekbo-san (C1), Acer pictum in Bogae-san (C2), Acer pseudosieboldianum in Geumwon-san (C3), Q. serrata in Jiri-san (C4), Zelkova serrata in Baegun-san (C5), and Q. acutissima in Duryun-san (C6). The Species diversity (H’) was 0.854~1.234, evenness (J’) was 0.654~0.993, and dominance (D) was found to be 0.067~0.346. Correlation coefficients analysis based on environmental factors, community structure and value of species diversity shows that growth of Cephalotaxus koreana is correlated with species diversity and evenness. This result show that Cephalotaxus koreana habitats located in mature stands.

        A diarylheptanoid, (5S)-1,7-bis-(3,4-dihydroxyphenyl)-5- hydroxyheptane-3-one-5-O-β-D-xylopyranoside, named oregonin (1), was isolated from the of Alnus japonica (A. japonica), which is a species of the genus Betulaceae, growing throughout Korea, Japan and China. The structure was elucidated by various spectroscopic methods including negative and positive LC/MS, 1H-NMR, and 13C-NMR techniques or by comparison with authentic samples. In order to evaluate the anti-oxidative activities of oregonin (1) isolated from A. japonica, 1,1-diphenyl-2-picryhydrazyl (DPPH) radical scavenging activity and 2,2'-azino-bis[3-ethylbenzothiazoline-6-sulphonic acid] (ABTS) radical scavenging activity were measured in vitro. Oregonin from A. japonica exhibited potent DPPH and ABTS radical scavenging activities. A. japonica shows not only 1,1-diphenyl-2-picryhydrazyl (DPPH) radical scavenging activity and ABTS radical scavenging activity, but also apoptosis modulative effects. The present results indicate that A. japonica could be a hair-growth-promoting agent for cosmetic products.

        In this study, a preliminary evaluation of the antioxidant and anti-inflammatory activity of the Ficus erecta var. sieboldii (Miq.) King (FES) leaf extract has been performed to assess its potential as a natural resource for food and medicinal materials. FES was extracted using 70% EtOH and then fractionated sequentially using n-hexane, CH2Cl2, EtOAc, and n-BuOH. To screen for antioxidant and anti-inflammatory agents effectively, the inhibitory effect of the FES extracts on the production of oxidant stresses (DPPH, xanthine oxidase, and superoxide) and pro-inflammatory factors (NO, iNOS, COX-2, PGE2, IL-6, and IL-β) in the murine macrophage cell line RAW 264.7 activated with lipopolysaccharide (LPS) was examined. Among the sequential solvent fractions of FES, the CH2Cl2 and EtOAc fractions showed decreased production of oxidant stresses (DPPH, xanthine oxidase and superoxide), and the hexane and CH2Cl2 fractions of FES inhibited the production of pro-inflammatory factors (NO, iNOS, COX-2, and PGE2). The CH2Cl2 fraction also inhibited the production of pro-inflammatory cytokines (TNF-α, IL-6, and IL-1β). These results suggest that FES has a significant effects on the production of oxidant stresses and pro-inflammatory factors and may be used a natural resource for antioxidant and anti-inflammatory agents.

        This study was conducted to compare anti-inflammatory effect of Robinia pseudoacacia L. using different extraction methods (water extraction, ethanol extraction and high temperature extraction). We investigated anti-inflammatory effect of Robinia pseudoacacia L. extract (RP1, water extract; RP2, ethanol extract; RP3, high temperature extract) on lipopolysaccharide (LPS)-stimulated inflammation using Raw 264.7 cell. Cells were treated with various concentrations (12.5, 25, 50, 100 or 200 ㎍/㎖) of water extract, ethanol extract and high temperature extract. Cytotoxicity was not observed on Raw 264.7 cells, LPS-stimulated production of NO (nitric oxide), PGE2 (prostaglandin E2) and cytokines (TNF-α, IL-6 and IL-1β) was reduced by RP3 treatment more than RP1 and RP2. In conclusion, these results indicated that inflammation on Raw 264.7 cells was improved by RP3. Treatment of RP3 could be used to natural medicine for improving inflammatory response. However, further experiment is required to observe how the high temperature extraction at 500℃ for 48 h influences on alteration of active ingredient in Robinia pseudoacacia L., and conducts the inflammation signal pathway on Raw 264.7 cells.

        Background: A soil-borne pathogenic fungus, Ilyonectria radicicola (Cylindrocarpon destructans) causes root rot on ginseng (Panax ginseng C. A. Meyer) and is known to attack many other plants. The Nectria/Neonectria radicicola complex has been renamed as the I. radicicola complex after analysis of its multi-gene relatedness and morphological characteristics. The fungi in this complex have been reclassified into 16 species under the genus Ilyonectria based on characteristics analysis Methods and Results: To obtain useful data from the Korean ginseng root rot, I. radicicola was isolated from the rhizosphere soils of the chestnut tree. They were identified through a pathogenicity test and a survey of the morphological features. The existence of I. radicicola in soil samples was confirmed by PCR detections using nested PCR with species-specific primer sets. These were subsequenctly isolated on semi-selective media from PCR-positive soils. Genetic analysis of the I. radicicola complex containing these pathogens was done by comparing the DNA sequences of the histone h3 region. These isolates originating from the rhizosphere soils of chestnut constituted a clade with other closely related species or I. radicicola isolates originating from ginseng or other host plants, respectively. Additionally, the pathogenicity tests to analyze the characteristics of these I. radicicola isolates revealed that they caused weakly virulent root rot on ginseng. Conclusions: This is the first study reporting that I. radicicola isolates from chestnut rhizosphere soils can attack ginseng plant in Korea. Thus, these results are expected to provide informations in the selection of suitable fields for ginseng cultivation.

        The study aims to investigate the effect of air temperature on growth date(bud bursting, flowering, blooming) of Citrus Unshiu in Jeju and Seogwipo, and estimate future growth date using air temperature data and growth date of Citrus Unshiu from 1998 to 2015. The trend and relationship between growth date of Citrus Unshiu and air temperature in Jeju and Seogwipo are analysed. As a result, The trend of growth date of Citrus Unshiu is delayed during study period. The correlation between Citrus Unshiu growth date and air temperature is negative. It means that air temperature related to the growth date of Citrus Unshui is decreasing. Future growth date of Citrus Unshiu is estimated to be earlier by future climate change scenarios. Air temperature expect to rise, but it can appear the low air temperature to be able to hurt citrus tree.

        Dendropanax morbifera Leveille (Araliaceae) is an endemic species growing in the south-western part of South Korea that has been used in folk medicine and health functional food. In this study, we investigated an extract of quercetin in Jeju D. morbifera by varying different parts (fruit, sprouts, leaves, sprigs, and branches), harvest times, and extraction solvents. In addition, we aimed to establish a simple and reliable HPLC/UV analytical method to determination of quercetin for the quality control and base line data of the Jeju D. morbifera extract as a health functional food ingredient. The analytical specificity was determined with retention time and photo diode array (PDA) spectrum by analyzing quercetin using HPLC and comparing the results to those of extracts. This analytical method for quercetin was validated for its limit of detection (LOD), limit of quantitation (LOQ), precision, and accuracy. A high linearity in the standard calibration curve was obtained, with a coefficient of determination (R2) of 0.9996. Also, the LOD and LOQ values were found to be 0.28 μg/mL and 0.85 μg/mL, respectively, and the recoveries of quantified compounds ranged from 97.91% to 104.10%. Furthermore, the relative standard deviation (RSD) values of data from the intra- and inter-day precision analyses were less than 1.36% and 3.65%, respectively. As a result, the highest quercetin content among the extracts of Jeju D. morbifera leaves was found to be 20.14 mg/g, which was extracted at harvest in May (cultivation period 10 years) with 60% EtOH. All in all, we believe that the results obtained would be helpful in the development of nutraceutics and natural medicines and for the quality control of D. morbifera.

        Background : Spiraea prunifolia var. simpliciflora (Rosaceae) called “Brial wreath” is a deciduous latifoliate shrub that is widely distributed throughout in Northeast Asia. Phytochemical and biological investigation of S. prunifolia have led to the discovery of biologically active compounds. Pharmacological studies revealed that the extract of the root of S. prunifolia possess antioxidant, antipyretic and anti-inflammatory activities. Some chemical constituents such as sterols, phenolics, terpene and fatty acid, as well as ethanolic extracts from the roots of S. prunifolia, have previously been reported to modulate the deleterious effects of diabetes, to prevent high-fat diet-induced obesity, and to prevent cisplatin-induced nephrotoxicity. Our continuing research was carried out to search for other phytochemical constituents from the leavess of S. prunifolia. The chemical structures of compounds were determined by NMR and FAB/MS spectroscopic data interpretation. Methods and Results : Multiple-preparative liquid chromatography (MPLC) purifications were carried out on YMC LC-forte/R instrument (YMC Kyoto, Japan) equipped with YMC-Pack ODS-A columns (ODS gel : 5 ㎛, 10 ㎜ × 250 ㎜). High-performance liquid chromatography (HPLC) was performed on Agilent Technologies instrument (Aglient Tec., Santa clara, CA, USA) equipped with YMC–Pack Pro C18 columns (ODS gel : 5 ㎛, 4.6 ㎜ × 250 ㎜). Next, quantitative analysis was carried out on UPLC-QqQ/MS 3200 Q-TRAP instrument (AB SCIEX Toronto, Canada) using a ACQUITY UPLC (waters corp.) with an ACQUITY BEH C18 column (2.1 ㎜ × 100 ㎜, 1.7 ㎛). The metabolite samples was applied to preparative reversed-phase HPLC and UPLC using gradient method, solvent A [water + 0.1% formic acid (v/v)] and solvent B [acetonitrile + 0.1% formic acid (v/v)]. Conclusion : In this study, we isolated the major metabolites from the stem of Spiraea prunifolia var. simpliciflora by using MPLC and HPLC. UPLC-QqQ/MS was also used to quantify target compounds. Finally, we established methodology and performed the quantitative analysis on target compounds from the stem of Spiraea prunifolia var. simpliciflora.

        Background : Mistletoe grown in eurasian continent range from England to northern Asia. It has abundant chlorophyll and phenolic compound in node and leaf which has anticancer activity. The purpose of the study is to improve nutritional quality and composition of the amino acids and mineral along with increasing seed set rates and productivity of Viscum album on Prunus mume. Methods and Results : This study was conducted to compare plant nutrients such as total nitrogen, Ca, Fe, K, Mg, Mn, Na, P and composition of amimo acids in leaves between Viscum album and its host tree, Prunus mume, which were artificially cultivated in field and green house with or without treatment of fertilizer. Total nirogen content in leaves of mistletoe and its host tree, Prunus mume was 3.90% and 2.09% in the fields fertilized and non-fertilized, respectively. In a greenhouse fertilized, mistletoe leaves contained higher nitrogen (3.78%) compared to the host tree (3.06%). NO3-N content in leaves was also higher in mistletoe (37.5 ppm & 20 ppm) than in host tree (22.5 ppm & 12 ppm) in a field fertilized and non-fertilized. Ca content (1.18%) were same in leaves of mistletoe and host tree in the field fertilized. Contents of Fe, K, and Mn in leaves of mistletoe were 61.37 ㎎/㎏, 3.33%, and 127.59 ㎎/㎏, which were 16%, 13%, and 48% lower than host tree, respectively. On the other hand, contents of Mg, N, and P were 0.22%, 127.95 ㎎/㎏, and 0.48%, which were 5%, 148%, and 92% higher than host tree, respectively. K contents were higher in host tree fertilized (3.33% in field & 2.55% in greenhouse) while higher K (4.07%) was contained in a field non-fertilized compare to host tree (3.96%). However, P content was higher in mistletoe grown in field (0.48%) and greenhouse (0.42%) fertilized. In non-fertilized field, mistletoe leaves contained higher P (0.65%) rather than in host tree (0.29%). Eighteen kinds of amino acids were investigated in leaves of host plant by 20 - 87%. Conclusion : This study indicates that nutrient management including fertilization is important to improve nutritional quality and productivity of mistletoe in the artificial cultivation on Prunus mume.

        Background : Collagen and elastin contribute to form a network under the epidermis and reduce the wrinkles. Collagen and elastin are degraded by collagenase and elastase. Therefore, inhibition of collagenase and elastase activity could be an excellent method for anti-wrinkle. Pinus Koraiensis leaves contain various flavonoid compounds such as quercetin and kaempferol. According to previous reports, kaempferol has increases procollagen synthesis and inhibitory effect on MMP-1 activity. This study was performed to investigate the anti-wrinkle effect of Pinus Koraiensis leaves extract and fractions. Methods and Results : The active components of Pinus Koraiensis leaves extracts and fractions were determined. Total phenol and total flavonoid contents measured using the Folin-Ciocalteu reagent and according with aluminum chloride colormetric method. The kaempferol content analysis using HPLC system. The total phenol, flavonoid and kaempferol contents of ethyl acetate fraction was the highest. The antioxidant activity measured in according with DPPH, ABTS and RP assay. Pinus Koraiensis leaves extract and factions were the highest ethyl acetate fraction in antioxidant activity. The inhibitory activity on collagenase and elastase of Pinus Koraiensis leaves extract and factions were determined. The results of inhibitory activity on enzymes associated with wrinkle formation was the highest of n-butanol fraction, the next higher was 80% ethanol extract. As a result of using MMP-1 content and procollagen type-I C-peptide content assay kit, the anti-wrinkle effect of n-hexane fraction was highest.
