        By using the Optical Wide-field Patrol (OWL) network developed by the Korea Astronomy and Space Science Institute (KASI) we generated the right ascension and declination angle data from optical observation of Low Earth Orbit (LEO) satellites. We performed an analysis to verify the optimum number of observations needed per arc for successful estimation of orbit. The currently functioning OWL observatories are located in Daejeon (South Korea), Songino (Mongolia), and Oukaïmeden (Morocco). The Daejeon Observatory is functioning as a test bed. In this study, the observed targets were Gravity Probe B, COSMOS 1455, COSMOS 1726, COSMOS 2428, SEASAT 1, ATV-5, and CryoSat-2 (all in LEO). These satellites were observed from the test bed and the Songino Observatory of the OWL network during 21 nights in 2014 and 2015. After we estimated the orbit from systematically selected sets of observation points (20, 50, 100, and 150) for each pass, we compared the difference between the orbit estimates for each case, and the Two Line Element set (TLE) from the Joint Space Operation Center (JSpOC). Then, we determined the average of the difference and selected the optimal observation points by comparing the average values.

        Brassinosteroids (BRs) control virtually every aspect of plant growth and development. BRs act alone or with other exogenous and endogenous signals including auxin and light. To screen for the novel player involved in BR signaling in Arabidopsis, we employed cDNA overexpression strategy. We created a cDNA library to be expressed under the 35S overexpression promoter, and introduced into a weak brassinosteroid insensitive 1 (bri1) mutant. The mutant dubbed bri1-5 with long petiole (blp) was identified to display bigger stature especially in hypocotyl and petiole length relative to bri1-5. Sequence analysis of the rescued transgene revealed that blp consisted of a chimeric DNA consisting of a 3’ half of PHYB, 2 bp insertion, and a part of a chloroplast ribosomal RNA. Re-introduction of chimeric DNA into bri1-5 recapitulated blp phenotype. The blp phenotypes being similar to phyB mutants led us to examine both the PHYB transcript and protein levels in the blp 35Spro:PHYB doubly homozygous line. Lower levels of both transcripts and proteins of PHYB suggested that introduction of the chimeric gene interfered with the stability of PHYB transcripts. Our results highlight that overexpression mutagenesis facilitates functional genomics to decipher a function of Arabidopsis genome.

        Much effort has been expended to find agronomically important QTLs for improving soybean yield. However, the complexity of genome, such as genome duplication, limits the utility of genome-wide association studies and linkage analyses to identify genes controlling yield traits. We propose the variation block method, a three-step process for recombination block detection and comparison. The first step is to detect variations by comparing short-read DNA sequences of the cultivar to a reference genome of the target crop. Next, sequence blocks with variation patterns are examined and defined. The boundaries between the variation-containing sequence blocks are regarded as recombination sites. All the assumed recombination sites in the cultivar set are used to split the genomes, and the resulting sequence regions are named as variation blocks. The practicality of this approach was demonstrated by the identification of a putative locus determining soybean hilum color and known genes such as flower color gene. We suggest that the variation block method is an efficient genomics method for recombination block-level comparison of crop genomes. We expect that this method holds the prospect of developing crop genomics by bringing genomics technology to the field of crop breeding.

        High yield is the most important trait in various agricultural characteristics. Many approaches to improve yield have been tried in conventional agricultural practice and recently biotechnological tools employed for same goal. Genetic transformation of key genes to increase yield is one way to overcome current limitation in the field. We are producing transgenic soybean plants through high efficient transformation method by introducing all gene member with AT-hook binding domain, hoping to obtain manageable delay of senescence. Many transgenic soybean plants are growing in greenhouse and GMO field, and will be evaluated their senescence and any association with yield increase.

        Soybean is a crop of importance economically and nutritionally in many parts of the world. Thanks to many new genes brought from genomic research, It is possible to introduce various candidate genes through genetic transformation to see the performance of the genes in field. In our lab, soybean transformations have been tried for last 10 years to probe the possibility of traits improvement by transformation of new gene into soybean. For this purpose, three different genes were transformed into Korean soybean variety, Kwangan. First, the gene that controls early flowering of plant was transformed into Kwangan. Second, a candidate gene for soybean mosaic virus (SMV) resistance was transformed to produce transgenic plants. Third, another candidate gene for drought tolerance was transformed. All the transgenic plants from three genes transformation were produced for their gene insertion and their expression using PCR, qRT-PCR. Further analysis including harvesting seeds is currently undertaken.

        This experiment was carried out to compare the morphological traits of Korean, Chinese, Japanese and Southeast Asian(SEA) soybeans from RDA genebank. Days to flowering were ranged from 51 to 125 days with an average of 75 days. Those of China were the shortest with an average of 58 days and those of SEA were the longest with an average of 99 days. Growth days were the shortest with 94 days from China, and longest with 188 days from Korea and SEA. The 100 seed weight of soybeans was ranged from 3.4g to 46.4g, with an average 22.2g. The 100 seed weight was the lightest with an average 11.8g from SEA and the heaviest with an average 24.6g from Korea. In growth habit, over 50% of being collected from Korea, Japan and China were erect type, but 94% from SEA were intermediate type. The highest percentage of seed coat color was yellow(66.1%), followed by yellowing green(10.0%). As a result of cotyledon color in 760 black seed was 76.1% with yellow, 23.9% with green. Green cotyledon was much more in Korea(38.6%) and Japan(33.3%) than other countries. One thousand seven accessions from Korea, Japan, China and SEA were analyzed using 7 SSR markers. One hundred eighty alleles were detected with a lowest 16 at the Satt537 and a highest 35 at the Satt390. The average polymorphism information content(PIC) was 0.68, the highest with 0.7 in Japan. Gene diversity was the highest with 0.73 in China and Japan, while the lowest in SEA with 0.68.

        The flower buds of Sophora japonica L (SF), as a well-known traditional Chinese medicinal herb, have been used to treat bleeding-related disorders such as hematochezia, hemorrhoidal bleeding, dysfunctional uterine bleeding, and diarrhea. However, no specific anti-cancer effect and its molecular mechanism of SF have been described. Thus, we performed in vitro study to investigate if treatment of SF affects activating transcription factor 3 (ATF3) expression and ATF3-mediated apoptosis in human colorectal cancer cells. The effects of SF on cell viability and apoptosis were measured by MTT assay and Western blot analysis against cleaved poly (ADP-ribose) polymerase (PARP). ATF3 activation induced by SF was evaluated using Western blot analysis, RT-PCR and ATF3 promoter assay. SF treatment caused decrease of cell viability and increase of apoptosis in a dose-dependent manner in HCT116 and SW480 cells. Exposure of SF activated the levels of ATF3 protein and mRNA via transcriptional regulation in HCT116 and SW480 cells. Inhibition of extracellular signal-regulated kinases (ERK) 1/2 by PD98059 and p38 by SB203580 attenuated SF-induced ATF3 expression and transcriptional activation. Ectopic ATF3 overexpression accelerated SF-induced cleavage of PARP. These findings suggest that SF-mediated apoptosis may be the result of ATF3 expression through ERK1/2 and p38-mediated transcriptional activation.

        This study was performed to investigate the growth characteristics and inorganic components of Codonopsis lanceolata regarding regional differences. The plant height of Japanese Codonopsis lanceolata was 373.6 cm, so it’s revealed that it has more vigorous growth than Korean won. The flowering time of Korean Codonopsis lanceolata was 2 weeks faster than Japanese one. Total fresh weight of root was 41.0 g and 39.0 g for Korean and Japanese respectively, thus, no significance difference was found. However, regarding fresh weight, Korean one had a more fresh weight (35.4 g) of main root parts, but Japanese one had a more fresh weight (9.6 g) of the lateral root part. Each inorganic component was found more in the aboveground parts, regardless of the region and the content of K was the largest. Regarding the content of macroelements for each part of Codonopsis lanceolata, the content of Na, Mg, P, S, and Ca in Korean Codonopsis lanceolata was found the highest on the leaf, followed by stem and root. In the case of Japanese Codonopsis lanceolata, same result was found on the content of Mg and Ca, however, the highest content of Na and P was found in the stem.

        This study was conducted to compare the growth, inorganic components, and proximate components of Codonopsis lanceolata grown in 10 regions of Korea for selecting superior species and breeding by crossing. Among the all tested lines, the shortest plant height (217.12 cm) was observed from the Ulleungdo region line (No. 4) while the longest (273.9 cm) was observed from Hwasun region line (No. 9). In addition, the lines of central and northern region (No. 1~No. 7) tend to have shorter plant height than those of southern region (No. 8~No. 9) except Jejudo region line (No. 10). Flowering tends to be late towards southern region, and lines in central and northern regions were started flowering about 2 weeks earlier than those in southern regions. However, the heaviest root weight was 13.1 g, found in only Jejudo line (No. 10) whereas there was no significant difference found in the other regions which have a range of 8.3~11.0 g. The inorganic components were varied in each line, however, proportion of macroelements, such as K, Ca, and P, was the largest for every line. Especially for Heongseong region line (No. 2), had larger proportion of macroelements than the others. There was a difference of proximate compositions of Codonopsis lanceolata, except the moisture content, among all regions, however, it was generally shown that the content of crude protein (1.31~3.76%) and crude fiber (2.18~3.12%) was the highest.

        This study investigated the effect of NH4 + concentrations on microalgae growth by appling mixotrophic microalgae chlorella vulgaris in order to treat anaerobic digested food waste leachate. The growth rate and final microalgae growth were an order as 400 > 100 > 800 > 1300 mg-N/L. As results, The growth rate and final growth of microalgae were highest at ammonia concentration of 400 mg-N/L, On the other hand microalgae growth was inhibited when ammonia concentrations were over 800 mg-N/L. high concentrations of nitrogen over 800 mg-N/L interrupt the growth of microalgae. All of nitrogen and phosphorus were removed by microalgae at the ammonia concentration of 100~400 mg-N/L. In addition, when ammonia concentration was over 800 mg-N/L, the removal of nitrogen and phosphorus was limited mainly due to the microalgae growth limit. It was possible to treat anaerobic digested food waste leachate with mixotrophic microalgae when the ammonia concentration was controlled below 400 mg-N/L.

        Climate change has been a global issue since the 19th century. The increase in rainfall variability, which covers the increase in the earth’s total precipitation, will definitely lead to frequent and more severe flood disasters. As the damage increases year after year with floods as the most chronic and costly disaster among these hazards, Korea has to improve its technological responses and countermeasures to better visualize the hazards brought about by such disasters. Gunsan City ranked number eight in the country’s most susceptible region to floods. From 2004 to 2013, Korea has experienced a total of 174 flood disasters which were estimated to cost USD 7.32 billion. But reports showed that the total expenditure of the government amounted to 1.4 times the estimated losses and damages and the private companies have spent twice the said estimated amount. To summarize, the post-disaster loss and damage reports showed underestimated values. This study aims to develop a semi-parametric geographically weighted regression which can implement a flood damage estimation model of Gunsan City. The model building process include parameters like flood depth, flood duration, inundated area, family income and land price. The datasets are composed of both untransformed and transformed data (using Box-Cox Method). Both Ordinary Least Squares (OLS) Regression and Geographically Weighted Regression (GWR) were evaluated in this study, but the search for best fit resulted to the use of GWR.

        From 2004 to 2013, Korea has experienced a total of 174 flood disasters and has a total estimated cost of USD 7.32 billion. However, reports showed that the total expenditure of the government amounted to 1.4 times the estimated losses and damages and the private companies have spent twice the said estimated amount. To summarize, the post-disaster loss and damage reports showed underestimated values. In this regard, the National Emergency Management Agency (NEMA), the government institution designated to assess and analyze the damages and losses as well as evaluate the disaster risks of the said areas in accordance to their disaster risk management plans, are now developing a new estimating method for damages and losses. This study aims to develop flood damage functions that will estimate the flood damages of Gunsan City based on the building type: residential, commercial and agricultural facilities, by utilizing the Ordinary Least Squares Regression and later on, the Geographically Weighted Regression. The model building process includes flood depth, flood duration, inundated area, family income and land price as the parameter variables. Due to normality issue, the datasets were transformed through Box-Cox Method. Both Ordinary Least Squares (OLS) Regression and Geographically Weighted Regression (GWR) were evaluated in this study, but the search for ‘best fit’ resulted to the use of GWR.

        AtRabG3b and CaMsrB2 genes incorporated into pPZP vetor were transformed to Korean soybean cultivar Kwangan using highly efficient transformation system. AtRabG3b gene plays a positive role in xylem development in Arabidopsis and 64 transgenic plants were produced. CaMsrB2 gene is known to confer drought tolerance in rice and 63 transgenic plants were produced. As a result of PPT leaf painting assay, about 20% of transformation efficiency was observed from 2 times of inoculation. These transgenic plants were confirmed for gene introduction using PCR. Currently, the copy number and the gene expression is investigating using qRT-PCR and RT-PCR. Moreover, 62 lines and 53 lines of T1 seeds from AtRabG3b and CaMsrB2, respectively, were sown in GMO field.

        ORE7 gene incorporated into 3 different promoters including pCKLSL-35S, pCKLSL-TP and pCSENIF was transformed to Korean soybean variety Kwangan using highly efficient soybean transformation system. The gene is known to exhibit a delayed leaf senescence phenotype in Arabidopsis. Fourteen, Fifteen and nine transgenic plants were produced from pCKLSL-35S::ORE7, pCKLSL-TP::ORE7 and pCSENIF::ORE7, respectively. Moreover, transgenic plants were confirmed for gene introduction and their expression using PCR, qRT-PCR and RT-PCR. To identify the transgene insertion events, the analysis of flanking sequence was determined. As a results, T-DNA was integrated intergenically in transgenic line 1 of pCKLSL-35S::ORE7 and line 1 of pCSENIF::ORE7. Currently, flanking sequence analysis with pCKLSL-35S::ORE7, pCKLSL-TP::ORE7 and pCSENIF::ORE7 is carrying out to investigate the stable T-DNA insertions.

        Insect resistant genes encode insecticidal δ-endotoxins that are widely used for the development of insect-resistant crops. Common soybean is a crop of economic and nutritious importance in many parts of the world. Korean soybean variety Kwangan was transformed with Insect resistant genes. These genes were transformed into Kwangan using highly efficient soybean transformation system. Transgenic plants harboring Insect resistant genes were confirmed for gene introduction and their expression using PCR, real-time PCR and RT-PCR. The confirmation of stable gene introduction with Insect resistant genes was also performing by Southern blot analysis.         Soybean mosaic virus (SMV), a member of Potyviridae family, is one of the most typical viral diseases and results in yield and quality loss of cultivated soybean. Due to the depletion of genetic resources for resistance breeding, a trial of genetic transformation to improve disease resistance has been performed by introducing SMV-CP and HC-Pro gene by RNA interference (RNAi) method via Agrobacterium-mediated transformation. Transgenic plants were infected with SMV strain G5 and investigated the viral response. As a result, two lines (3 and 4) of SMV-CP(RNAi) transgenic plants and three lines (2, 5 and 6) of HC-Pro(RNAi) transgenic plants showed viral resistance. In genomic Southern blot analysis, most of lines contained at least one T-DNA insertion in both SMV-CP(RNAi) and HC-Pro(RNAi) transgenic plants. Subsequent investigation confirmed that no viral CP and HC-Pro gene expression was detected in two SMV-resistant lines of SMV-CP(RNAi) and three lines of HC-Pro(RNAi) transgenic plants, respectively. On the other hand, non-transgenic plants and other lines showed viral RNA expression. Viral symptoms affected seed morphology, and clean seeds were harvested from SMV-resistant line of SMV-CP(RNAi) and HC-Pro(RNAi) transgenic plants. In addition, strong viral gene expression was detected from seeds of SMV-susceptible non-transgenic plants and SMV-susceptible transgenic lines. When compared the viral resistance between SMV-CP(RNAi) and HC-Pro(RNAi) transgenic plants, soybean transgenic plants with the HC-Pro gene using RNAi strategy showed much stronger and higher frequency of viral resistance.

        Acanthopanax species is known commonly as Siberian ginseng, touch-me-not, devil’s shrub, prickly eleutherococc, eleutherococc and wild pepper. A diverse group of chemical compounds isolated from Acanthopanax species was named ‘eleutherosides’. Among eleutherosides, eleutherosides B and E were widely known in Acanthopanax species. Acanthopanax species are cultivated and grow wild in a various area of Korea and have a variety of pharmacological effects. But, there are a lot of difficulties on producing excellent Acanthopanax species, according to the cultivated method is different pharmacological ingredients. This study, therefore, analyzed eleutherosides B and E in A. divaricatus and A. koreanum by different fertilizer ratio using HPLC. We will be investigated a high content of eleutherosides B and E by different fertilizer ratio and suggest an efficient fertilizer ratio of A. divaticatus and A. koreanum. All samples of A. divaricatus and A. koreanum were collected at Yeongcheon Agricultural Technology & Extension Center, Yeongcheon, Korea. The sample was prepared by upper and lower parts. The fertilizer ratio are N-P-K(10.5-8.5-8.5: 50 kg/10a), 2N-P-K (21-8.5-8.5: 50 kg/10a), N-2P-K (10.5-17-8.5: 50 kg/10a), N-P-2K (10.5-8.5-17: 50 kg/10a), and 2N-2P-2K (21-17-17: 50 kg/10a), respectively. To analyze eleutherosides B and E, 5 g of A. divaricatus and A. koreanum was extracted with 50% MeOH (3 × 100 ml) by reflux and evaporated in vacuo. The residue was dissolved in 1 ml of MeOH. The resulting solution was used for HPLC analysis. HPLC separation of eleutherosides B and E for qualitative and quantitative analysis was performed using a reverse phase system. A Discovery®C18 (4.6 × 250 mm, 5 μm) column was used with a mobile phase that consisted of water and acetonitrile. A gradient solvent system of water and acetonitrile (90:10 to 70:30 for 20 min) was used for the elution program. UV detection was conducted at 350 nm. The injection volume was 10 μl and the flow rate was 1 ml/min. All injections were performed in triplicate. The different fertilizer ratio yielded total eleutherosides B and E contents of 4.417-6.905 and 3.652-7.227 mg/g in the upper and lower parts of A. divaricatus, respectively. In A. koreanum, the total eleutherosides B and E contents were 4.591-10.108 and 3.834-9.079 mg/g in the upper and lower parts, respectively. The best conditions to increase eleutherosides B and E content in A. divaricatus was determined to be with N-2P-K fertilizer ratio, on the other hand, in A. koreanum was 2N-2P-2K fertilizer ratio.

        Marked neutrophilia associated with neoplasia is a relatively rare finding and has been considered as a paraneoplastic manifestation. Thyroid cancer seldom presents with paraneoplastic leukocytosis. We report on a case of a 69-year-old man who presented with paraneoplastic leukocytosis seven months after undergoing total thyroidectomy and I-131 therapy for treatment of papillary thyroidcarcinoma. We found neither bone marrow involvement of malignant cells nor hematologic malignancy. Based on elevated levels of serum granulocyte colony-stimulating factor, we concluded that the cause was paraneoplastic leukocytosis. Another interesting point was the anaplastic transformation in the pleural metastatic site. It usually occurs in the intrathyroid or regional lymph node.

        Mesenchymal stem cells (MSCs) are considered to be attractive approaching in gene or drug delivery for cancer therapeutic strategies. In this study, the ability and feasibility of human bone marrow derived MSCs expressing the cytosine deaminase (CD)/5-Fluorocytosin (5-FC) prodrug was evaluated to target human osteosarcoma cell line Cal-72. At first, the fibroblast-like cells were successfully obtained from human bone marrow and demonstrated that they contained full of stem characteristics by the ability of differentiation into adipocyte/osteocyte and expression of typical mesenchymal markers CD90, CD44, while negative for CD34 and CD133 markers. We established the stable CD-expressing MSCs cell line (CD-MSCs) by transfection of pEGFP-C3 containing cytosine deaminase::uracil phos-phoribosyltransferase (CD::UPRT) gene into MSCs, and confirmed that the manipulated MSCs still remained full characteristics of multipotent cells and shown migration toward human osteosarcoma cancer cells Cal-72 as high as origin MSCs. Based on bystander effect, the therapeutic CD-MSCs significantly augmented the cytotoxicity on cancer cell Cal72 in either direct co-culture or conditioned medium in the presence of 5-FC. Moreover, in osteosarcoma cancer- bearing mice, the therapeutic CD/5-FC MSCs showed the inhibition of tumor growth compared with control mice which was s.c injected with only Cal72. Our findings suggest that these therapeutic CD-MSCs may be suitable and viable cellular vehicles for targeting human osteosarcoma cancer.
