        Historically the rise of contagious diseases and epidemics has been mainly caused by lack of food safety knowledge and negligence in personal hygiene. Food poisoning is an example that have been caused by lack of proper knowledge in food safety and personal hygiene. In order to prove this hypothesis, our team distributed a total of 225 survey sheets to middle school students. Survey questions mainly focused on the students' food safety knowledge, personal hygiene and regular dietary habits. We used 12 items to determine the stages of change in teenagers and divided into 3 stages of change: precontemplation stage (poor problem recognition), contemplation and preparation stage (some problem recognition), and action stage. Out of the 225 students surveyed, 105(47.1%) were male and 118 (52.9%) were female; 44.0% were in 7th grade, 33.0% in 8th grade, and 24.0% in 9th grade. In stages of change model, 3.8% were classified in pre-contemplation stage, 31.1% in preparation stage, and 65.1% in action stage. The awareness of the significance of food safety has a strong positive correlation with the stages of change food safety behavior (P<0.001), which suggesting our questionnaires regarding stages of change and food safety behavior were quite credential. Stages of change were also correlated with the consumption of unsanitary foods (P<0.01) and with the frequent hand washing (P<0.05). The stages of change showed a positive correlation with the food safety knowledge level (P<0.05). The four questions (out of 10) with the least percentage of correct answers were questions about the meaning of Norovirus (23.6%), the meaning of expiration date (43.6%), the meaning of food poisoning (36.4%), and proper food-handling (36.0%).

        Bacillus thuringiensis (Bt) is characterized by its ability to synthesize crystal toxins and also able to produce bacteriocins such as thuricin, tochicin, entomocin and bacthuricin. The present work, for the first time, describes the biological activity of bacteriocins from B. thuringiensis subsp. cameroun (Btc). Supernatant which was produced from a liquid culture of Btc had antimicrobial activity against Bacillus cereus, ending up to making a inhibition zone on an agar medium. A significant reduction in antimicrobial activity was observed when the supernatant was exposed to heat at 75~100°C for 15 min. Proteins were separated from the supernatant by a fast protein liquid chromatography (FPLC) given the thermal instability. A group of FPLC fractions had antimicrobial activity against Bt subsp. palmanyolensis, israelensis, 1-3, morrisoni, toguchini and kurstaki, and B. cereus ACTC21768, ATCC14579 and NRRLB-569. Interestingly, when the supernatant was individually incorporated into the liquid cultures of Bt subsp. israelensis (Bti) and mogi (Btm) with mosquitocidal activity, a vegetative cell growth was observed only in the Btm culture 10 h post-incubation. A possible recovery of vegetative Btm cell growth was observed, compared to a control without the supernatant. These results suggest that Btc produced proteinous antimicrobial substances, one of which may be used as a selection marker to separate Btm after possibly conjugating the two mosquitocidal strains.

        xylanolytic gut bacterium isolated from Eisenia fetida, Cellulosimicrobium sp. strain HY-13, produced an extracellular glycoside hydrolase capable of efficiently degrading mannose-based substrates such as locust bean gum (LBG), guar gum, mannotetraose, and mannopentaose. The purified mannan-degrading enzyme (ManS, 34,926 Da) from strain HY-13 was found to have an N-terminal amino acid sequence of DEATTDGLHVVDD, which has not yet been identified. Under the optimized reaction conditions of 50℃ and pH 7.0, ManK exhibited extraordinary high specific activities of 7,109 IU/mg and 5,158 IU/mg toward LBG and guar gum, respectively, while the enzyme showed no effect on sugars substituted with p-nitrophenol and various non-mannose carbohydrates. ManK strongly attached to Avicel, lignin, β-cyclodextrin, and poly(3-hydroxybutyrate) granules, but not bound to chitin, chitosan, curdlan, or insoluble oat spelt xylan. The aforementioned characteristics of ManS suggest that it is a unique endo-β -1,4-mannanase with out additional carbohydrolase activities, which differentiates it from other well-known carbohydrolases.

        Bacillus thuringiensis (Bt) strain K4 was isolated from fallen leaves which had been collected at a forest stand in Mungyeong city, Republic of Korea. The flagellated vegetative cells of Bt K4 were agglutinated with the H3 reference antiserum among 55 reference H-antisera. In a further test to identify subfactors, 3b and 3d monospecific antisera were reactive to the cells, followed up with introducing a novel serogroup of 3a3b3d, designated as serovar mogi. The strain K4 had mosquitocidal activity against Dipteran larvae, Anopheles sinensis and Culex pipiens pallens, with no Lepidopteran toxicity observed. The SDS-PAGE profile of K4 crystal protein, ovoidal-shaped, included several bands ranging from 30-75 kDa. Four putative peptides, Cry19Ba, Cry40ORF2, Cry27Aa and Cry20Aa were detected from the bands by a nano-LC-ESI-IT MS analysis. Through a thermal asymmetric interlaced PCR, cry19Ba, cry40ORF2 and cry27Aa genes were partially cloned from K4 strain. Three cry genes were further found in the strain by a 454 pyrosequencing, ending up to showing 58%, 39% and 84% homology in amino acids with Cry56Aa, Cry8Ba and Cry39ORF2 toxins, respectively. This novel 3a3b3d type strain, B. thuringiensis subsp. mogi, can be used as a good resource for studying unknown mosquitocidal cry genes.

        Paecilomyces lilacinus HY-4 is an entomopathogenic filamentous fungus that has exhibited insecticidal activity against Adoretus tenuimaculatus and Tetranychus urticae. Strain HY-4 has attracted a great deal of industrial concerns because this organism can be applicable as a potent bio-pesticide. In this study, we developed an optimal diphasic fermentation technique for HY-4 conidial production. The substrate prescription which was made up of hulls or brans was obtained by screening of agricultural products, and the conidial production could reach a minimum of 2.5×1010 conidia/g after solid fermentation for 12 days in a given condition. It was interesting to note that strain HY-4 was propitious to sporulate more efficiently and productively in liquid fermentation process in the presence of insect pupa. For the industrial application of the HY-4 spores, their pH and thermal stabilities were evaluated on SDA agar plate as well. Additionally, the germination rate of HY-4 spores was still above 90% even when they were stored for 5 months at ambient temperature.

        This study was conducted to investigate the occurrence patterns of sporadic and major insect pests in Gyeonggi Province Area. Lantern fly (Lycorma delicatula) damages to reduce the yield and quality of commodities in grape orchard begin to emergence from the early May, and new adults have appeared from the middle July lay eggs till the early November. The hatchability of eggs laid in last year was influenced by the extremely low temperature (especially -20℃ and under) in over-wintering season. Sweet-potato whitefly (Bemisia tabaci) is widely distributed in the southern part of gyeonggi province, and the biotype of most regional populations is Q-type except the partial Goyang area population (B-type). Potato tuber moth (Phthorimaea operculella) had been reported firstly in 2009 expanded and was confirmed in the northern part of gyeonggi province in 2010. Citrus flatid planthopper (Metcalfa pruinosa), becoming an issue in Korea, have been detected apple (Paju), pear (Yongin, paju), grape (Paju) orchards in 2010.

        Cambodia covers an area of 181,035 sq km, among them, forests cover 53 percent, so there are abundant Insect diversities. Collecting expeditions of Insect were conducted from towns of Osam (first base camp, N 12° 04' 51.7" E 103° 12' 30.5" alt. : 549m) and Roveang (second base camp N 12° 19' 06.5" E 103° 3' 03.8" alt. : 53m) in Pursat Province, western part of Cambodia. As protected forest from government, these areas were almost natural conservation field. Pitfall trap, sweeping method, light trap and searching etc. were used to collect insect. Specially in this expedition, large or big sized beetles, more than 5cm body length, for example, Lucanidae, Cerambycidae, Elateridae etc., mostly collected from light trap in deep forest. Materials are expected more than 100 species belonging to 20 families but it was difficult to identification because of lack of references of this areas. Maybe lots of new or new recorded species will found from Cambodia. Among Coleoptera species, with big sized beetles as the central figure, photos for species of dominant, rare, nominated species of new or new recorded were introduced, and also collecting sites and activities were presented.

        A new spray chrysanthemum cultivar ‘Snow Bowl’ was released by National Institute of Horticultural & Herbal Science(NIHHS), Rural Development Administration (RDA), in 2008. The cross was made in 2005 between ‘03B1-230’, breeding lines of NIHHS and ‘Sei-Alps’. Trials were conducted from 2006 to 2008 for the evaluation and selection of this cultivar, including shading cultures in summer and retarding cultures in spring. The natural flowering time of ‘Snow Bowl’ is late October, but year-round flowering is possible by photo-periodic control. It has single type flowers with white petals. The growth of plant is very vigorous and it is resistant to white rust. The diameter of flower is 6.3cm. Number of flowers per stem and petals per flower are 12 and 31, respectively. Days to flowering under the short day treatment is about 59 and its vase life is 24.1 days in autumn season. ‘Snow Bowl’ was applied as No. 2009-179 on February 18, 2009 for variety protection and the plant variety protection rights have been registered as No. 3239 on August 3, 2010 at the Korea Seed and Variety Service

        A Truncatipennes species, Pentagonica subcordicollis Bates is newly recorded in Korea. Here we provide a diagnosis and systematic accounts of P. subcordicollis. Consequently, a total of three species of Pentagonia have been recorded in the Korea, including the present species.

        The purpose of this study was to analyze and compare the effect of resistance exercise and balance exercise on proprioception and WOMAC index of patients with degenerative knee osteoarthritis. A total of 40 subjects participated in this study. The subjects were diagnosed with degenerative knee osteoarthritis and all were more than 60 years old. They were divided into three groups. Group Ⅰ(n=8) was trained with resistance exercise, Group Ⅱ(n=6) was trained with balance exercise and GroupⅢ(n=6) was trained with range of motion as a control. The results of this study were as follows. It was significantly indicated that the resistance exercise group and balance exercise group elicited error-reduction on proprioception goal-angle (p<.05). There was a statistically significant difference on proprioception between resistance exercise group and control(range of motion) group. There was a statistically significant reduction on WOMAC index between resistance exercise group and balance exercise group (p<.05) and on the WOMAC index between resistance exercise group and range of motion group(p<.05). In conclusion, resistance exercise and balance exercise are effective on degenerative knee osteoarthritis and resistance exercise is the most effective for improving proprioception and WOMAC index. More research on the intervention according to the degree of degenerative knee osteoarthritis is needed.

        Bee venom contains a variety of peptides and enzymes, including serine proteases. While the presence of serine proteases in bee venom has been demonstrated, the role of these proteins in bee venom has not been elucidated. Furthermore, there is currently no information available regarding the melanization response or the fibrin(ogen)olytic activity of bee venom serine protease, and the molecular mechanism of its action remains unknown. Here we show that bee venom serine protease (Bi-VSP) is a multifunctional enzyme. In insects, Bi-VSP acts as an arthropod prophenoloxidase (proPO)-activating factor (PPAF), thereby triggering the phenoloxidase (PO) cascade. Bi-VSP injected through the stinger induces a lethal melanization response in target insects by modulating the innate immune response. In mammals, Bi-VSP acts similarly to snake venom serine protease, which exhibits fibrin(ogen)olytic activity. Bi-VSP activates prothrombin and directly degrades fibrinogen into fibrin degradation products, defining roles forBi-VSP as a prothrombin activator, a thrombin-like protease, and a plasmin-like protease. These findings provide a novel view of the mechanism of bee venom in which the bee venom serine protease kills target insects via a melanization strategy and exhibits fibrin(ogen)olytic activity.

        Rice black-streaked dwarf virus (RBSDV), a member of the genus Fijivirus within the family Reoviridae, is the causative agent of maize rough dwarf and rice black-streaked dwarf diseases, both of which can lead to severe yield losses in east Asia. Although molecular approaches such as RT-PCR have potential for detection and diagnosis of this virus infections, their impact on high throughput certification is still limited. Therefore, the development of an antibody-based assay for rapid and effective diagnosis of RBSDV is preferable. In this study, we collected RBSDV from rice with rough dwarf disease and its complete nucleotide sequences of 10 genomic segments encoding 12 non-overlapping ORFs were determined. Among 12 ORFs, ORF1, 2 and 12 showed high level of similarities with the RdRp, major core protein and major outer shell protein, respectively. These ORFs were expressed as polyhedrin fusion protein or full-length soluble protein using baculovirus expression system for the preparation of specific antibody against RBSDV, which could be useful for the detection and diagnosis of this virus.

        We isolated two baculoviruses, Spodoptera litura granulovirus (SlGV) and S. litura nucleopolyhedrovirus (SlNPV) in the dead larvae of S. litura. The granule of SlGV were ovoidal shape with an approximate measure of 240-340 nm×140-180 nm, and each granule contained one single rod-shape virion with a mean size of 180-200 nm×20-40 nm. Whereas, the polyhedra of SlNPV were irregular in shape with a approximate diameter of 1.0-1.5 ㎛, and numerous virions comprised of the multinucleocapsid were contained in each polyhedra. The major component of occlusion bodies produced by SlGV and SlNPV were about 29 and 30 kDa, respectively. When the phylogenic relationship between these viruses were analyzed using the nucleotide sequences of granulin gene from SlGV and polyhedrin gene from SlNPV, they were not closely related to each other. We also found that the two viruses showed similar insecticidal activity against 2nd instar larvae of Spodotera litura in terms of dose-response, but SlGV showed much longer LT50 than that of SlNPV. The two baculoviruses might be cooperatively be applied as biological control agent for the control of S. litura

        Classical swine fever virus (CSFV) envelope glycoprotein E2 is the main target for inducing neutralizing antibodies and protective immunity in swine. Here, we report a novel strategy forthe large-scale production of a CSFV E2 subunit vaccine that demonstrates a high immunogenic capability in the larvae of a baculovirus-infected silkworm, Bombyx mori. We constructed a recombinant B. mori nucleopolyhedrovirus (BmNPV) that expressed recombinant polyhedra together with the N-terminal 179 amino acids of CSFV E2 (CSFV E2ΔC). BmNPV-E2ΔC-infected silkworm larvae expressed an approximately 44-kDa fusion protein that was detected using both anti-polyhedrin and anti-CSFV E2 antibodies. Electron and confocal microscopy both demonstrated that the recombinant polyhedra were morphologically normal and contained CSFV E2ΔC. The CSFV E2ΔC antigen produced in BmNPV-E2ΔC-infected silkworm larvae reached 0.68 mg per ml of hemolymph and 0.53 mg per larva at 6 days post-infection. Mice that were immunized with the granule form of recombinant polyhedra or the soluble form of the fusion protein elicited CSFV E2 antibodies, which indicated that the recombinant polyhedra carrying CSFV E2ΔC were immunogenic. The virus neutralization test showed that the serum from mice that were treated with recombinant polyhedra or the soluble form of the fusion protein contained significant levels of virus neutralization activity. These results demonstrate that the present strategy can be used for the large-scale production of CSFV E2 antigen and that the recombinant polyhedra containing CSFV E2ΔC as a granule antigen can be used as a potential subunit vaccine against CSFV.

        Autographa californica nucleopolyhedrovirus (AcMNPV) has a large doublestrand DNA genome of approximately 134 kbp and harbors 156 open reading frames (ORFs). To elucidate DNA replication cascade of AcMNPV, we developed a novel baculovirus genome that can be maintained in Escherichia coli as a plasmid and can infect susceptible lepidopteran insect cells. This genome, named bAc-MK, contains a mini-F replicon and a kanamycin resistance marker. Using a convenient Tn7 transposon-based system, pPCS-S, 55 single ORF-truncated mutants were generated by random insertion into bAc-MK genome. These single ORF-truncated mutants were independently transfected into Sf9 cells, 16 of them were found affecting viral replication since they defected in producing polyhedra. Furthermore, to verify the pathogenicity of the single ORF-truncated mutants, the remaining 39 mutants were subjected to bioassay to Spodoptera exigua 3rd instar larvae. Among them, ac9-, ac49-, ac103- and ac105-knockout mutants showed higher mortality compared to that of bAc-MK. These results suggested that these ORFs could be related to pathogenicity of AcMNPV.

        The Bacillus thuringiensis strain K4 was isolated from fallen leaves, sampled in a forest region of the city of Mungyeong, Korea. The flagellated vegetative cells of B. thuringiensis strain K4 were agglutinated with the H3 reference antiserum and further, agglutinated with 3b and 3d monospecific antisera but non-reactive for 3c and 3e factor sera. These results create a new serogroup with flagellar antigenic structure of 3a3b3d, designated serovar mogi. The strain K4 showed high activity against dipteran larvae, Anopheles sinensis and Culex pipiens pallens while no lepidopteran toxicity. It produced a single ovoidal-shaped parasporal crystal whose SDS-PAGE protein profile consisted of several bands ranging from 75 to 30 kDa. Through the protein identification by nano-LC-ESI-IT MS analysis, the putative peptides of Cry19Ba, Cry40ORF2, Cry27Aa and Cry20Aa were detected. In contrast to the plasmid profile of B. thuringiensis H3 serotype strains, the strain K4 contained only a large plasmid (~100 kb) and we cloned partial cry27Aa, cry19Ba and cry40ORF2 genes from it by thermal asymmetric interlaced PCR. Sequencing analysis showed 87%, 88% and 88% homologous with known cry27Aa, cry19Ba and cry40ORF2 genes, respectively. The new type strain, B. thuringiensis subsp. mogi (H3a3b3d) will be a good resource for new mosquitocidal cry genes.

        Ascotis selenaria, a major geometridae moth in citrus trees, annually damages the citrus leaves and fruits. The surface of young citrus fruit were usually fed by 1st larva of A. selenaria after landing or stepping onto the citrus fruits. To protect the larval damages of citrus fruits needs to predict the accurate occurrence time of the 1st larva for spraying. Because larval occurrences is dependent on the oviposition of adult female and the eggs were not found in/on citrus trees, oviposition model of A. selenaria linked with the egg development model will be helpful of protecting larval damages on citrus fruits. Adult longevity, survival and fecundity of A.selenaria was investigated at 13, 16, 20, 24, 28, 30, 32, and 35℃. The longevity decreased as the temperature increased and the female development rates (1/median longevity) were well described by a modified sigmoid model, which was used to calculate the adult physiological age. Description of the total fecundity was used by a non-linear model: The maximum fecundity of A. selenaria was estimated as 2490 eggs and peaked temperature was 19.7℃ according to the fecundity model. The cumulative age-specific oviposition rate and the age-specific survival rate was well described by 2 parameters Weibull function and a reverse logistic curve respectively. Total fecundity model, age-sepecific oviposition model, and age-specific survival model were incorporated into the oviposition model.

        Although the greater part of Coccinellidae are all pure entomophagous insects (Weber and Lundgren, 2009), many species of phytophagous, mycophagous and polyphagous ladybirds have been reported in worldwide. This study was conducted to investigate the morphological characteristics of mouthparts of mycophagous ladybird, I. koebelei, comparison with aphidophagous species, Propylea japonica (Coccinellidae: Coccidellini), and developmental charicteristics of I. koebelei at different temperature under incubation conditions. Four species of powdery mildews that I. koebelei fed were observed, and their host plants were pear tree, persimmon tree, red pepper and cucumber. The most conspicuous elements of I. koebelei mouthparts is the mandible. The distal incisor region of the mandible of both species is formed by two teeth, giving them a bifid apex. and the terminal teeth spilt near top and the ventral tooth is smaller than the dorsal one. but the ventral tooth of I. koebelei divided into a row of additional teeth was different with P. japonica. Furthermore, while the inner mandibular cutting edge of P. japonica is smooth, that of I. koebelei is covered in minute teeth, forming a comb. These structures are presumed to help I. koebelei rake fungal spores growing on leaf surface. The developmental period and reproduction ability of were significantly different according to temperature conditions when I. koebelei was fed the powdery mildew from cucumber leaves. The optimum temperature for I. koebelei rearing was from 23 to 25℃ that is the similar condition of cucumber powdery mildew occurrence.
