        Background : Perilla frutescens L. is valuable as a medicinal plant as well as a natural medicine and functional food. Limonene perilla collected from various places showed 60% limonene compounds. However biological activity of these accession has not been reported before. Therefore, this study was conducted to investigate the biological activity of limonene perilla. Methods and Results : Fractional solvent extracts were obtained by using organic solvents such as n-hexane, chloroform, ethyl acetate, n-BuOH, and aqueous solvent from different parts of limonene perilla extracted initially in 70% EtOH. We investigated the effects of limonene perilla on total phenol and flavonoid contents, FRAP (Ferric Reducing Antioxidant Power), total saponin contents and tyrosinase inhibition activity. Leaves of limonene perilla produced the highest total phenolic contents (29.88 mg·CAE/g), flavonoid (8.39 mg·QE/g) and saponin contents (47.77 mg·GIE/g) than stems and roots of limonene perilla. FRAP of leaves was 823.00±3.58 μM·FeSO4·E/mg. Tyrosinase inhibition activity rate was 40.31% in 70% ethanol extracts from leaves of limonene perilla. Conclusion : This results suggest that leaf of limonene perilla fractions has significant antioxidant activity. Also, limonene perilla could be used as a functional biomaterial in developing cosmetics and functional foods.

        Background : This study, the fraction for testing the efficacy of the Astragalus extract was concentrated active ingredient. The concentrated fraction was applied to a cosmetic material that Astragalus testing results confirmed that the improved efficacy. Methods and Results : The fractions were performed using an n-butanol solvent for increasing the efficacy of the Astragalus extract, by using the material fractions collected to compare and ultimately an increase in whitening and wrinkle efficacy. The solvent to be used in the fractions was used for the n-butanol good dissolution to the effective substance(Astragaloside, Isoflavonoid). It increased approximately 6.5 times the sample extract and the n-butanol fraction of the Astragalus as a result Astragaloside 15 ppm, 97 ppm respectively analyzed by HPLC equipment, isoflavonoid content was confirmed by an increase in the content of the fractions increased 4.5 times to 280 ppm, 1,260 ppm. Tyrosinase inhibitory effect, respectively IC50 5.70 mg/mL, IC50 1.02 mg/mL to, Collagenase producing ability is IC50 4.88 mg/mL, IC50 0.93 mg/mL with n-butanol fraction was good whitening, anti-wrinkle efficacy than the extract. Sensory evaluation was conducted in the same amount of sample, using a purified Astragalus cosmetics received high marks. Stability evaluation(MTT assay) was checked for more than 100% cell viability at the concentration 2,000 ppm. Conclusion : n-butanol fraction of Astragalus was subjected to a component analysis and In vitro test, it was confirmed an increase active ingredient content. The results of sensory evaluation and stability evaluation, it was confirmed been made to improve qualities as a cosmetic materials.

        Background : Bitter gourd (Momordica charantia) is a vegetable with pantropical distribution and contains the active ingredient, such as momordicine and charantin. Moreover, bitter gourd has been reported to exhibit antidiabetic, anticancer, cardiovascular-protective and antioxidant effects. But, the distinctive bitter taste of bitter gourd was not suitable for food preference. This study was conducted to evaluate the improvement effects of bitter taste of bitter gourd with natural fermentation and roasting process. Methods and Results : Bitter gourd fruits were obtained from Headeulnyeokae Co., Ltd.(Gang-Jin 59253, Repubilc Korea). Fruits of bitter gourd were prepared by natural fermentation process for 4h at 25℃. After natural fermentation, the fruits were dried in hot-air dryer for 15h at 49-55℃. The dried fruits were roasted in 500 g batches at 120℃ for 10 min in the electric rotisserie oven. After roasting, fruit pieces were extracted with hot water and cold water. One of them was cool-type, and the other was hot-type bitter gourd tea. In the sensory evaluation, hot tea and cold tea showed the high scores on color, flavor and overall acceptability. Conclusion : These mean that roasted bitter gourd had less bitter, and it could be utilized widely as drink and food material.

        Background : This text was conducted to compare the difference of fatty acid by part and ha bitat of Glehnia littoralis in Korea Midwest. Methods and Results : In Glehnia littoralis, Saturated fatty acid consisted of palmitic acid, ste aric acid, and unsaturated fatty acid was oleic acid, linoleic acid, linolenic acid. Content of w hole fatty acid was the most as 42.8% linoleic acid, and stearic acid was less best as 2.9%. Other fatty acid were oleic acid 27.1%, palmitic acid 15.3%, linolenic acid 11.9%. Fatty acid by plant part it was the most in leaf that linoleic acid (31.4%), and it was the most in root t hat linoleic acid (68.3%), and it was the most in breed that oleic acid (65.5%). Fatty acid by natural habitat contained in Incheon area located in high latitude more than Taean area. Conclusions : In Glehnia littoralis of Korea Midwest, The collection and cultivation of medici nal scope was broader, because difference of fatty acid content by natural habitat was light.

        Background : This study was performed to investigate by antioxidant activity, total phenolic contents, total flavonoid contents, and effective component of Astragalus membranaceus treated with different artificial light Sources (fluorescent lamp, red, blue, green, white, LEP). Methods and Results : We investigated the effects of various artificial light sources on the DPPH radical activity, total phenol and flavonoid contents, tyrosinase activity and main flavonoid compounds contents (formononetin and calycosin) and other biological activities in A. membranaceus. Antioxidant activities were 53.6% as the highest level of activity under LEP light. Growth under LEP light also produced the highest total phenolic and flavonoid contents of 36.05 and 5.94 mg/ml, respectively. Extracts from plants grown under LEP light caused the highest inhibition of tyrosinase activity with inhibition of 35.37, 61.87, and 65.49%, respectively, for extract concentrations of 100 μg/ml, 500 μg/ml, and 1000 μg/ml compared with other artificial light treatments. Conclusion : Little information is available on the influence of LED and LEP light sources on antioxidant production or other biological activities in A. membranaceus. Our goal in this study was to determine the effects of LED and LEP artificial light sources on the production of new functional compounds in A. membranaceus.

        A double-cropping system with soybean (Glycine max) following the cultivation of potato, garlic, and onion is widely adopted in the southern region of Korea. For this system, marginal dates for planting must be determined for profitable soybean yields, because the decision to plant soybean as a second crop is occasionally delayed by harvest of the first crop and weather conditions. In order to investigate the effect of planting date on soybean yield, three cultivars (early and late maturity) were planted on seven different dates from May 1 to July 30 in both paddy and upland fields across 2012 and 2013. Soybean yields were significantly different among the planting dates and the cultivars; however, the interaction between cultivar and planting date was not significant. Based on linear regression, the maximum yield of soybean was reached with a June 10 planting date, with a sharp decline in yield for crops planted after this date. The results of this study were consistent with those of a previous one that recommends early and mid-June as the optimum planting period. Regardless of soybean ecotype, a reduction in yield of greater than 20% occurred when soybean was planted after mid-July. Frost during soybean growth can reduce yields, and the late maturity cultivars planted on July 30 were damaged by frost before completing maturation and harvest; however, early maturity cultivars were safely harvested. For sufficient time to develop and reach profitable yields, the planting of soybean before mid-July is recommended.

        The effect of sudden changes of water temperature (WT) on the survival rate and physiological responses of the red spotted grouper (Epinephelus akaara) were examined by manipulating WT control system for 9 days. Experimental condition was divided in two different regimes at low (from 10°C to 4°C, decreased 1℃/d) and high (from 28°C to 34°C, increased 1°C/d) WT. Survival rate of experimental fishes were observed, and determined the changes of hematological characteristics by analyzing plasma levels of cortisol, glucose, total protein, and electrolytes (Na+, Cl–, K+). No mortality was observed until low WT 6°C (144 h) and high WT 32°C (96 h), and 100% mortality was observed at low WT 4°C (216 h) and high WT 35°C (171 h). Plasma levels of cortisol and glucose increased rapidly as decreasing WT, and the loss of swimming ability and respiration response was observed at low WT 7°C and high WT 34°C conditions.

        Background : The research is designed to investigate the optimal cultivation technology and the growth of above-ground and below-ground sections as well as the photosynthetic characteristics for new ginseng variety “K-1” by differentiating the planting density under the conditions of transplanting and direct seedling. Methods and Results : The K-1 variety and hybrid variety (Jakyungjong) were selected for the research and the ginseng varieties were transplanted and directly sown in Yeoncheon area in 2013. The transplanting was made in the form of 5 lines × 9 rows (45 plants), 6 lines x 9 rows (54 plants), 7 lines × 9 rows (63 plants) and 8 lines × 9 rows (72 plants) in each lot (1.65㎡) while the direct seedling for testing was conducted three times in randomly blocked design in the form of 11 lines × 14 rows (154 plants), 12 lines × 14 rows (168 plants), 13 lines × 14 rows (182 plants). Various measures were collected from the 4-year transplanted ginseng and 3-year direct seedling ginseng in 2015 to find out the growth features and photosynthesis of above-ground section (rate of germination, leaf length, leaf width, stem length and leaf area index (LAI)) and the below-ground section (length, diameter, weight and class of roots). Conclusion : After the planting of the ginseng, the germination rate of K-1 for the transplanting was 85.1 ~ 92.0% across different plantation densities while that for the direct seedling was 67.7% ~ 77.9% across plantation densities, thus showing no significant difference between the two planting methods. LAI was higher for the higher planation density for both transplanting and direct seedling. As for the photosynthesis speed, the form of 6 lines × 9 rows showed the higher speed in transplanting while the form of 12 lines x 14 rows showed the higher speed in direct seedling. The photosynthesis of K-1 was higher than that of Jakyungjong. In the 4-year ginseng cultivated under the transplanting, diameter of roots, number of branch roots and weight of raw ginseng were the highest in the plantation density of 5 lines × 9 rows. The distribution of root weight was high with 23.3% and 20.0% for the 51~70g group and the 71g or above group, respectively, for the 4 year transplanted plants in the form of 5 lines × 9 rows. The growth for above-ground and below-ground sections for K-1 was better than that for Jakyungjong. As a result, it was found that the proper plantation density for the 4-year root in the transplanted K-1 was 5 lines × 9 rows considering the growth of the above-ground section, quantity and distribution of root weight.

        Safflower (Carthamus tinctorius L.) seeds have long been clinically used in Korea to promote bone formation and prevent osteoporosis. In addition, the safflower buds (SB) were found to have more useful functional ingredients than safflower seed. Thus, we investigated the preventive effects of SB diet in ovariectomized (OVX) rats. The rats were divided into five groups; sham operated group, OVX alone group, OVX plus 17β-estradiol (E2 10 ㎍/㎏, i.p.) and OVX plus SB diet feeding group (0.3% or 1%). Feeding of SB diet (0.3% or 3%) to OVX rats markedly increased trabecular formation in femur compared to OVX rats. Feeding of SB diet (0.3% or 3%) to OVX rats also decreased TRAP activity compared to OVX rats. These results suggest that SB diets have bone sparing effects by the decrease of osteoclast activity. We also observed that OVX rats fed with SB diet (0.3% or 3%) exhibited the decrease of calcium and phosphorus in serum compared to OVX-induced rats. Therefore, SB may be beneficial for the patients of osteoporosis, especially in postmenopausal women.

        The microbial composition in Nuruk, a Korean cereal fermentation starter, is a critical factor for the quality and organoleptic properties of traditional alcoholic beverages. This study was aimed at monitoring the compositional change and enzyme activity of culturable lactic acid bacteria (LAB) in two types of Nuruk fermented at different temperatures. All culturable LAB were isolated at various time points (0, 3, 6, 10, 20, and 30 days) and identified by 16S rRNA sequencing. In traditional Nuruk type A (TN-A), which was fermented at 36℃, the population of total culturable LAB during the fermentation period was between 104 and 105 log CFU/mL. On the other hand, the LAB population in traditional Nuruk type B (TN-B) fermented at 45℃ (primary fermentation for 10 days) and 35℃ (secondary fermentation for 20 days) was 102 log CFU/mL; however, these bacteria could not be detected after 6 days. Major LAB strains were identified in both Nuruk types: (1) from the MRS-culture of TN-A, Pediococcus pentosaceus at 3-30 days; (2) from MRS-culture of TN-B, P. pentosaceus at 3 days and Enterococcus hirae at 6 days. The protease activities of the dominant LAB isolated from the TN-A and TN-B cultures were within the ranges of 0.64~1.03 mg/mL and 0.74~0.81 mg/mL (tyrosine content), respectively, whereas the α-amylase activities were 0.75~0.98 mg/mL and 0.78~0.79 mg/mL (amylose content), respectively.

        In this study, antioxidant and anti-inflammatory activities of water, methanol, and ethanol extracts obtained from Allium hookeri root were evaluated. The ethanol extract of A. hookeri was found to possess the strongest reducing power and also exhibited dominant effects on scavenging of nitrites, DPPH radicals, and superoxide radicals. The water extract showed more efficient DPPH and hydroxyl radical-scavenging activities than those of the methanol extract. Furthermore, the inhibitory activity against nitric oxide (NO) production in RAW 264.7 macrophages was evaluated to elucidate the anti-inflammatory properties of the extracts. Results indicated that all the extracts of A. hookeri exerted inhibitory activities against NO production, especially the ethanol extract (IC5029.13μg/mL). Total phenolic and total flavonoid contents were found to be abundant in the ethanol extract, with values of 24.96 mg gallic acid equivalent/g extract and 4.27 mg rutin equivalent/g extract, respectively. Total thiosulfinate content was determined for the first time and a high amount was present in the ethanol extract (14.2 μM/g extract). These results suggest that A. hookeri root has antioxidant and anti-inflammatory properties and could be used as a natural source for the development of pharmaceutical agents or functional foods.

        Environment-friendly growth enhancers for rice are being promoted to reverse the negative impact of intensive chemical-based and conventional rice farming on yield sustainability and environmental problems. Several rhizosphere microorganisms and pyroligneous acids (PA) had demonstrated beneficial influence on growth, yield and grain quality of rice. Since most of the previous study had evaluated the effect of PGPR and PA on paddy rice singly, the effect of combined application of these on the growth and yield of paddy rice and on some soil chemical properties were determined. A four factorial pot experiment was conducted to evaluate the effect of PGPR, PA in combination with fertilizers and on different soil types. There were 54 treatment combinations including the control with three replications under complete randomized design. Plant growth parameters were evaluated using standard procedures during tillering and heading stages. Rice yield and some soil chemical properties were determined at harvest. Results showed that inoculation of Bacillus licheniformis and Fusarium fujikuroi enhanced plant growth by increasing the plant height which could be ascribe to its ability to promote IAA and GA production in plants. Inoculation of Rhizobium phaseoli enhanced chlorophyll content indicative to its ability to improve the N nutrition. However, these plant growth benefits during the vegetative stage were override by the fertilizer application effect especially during the maturity stage and grain yield. High fertilization rates on coarse-textured soil without nutrient loss resulted to high available nutrients and consequently high yield. Wood vinegar application however improved nutrient availability in soil which could be beneficial for improving soil quality. Further evaluation is necessary to fully assess the potential benefits that could be derived from inoculation of these organisms and wood vinegar application in different soil environment especially under different field conditions.

        The transposable element is a DNA sequence that can be changed its position within the genome, sometimes it can create or reverse mutations and altering the cell's genome size. Target region amplification polymorphism (TRAP) is a rapid and efficient PCR-based marker technique, which uses bioinformatics tools and expressed sequence tag (EST) database information to generate polymorphic markers around targeted candidate gene sequences. TE-TRAP is a new marker system which used terminal inverted repeat (TIR) instead of targeted candidate gene sequences. Sorghum holds a good potential plant organism for transposon tagging due to its small genome size, low amount of repetitive DNA and co-linearity with other cereal genomes, which allows the use of information derived from sorghum in other cereal grasses. IS2868 of sorghum accession was treated Gamma irradiation on seed. To define availability and utilization of TE-TRAP, twenty-one accessions were used to evaluate the genetic diversity and underlying relationships. One-thousand thirty-three TE-TRAP markers were amplified by thirty-one primer combination. Altogether, 712 (62.8%) markers were observed polymorphic segregation, whereas 421 (37.2%) showed monomorphic patterns. To estimate genetic differentiation of population by various gamma radiation doses, the analysis of molecular variance (AMOVA) was performed using 4 to 5 different radiation doses population of M1 sorghum individuals. This study and marker system will provide valuable information to assist radiation mutation breeding.

        Space has many distinguishable characteristics from earth such as strong cosmic radiation, microgravity, supervaccum and weak magnetic field. For this reason, space environments can be used an efficient mutagen for plant breeding nowadays. To identify the affected genes by condition in space with outer space, Brachypodium seeds were placed in the Russia Segment (RS) Biorisk module of International Space Station (ISS). Brachypodium distachyon is a model system for temperature grass, because they represent the characteristics for annual winter grass. Seeds and organs of plants carried by satellite or spacecraft to space can be genetically mutated by exposing space environment. We performed a duplicated RNA sequencing to profile the differentially expressed genes. As a results, about 700 genes were upregulated and 250 genes were downregulated by cosmic environments, respectively. In the molecular function category, protein kinase and transcription activity related genes were upregulated. Among the many transcription factors (TFs), stress related TFs such as ERF, NAC and WRKY were differentially expressed in space exposed samples. In the future, their expression will be identified by using qRT_PCR.

        Phytophthora capsici an Oomycete pathogen is a major challenge to the pepper (Capsicum spp.) production around the world. Control measures are proved ineffective, so breeding resistant cultivars are the most promising strategy against the pathogen. Resistance against P. capsici is governed by quantitative trait loci (QTL). According to previous studies on QTL detection, the QTL on pepper chromosome 5 is a major contributor to resistance. In this study, to exploit the involvement of this QTL and identify its contributing genes, the F2 population derived from a cross between ECW30R and CM334 was inoculated with a medium virulence P. capsici strain JHAI1-7 zoospores at the 6-8 leaf stage. Composite interval mapping revealed two major QTLs; QTL5-1 from 7 days post inoculation (dpi) and QTL5-2 from 16 dpi on chromosome 5. To characterize and detect interactions of the two QTLs, near isogenic lines (NIL) were constructed by crossing Tean and recombinant inbred line (RIL) derived from a cross between YCM334 and Tean. RILs were screened with P. capsici strain MY-1 and resistant lines were selected. Among the resistance RILs most closely related to Tean were selected using AFLP and SSR genotyping data. These RILs were named as YT39-2 and YT143-2. To develop more advanced NILs, two rounds of marker-assisted backcrossing were done using a high-throughput SNP genotyping system (EPI Fluidigm, USA). Among the NILs derived from YT39-2, YT39-2-64 contains only QTL5-1 whereas YT39-2-61 and YT39-2-69 were identified to have both QTLs. On the other hand, YT143-2-55-7 with the highest Tean genetic background contains QTL5-1 only. In the next step, the 3 different NILs having QTL5-1, QTL5-2 individually and both QTLs will be identified. Furthermore, phenotyping and fine mapping will be done for the analysis of individual and interaction effects of QTLs.

        UDP-glucose 4-epimerase (UGE; EC 5.1.3.2) is an enzyme that plays an essential role in the interconverts UDP-D-glucose (UDP-Glc) and UDP-Dgalactose (UDP-Gal). Five members of the Chinese cabbage (Brassica rapa) UDP-glucose 4-epimerase gene family, designated BrUGE1 to BrUGE5, have been cloned and characterized. Quantitative PCR shows that the BrUGE1and BrUGE4 mRNA are most abundant among other BrUGE genes, accounting for more than 55% of total BrUGE transcripts in most of the tissues examined. All genes showed organ specific expression pattern, two of which (BrUGE1 and 4) actively responded after Pectobacterium carotovorum subsp. carotovorum infection, while four genes (BrUGE-1, -3, -4 and -5)were shown to respond considerably against salt, drought and abscisic acid (ABA) treatments. To better understand the function of the UGE gene, we constructed a recombinant pART vector carrying the BrUGE1 gene under the control of the CaMV 35S promoter and nos terminator and transformed using Agrobacterium tumefaciens. We then investigated BrUGE1 overexpressing rice lines at the physiological and molecular levels under biotic and abiotic stress conditions. Bioassay of T3 progeny lines of the transgenic plants in Yoshida solution containing 120 mM Nacl for 2 weeks, confirmed that the BrUGE1 enhances salt tolerance to transgenic rice plants. Also T3 progeny lines of the transgenic plants, when exposed to infection caused by Xanthomonas oryzae pv oryzae, showed tolerance to bacterial blight. These results showed that BrUGE1 can be used as potential genetic resource for engineering Brassica with multiple stress resistance.

        This study was carried out to determine the amount of lignin, cellulose, and hemicellulose in six kenaf cultivars during different harvesting stages. Three mutant cultivars (Jangdae, Jeokbong and Baekma), two original cultivars (Jinju, C14), and one Chinese cultivar (Auxu) were planted on May 14, 2013. Four harvesting times were made at intervals of 20 days from 15 July to 16 September, 2013. The overall growth characters of mutant cultivar ‘Jeokbong’ such as plant height, stem diameter, flowering time, and dry mass were similar with those of the original variety. The mutant cultivar ‘Baekma’ occurred 10-day late flowering in comparison with the original variety and also displayed higher dry mass than the original variety. Jinju, Auxu and Jangdae, mid-late maturing kenaf cultivars, had high dry weight compared to early maturing cultivars such as Jeokbong, Baekma and C14. In all cultivars, the lignin contents were increased by a late harvest. The Mid-late maturing kenaf cultivars showed high lignin content in comparison with those of the early maturity cultivars. There were no significant differences of cellulose, and hemicellulose content between the cultivars, however cellulose content in stems of these kenaf cultivars were significantly decreased by a late harvest. These results may provide valuable information to assist the parental selection of kenaf breeding.

        Kenaf (Hibiscus cannabinus L.) native to Africa can be used as fiber, food, feedstock and bio plastic. This study was carried out to evaluate the mineral, amino acid and vitamin contents of six selected kenaf cultivars which are enable to produce seed under Korean circumstance. The leaves of three mutant cultivars (Jangdae, Jeokbong and Baekma), two original cultivars (Jinju, C14) and one Chinese cultivar (Auxu) were harvested at flowering time. Mineral components of kenaf leaves, such as calcium, potassium, and mineral, did not showed significant differences among the cultivars. As major amino acids including proline and phenylalanine, significant differences were found in these kenaf cultivars. The Auxu cultivar contained the highest amount of essential amino acid (Phenylalanine, Leucine, Isoleucine, Valine, Methionine and Lysine). The amount of vitamin displayed significant differences such as vitamin E and vitamin K among these cultivars. Especially, Jangdae cultivar contained the highest amount of vitamin E and vitamin K. Thus, these data suggested that Jangdae and Auxu is the most desirable cultivar containing high amount of vitamin and amino acid.

        Foxglove aphid, Aulacorthum solani (Kaltenbach), is a Hemipteran insect that infected a wide variety of plants worldwide and caused serious yield losses in crops. The objective of this study was to identify the putative QTL for foxglove aphid resistance in wild soybean, PI 366121, (Glycine soja Sieb. and Zucc.). One hundred and forty one F2-derived F8 recombinant inbred lines developed from a cross of susceptible Williams 82 and resistant PI 366121, were used. The phenotyping of antibiosis and antixenosis was done through choice and no-choice assays with total plant damage (TPD) and primary infestation leaf damage (PLD); a genome-wide molecular linkage map was constructed with 504 single nucleotide polymorphism markers utilizing a GoldenGate assay. Using inclusive composite interval mapping analysis for foxglove aphid resistance, one major candidate QTL on chromosome 7 and 3 minor QTL regions on chromosome 3, 6 and 18 were identified. The major QTL on chromosome 7 showed both antixenosis and antibiosis resistance responses. However, the minor QTLs showed only antixenosis resistance response. The major QTL mapped to a different chromosome than the previously identified foxglove aphid resistance QTL, Raso1, from the cultivar Adams. Also, the responses to the Korea biotype foxglove aphid were different for Raso1, and the gene from PI 366121 against the Korea biotype foxglove aphid were different. Thus the foxglove aphid resistance gene from PI 366121 was determined to be an independent gene to Raso1 and designated to Raso2. This result could be useful in breeding for new foxglove aphid resistant soybean cultivars.

        Foxglove aphid, Aulacorthum solani (Kaltenbach), is a Hemipteran insect that infected a wide variety of plants worldwide and caused serious yield losses in crops. The foxglove aphid resistance gene, Raso2 was previously mapped from PI 366121 (Glycine soja Sieb. and Zucc.) to a 26cM marker interval on soybean chromosome 7. The development of additional genetic markers, which are mapped closer to Raso2 were required to accurately position the gene to improve the effectiveness of marker assisted selection. The objective of this study was to narrow down the putative QTL region, which is responsible to foxglove aphid resistance in PI366121 using recently developed high-density 180K Axiom SoyaSNP genotyping array. One hundred and forty one F8-derived F12 recombinant inbred lines developed from a cross of susceptible Williams 82 and resistant PI 366121, were used to generate a fine map of Raso2 interval. The phenotyping of antibiosis and antixenosis was done through choice and no-choice assays with total plant damage (TPD) and primary infestation leaf damage (PLD). The composite interval mapping analysis showed that the physical interval between two flanking makers, which was corresponding to Raso2, was narrowed down to 500kb on the Williams 82 genome assembly (Glyma2.0), instead of 4Mb in the previous report using Goldengate assay. In the Raso2 interval, there are about 60 candidate genes, including 4 of NBS-containing putative R genes. This result could be useful in breeding for new foxglove aphid resistant soybean cultivars.
