Eating and cooking qualities are the most important trait in japonica rice breeding program in Korea. More improvements in grain quality to meet the demand of consumers are needed to develop new rice germplasm of high grain quality. In this study, we performed genetic analysis and grain quality evaluation in 96 Korean japonica rice germplasm including 26 varieties with 4 Japanese japonica high eating quality, 24 landraces, 22 weedy rices, and 14 breeding lines. These germplasm were analyzed using 13 DNA markers related to eating quality to conjecture the palatability of cooked rice (Lestari et al. 2009). Most varieties of high eating quality were clustered with germplasm of high expected quality (eq) varieties of similar genetic background of pedigree. The expected quality (eq) values of high eating quality varieties, Gopum, Ilpum, Samgwang, and Sugwang were 99.6∼104.5, and Koshihikari was 103.5. The eq of two weedy rices, Hoengseongaengmi 3 and Namjejuaengmi 6 were 101.9 and 101.6, respectively. However, Haiami of high eating quality was clustered with 15 weedy rice and 11 landrace germplasm of low eq value. The eq values of Haiami and Wandoaengmi 6 were 66.9 and 40.8, respectively, but they has 2 and 3 of palatability of cooked rice, and 73.6 and 78.6 of glossiness of cooked rice, respectively. We expect these germplasm would be new source for rice grain quality to develop japonica rice of high eating quality.

Stem rust of wheat caused by Puccinia graminis f. sp. Tritici is historically one of the most important diseases of crops worldwide. The objective of this study was to screen the growth habits and yield of wheat accessions in Korea winter condition. We collected ninety stem rust resistant germplasms from USDA-Grain and planted for the characteristics of growth and yield in 2013. We measured the growth characteristics such as the plant type, leaf color, heading date, stem length, spike length, awn length, number of spikelet, number of tiller, and 1,000 kernel weight (g). Early maturity is most important wheat breeding target in double cropping system such as Korea. Heading dates of ninety cultivars ranged from May 17 to June 7. The Keumkang control cultivar headed on May 23. Heading dates of seventeen accessions had as early as Keumkang control (May 23). The numbers of semi-erect and intermediate type for growth habit were 45 and 33 respectively. The 1000 kernel weights were also measured. Mean 1000 kernel weights ranged from 16.7g (Webster) to 41.8g (123 A1C2). The average of stem length was 100.2cm and Keumkang was 58.4cm. These results showed that A4849-60P-7P-1P (Chile), MG 31492 (Ethiopia), 1338.F.6.C.3.B (Kenya), 1203 A1C2 (Kenya), and II-90-20/3-1R-1M- 3R (Mexico) were best lines for agronomic characteristic such as early heading date and grain yield

Fusarium head blight (FHB), primarily caused by Fusarium graminearum is a major disease problem on wheat and barley around the world. The objectives of this study were to evaluate for FHB resistance under greenhouse and filed condition in 2012 and 2013. We screened for Type I resistance in the field using spray inoculation and for Type II resistance in the greenhouse using single floret inoculation. Sumai 3 and Ning7840 were used the FHB resistant check. Five hundred and twelve lines were evaluated for resistance to initial infection (type I) and to spread of symptoms within spike (type II). The inoculum was F. graminearum (GZ3639) prepared at a concentration of 1 X 105 macroconidai/ml with Tween 20. The field screening plots were inoculated twice and mist-irrigation was applied to facilitate FHB development. FHB severity was assessed visually 21 days and determined as the percentage of symptomatic spikelets from the total of all spikelets observed in 20 spikes. Both in the greenhouse and field test, we could observe five categories of FHB severity: resistant (R: 0-20%), moderately resistant (MR: 21-40%), moderate (M: 41-60%), moderately susceptible (MS: 61-80%), and susceptible (S: 81-100%). The results showed that seventy seven lines showed the R and MR category on FHB severity. In addition, nine lines showed similar FHB severity compared to Sumai 3 (13.3%).

A field study was conducted to understand nitrogen use efficiency of high yielding Japonica rice varieties under three levels of nitrogen fertilizer (90, 150 and 210 kg N ha -1 ) in Iksan, Korea. Two high yielding rice varieties, Boramchan and Deuraechan, and an control variety, Dongjin2, were grown in fine silty paddy. Nitrogen use efficiencies (NUE) were 83.3, 56.3, and 41.2 in 90, 150, and 210 kg N ha -1 fertilizer level, respectively. Total nitrogen uptake varied significantly among nitrogen levels and varieties. Variety Dongjin2 showed the highest nitrogen uptake efficiency (NUpE), while Boramchan and Deuraechan showed higher nitrogen utilization efficiency (NUtE). However, Nitrogen harvest index (NHI) was higher in Boramchan (0.58) than Deuraechan (0.57) and Dongjin2 (0.53). Rough rice yield showed linear relationship with total nitrogen uptake (R 2 =0.72) within the range of nitrogen treatments. Boramchan produced significantly higher rough rice yield (8546 kg ha -1 ) which mainly due to higher number of panicles per m 2 compared to Deuraechan (7714 kg ha -1 ). Deuraechan showed higher number of spikelets per panicle, but showed lower yield due to lower number of panicle per m 2 . Rice varieties showed different nitrogen uptake ability and NUE at different nitrogen level. Plant breeders and agronomist should take advantage of the significant variations and relationships among grain yield, NUpE, and NUE.

Although much effort has been made to find agronomically important loci in the soybean plant, extensive linkage disequilibrium and genome duplication have limited efficient genome-wide linkage analyses that can identify important regulatory genes. In this respect, recombination block-based analysis of cultivated plant genomes is a potential critical step for molecular breeding and target locus screening. We propose a new three-step method of detecting recombination blocks and comparative genomics of bred cultivars. It utilizes typical reshuffling features of their genomes, which have been generated by the recombination processes of breeding ancestral genomes. To begin with, mutations were detected by comparing genomes to a reference genome. Next, sequence blocks were examined for likenesses and difference with respect to the reference genome. The boundaries between the blocks were taken as recombination sites. All recombination sites found in the cultivar set were used to split the genomes, and the resulting sequence fragments were named as core recombination blocks (CRBs). Finally, the genomes were compared at the CRB level, instead of at the sequence level. In the genomes of the five Korean soybean cultivars used, the CRB-based comparative genomics method produced long and distinct CRBs that are as large as 22.9 Mb. We also demonstrated efficiency in detecting functionally useful target loci by using indel markers, each of which represents a CRB. We further showed that the CRB method is generally applicable to both monocot and dicot crops, by analyzing publicly available genomes of 31 soybeans and 23 rice accessions.

Sea Buckthorn (Hippophae rhamnoides L.) has been used in traditional medicine for the treatment of cough, indigestion, circulatory problems and pain. The associated anti-inflammatory effect of this agent is achieved via the inhibition of Nf-kB signaling, a property that has been demonstrated to effectively control the symptoms of various skin disorders, including atopic dermatitis. Accordingly, the purpose of this study was to assess the efficacy of Sea Buckthorn in reducing the production of lipopolysaccharide (LPS) activated nitric oxide (NO) by inhibiting the Nf-kB pathway, as measured by the symptoms of atopic dermatitis (AD) occurring secondarily to inflammation and immune dysregulation. Our data demonstrate that Sea Buckthorn significantly decreased the LPS-induced production of NO (p〈0.001). Atopic dermatitis was induced by repeated application of 2,4-dinitrochlorobenzene to the dorsal skin of mice. Topical application of 5% Sea Buckthorn extract improved the symptoms of AD, specifically reducing disease severity scores, scratching behaviors and epidermal thickness. When compared to the control group, animals treated with Sea Buckthorn exhibited increased serum IL-12 levels and decreased serum TNF-α, IL-4 and IL-5 levels. Such a modulation of biphasic T-helper (Th)1/Th2 cytokines may result in a reduction in serum IgE levels. Our findings suggest that mechanism of action of Sea Buckthorn in the treatment of AD is associated with a marked anti-inflammatory effect as well as an inhibition of Th2-mediated IgE overproduction via the modulation of biphasic Th1/Th2 cytokines. Such results suggest that topical Sea Buckthorn extract may prove to be a novel therapy for AD symptoms with few side effects.

Resequencing data is actively used for searching QTL or analyzing genetic diversity in the crops. However, the complexity of genome, caused by genome duplication, limits the utility of genome-wide association studies and linkage analyses to identify genes that regulate agronomically valuable traits. Here, we propose a comparative genomics approach based on core or common variation-based recombination blocks (CRB) using single nucleotide variation (SNV) density information. We found that the soybean genomes are assembled with long and distinct CRBs as large as 10Mb. CRB-based comparative genomics enabled us to accurately identify recombination blocks at the whole-chromosome level. We identified the Ih locus that determines the yellow hilum color in soybeans using CRB-based mapping with representative indel markers. These results suggest that the CRB-based comparison method is a promising platform for molecular breeding and map-based cloning.

This experiment was carried out to investigate the appropriate chilling requirements for breaking dormancy by treating the dormant plant of Hanabusaya asiatica with low temperature (4℃) for different time periods. The rates of sprouting and flowering were higher with longer treatment periods at low temperature. In addition, the growth and flowering of the plant were better when it was potted after treatment at a low temperature for 90 days. The abscisic acid levels and polyphenoloxidase activity of the dormant plant increased during the low temperature treatment, reached a climax 90 days and decreased thereafter. The catalase activity was the lowest after the low temperature treatment for 90 days and increased subsequently. The peroxidase activity increased and showed a sharp rise after the low temperature treatment for more than 90 days. Considering the physiological activities of the enzymes, the changes in the abscisic acid levels, and the characteristics of growth and flowering after sprouting of the plant, the appropriate cold periods required for breaking dormancy could be 90 days.

Butyltin compounds in seawater, sediments and organisms inside Jeju Harbor of Jeju Island, Korea, were quantitatively determined to assess the extent of contamination and to evaluate the bioaccumulation in Thais clavigera (gastropod), Liolophura japonica and Cellana nigrolineata. In addition, imposex in T. clavigera was used to assess the sublethal effect of tributyltin compounds and its usage as a specific biomarker for these compounds was also studied. The main species in seawater was tributyltin (TBT) in March and June, monobutyltin (MBT) in August, but in sediments and organisms it was dibutyltin (DBT) and TBT, respectively, irrespective of survey time. TBT, the most toxic to aquatic organisms among BTs, was found at concentrations which were sufficient to have a serous effect on the sensitive organisms upon chronic exposure. The high correlations (r>0.7) between BTs indicated that DBT and MBT were mainly degraded from TBT based on antifouling paints and their sources were negligible. The sedimentary organic matters have little influence on the distribution of BTs in marine environment and the lipid content in T. clavigera showed a significant correlation with TBT concentrations. Measurement of imposex T. clavigera was expected to be a very helpful tool for preliminary survey of BTs prior to trace analysis of those.

The objective of this study was to rapidly evaluate fatty acids in a collection of foxtail millet (Setaria italica (L.) P. Beauv) of different origins so that this information could be disseminated to breeders to advance germplasm use and breeding. To develop the calibration equations for rapid and nondestructive evaluation of fatty acid content, near-infrared reflectance spectroscopy (NIRs) spectra (1104-2494 nm) of samples ground into flour (n=100) were obtained using a dispersive spectrometer. A modified partial least-squares model was developed to predict each component. For foxtail millet germplasm, our models returned coefficients of determination (R2) of 0.91, 0.89, 0.98 and 0.98 for strearic acid, oleic acid, linoleic acid, and total fatty acids, respectively. The prediction of the external validation set (n=10) showed significant correlation between references values and NIRs values (r2=0.97, 0.91, 0.99 for oleic, linoleic, and total fatty acids, respectively). Standard deviation/standard error of cross-validation (SD/SECV) values were greater than 3 (3.11, 5.45, and 7.50 for oleic, linoleic, and total fatty acids, respectively). These results indicate that these NIRs equations are functional for the mass screening and rapid quantification of the oleic, linolenic, and total fatty acids characterizing foxtail millet germplasm. Among the samples, IT153491 showed an especially high content of fatty acids (84.06 mg g-1), whereas IT188096 had a very low content (29.92 mg g-1).

This study was performed at Rice and Winter Cereal Crops Department of NICS during 2007 and 2008 to investigate the characteristics of rice leaf emergence and to obtain basic data which can be used for rice growth simulation model by which we can forecast rice growth stage and heading date accurately under different cultivars, transplanting date, and climatic conditions. To confirm leaf emergence rate according to rice maturing ecotype, we surveyed the leaf emergence rate and heading date of Unkwangbyeo, Hwayoungbyeo and Nampyeongbyeo which are early maturing, medium maturing and medium-late maturing cultivars, respectively, according to seedling raising duration and transplanting time. When seedling duration was 15 days, the growth duration between transplanting time and completion of flag leaf emergence on main culm were 51.5~78.3 days in Unkwangbyeo, 55.3~87.9 days in Hwayoungbyeo and 58.4~98.4 days in Nampyeongbyeo, respectively. When seedling duration was 30 days, they were 50.1~75.5 days in Unkwangbyeo, 52.4~84.7 days in Hwayoungbyeo and 56.4~93.8 days in Nampyeongbyeo, respectively. As transplanting time delayed, the emerged leaf number after transplanting decreased in all rice cultivars. The cumulative temperature between transplanting time to completion of flag leaf elongation on main culm were 1,281~circC~1,650~circC in Unkwangbyeo, 1,344~circC~1,891~circC in Hwayoungbyeo and 1,454~circC~2,173~circC in Nampyeongbyeo, respectively. Leaf emergence rate on main culm were precisely represented by equation, y = y0 + a / [1 + exp( - (x - x0 ) / b)]c, when we used daily mean temperature as variable.
