        The present study was conducted to investigate the effect of different heights from liquid nitrogen (LN2) vapor on sperm motility and morphology after frozen-thawing. Two ejaculates were collected from 2 fertile Hanwoo bulls (A and B) by using artificial vagina at Hanwoo Research Institute. After collection, ejaculates were transferred to laboratory immediately and diluted with semen extender (Optixcell, France). Sperm dilutions were extended to a final concentration of 40 x 106 sperm/ml, and cooled at 4°C for 4 h and loaded to 0.5 ml straws. The straws were divided into 2 groups. Straws were placed in 3 or 9 cm of LN2 vapor for 14 min and then plunged into LN2 tank and cryopreserved until evaluation. Sperm motility and motility parameters (total motility, VSL with 25μm≥, VCL, VSL, VAP, LIN, STR, WOB, ALH and BCF) were evaluated by sperm class analysis (SCA, IVOS, Spain) after frozen-thawed. In bull A, 3cm group showed higher percentages of total motility, VSL with 25μm and VAP compared those with 9cm group (98.0 vs. 93.4%, 62.4 vs. 54.0% and 98.6 vs. 93.2%, 3 vs. 9 cm, irrespectively; p<0.001). In bull B, frozen-thawed sperm of 3cm group showed higher percentages of VSL with 25μm, VCL, VSL, VAP and BCF compared with those of 9cm group (43.5 vs. 26.0%, 123.8 vs. 111.6 μm, 62.9 vs. 57.3 μm and 81.5 vs. 72.5 μm; 3 vs. 9 cm, irrespectively; p<0.001). The viability and acrosomal integrity of spermatozoa were evaluated by Trypanblue/Giemsa staining method divided into 4 groups; live and intact acrosome (LIA), live and damaged acrosome (LDA), dead intact acrosome integrity (DIA), dead damaged acrosome (DDA). In bull A, frozen-thawed sperm of 3 and 9cm groups showed no significant difference in LIA, LDA, DIA and DDA. In bull B, 3 cm group showed higher LIA and lower DIA compared with those of 9 cm group (73.2 vs. 23.7% and 23.7 vs. 32.2%, 3 vs. 9 cm, irrespectively; p<0.001). We suspected that 3 cm vapor on LN2 vapor might be affected positively spermatozoa viability and acrosomal integrity compared with 9 cm group. In conclusion, semen freezing procedure in the present study will improve sperm quality after frozen thawing.

        Although assisted reproductive technology (ART) has been developed in many mammalian species including cows, the only embryo preservation technology that is available is cryopreservation. In the present study, small molecules were used to preserve embryos at room temperature. The basic medium for embryo preservation consisted of 1% BSA non-cryopreservation medium (BNC) instead of fetal bovine serum (FBS). To maintain survival and prevent damage during embryo storage, three candidate small molecules were selected—CHIR99021, Y-27632 and Thiazovivin—and their concentrations were optimized. Then, the embryos in the small molecule supplemented preservation medium were stored at room temperature. The viability and hatching rate of embryos stored at 10°C were greater for Y-27632-BNC and CHIR99021+Y-27632-BNC compared to BNC. However, the rate was lower for Thiazovivin-BNC compared to BNC. Although there were no surviving embryos after storage at 20°C, the viability and hatching rate of embryos significantly increased in Y-27632-BNC and CHIR99021+Y-27632-BNC compared to BNC. The mechanism by which small molecules enhance survival of embryos during storage was investigated, and expression of heat shock protein 70 was observed to increase. The findings of this work may be useful in improving ART in the agricultural field.

        Bourbon is the major coffee variety grown in Rwanda. Fully washed bourbon produced in Rwanda is a nascent specialty coffee and no investigation of its quality characteristics has been reported. Thus, this study aimed at providing information about intrinsic flavor characteristics and identifying the most discriminating aroma compounds of this coffee from the major growing areas in four provinces (considered by this study as geographical sub-regions) in comparison with Arabica coffee from Ethiopia and Brazil using electronic nose and electronic tongue. Samples were discriminated and characterized by performing a principal component analysis (PCA). A discriminant factorial analysis (DFA) model was also used to predict the geographic origin of coffee samples based on their flavor characteristics. A similarity was observed between flavor characteristics of coffee samples from western and southern sub-regions as well as between northern and eastern sub-regions of Rwanda. Twenty one most discriminating aroma compounds were found but the identified and confirmed ones are: Butan-2-one, Acetic acid, n-Butanol, 2, 3- Pentanedione, Ethyl-2-methybutyrate, Dimethylsulfide, 2-Prpanol, 2-Methylfuran, Ethylacetate, Ethyl propionate and Heptanal. A DFA model using e-nose was successful in predicting the geographic origin of coffee samples but not with e-tongue. This means that aroma can reliably be used to predict the geographic origin of coffee samples than their taste characteristics. Further investigation is required to test the possibility of origin recognition based on taste characteristics.

        에너지 음료는 카페인을 주성분으로 타우린, 비타민 같은 다른 energy-enhancing 성분을 함유하고 있다. 미국과 유럽에서는 글루쿠로노락톤이 에너지 음료에 첨가될수 있으나, 국내에서 의약품으로는 허가되어 있다. 따라서 식품 첨가물로는 그 사용이 금지 되어 있어, 지속적으로 수입 및 유통 음료에서 시험검사를 하여 규제하고 있다. 현재 분석법으로 사용하는 LC-PDA 법은 복잡한 유도체화 과 정을 거치고, 음료 중에 당류들이 위양성 결과를 나타내 기도 한다. 이런 기존 방법의 단점을 개선하기 위해 HILICESI- MS/MS (hydrophilic interaction liquid chromatography coupled to electrospray ionization tandem mass spectrometry) 를 이용한 분석법을 개발하고, 선택성, 직선성, 검출한계, 정량한계, 정밀도, 정확성, 재현성에 대하여 분석법 유효성 검증을 수행했고, AOAC, EURACHEM 가이드라인에 부합되는 결과를 얻었다.

        To replace methyl bromide (MeBr) due to its ozone depletion properties and the life-threatening to related worker as well as fumigators, ethyl formate (EF) fumigation on exported and imported fruits and vegetables has been developed by QIA in Korea since 2005. The benefits of using ethyl formate (EF) fumigant is considered as the safest practice among the commercially available chemical alternatives. The issues was that the cylinderized EF with carbon doxide (CO2) is not enough to extend their applications in terms of fumigation cost. We have developed the cost-effective liquid EF applications with nitrogen gas and demonstrated in quarantine trials (10m3) on imported bananas. We showed the new 35 gm-3 of EF applications with inert gas (nitrogen), were completely control all stage of citrus mealybug (Planoccous citri) for 4hr fumigation at 13°C. These applications meet the quarantine guidelines and protocols in terms of efficacies to target pest and safety concern as well as satisfactions of consumers showing no damages to imported bananas. Extensively, this cost effective EF technology could be acceptable to other fruit and vegetable commodities to reduce the fumigation costs and green house gas (CO2).

        The green peach aphid, Myzus persicae, is one of the most important pests of vegetable crops worldwide. This species cause direct damage by feeding on plant nutrients and indirect damage as transmits many virus vectors. The control of aphid has relied on the use of chemical insecticides and their intensive use over many years has led to the development of resistance. To surmount or avoid these problems, we screened the entomopathogenic fungi against green peach aphid and evaluated their virulences. Several entomopathogenic fungal isolates were selected with high virulence to green peach aphid. Additionally, the antimicrobial activity of the selected fungal isolates was tested and analyzed to phytopathogens. Consequently, these entomopathogenic fungi would be used effectively for dual control agents for the green peach aphid and phytopathogen.

        To investigate unknown molecualr and cellular mechanisms underlying polyphenisms in the migratory locusts, we performed whole transcriptome analysis from 12 different groups. Significantly differentially expressed coding and long non-coding (LNC) RNAs were present according to habitats, phases, and genders. Those differentially expressed coding and LNC RNAs were confirmed by quantitative-realtime-reverse transcriptase PCR. Those identified coding and LNC RNAs could be a target for developing environmental friendly management tools in the near future.

        Polyphenism is one of the most intriguing phenomenon observed from various insect species. Polyphenism is changing morphological, behavioral, and physiological phenotypes without changing genome informations. Therefore, polyphenism is thought to be controlled by epigenetic methods. Epigenetic controls are including DNA methylation, histone acetylation, small RNAs, long non-coding RNAs, and protein phosphorylation. To investigate the mechanism involved in polyphenism, various genomic researches have been completed. However, metabolite changes in different phases have not been reported yet. Thus, we studied the metabolite changes and found the changed metabolites. These studies will make an important contribution to reveal the difference between gregrious and solitary phases and reveal eco - friendly control techniques.

        The leafhopper subfamily, Typhlocybinae (Hemiptera: Auchenorrhyncha: Cicadellidae), caused direct damage by sap-sucking or indirect damage by transmission of viruses or diseases to the host plants, so Typhlocybinae is one of the most important insect pests in agriculture, urban ecology, forest, etc. in the world. However, their identifications are very difficult due to small size and morphological similarities, and their exact biological information such as host plants and life cycle are not reported yet for most species. So, we construct an integrated biological database that presents accurate diagnostic methods (illustrated key characters, DNA barcoding data), patterns and seasons of occurrences, and some ecological photographs such as dorsal habitus of specimens and plant damages

        The subfamily Iassinae Walker belongs to the family Cicadellidae (Hemiptera: Auchenorrhyncha). A total of 124 genera with 1,999 species have been described in the world. To date, two genera, Batracomorphus Lewis and Iassus Fabricius, and 5 species of the subfamily have been recorded in Korea. In this talk, one new species and one new record of the genus Batracomorphus and one new species and two new records of the genus Iassus from Korea are presented. And we also provide biological information such as host plants and distributional records of the Korean Iassine species.

        Maruca vitrata larvae feed on flowers and pods of several leguminous crops, and can severely reduce seed yield. Adult emergence, mating and oviposition behaviors were observed in a 15h/9h=light/dark and 25℃ condition as a base study for monitoring. Emergence occurred mainly during the first five hours of scotophase. Mating occurred from the two days after emergence, and the mated females started to lay eggs from the next day. A maximal mating rate was observed in the night of five days. Mating occurred significantly more often during the time from 2 hours before to 3 hours after lights-off, but older females mated more frequently during the photophase.         Dermanyssus gallinae is parasitic to chicken, and they cause many damages such as disturbing sleep reducing for body weight and egg production by blood-sucking. To develop acaricide against D. gallinae, The acaricidal activity of 40 species plant extract were examined. Cnidium officinale extracted by Me-OH showed 82.0% acaricidal activity after treated 48 hrs at 4,000 ppm. The hexane fraction showed 92.4% mortality against D. gallinae at 48 hrs at 2,000 ppm. Purification of the biologically active constituents from the hexane extraction with acaricidal activity was done using silica gel open column chromatography and HPLC. H1122 fraction gave 80.9% mortality to D. gallinae at 400 ppm after treated 48 hrs. H1122 fraction was analyzed by GC-MS and NMR.

        Utilizing a unique capability of AKARI that allows deep spectroscopy at 2.5 - 5.0 μm, we performed a spectroscopy study of more than 200 quasars through one of the AKARI mission programs, QSONG (Quasar Spectroscopic Observation with NIR Grism). QSONG targeted 155 high redshift (3:3 < z < 6:42) quasars and 90 low redshift active galactic nuclei (0:002 < z < 0:48). In order to provide black hole mass estimates based on the rest-frame optical spectra, the high redshift part of QSONG is designed to detect the Hα line and the rest-frame optical spectra of quasars at z > 3:3. The low redshift part of QSONG is geared to uncover the rest-frame 2.5 - 5.0 μm spectral features of active galactic nuclei to gain useful information such as the dust-extinction-free black hole mass estimators based on the Brackett lines and the temperatures of the hot dust torus. We outline the program strategy, and present some of the scientific highlights from QSONG, including the detection of the Hα line from a quasar at z > 4:5 which indicates a rigorous growth of black holes in the early universe, and the Brβ-based black hole mass estimators and the hot dust temperatures (~ 1100 K) of low redshift AGNs.

        국내에서 개발중인 콘크리트 저장용기는 방사성 물질의 격납 건전성을 유지하기 위하여 내부에 캐니스터를 포함하고 있다. 본 논문에서는 콘크리트 저장용기 내부 캐니스터의 뚜껑 용접시, 용접시간 저감과 이에 따른 캐니스터 용접부의 구조적 건 전성을 확보하기 위한 방안으로, 정상, 비정상 및 사고조건에서 캐니스터 용접부 균열을 진전시키는 하중에 의해 발생되는 균열 깊이를 분석하여, 용접부의 최대 허용결함깊이를 평가하였다. 정상, 비정상 및 사고조건에서의 구조해석은 범용 유한 요소해석 프로그램인 ABAQUS를 사용하였으며, 허용결함깊이는 ASME B&PV Code Section XI에 따라 막응력과 조합하중 에 대해 평가하였다. 평가결과 콘크리트 저장용기의 캐니스터 용접부의 허용결함깊이는 18.75 mm로 평가되었으며, 이는 NUREG-1536에서 권고하고 있는 임계결함깊이를 만족하고 있는 것으로 나타났다.

        Background: The wall squat is considered an effective exercise because it can reduce the knee load and prevent excessive lumbar movement. However, the relationship between wall squat performance and strength of knee extensors and hip extensors remained unclear. Objects: The purpose of this study was to compare the strengths of the knee extensors and hip extensors between groups with low and high wall squat performance. Method: Nineteen males (low performance group: 9 subjects, high performance group: 10 subjects) participated in this study and performed wall squats. The subjects who were performing less than 30% of the average wall squat count were classified into the low wall squat performance group (less than or equal to 4 times) and the subjects who performed more than 30% of the average wall squat count were classified into the high wall squat performance group (greater than or equal to 8 times). Knee extensor and hip extensor strength were measured with a strength measurement system. An independent t-test was used to compare the strengths of the knee extensors and hip extensors between the groups with low and high wall squat performance. Results: The ratios of knee extensor and hip extensor strength to bodyweight were greater in the high wall squat performance group than in the low wall squat performance group (knee extensors: p<.001; hip extensors: p=.03). In the high- and low-performance groups, the ratios of knee extensor strength to bodyweight were 42.74±5.72 and 30.76±8.54, respectively, and the ratios of hip extensor strength to bodyweight were 31.95±10.61 and 20.66±11.25, respectively. Conclusion: Our findings suggest that knee extensor and hip extensor strength are needed for high wall squat performance. Thus, exercise to increase the knee and hip extensors strength can be recommended to improve squat performance.
