        Method for Protect of the river levee was method for installing concrete revetment block and concrete mat method in Korea. But this method is non-environmental approach because the vegetation can not take at all. To solve these problem, the method has been applied using porous vegetation concretes. Porous vegetation concrete has filler technique to provide water retention and nutrient a into the porous interior in order to facilitate plant growth. But filler used increasing the cost and the construction period. Therefore in order to not use a filler, a high absorption rate and good absorption capacity needed aggregate. Current, Blast furnace slag aggregate has been used in architectural and civil engineering field as an alternative aggregate resources. Blast furnace slag aggregate is high absorption rate and lighter in weight per unit. This study evaluated the absorption capacity of the blast furnace slag aggregate. Phosphorus Sorption experiment was carried out to produce a cylindrical acrylic(diameter 11cm, height 90cm). There filling the aggregate, for supplying the test solution was using as a pump from bottom to top. And The water of having passed through the solution was analyzed. Results, blast furnace slag aggregate showed absorption capacity of 64~77% of PO43-P and T-P. Crushed aggregate show absorption capacity of 6~24% of PO43-P and T-P. Blast furnace slag aggregate showed excellent result of good absorption capacity compared to the crushed aggregate showed excellent result.

        This study aims to increase the organic carbon, nitrogen and phosphorus removal efficiencies than the conventional method and meet higher effluent water quality standards by co-culture with bacteria in activated sludge in the aeration tank. By the co-culture in mixotrophic conditions through metabolic characterization of Scenedesmus dimorphus and bacteria in the aeration activated sludge, assessed how the impact on the organic matter, nitrogen and phosphorus removal rate. In addition, the study was to determine if it is possible to supply the oxygen necessary for the bacteria through the photosynthesis of algae without aeration. To test the synergistic effects of the co-culture, we compared co culture with S. dimorphus - only, Bacteria - only as a controls. In the co-culture condition inoculation ratio is based on the TSS concentration S. dimorphus and bacteria 1 : 5 (w / w), 5 : 5 (w / w) at a ratio. The growth rate of S. dimorphus - only condition was higher, which was 10 times and 2 times lower compared to B : A (5 : 1), B : A (5 : 5) respectively. Organic carbon removal rate of S. dimorphus - only condition was lower than other conditions, the remaining conditions were consumed at a similar rate. In the co-culture of Mixed bacteria and S. dimorphus, ammonium and phosphate removal rate has been high. But, nitrate removal rate showed a tendency to decrease compared to the Bacteria-only condition. When considering the Ammonium, nitrate and phosphate removal rate, N, P removal efficiency which is most excellent in terms of bacteria, such as S. dimorphus ratio (5 : 5) were co-cultured in the conditions. DO was maintained at 3 mg/L or more under all conditions. So, implying co-culture has the potential to decrease aeration costs.

        Abscission is an important developmental process used to shed organs such as leaves, flowers and fruits. Despite the detailed characterization of growth dynamics and hormonal balance during the early steps of fruit development, the molecular aspects remain unclear. Abscission of young fruit occurs by separation of cells in anatomically distinct regions between the pedicel and junction. Differences of gene expression between central pedicel and lateral pedicel were investigated by NGS. Partial cDNAs from 15 clones from both the central pedicel and lateral pedicel were selected for nucleotide sequence determination and homology searches, and 12 clones were subsequently selected for further analysis. In preliminary series of Real Time PCR analysis, 9 genes were confirmed as showing a higher expression level in lateral pedicel than in central pedicel. Many of these genes are expressed in a central or lateral pedicel in specific manner, and the expression profiles of the representative genes were confirmed. To clarify the mechanism of MdIAA14 transcription factor gene underlying abscission zone development, we are investigating the expression patterns between central and lateral pedicels in different apple cultivar using real-time PCR and constructing the vector for transformation into apple.

        The precise, fast, and cost-effective identification of important fruit crop cultivars is essential for practical breeding and plant breeder’s rights. Traditional methods for identification of persimmon cultivars are based on the evaluation of sets of morphological characteristics. However, the identification using only morphological traits is difficult to distinguish among genetically closely related cultivars. This study was conducted to develop more reliable DNA markers for identification of the 32 persimmon cultivars in Korea and Japan. In total, 309 random amplified polymorphic DNA (RAPD) markers were identified using 40 different random primers. The 4 (OPP-08) to 14 (UBD159) polymorphic bands were detected with an average of 7.7. The resulting 57 RAPD fragments were selected, and their sequences were determined for developing sequence-characterized amplified region (SCAR) markers. As a result, 15 of 57 RAPD fragments were successfully converted to SCAR markers. A single polymorphic band of the same size as the RAPD fragments or smaller DNA fragments were amplified depending on primer combinations in the 15 SCAR markers. Among these markers, a combination of eight SCAR markers (PS225_200, PSN05_420, PSF13_523, PSN11_540, PS372_567, PS485_569, PSP08_635, and PS631_735) provided sufficient polymorphisms to identify 32 persimmon cultivars depending on number and size of amplicons. These newly developed markers will be useful as a fast and reliable tool to identify persimmon cultivars.

        In higher plants, formate dehydrogenase (FDH; EC 1.2.1.2) is a NAD-dependent enzyme that catalyzes the oxidation of formate into carbon dioxide. Although FDHs have been reported to be induced by various abiotic stresses, the function of FDHs in biotic stress is rarely known. In this work, we describe the identification of pepper mitochondrial FORMATE DEHYDROGENASE1 (CaFDH1) as a positive regulator of cell death and disease resistance. Transient expression of CaFDH1 in Nicotiana benthamiana leaves caused hypersensitive response (HR)-like cell death, where D-isomer specific 2-hydroxyacid dehydrogenase signature was crucial for the induction of the cell death. Localization analysis using green fluorescence protein showed that CaFDH1 targeted to the mitochondria with the mitochondria targeting sequence at N-terminal region. However, the mitochondrial localization of CaFDH1 is not essential for the induction of HR-like cell death, because the cytosolic CaFDH1 could elicit cell death response. Silencing of CaFDH1 in pepper significantly compromised the cell death response and salicylic acid (SA) levels, but enhanced growth of Xanthomonas campestris pv. vesicatoria. In contrast, transgenic Arabidopsis overexpressing FDH1 exhibited enhanced resistance to Pseudomonas syringae pv. tomato. Taken together, these data suggest that CaFDH1 has a role in HR cell death and defense response to microbial pathogens.

        Mitogen-activated protein kinase (MAPK) signaling cascades play critical roles in various cellular events including abiotic/biotic stress responses, innate immunity, hormone signaling and cell specificity in plants. The MAPK-mediated stress and ethylene signaling are recently known to be involved in nitogen-fixing symbiotic interactions; however, the biological role of MAPK for nodule development in legume plants is largely unknown. We here elucidated that MtMKK5-MtMPK3/6 cascade negatively regulate the nitrogen fixing nodule formation in Medicago truncatula. MtMKK5, an ortholog of SIMKK, overexpression significantly reduces the nodule formation in M. truncatula roots. MtMKK5 directly activates MtMPK3/6 by phosphorylation on the TEY motif within the activation loop in the cytoplasm, which might link to EFD as a negative regulator for nodule formation. EFD has a putative MAPK phosphorylation Thr residue and could be a target of the activated MtMPK3/6 in the nucleus. Consistently, a MAPK specific inhibitor U0126 enhances nodule formation and confers similar nodule phenotypes to the efd-1 mutant such as lower proliferation and differentiation to symbiotic tissues. Our works thus reveal a key negative signaling module mediated by MtMKK5-MtMPK3/6-EFD for symbiotic nitrogen fixing nodule organogenesis.

        To verify the sex steroids which are involved in oocyte maturation of the blacktip grouper, , we incubated vitellogenic oocytes (0.41 and 0.50 mm in average diameter) in the presence of exogenous steroid precursor (-hydroxyprogesterone). Steroids were extracted, separated and identified by thin layer chromatography. The major metabolites produced were androstenedione, estradiol-, estrone and progestogens. Progestogen metabolites in the oocytes of 0.50 mm were more abundant than those of 0.41 mm. Also, we investigated the effects of human chorionic gonadotropin (HCG; 5, 50 and 500 ), -dihydroxy-4-pregnen-3-one () and -trihydroxy-4-pregnen-3-one (; 5, 50 and 500 , respectively) on oocyte maturation. In the oocytes of 0.41 mm, treatment with 50 IU HCG stimulated GVBD () compared with controls (, <0.05). In the oocytes of 0.50 mm, treatment of (50 and 500 ) stimulated GVBD ( and , respectively) compared with controls (, <0.05). Treatment with 500 IU HCG also stimulated GVBD () compared with controls (<0.05). Taken together, these results suggested that both HCG and were effective on in vitro oocyte maturation and may act as a maturation inducing hormone in blacktip grouper.

        The ripening behavior of three apple cultivars, ‘Tsugaru’, ‘Hongro’ and ‘Fuji’ was distinctive and the involvement of POLYGALACTURONASE(PG) in the fruit softening process was confirmed to be ethylene dependent. Fruit softening is genetically coordinated by the action of several cell wall enzymes, including PG which depolymerizes cell wall pectin. Also, loss of firmness is associated with increasing of the ripening hormone, ethylene. In this work, climacteric ripening of three apple cultivars, Tsugaru, Hongro and Fuji, producing different ethylene levels and ripening responses, was examined. Correspondingly in Fuji, a linear and basal ethylene level was observed over the entire period of measurements, and Tsugaru and Hongro displayed a typical climacteric rise in ethylene production. Transcript accumulation of genes involved in ethylene biosynthesis (MdACS3 and MdACO1) and MdPG1 was studied in Tsugaru, Hongro and Fuji cultivars. Expression of MdACO1 transcripts was shown in all three ripened apple fruits. However, the MdACS3 and MdPG1 were transcribed differently in these cultivars. Comparing the MdPG1 of ‘Tsugaru’, ‘Hongro’ and ‘Fuji’, structural difference was discovered by genomic Southern analysis. Overall results pointed out that MdACS3 and MdPG1 play an important role in regulation of fruit ripening in apple cultivar.

        The Hardy Kiwifruit (A. arguta (Sieb. & Zucc.) Planch. ex Miq.) is one of the valuable species due to their edible fruit, high content of nutritious substances, especially abundant of vitamin C, and distinctive flavor and medicinal usage. In order to develop a new A. arguta cultivar with larger fruit and high yielding, 168 candidate plants were collected from wild population in 12 locations distributed 4 provinces (Chungbuk, Gangwon, Gyeongnam, Jeonbuk) from 1985 to 1987. A clone bank that contained highly productive, superior genotypes of A. arguta was assembled in 1988, and 32 excellent clones were selected in 1996 through the clonal test for the growth and fruiting. Evaluation on these clones was done for the fruiting characteristics, yield trial, and major agronomic traits, such as Average Fruit Length (FL) and Width (FW), and Weight of Fruit (WF) and Individual Yields (IY), during 6 years (1997~2002). After the final selection, a new A. arguta cultivar, “Sae-Han” with larger fruit and high yielding was registered as a new variety denomination and certificated variety production and merchandising in 2006 (Table 1). “Sae-Han” is a tetraploid plant and has ellipsoid fruit shape, and has particularly large fruit size (Fig. 1, Table 3). The major agronomic traits of “Sae-Han” showed the large values and selection effect with an average of 43.6 mm (FL), 36.1 mm (FW), 29.4 g (WF), and 17.5 kg (IY), which are 35.0%, 19.9%, 81.5%, and 124.4% compared to the mean of 29 sample trees, respectively (Table 2).

Finally, we have selected the new A.arguta cultivar, “Chil-Bo” with large fruits and high yielding and registered as a new variety denomination and certificated for variety production and merchandising in 2006 (Table 1). This cultivar is characterized by a tetraploid plant and spheroid type in the fruit shape, and is particularly characterized by large fruit size (Fig. 1). The major agronomic traits of this cultivar showed the large selection effect with an average of 28.4 mm (FL), 36.9 mm (FW), 18.2 g (WF), and 24.2 kg (IY), which are -12.1%, 22.6%, 12.4%, and 210.3% compared to the mean of 29 sample trees, respectively (Table 2).
