        Background : Although ginseng has various bioactive compounds in it, there is lack of study on the variations of bioactive compounds in ginseng according to the cultivation soil and the applied fertilizer types (or amount). Therefore, this study aims to examine the variations of 37 fatty acids (FA) and 8 vitamin E (Vit-E) vitamers in 6-year-old ginseng root cultivated in different soil types with different fertilizers regimes. Methods and Results : The profiling of 37 FAs and 8 Vit-E vitamers in 6-year-old ginseng roots was measured by gas chromatography coupled with a flame ionization detector, and then these results were statistically analyzed with chemometrics. The FA and Vit-E content in ginseng roots varied significantly with respect to soil cultivation conditions due to organic fertilizer types and amounts used. Unsaturated FA in ginseng is approximately 2.7 fold higher than the saturated FA. Linoleic, palmitic, and oleic acids were the most abundant FAs found in the ginseng roots. Also, the major Vit-E vitamer found in ginseng root is α-tocopherol. In particular, the application of rice straw compost or food waste fertilizer was increased to create nutritionally desirable FAs and bioactive Vit-E in ginseng root. In addition, phytonutrient profiling coupled with chemometrics can be used to discriminate the cultivation conditions of ginseng. Conclusion : This study extends our understanding about the variations of FA and Vit-E in ginseng root depending on cultivation conditions. Hence, these results can be useful as basic information for reliable ginseng production containing high amounts of phytonutrients in a paddy-converted field.

        Background : Corrosion is one of the most devastating problems faced by most industries. Mild steel has played a vital role in various fields due to the excellent mechanical properties of mild steel such as low density, high strength-to-weight ratios, good environmental stability, high thermal conductivity, and corrosion resistance. Methods and Results : The total phenolic contents (TPC) and total flavonoid contents (TFC) of the methanolic extract of C. grandiflora and R. verniciflua leaf have been examined, and its corrosion inhibition performance was investigated by weight loss and electrochemical impedance spectroscopy (EIS) and polarization measurements. The surface morphology of mild steel was analyzed by scanning electron microscopy (SEM) equipped with energy dispersive X-ray spectroscopy (EDX) and by atomic force microscopy (AFM). The percentage composition of polyphenolic compounds was found to be higher in C. grandiflora and R. verniciflua plant extracts, and it was proved to be a superior, eco-friendly, and anti-corrosive inhibitor for mild steel in 1M of H2SO4. The Tafel polarization studies indicate that the plant extract is a mixed-type inhibitor. Scanning electron microscopy/energy -dispersive X-ray spectroscopy (SEM-EDS), and atomic force microscopy (AFM) studies confirmed the formation of a protective film on the metal surface. The corrosion inhibition of the C. grandiflora and R. verniciflua plant extracts was characterized by Fourier transform infrared (FT-IR), UV-visible spectra, and wide-angle X-ray diffraction (XRD) studies; these show the strong interaction between the metal surface and the inhibitor. Conclusion : The methanolic extract was prepared the two different plants like C. grandiflora, and R. verniciflua was studied the corrosion inhibition on the mild steel specimen in acidic medium through various methods involving weight loss measurements, EIS, and potentiodynamic polarization. The results shows that the C. grandiflora, and R. verniciflua plant extracts illustrate an effective corrosion inhibitor for mild steel with good anticorrosion properties in acidic environmen

        Sexual dimorphism is the most conspicuous difference between the sexes. This study examines possible sexual dimorphism and the relative growth patterns of morphometric characteristics in the marine medaka, Oryzias dancena for their potential to help differentiate between males and females of this species. The von Bertalanffy growth parameters estimated by a non-linear regression method were L∞=30.2 mm, K=3.22/year, and τ0=-0.05. All 18 characteristics measured showed a difference between males and females from 70 days after hatching. Each of these characteristics were significantly different between sexes (ANCOVA, P<0.05), and the ratio of standard length between sexes showed that males were larger than females for all five morphometric measurements. Fin length measurements were taken for 21 distances of anal fin and 7 distances of dorsal fin between landmarks. There were all differences for all dorsal fin rays between the males and the females and there is significant difference in 70 days after their hatch when the sexual dimorphism is presented. The significant difference (P<0.05) in fin ray for male and female was more greatly seen as they grow. Male marine medaka showed more rapid growth than females, with longer length, dorsal fins and anal fins. Differences in these characteristics will be useful during experiments when it is necessary to differentiate between sexes of marine medaka.

        Background : Some of invasive plants, which were introduced from foreign countries, have caused problems in Korea. Invasion of these invasive plants in the ecosystem threatens the habitat of endemic species, reducing biodiversity, and causing a disturbance in the ecological system. Hypochaeris radicata L. (Asteraceae), the most invasive plants in Korea, particularly in Jeju Island, invade farmland, and autochthonous forest, establishing monocultures and modifying the ecosystem structure. This invasive species has become a serious environmental problem because they displace the indigenous plant species. This study was conducted to evaluate the antioxidantive effects of ethanolic extracts from different parts (root, stem, seed and leaf) of the invasive exotic species Hypochaeris radicata L. Methods and Results : The aim of present study was to estimate the total phenolic and flavonoid contents and to investigate in vitro antioxidant potential of ethanolic leaf, root, seed, and stem extracts of the Hypochaeris radicata. Antioxidant activity was assessed by using 2,2-diphenyl-1-picryl-hydrazyl (DPPH) assay, reducing power activity, [2,2-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid)] ABTS+ assay and ferrous ion chelating activity. The total phenolic and flavonoid contents were also determined and expressed in gallic acid and quercetin equivalent respectively. The results of the study indicate that the ethanolic extracts of the leaf, root, seed, and stem of H. radicata posses significant scavenging activity against DPPH (21.25% for leaf, 34.98% for root, 60.76% for seed and 45.25% for stem at 250 μg/ml each) and ABTS+ radical scavenging activity (14.85% for leaf, 17.40% for root, 35.91% for seed and 24.70% for stem at 250 μg/ml each), reducing power activity (0.178 absorbance at 300 μg/ml for leaf, 0.211 absorbance at 300 μg/ml for root, 0.447 absorbance at 300 μg/ml for seed, 0.276 absorbance at 300 μg/ml for stem). The free radical scavenging and antioxidant activities may be attributed to the presence of adequate phenolic (gallic acid content is 361.92.98 μg/g in leaf, 356.59μg/g in root, 719.72 μg/g in seed and 512.08 μg/g stem) and flavonoid compounds (219.52 μg/g in leaf, 75.67μg/g in root, 281.39 μg/g in seed and 215.66 μg/g stem). This study revealed that the ethanolic extracts of both leaf, root, seed and stem of H. radicata has demonstrated significant antioxidant activity. Conclusion : In conclusion, the present study has demonstrated that Hypochaeris radicata seed ethanol extracts are rich in phenolics and have a strong antioxidant activity and a radical-scavenging action in all of the tested methods. This suggests that Hypochaeris radicata is a good source of natural antioxidants.

        Background : More than 1250 bamboo species, belonging to 75 genera, are distributed all over the world. Sasa quelpaertensis Nakai is a type of bamboo grass widely distributed in Halla mountain, Jeju Island, which has been used as antidiabetic, anti-cancer and anti-inflammatory drugs. In this study, we investigated the antioxidant and antimicrobial activities of Sasa quelpaertensis Nakai leaf extracted with different ethanol concentration and demonstrated the potent bioactivities of the extracts suitable to be used as natural antioxidant compounds or pharmaceutical supplements. Methods and Results : Antioxidant and anti-microbial activities of Sasa quelpaertensis Nakai extracts were studied. At first, different ethanol concentrations (0, 20, 40, 60 and 80%) were compared for determining of the best solvent for extraction of phenolic compounds from Sasa quelpaertensis Nakai. Forty percent Ethanol extract with 990.01±28.9 (mg of gallic acid equivalents/g sample) were the best solvent in the extraction of phenolic compounds. But, 60% ethanolic extracts were highest antioxidant activity appeared such as DPPH radical scavenging (IC50 21.20±0.42 μg/ml), ABTS radical scavenging (IC50 49.85±1.27 μg/ml) and reducing power. However, 80% ethanol extracts showed the strongest SOD like activity. The anti-microbial capacity was screened against Gram positive and Gram negative bacteria, and yeast. Sixty percent and 80% ethanol extracts inhibited the growth of Gram positive bacteria; Bacillus cereus was the most susceptible one with MIC of 125 μ g/ml and 250 μg/ml for the 60% and 80% extracts, respectively. Conclusion : The results of this study show that the extract of Sasa quelpaertensis Nakai can be used as easily accessible source of natural antioxidants and as a possible food supplement or in pharmaceutical industry. However, the components responsible for the antioxidant and antimicrobial activities of both extracts of Sasa quelpaertensis Nakai are currently unclear. Therefore, it is suggested that further works should be performed on the isolation and identification of the antioxidant components in Sasa quelpaertensis Nakai.

        Backgrounds : Pomelo (Citrus grandis Osbeck) is the kind of citrus fruit which is Dicotyledoneae belongs to Rutaceae and special product in Jeju island. According to the previous researches, coumarin, eliocitrin, naringin are identified and these kinds of constituents revealed to be effective as anticancer, antioxidant, and antidiabetic. Until recently, there are many investigations about its functional properties were reported, but investigation about biological activities depend on extraction conditions are not sufficient. Methods and Results : 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was performed by the method of Blois with minor modification. After adding DPPH radical to each sample solutions, the mixtures were incubated in 30 minutes at aphotic place. Then, the degree of scavenging activity was recorded by microplate reader at 490 nm. The scavenging activity was expressed by RC50, which is the concentration of sample solution necessitated to scavenge 50% DPPH radical against negative control. For α-glucosidase inhibitory activity, the method using p-nitrophenyl-α-D-glucopyranoside (pNPG) was applied. To each samples, α-glucosidase and pNPG were mixed and incubated, consequently, Na2CO3 was added to terminate the reaction. Finally, absorbance was read at 450 nm. The same as DPPH radical scavenging activity, the inhibition was explicitly expressed by the amount of sample solution to inhibit 50% α-glucosidase, IC50. The 80% MeOH extract demonstrated the highest radical scavenging activity with 74.32±8.45 μg/ml. α-Glucosidase inhibitory activity was carried out to evaluate the inhibitory activity of sugar digestion and absorption and 60, 80% MeOH extract exhibited 416.35±11.07 μg/ml and 336.57±2.03 μg/ml, respectively. Conclusion : The DPPH radical scavenging and α-glucosidase inhibitory activity were evaluated in this study. The highest DPPH radical scavenging activity was seen in 80% Citrus grandis Osbeck MeOH extract, following 60% MeOH extract exhibited the second highest scavenging activity. Also, 80% MeOH extract showed 336.57±2.03 μg/ml, which was the highest α-glucosidase inhibition among all extracts.

        This study was conducted to develop a fast and efficient screening method to determine the quantity of fatty acid in peanut oil for high oleate breeding program. A total of 329 peanut samples were used in this study, 227 of which were considered in the calibration equation development and 102 were utilized for validation, using near infrared reflectance spectroscopy (NIRS). The NIRS equations for all the seven fatty acids had low standard error of calibration (SEC) values, while high R2 values of 0.983 and 0.991 were obtained for oleic and linoleic acids, respectively in the calibration equation. Furthermore, the predicted means of the two main fatty acids in the calibration equation were very similar to the means based on gas chromatography (GC) analysis, ranging from 36.7 to 77.1% for oleic acid and 7.1 to 42.7% for linoleic acid. Based on the standard error of prediction (SEP), bias values, and R 2 statistics, the NIRS fatty acid equations were accurately predicted the concentrations of oleic and linoleic acids of the validation sample set. These results suggest that NIRS equations of oleic and linoleic acid can be used as a rapid mass screening method for fatty acid content analysis in peanut breeding program.

        Heat shock protein (HSP) 70, the highly conserved stress protein families, plays important roles in protecting cells against heat and other stresses in most animal species. In the present study, we identified and characterized four Hsp70 (RuHSP4, RuHSC70, RuHSP12A, RuGRP78) family proteins based on the expressed sequence tag (EST) analysis of the Korean rose bitterling R. uyekii cDNA library. The deduced RuHSP70 family has high amino acid identities of 72-99% with those of other species. Phylogenetic analysis revealed that RuHsp70 family clustered with fish groups (HSP4, HSC70, HSP12A, GRP78) proteins. Quantitative RT-PCR analysis showed the specific expression patterns of RuHsp70 family members in the early developmental stages and several tissues in Korean rose bitterling. The expression of 4 groups of Hsp70 family was detected in all tested tissue. Particularly, Hsp70 family of Korean rose bitterling is highly expressed in hepatopancreas and sexual gonad (testis and ovary). The expression of Hsp70 family was differentially regulated in accordance with early development stage of Rhodeus uyekii.

        The flower buds of Sophora japonica L (SF), as a well-known traditional Chinese medicinal herb, have been used to treat bleeding-related disorders such as hematochezia, hemorrhoidal bleeding, dysfunctional uterine bleeding, and diarrhea. However, no specific anti-cancer effect and its molecular mechanism of SF have been described. Thus, we performed in vitro study to investigate if treatment of SF affects activating transcription factor 3 (ATF3) expression and ATF3-mediated apoptosis in human colorectal cancer cells. The effects of SF on cell viability and apoptosis were measured by MTT assay and Western blot analysis against cleaved poly (ADP-ribose) polymerase (PARP). ATF3 activation induced by SF was evaluated using Western blot analysis, RT-PCR and ATF3 promoter assay. SF treatment caused decrease of cell viability and increase of apoptosis in a dose-dependent manner in HCT116 and SW480 cells. Exposure of SF activated the levels of ATF3 protein and mRNA via transcriptional regulation in HCT116 and SW480 cells. Inhibition of extracellular signal-regulated kinases (ERK) 1/2 by PD98059 and p38 by SB203580 attenuated SF-induced ATF3 expression and transcriptional activation. Ectopic ATF3 overexpression accelerated SF-induced cleavage of PARP. These findings suggest that SF-mediated apoptosis may be the result of ATF3 expression through ERK1/2 and p38-mediated transcriptional activation.

        Serine-arginine-rich nuclear protein LUC7L plays an important role in the regulation of myogenesis in mice. In the present study, we isolated and characterized the Korean rose bitterling Rhodeus uyekii Luc7l cDNA, designated RuLuc7l. The RuLuc7l cDNA is 1,688 bp long and encodes a 364-amino-acid polypeptide containing serine/arginine-rich region at the C-terminus. The deduced RuLuc7l protein has high amino acid identity (71-97%) with those of other species including human. Phylogenetic analysis revealed that RuLUC7L clustered with fish LUC7L proteins. The expression of RuLuc7l mRNA was high in the brain, kidney, and stomach of Korean rose bitterling. Expression of the RuLuc7l mRNA was detected from 1 day post-fertilization (dpf) and moderately increased until 21 dpf during the early development. Further investigations are required to elucidate the functional role of RuLUC7L in myogenesis in R. uyekii.

        A 63-year-old woman with a history of nephrolithiasis and long-term hemodialysis (HD) presented with abdominal pain. Abdominal computed tomography (CT) showed urinoma induced by renal forniceal rupture, and three causative calculi. No active stone growth or new stones were observed when compared with previous CT, however, fragmentation and migration of an existing stone was observed. After ureteroscopic lithotripsy and stenting with a double-J catheter, the patient made a full recovery. To the best of our knowledge, this is the first case of spontaneous renal forniceal rupture in an anuric HD patient caused by calculi. Renal forniceal rupture can occur, even in anuric HD patients.

        Doenjang is one of the most favorite traditional food which was produced by the fermentation process of soybean with various microbes such as Aspergillus oryzae, Bacillus subtilis and so on. The aim of this research is to analyse the antioxidant and anti-α-glucosidase inhibitory activities of doenjang made by 9 different cultivar (Daewonkong, Taekwangkong, Daepungkong, Hwangkeumkong, Saedanbaek, Hanolkong, Pungsannamulkong, Cheongja-3 and Cheongdu-1). Doenjang samples were made by traditional manner using rice straw. Each samples were extracted with 40% methanol and extract were used Among them, cheongja-3‘s doenjang showed a significant highest antioxidant and anti-α-glucosidase inhibitory activities because of its anthocyanin. Except for cheongja-3's Doenjang, antioxidant activity had positive correlation with isoflavone content. But no relationship observed between isoflavone content and anti-α-glucosidase activity. The highest anti-α-glucosidase activity was observed in Saedanbaek's Doenjang with highest protein content.

        The change of quality characteristics with storage temperature (room (25°C), low (15°C) and cold (4°C) temperature) and period of unhulled foxtail millet (Setaria italica Beauv. cv. Samdame and Gyeongkwan 1), proso millet (Panicum miliaceum L. cv. Hwanggeum-gijang and Manhongchal) and sorghum (Sorghum bicolor (L.) Moench cv. Hwanggeumchal and Donganme) were evaluated. The 1,000 grain weight, lightness, redness, yellowness and moisture content with storage temperature and period of unhulled foxtail millet, proso millet and sorghum were not showed difference. Germination percent and milling recovery of foxtail millet, proso millet and sorghum decreased with increasing storage temperature and period. Fat acidity of stored foxtail millet, proso millet and sorghum was increased with increasing storage period, and the higher temperature increased more. The results of this study show that the storage of foxtail millet, proso millet and sorghum at the low and cold temperature is better than the room temperature. Especially, in mind of the economics, the storage method is good low temperature than cold temperature.

        Soybean isoflavones include daidzein, genistein and glycitein with their glycosides, and their malonated derivatives are the main polyphenolic compounds that are helpful for human health. Our research objective was to investigate the differentitation of soybean isoflavones contents of breeding populations between yellow and black soybean. Isoflavones contents in soybean are a wide range from 500 to 7000 ㎍/g. In this study, we used Ilmi (Isoflavones content, 3.612 ㎍/g) as male parent and 04GAYT-4 (Isoflavones content, 1,648 ㎍/g) as female parent. From these varieties, we obtained 94 breeding lines (yellow, 48 lines; black, 25 lines; brown, 21 lines) which have isoflavones content range from 1000 to 6000 ㎍/g. Highest isflavone contents of three different breeding lines was yellow seed coated lines (average isoflavone content, yellow 3,046 ㎍/g, brown 2,935 ㎍/g, black 2,813 ㎍/g).

        Cereal seeds, sorghum, foxtail millet, hog millet, adlay, and corn are traditionally used as health assistant as well as energy supplying food in Korea. While beneficial phytochemicals to human have revealed in cereals, the information on peptides from cereals is far less accumulated than major reserve protein. Here, we analyzed peptide profiles using surface enhanced laser desorption/ionization time of flight mass spectrometry (SELDI-TOF MS) in cereal seeds for construction of peptide information and attempted to develop peptide biomarkers for cereal identification. To optimize the analysis condition of SELDI-TOF MS, the effect of dilution factor on binding affinity to protein chips was tested using CM10 and Q10 arrays. Peptide clusters were significantly different at the level of 0.01 p-value. Peak spectra were the most stable in 1:50 of dilution factor in both chip arrays. Numbers of detected peak of 5 cereal seeds were 131 in CM10 and 74 in Q10 array. Each cereal was grouped as a cluster and well discriminated into different cluster in the level of 0.01 p-value. Numbers of potentially identified peptide biomarkers are 11, 13, 9, 5 and 12 in sorghum, foxtail millet, hog millet, adlay and corn, respectively. This study demonstrates that each cereal seed have own distinguishable specific peptides although their function are not identified yet in this study. In addition, the proteomic profiling using SELDI-TOF MS techniques could be a useful and powerful tool to discover peptide biomarker for discrimination and assess crop species, especially under 20 kDa.

        Soybean isoflavones include daidzein, genistein and glycitein with their glycosides, and their malonated derivatives are the main polyphenolic compounds that are helpful for human health. Our research objective was to investigate the differentitation of soybean isoflavones contents of breeding populations between high and low isoflavones contained soybean. Isoflavones contents in soybean are a wide range from 500 to 7000 ㎍/g. In this study, we used Ilmi (Isoflavones content, 3.108 ㎍/g) as male parent and Dajin (Isoflavones content, 578 ㎍/g) as female parent. From these varieties, we got 165 breeding lines which have isoflavones content range from 472 to 2973 ㎍/g. Isflavone contents in breeding lines showed normal distribution.
