        Flowering time is a important agronomic trait for grain production in rice. So the control of flowering time is a critical step. In Arabidopsis, expression of certain key flowering gene such as FLOWERING LOCUS C (FLC) is known to be epigenetically regulated by chromatin modification through Enhancer of Zeste[E(z)], a histone methyltransferase, that core component of repressive complex, polycomb repressive complex2(PRC2). However, the chromatin mechanism involved in the regulation of rice flowering genes is presently not well known. Here we show that predict coding region of a intronic LncRNA[termed rice COLDAIR(OsCOLDAIR)], which is expected to associate with a component of PRC2, is predicted at rice FLC gene. And additionally we suggest interaction of histone methyltransferase and E3 SUMO ligase that indicate possibility of interaction with rice E(z) gene and rice E3 SUMO ligase. Our study contribute to control of rice flowering time by observing two factor that can regulate expression of related of rice FLC gene.

        In order to improve high persistence and forage quality, through selection of various superior parental varieties for breeding and synthesis of them with new lines, there are ongoing worldwide studies aiming to enhance the quality of tall fescue through a traditional breeding method by selection and hybridization. A new tall fescue variety (Festuca arundinacea Schreb.), named Greenmaster3ho, was developed by the National Institute of Animal Science, Rural Development Administration in Korea from 2010 to 2014. For synthetic seed production of this new variety, five superior clones, 09XFa02, 09XFa03, 09XFa11, 09XFa13, and 09XFa14 were selected and polycrossed. The agronomic growth characteristics and forage production capability of the seeds were studied at Cheonan from 2010, and regional trials were conducted in Cheonan, Hoengseong, Jeju, and Jinju from 2012 to 2014. Greenmaster3ho showed enhanced disease resistance, persistence, and regrowth ability as compared to Fawn. The dry matter yield of Greenmaster 2 was 29% higher (15,119 kg/ha) than that of Fawn. However, the nutritive value of both varieties was similar. This study developed a new tall fescue variety with excellent environmental adaptability, aiming to make a contribution to the vitalization of the Korean grassland industry.

        This study was performed to investigate the growth characteristics and inorganic components of Codonopsis lanceolata regarding regional differences. The plant height of Japanese Codonopsis lanceolata was 373.6 cm, so it’s revealed that it has more vigorous growth than Korean won. The flowering time of Korean Codonopsis lanceolata was 2 weeks faster than Japanese one. Total fresh weight of root was 41.0 g and 39.0 g for Korean and Japanese respectively, thus, no significance difference was found. However, regarding fresh weight, Korean one had a more fresh weight (35.4 g) of main root parts, but Japanese one had a more fresh weight (9.6 g) of the lateral root part. Each inorganic component was found more in the aboveground parts, regardless of the region and the content of K was the largest. Regarding the content of macroelements for each part of Codonopsis lanceolata, the content of Na, Mg, P, S, and Ca in Korean Codonopsis lanceolata was found the highest on the leaf, followed by stem and root. In the case of Japanese Codonopsis lanceolata, same result was found on the content of Mg and Ca, however, the highest content of Na and P was found in the stem.

        This study was conducted to compare the growth, inorganic components, and proximate components of Codonopsis lanceolata grown in 10 regions of Korea for selecting superior species and breeding by crossing. Among the all tested lines, the shortest plant height (217.12 cm) was observed from the Ulleungdo region line (No. 4) while the longest (273.9 cm) was observed from Hwasun region line (No. 9). In addition, the lines of central and northern region (No. 1~No. 7) tend to have shorter plant height than those of southern region (No. 8~No. 9) except Jejudo region line (No. 10). Flowering tends to be late towards southern region, and lines in central and northern regions were started flowering about 2 weeks earlier than those in southern regions. However, the heaviest root weight was 13.1 g, found in only Jejudo line (No. 10) whereas there was no significant difference found in the other regions which have a range of 8.3~11.0 g. The inorganic components were varied in each line, however, proportion of macroelements, such as K, Ca, and P, was the largest for every line. Especially for Heongseong region line (No. 2), had larger proportion of macroelements than the others. There was a difference of proximate compositions of Codonopsis lanceolata, except the moisture content, among all regions, however, it was generally shown that the content of crude protein (1.31~3.76%) and crude fiber (2.18~3.12%) was the highest.

        Watermelon (Citrullus lanatus) is one of the most economically important cucurbitaceous crop over the world. Anthracnose disease caused by Colletotrichum orbiculare, result to severe damage to cucurbits worldwide. Anthracnose is a typical plant disease which significantly affecting the yield of cucurbit crop. Pathogeneis-related (PR) proteins are well-known plant defense proteins against pathogens. Therefore, we observed PR genes expression patterns when watermelon got anthracnose disease. We did RT-PCR experiment to evaluate differences of PR genes expression pattern among Au-Producer(R), one of the representative of resistance watermelon varieties against anthracnose, and 920533(S), one of the representative of susceptible watermelon varieties against anthracnose. As a result, there were differences of the expression of several PR genes between the R and S watermelon. Analysis of the function of these genes is expected to perform in the future.

        Mungbean (Vigna radiata) is a fast-growing, warm-season legume crop that is primarily cultivated in developing countries of Asia. We constructed a draft genome sequence of mungbean to facilitate genome research into the subgenus Ceratotropis and to enable a better understanding of the evolution of leguminous species. The draft genome sequence covers 80% of the estimated genome, of which 50.1% consists of repetitive sequences. In total, 22,427 high confidence protein-coding genes were predicted. Based on the de novo assembly of additional wild mungbean species, the divergence of what was eventually domesticated and the sampled wild mungbean species appears to have predated domestication. Moreover, the de novo assembly of a tetraploid Vigna species (Vigna reflexo-pilosa var. glabra) provided genomic evidence of a recent allopolyploid event. To further study speciation, we compared de novo RNA-seq assemblies of 22 accessions of 18 Vigna species and protein sets of Glycine max and Cajanus cajan. The species tree was constructed by a Bayesian Markov chain Monte Carlo method using highly confident orthologs shared by all 24 accessions. The present assembly of V. radiata var. radiata will facilitate genome research and accelerate molecular breeding of the subgenus Ceratotropis.

        Biological resources including proteins, cells, and tissues were confronted with both safe and stable preservation for practical use in biotechnological industry. Particularly, cell therapy for regenerative engineering is needed to restricted regulation and accurate preservation. Therefore, this study was investigated improved conditions of mesenchymal stem cells from human umbilical cord (hUCs) or aspirated adipose tissues (hATs) for clinical cell banks. Both cells were isolated according to standard operation procedure of Hurim BioCell Inc. and analyzed the inherent characteristics in passage 4. To compare the ability of experimental groups after cryopreservation, proliferation ability using calculated values and cytomorphological patterns of each experimental step were analyzed. Also proteins such as ice-binding protein or caspase inhibitor were applied to add the preservation medium of hUCs or hATs. Result of preservation solution with 20% serum was considered a positive group. Recovery rate and expansion results showed specific dosage and cell type-dependent differences in the experimental group. Chromosomal stability and multipotency of hUCs or hATs were expressed stable pattern after cryopreservation using advanced medium. As a result, these additives could be substituted for xenogenic sources in banking of hUCs or hATs.

        Recently, human mesenchymal stem cells (MSCs) are attracting attention as a useful source for regenerative therapy. Controlled production of cell therapy requires the establishment and management of an accurate isolation, characterization and monitoring for quality assurance of developing MSCs mediated. In this study, we were confirmed maintenance of potency of isolated and cultured human umbilical cord (hUC)-MSCs during ex vivo expansion or after cryopreservation. Expression of their cell specific marker was analyzed by flow cytometry and the differentiation potency was confirmed by guided differentiation of adipocyte, osteocyte, chondrocyte and hepatocyte after expanding over 15 doublings in vitro. Safe production of developing a cell therapy was proved by testing for microbial, mycoplasma, endotoxin, and adventitious agents. Also stability of cells in cultivation, preservation and/or differentiation was determined chromosomal assay. In developing using hUC-MSCs, cells showed an accurate isolation and stable expansion in ex vivo condition. The results of several management assay showed that the stem cell marker expression of CD31, CD34 and CD45 were under 10%, however CD90 was over 90% by FACS analysis. Any contamination and mutation in all tests weren't detected in specific points for safe or stable production of hMC-MSCs. Also the proliferation and differentiation potency maintains during in vitro culture and after cryopreservation of hUC-MSCs. These results could be used as standard methods of maintenance of hUC-MSCs for cell therapy products and clinical application.

        “Hojoong”, a winter wheat (Triticum aestivum L.) cultivar was developed by the National Institute of Crop Science, RDA, Iksan, Korea, during the period from 2006 to 2012. The heading and maturing dates of this variety were May 2 and June 10 in upland, and May 3 and June 7 in paddy field, respectively. It is an It is an awned, semi-dwarf and soft red winter wheat. Culm and spike length of “Hojoong” were 79cm and 9.1cm. It had lower test weight (803 g/ℓ) and 1,000 grain weight (39.5g) than “Keumkang”. It showed resistance to pre-harvest sprouting, which lower rate of pre-harvest sprouting (5.5%) than “Keumkang”. “Hojoong” had lower flour yield (66.7%) and ash content (0.38%) than “Keumkang”. It showed lower protein content (11.3%) and lower SDS-sedimentation volume (34.0㎖) and amylose content (20.5%) than “Keumkang”. It showed higher high viscosity (204BU) and lightness of noodle dough sheet (80.63). It showed different composition in HMW-GS compositios (2.2+12), PPO18 (876bp), GBSS B (null type) and Puroindolines composition compared th “Keumkang”. Average yield of “Hojoong” in the regional adaptation yield trial test was 5.51 ton/ha in upland and 5.10 ton/ha in paddy field. “Hojoong” would be suitable for the area above –10℃ of daily minimun temperature in January in Korean peninsula

It showed moderate to pre-harvest sprouting, which lower rate of pre-harvest sprouting (13.7%) than “Keumkang”. “Baekchal” had similar ash content (0.47%) and protein content (13.1%) to “Keumkang”. It showed lower gluten content (9.6%), SDS-sedimentation volume (38.2㎖) and amylose content (6.5%) than “Keumkang”. It showed higher high viscosity (643BU), water absorption (213%), expansibility of cooking (357%). It showed different composition in HMW-GS compositios (2*), PPO18 (876bp), GBSS (waxy type) and Puroindolines composition compared th “Keumkang”. Average yield of “Baekchal” in the regional adaptation yield trial test was 4.89 ton/ha in upland and 4.83 ton/ha in paddy field. “Baekchal” would be suitable for the area above –10℃ of daily minimun temperature in January in Korean peninsula

        Fusarium head blight (FHB), caused by Fusarium graminearum is a major disease problem on wheat and barley in Korea. The objectives of this study were to evaluation of korean wheat cultivars for Type I and Type II resistance to FHB. We screened for Type II resistance in the greenhouse using single floret inoculation and for Type I resistance in the field using spray inoculation. Sumai 3 was used the FHB resistant check. Thirty-two korean wheat cultivars were evaluated for resistance to spread of symptoms within spike. The 2013 field screening with wheat cultivar was located in Kimjae-si Joeonbuk Korea. All plots were inoculated twice. Mist-irrigation was applied to facilitate FHB development. FHB severity was assessed visually 21 days after inoculation on 20 arbitrarily selected spikes per plot. FHB severity was determined as the percentage of symptomatic spikelets from the total of all spikelets. For FHB resistance, the average of FHB severity of Type I exhibited ranging from 21.9% to 77.2% and FHB severity of Type II ranging from 20.8% to 100%. Namhae, Milseong, Geuru, Joen, Anbaek and Sukang were the moderately resistant cultivars while Gobun, Alchan, Dajoong, Eunpa, Shinmichal1, Eunpa and Uri were the most susceptible cultivars for Type II resistance.

        This study histologically describes the intersexuality of Scomberomorus niphonius collected from the coastal area near Jeju Island. A total of 126 S. niphonius, collected from March to July 2012 with a total length of 62.4 cm (±17.5) and a total weight of 1,701.9 g (±1,528.9) were used for analyses. From a histological perspective, two types of intersex were confirmed. One type had scattered germ cells from the opposite sex within the gonad. The second type developed germ cells from the opposite sex in the connective tissue of the outer gonadal membrane. The intersexuality was 14.3% (n=18/126), with females (21.3%; n=16/75) exhibiting a higher rate than males (3.9%; n=2/51). There was no displayed correlation between intersexuality and the total length and weight.

        Manganese () is a trace element that is essential for normal physiology, and is predominantly obtained from food. Several lines of evidence, however, demonstrated that overexposure to exerts serious neurotoxicity, immunotoxicity and developmental toxicity, particularly in male. The present study aimed to evaluate the effect of 0, 1.0, 3.3, and 10 mg/kg/day doses of on the reproductive organs in the immature female rats. Rats (PND 22; S.D. strain) were exposed to () dissolved in drinking water for 2 weeks. The animals were sacrificed on PND 35, then the tissues were immediately removed and weighed. Histological studies were performed using the uteri tissue samples. Serum LH and FSH levels were measured with the specific ELISA kits. Body weights of the experimental group animals were not significantly different from those of control group animals. However, ovarian tissue weights in 1 mg and 3.3 mg dose groups were significantly lower than those of control animals (p<0.05 and p<0.01, respectively). Uterine tissue weights of 3.3 mg dose groups were significantly lower than those of control animals (p<0.01), while the 1 mg dose and 10 mg dose failed to induce any change in uterine weight. Similarly, only 3.3 mg dose could induce the significant decrease in the oviduct weight compared to the control group (p<0.05). Non-reproductive tissues such as adrenal and kidney failed to respond to all doses of exposure. The uterine histology revealed that the exposure could affect the myometrial cell proliferation particularly in 3.3 mg dose and 10mg dose group. Serum FSH levels were significantly decreased in 1mg dose and 10 mg groups (p<0.05 and p<0.01, respectively). In contrast, treatment with 1 mg dose induced a significant increment of serum LH level (p<0.05). The present study demonstrated that exposure is capable of inducing abnormal development of reproductive tissues, at least to some extent, and altered gonadotropin secretions in immature female rats. Combined with the well-defined actions of this metal on GnRH and prolactin secretion, one can suggest the might be a potential environmental mediator which is involved in the female pubertal process.

        Bacillus thuringiensis (Bt) crystal protein genes encode insecticidal δ-endotoxins that are widely used for the development of insect-resistant crops. Common soybean is a crop of economic and nutritious importance in many parts of the world. Korea soybean variety Kwangan was transformed with Bacillus thuringiensis (Bt) crystal protein genes. These genes were transformed into Kwangan using highly efficient soybean transformation system. Transgenic plants with Bt crystal protein genes were confirmed for gene introduction and their expression using PCR, real-time PCR and RT-PCR. Currently, the confirmation of stable gene introduction with Bt genes is also performing by southern blot analysis and physiology test and agronomic characters are investigating.

        Cry1Ac protein is known as one of toxin crystal proteins synthesized from Bacillus thuringenesis that plays a critical role for the insect resistance. Recently, cry1Ac genes have introduced into many plants in general and soybean as well. However, the gene expression of cry1Ac genes in transgenic plants remains low that need to be improved. Several mutations we reintroduced into the cry1Ac genes in order to enhance the insecticidal effect. In this study, the cry1Ac with mutant #2, #11 and #16 were transformed into Kwangan, a Korean soybean variety by using the “half-seed” method. The plant lets carrying modified cry1Ac genes were primarily selected on media containing Phosphinothricine (PPT), a bar selective agent and Basta leaf painting. Then, the presence of introduced genes in T0 plants and the gene expression were investigated by PCR, RT-PCR and Real-time PCR. PCR and RT-PCR analysis showed expression of bar and cry1Ac genes from tested transgenic soybean plants. The number of copy of bar gene ranged from 1 to 3 by Real-time PCR analysis. These results provided a fundamental back ground for our further experiments: Confirmation of the gene expression by Southern blot and identification of the function of modified cry1Ac by insect bioessays.

        Korean soybean variety Kwangan was transformed with ORE7 gene using highly efficient soybean transformation system. The gene is known to exhibit a delayed leaf senescence phenotype in Arabidopsis. To confirm phenotypic characterization of leaf senescence for non-transgenic (NT) and transgenic plants, we transplanted T1 transgenic lines 7, 9, 14 and 15 together with two negative controls (NT and EV) in greenhouse. As a result, line 15 showed dramatic phenotypic characterization of yield increase and senescence delay. In addition, to investigate the agriculture traits for transgenic plants with leaf senescence delaying, T2 transgenic lines and two negative controls were transplanted on GMO fields in Ochang and harvested T3 seeds (2010). Most transgenic lines showed higher total seed weigh than NT. Especially, total seed weight of line 15 was increased by about 180% and 120% compared with the NT and EV, respectively. Therefore, we carried out the second field experiments with T3 transgenic line 15 and NT in Ochang (2011). A total of 117 transgenic plants were divided into two groups, senescence delaying (64 out of 117 plants) and increased yield (53 out of 117 plants), by transcript level of ORE7 gene. Interestingly, among increased yield plants, total seed weight of each 7 plants were increased by more than 200% compared with NT.

        Korean soybean variety Kwangan was transformed with coat protein (CP), helper component-proteinase (HC-Pro), and ABRE binding factor 3 (ABF3) genes using highly efficient soybean transformation system. Among these genes, CP and HC-Pro were transformed using RNAi technology. Transgenic plants with CP were confirmed for gene introduction and their expression using PCR, real-time PCR, RT-PCR, Southern blot, and Northern blot. To investigate the response of viral infection with CP, T1 plants were inoculated with SMV-infected leaves and confirmed the existence of mosaic symptom in both leaves and seeds. Two transgenic lines with CP were highly resistant to SMV with clear leaves and seeds while SMV-susceptible lines showed mosaic symptom with seed mottling. The transcript levels of T1 plants with CP were also determined by northern blot, suggesting that SMV-resistant T1 plants did not show viral RNA expression whereas SMV-susceptible T1 plants showed viral RNA expression. Currently, the response of viral infection with HC-Pro is investigating to produce SMV-resistant soybean transgenic plants, and the physiological experiment with ABF3 is also carrying out to produce drought-tolerant soybean transgenic plants.

        “Joah”, a winter wheat (Triticum aestivum L.) cultivar was developed by the National Institute of Crop Science, RDA, Iksan, Korea, during the period from 1997 to 2011. The heading and maturing dates of this variety were April 26 and June 8 in upland, and April 28 and June 6 in paddy field, respectively. It is an awned, semi-dwarf and soft winter wheat. Culm and spike length of “Joah” were 78 cm and 8.4 cm. It had lower test weight (763g/L) and l,000-grain weight (43.0 g) than “Keumkang”. It showed moderate to pre-harvest sprouting, which lower rate of pre-harvest sprouting (12.3%) than “Keumkang”. “Joah” had lower flour yield (66.3%) and ash content (0.33%) than “Keumkang”. It showed higher lightness (92.29) of flour color than “Keumkang”. It showed lower protein content (11.5%) and gluten content (10.7%) and lower SDS-sedimentation volume(29㎖) and mixing time of mixograph than “Keumkang”. It was showed higher amylose content and high viscosity than “Keumkang”. It showed different compositions in HMW-GS compositions (2.2+12), PPO 18 (876bp) and Puroindolines composition compared to “Keumkang”. “Joah” showed similar firmness (2.20N) and higher volume of sponge cake to “Keumkang”. Average yield of “Joah” in the regional adaptation yield trial was 6.44 ton/ha in upland and 5.26 ton/ha in paddy field. “Joah” would be suitable for the area above -10℃ of daily minimum temperature in January in Korean peninsula.
