A solid-phase competition enzyme-linked immunosorbent assay (ELISA), recombinant VP2 (rVP2) protein, and monoclonal antibody (mAb) were developed for the specific and sensitive detection of porcine parvovirus (PPV) antibodies in pig sera. A total of 1,544 sera samples were collected from breeding pig farms located in the Gyeongsangbuk-do Province in the Republic of Korea. The optimal operating conditions of SC-ELISA were as follows. The concentration of rVP2 proteins coated on the wells was 4 μg/mL, the swine sera were diluted 1:2, and the HRP-conjugated PPV VP2 mAb (9A8 clone) was used at 500 ng/mL. These results suggest that the SC-rVP-ELISA assay may be a valuable alternative to the current diagnostic tools used to detect PPV-specific monoclonal antibodies and broadly monitor PPV infections in domestic pigs at different breeding stages.
In this study, the near-complete genome sequence of the novel reassortant H1N2 influenza A virus strain A/swine/Korea/KS60/2016 is reported. Sequences of the hemagglutinin (HA), neuraminidase (NA), and polymerase basic 2 (PB2) genes were analyzed, revealing that the isolates contain segments from previous Korean swine H1N2 strains. Additionally, the remaining genes of this strain originated from human H1N1 strains in 2009.
과수농가에서 수행되는 수확작업은 노동부하가 큰 주요작업으로 수확된 과실의 적재 및 운반작업에 소모되는 노동력이 크다. 본 연구에서는 과수농가를 대상으로 수확된 열매의 적재 및 운반작업에 사용 가능한 간이형 보행 동력운반차를 개발하였다. 과수 농민들의 요구조건을 고려하여 동력운반차의 이동속도 및 적재 용량을 선정하였다. 이에 따라, 동력운반차의 주요 구성요소를 선정하고 동력전달경로를 분석하였다. 또한, 개발된 동력운반차의 성능시험을 수행하였으며 상용 소프트웨어를 활용하여 구조해석을 수행하였다. 설계된 간이형 보행 동력운반차는 전·후진이 가능하고 이동속도는 최대 1 m/s이며, 최대적재용량은 500 kg이다. 본 연구에서 개발된 간이형 보행 동력운반차를 활용하면 농민들의 작업효율을 높이고 신체적인 부담을 경감시킬 수 있을 것으로 판단된다.
Canine parvovirus (CPV) remains a leading infectious cause of death in canines, especially in young puppies. Though vaccination is being carried out regularly, immunization failures occur, and puppies may be exposed to infection. Virus-like particles (VLPs) act like a subunit vaccine, mimicking the structure of authentic viruses. Therefore, VLPs have the potential to be used as vaccine candidates. Since Viral Protein 2 (VP2), a major structural protein of CPV, is the crucial antigen for CPV, the purpose of this study was to produce a recombinant VP2 of new canine parvovirus-2a using the baculovirus expression system in SF9 insect cells. The results revealed that recombinant VP2 assembles to form VLPs with antigenic properties similar to those of natural CPV, the recombinant VLP can produce a hemagglutination assay (HA) titer (1:210) in SF9 cells. Expression of the recombinant 6-His-tagged VP2 in SF9 cells was confirmed by western blotting. These findings suggest that the recombinant VP2 expressed in this study could be used as an efficient subunit vaccine against CPV infection.
Porcine parvovirus 2 (PPV2) was recently detected in the Republic of Korea. This paper reports two near-complete genome sequences of PPV2 identified for the first time in the lung tissue of aborted pig fetuses.
Viral protein 2 (VP2), which is the structural protein of parvovirus, can produce virus-like particles (VLPs) by a self-assembly process in vitro, making VLPs attractive vaccine candidates. VP2 of canine parvovirus (CPV) is responsible for neutralizing antibodies in immunized animals. In this study, VP2 protein of canine parvovirus-2c was expressed using a baculovirus expression system and assembled into parvovirus-like particles in insect cells. The results show that VP2 proteins assembled into virus-like particles (VLPs) with antigenic properties similar to those of natural CPV and a high hemagglutination (HA) titer (1:27). The recombinant 6-His-tagged VP2 protein with a molecular mass of about 65 kDa was detected by anti-His antibody and anti-PPV serum. This study provides a foundation for the application of VP2 protein in the clinical diagnosis of CPV and in the vaccination against CPV.
The number of those dreaming of becoming idols is increasing every year. It is estimated that, including those trainees at management agencies, about one million are aspiring to be entertainers. According to the status analysis of youth entertainers conducted by Ministry of Gender Equality and Family, however, many young rights, the right to study, etc. This study, therefore, looked into the real situation of idol trainees by analyzing media texts. In doing so, the study examined the trainee training systems of different management agencies and the pre-debut process that young trainees aspiring to be entertainers go through, and gained an understanding of the psychological maladjustment they experience. Based on these, the study discusses the followings. First, as one axis of entertainer training systems of management agencies, personality development education and psychiatric counseling need to be systemized. Second, the existing training system needs to be changed. As it requires trainees to stay together, their relationships with their families and friends can be hurt. Third, trainees themselves should make an effort to find their own identities to have mental stability. Lastly, all the sectors in the entertainment industry need to pitch in to find different and realistic ways to support them, and the current broadcasting environment where competition is encouraged needs to be changed as well.
Viral protein 2 (VP2) of porcine parvovirus (PPV) is responsible for inducing neutralizing antibodies in immunized animals. It is the major viral structural protein. In this study, novel subunit vaccines against PPV based on virus-like particles (VLPs) formed from VP2 proteins (PPV 13-7 Korean strain) were expressed in an insect baculovirus cell system and purified using Ni-NTA affinity column chromatography. These VP2 proteins assembled into virus-like particles (VLPs). They showed antigenic properties similar to those of natural PPV. In addition, they showed high hemagglutination (HA) titers (211 for PPV 13-7 Korean strain). This study provides a foundation for the application of the difference immunization of recombinant protein in the diversity of PPV VP2 genes and in vaccination against PPV in the future.
Canine parvovirus type-2 (CPV-2) has been reported worldwide as the main agent associated with acute hemorrhagic enteritis, resulting in high morbidity, especially in young dogs. CPV-2 has three genetic variants, 2a, 2b, and 2c. Here, we report three cases of canine parvovirus enteritis associated with CPV-2a (2 samples) and -2c (1 sample) infections that occurred in three young dogs suffering from enteritis. Isolates from dog diarrheic fecal samples were sequenced by polymerase chain reaction (PCR) and identified as two types of CPV-2a and one type of CPV-2c. This work constitutes the first isolation and genetic characterization of CPV-2c in the Republic of Korea.
One of the characteristics of suicide phenomena in the Korean society is that it witnesses suicide committed by entertainers quite often. Some entertainers choose suicide as a way to resolve crises and conflicts, and those suffering from maladjustments, when seeing this, are highly likely to follow their suit, committing copycat suicide. For this reason, society needs to pay close attention to entertainers’ suicide. Many entertainers with depressive, negative sentiments relate themselves easily to other entertainers’ suicide in terms of both their jobs and feelings, which in turn make them choose suicide. This study explores what psychological effect entertainers’ suicide has on those entertainers suffering from psychological maladjustment. This is done through analyzing texts appearing on different kinds of media. At the same time, the study discusses possible ways to help them think less about suicide, at a preventive level.
The blending schemes of Saengshik powder with water were established to retain the dispersion stability of the Saengshik beverage in order to develop a ready-to-drink type product. The effect of the blending on the rheological properties and the dispersion stability of Saengshik beverage were determined. The Saengshik beverages, blended with low shear force for a short time, showed dilatant fluid characteristics, while those with high shear force for a long time had pseudoplastic fluid ones. The consistency of Saengshik beverage increased with the blending speed and time. Saengshik beverage blended at low shear showed rheopexy while becoming time independent by a highshear blending action. A logarithmic model fitted well to the changes in backscattering light flux with the rest time of Saengshik beverage. Based on sedimentation kinetics, the migration speed of clear front, and the thickness of sedimented layer, Saengshik beverage with maximum dispersion stability was obtained by a high shear blending at 15,120 rpm for 25 s.
본 연구는 수분함량 22, 25, 28%, 스크루 회전속도 200, 250 rpm, 용융물 온도 110, 120, 130℃의 조건범위로 조절하고 원료 사입량 100 g/min으로 고정하여 압출 성형한 밤과육의 물리적 특성(직경 팽화율, 비길이, 체적밀도), 수분용해지수 및 수분흡착지수, 환원당 함량과 항산화 활성에대해 분석하였다. 밤 압출성형물은 직경 팽화율이 증가할수록 비길이와 체적밀도는 감소하였고, 수분용해지수와 수분흡착지수는 수분함량이 낮아질수록 증가하였다. 압출성형을 통하여 환원당 함량, DPPH 라디칼 소거능, 총 페놀함량 및 플라보노이드 함량은 감소하였으며, 탄닌함량은 압출성형을 통해 2.94±0.10−4.23±0.12 mg/g 범위로 낮아지는 경향을 나타내었다. 결론적으로 압출성형 후 수분용해지수의 증가는 밤 음료의 용해도 개선의 효과를 나타낼 수 있으며, 또한 압출성형공정은 소비자 기호성에 영향을 미치는 탄닌의 쓴맛을 감소시키는 가능성을 제시하였다. 이를 통해 다양한 식품의 중간소재로써 압출성형 밤이 활용 될 수 있다고 판단된다. 밤의 식품중간소재 기초자료로서 응용될 것으로 생각되지만, 압출성형 최적조건을 보다 효과적으로 도출하기 위한 연구가 필요할 것으로 판단된다.

The presence of distinct bacterial species is found to be dependent on age, diet, and disease. We compared the detection rate of several oral bacterial strains in a cohort of 36 subjects including healthy volunteers, periodontal patients, and oral cancer patients. Gargling samples were obtained from these subjects from which DNA was then extracted. Specific primers for 29 bacterial species were used for PCR detection. In the oral cancer patients, Capnocytophaga ochracea, Gemella morbillorum, and Streptococcus salivarius were detected more frequently compared with the healthy volunteers and periodontitis patients. Fusobacterium nucleatum/ polymorphym and Prevotella nigrescens were significantly less prevalent in oral cancer patients than the other groups. In periodontitis patients, Porphyromonas gingivalis and Treponema denticola were more frequently found compared with the healthy volunteers. In the healthy volunteer group, Peptostreptococcus anaerobius was more frequently found than the other groups. The detection rate of several oral bacterial species was thus found to differ between healthy volunteers, periodontitis patients and oral cancer patients.
Cholinesterase (ChE) is one of the most ubiquitous enzymes and in addition to its well characterized catalytic function, the morphogenetic involvement of ChE has also been demonstrated in neuronal tissues and in non-neuronal tissues such as bone and cartilage. We have previously reported that during mouse tooth development, acetylcholinesterase (AChE) activity is dynamically localized in the dental epithelium and its derivatives whereas butyrylcholinesterase (BuChE) activity is localized in the dental follicles. To test the functional conservation of ChE in tooth morphogenesis among different species, we performed cholinesterase histochemistry following the use of specific inhibitors of developing molar and incisors in the hamster from embryonic day 11 (E11) to postnatal day 1 (P1). In the developing molar in hamster, the localization of ChE activity was found to be very similar to that of the mouse. At the bud stage, no ChE activity was found in the tooth buds, but was first detectable in the dental epithelium and dental follicles at the cap and bell stages. AChE activity was found to be principally localized in the dental epithelium whereas BuChE activity was observed in the dental follicle. In contrast to the ChE activity in the molars, BuChE activity was specifically observed in the secretory ameloblasts of the incisors, whilst no AChE activity was found in the dental epithelium of incisors. The subtype and localization of ChE activity in the dental epithelium of the incisor thus differed from those of the molar in hamster. In addition, these patterns also differed from the ChE activity in the mouse incisor. These results strongly suggest that ChE may play roles in the differentiation of the dental epithelium and dental follicle in hamster, and that morphogenetic subtypes of ChE may be variable among species and tooth types.
저염 된장 제조를 위하여 염농도 8-14%로 달리하여 제조한 된장을 20oC에서 16주 동안 발효하여 이화학적 특성의 변화를 조사하였다. 된장의 pH는 발효기간에 따라 전반적으로 감소하였고, 발효 4주 후부터 염농도 8%와 10%의 저염 된장이 염농도 12%와 14%인 고염 된장에 비하여 낮은 pH를 보였다. 적정산도는 제조 직후 1주간 급격히 높아진 후 발효기간이 경과함에 따라 완만히 증가하는 경향을 보였으며, 염함량이 높은 된장일수록 낮은 값을 보였다. 단백질 함량은 염농도에 관계없이 발효기간에 따른 변화 없이 일정한 수준을 유지하였다. 아미노태 질소 함량은 발효기간에 따라 지속적으로 증가하는 경향을 보였으며, 저염 된장에서 높은 값을 보였다. 휘발성 염기질소는 발효 중반까지 일정한 수준으로 유지되다가 발효 후기에 급격히 증가하였으며, 염농도가 낮을수록 높은 경향을 보였다. 환원당은 발효 중기에 증가하다가 발효 후기에는 감소하였다. 염 함량을 달리하여 제조한 된장의 적정 발효기간은 염농도 8%에서 2-11주, 10%에서 3-12주, 12%에서 6-14주, 14%에서 9-15주로 염 함량에 따라 길어졌다. 된장의 관능적 품질은 낮은 염농도에서 단기간 발효한 된장이 높은 염농도로 장기간 발효한 된장에 비하여 우수하였다.
생식 첨가량을 3.7-11.7%로 조절한 생식음료의 분산안정성을 터비스캔을 사용하여 광학적 방법으로 측정하였다. 생식음료는 제조 후 시간이 경과함에 따라 음료 하부에서는 침전이 생성되어 후방산란광플럭스(이후 후광플럭스로 약칭)가 강해진 반면, 중간부에서는 청징현상에 의해 약해졌으며, 최상층부에서는 거품이 생성되어 다시 강해졌다. 생식 첨가량이 증가할수록 침전이 서서히 진행되었으며 전체 침전층은 두텁게 형성되었다. 침전에 의한 후광플럭스의 퍼센트변화율의 시간에 따른 변화는 로그 모델이 적용되었으며 결정계수는 0.979-0.988로 높은 값을 보였다. 생식 첨가량은 생식음료의 상층부에 형성되는 청징층과 하층부의 침전층의 경계면, 즉 청징층 선단의 이동속도 뿐 만 아니라 분리된 층의 수에도 영향을 미쳤다. 생식 첨가량 3.7%의 경우는 2층으로, 5.7-9.7%의 경우는 3개 층으로, 11.7%는 4개 층으로 분리되었다. 생식음료 제조 후 17-29분 사이에 1차 청징층 발생이 시작되었고 22-53분간 지속되었으며, 생식첨가량이 낮을수록 청징층 두께는 증가하였다. 청징층 발생은 생식 첨가량 7.7%인 음료에서 가장 늦게 나타나 최대 분산안정성을 보였다.
다양한 식품군을 동결건조 또는 저온에서 건조하여 분말화한 생식은 물과 혼합 시 분산성이 저조하고 섭취방법이 불편하여 생식 소비 및 시장 확대에 제약이 있다. 생식의 섭취를 편의화하기 위하여 음료 타입의 생식을 개발하고자 생식 3종(BS, ES, SS)을 음료로 제조하여 품질을 비교하였다. 생식음료의 이화학적 특성과 관능적 특성을 조사한 결과 BS의 기호도가 가장 높았으며, 생식첨가량은 7.7%에서 기호도가 가장 우수하였다. 생식 BS를 사용한 음료의 경우 생식 함량이 증가함에 따라 점조도는 증가하였으며 함량 5.7% 이하의 음료는 팽창유체 양상을 보이다가 7.7%와 9.7%에서는 뉴튼유체와 유사한 특성을 나타내었고 11.7%에서는 뚜렷한 의가소성을 보였다. 생식음료 점조도의 시간의존성은 생식함량 5.7-9.7% 사이에서는 레오펙틱형 시간의존성이 관찰되었으나 3.7%와 11.7%에서는 시간 독립적이었다.
