        Mushroom is cultivated as one of the major economical crops in many areas in Korea. The total production has steadily increased from approximately 186,400 M/T in 2007 to 190,104 M/T in 2009. The oyster mushroom, Pleurotus ostreatus, showed the 2th production to 46,598 M/T in 2011. This study was carried out to investigated optimum mixing ratio of Korean natural Acer tegmentosum for production of functional oyster mushroom. Mycelial growth of Pleurotus ostreatus showed the highest growth at the media of 10% Acer tegmentosum and was decreased to increase of Acer tegmentosum addition ratio. But mushroom mycelial growth at Acer tegmentosum media was slower than that of the control. Total nitrogen and carbon source of Acer tegmentosum was 0.19% and 44.4%, respectively and C/N ratio was 234. Total nitrogen source and pH of substrates mixed with Acer tegmentosum were 2.7∼3.0 and 4.8∼5.0, respectively. The contents of P2O5 and K2O were decreased by increasing Acer tegmentosum but the contents of CaO, MgO and Na2O were no significant difference. The contents of Fe and Mn were decreased according to increasing of Acer tegmentosum, but the contents of Zn and Mn were no significant difference. Yields of fruiting body and diameter and thick of pileus were the highest at 10% Acer tegmentosum. The L value of pileus was the highest at the 30% Acer tegmentosum during mushroom harvest, but there was no significant difference in the a-value and the b-value. The contents of P2O5,, CaO, MgO and Na2O of fruiting body were no significant difference, but contents of Cu was decreased and Fe was increased according to increasing of Acer tegmentosum.

        팽이버섯의 품종, 저장 온도 및 저장기간에 따른 자실체의 유기산의 변화를 조사하여 저장기간을 판단하기위한 기초자료로 사용하고자 HPLC을 이용하여 유기산을 분석·정량하였다. 팽이버섯 자실체에 존재하는 acetic acid, butyric acid, citric acid, fumaric acid, DL-isocitric acid, L(+) lactic acid, D-malic acid, propionic acid, succinic acid, D-tartaric acid 등 총 10종류의 유기산을 분리하였다. 대부분의 품종에서 acetic acid 함량이 가장 높게 나타났으며, fumaric acid 함량이 가장 낮았다. mono-carboxyl group의 유기산인 acetic acid는 품종 및 저장온도에 따른 함량에 차이도 있으면서 저장기간에 따라 일률적으로 증가하는 경향을 보였다. 그러나 butyric acid 등은 저장온도 및 품종에 따라 함량의 차이가 심하였다. Di-carboxyl group의 유기산인 fumaric acid는 저장 및 품종에 따른 차이 없이 감소하였으나, 저장기간 동안에 확인되는 함량이 1.5mg/g으로 그 함량이 적고, ASI 4149, 4166균주에서는 약간 다른 경향을 보이고 있다. 그 외에 malic acid 등은 저장온도 및 품종에 따라 함량의 차이가 심하였다. Tri-carboxyl group의 citric acid는 저장온도 및 품종에 상관없이 저장기간에 따라 증가하여 효과적으로 저장기간을 평가할 수 있는 지표로 사용할 수 있을 것으로 추정된다. isocitric acid는 저장온도, 품종 및 저장기간에 따라 성분함량의 변화가 심하였다.

        Mushrooms has been used for food ingredients since early times. Recently it is being used as a good pharmaceutical material or functional bio-material. Especially Ganoderma lucidum, Phellinus and Inonotus are used for medicinal purposes. Ganoderma is a basidiomycete white rot fungus which has been used for medicinal purposes for centuries particularly in China, Japan and Korea. A great deal of work has been carried out on Ganoderma lucidum. The metabolites consist of mainly polysaccharides and terpenoids. At present, Ganoderma is a health food supplement to support cancer patients, yet the evidence supporting the potential of direct in vivo anticancer effects should not be underestimated. the aim of this study was to investigate the anti-cancer effects and anti-inflammatory of Ganoderma lucidum in some kinds of human cell lines (Gastric:AGS, liver:HepG2, breast:MCF-7, lung:A549, uterine:Hela, Raw 264.7) we are examined the cell viability by MTT assay and effects of anti-inflammatory by NO assay. Anticancer and anti-inflammatory effect in the 10strains were selected.

        Ganoderma lucidum, a popular medicinal mushroom, has been used in China for longevity and health promotion since ancient times. Recently it is being used as a good pharmaceutical material or functional bio-material. Total 106 strains was cultured at temperature(25 ℃, 30 ℃ ,35 ℃) to study cultural properties of Ganoderma lucidum. Then the mycelium growth was digitized in the average diameter. With the result, Ganoderma lucidum strain has the optimum growing temperature at 25 ℃ with 25 strains, 30 ℃ with 70 strains and 35 ℃ with 11 strains. The 84strains were cultivated in wood for investigating the cultivation properties of Ganoderma lucidum. With the result, 70mm~90mm of the pileus diameter (major axis, mm) and 70mm-80mm of pileus diameter (minor axis, mm) was measured after one year cultivation. And 1,000-2,000 ㎟ of strain took the most the area (major axis x minor axis). The 30-40mm of stipe length,1time of harvesting and volume was 200g~400g took the most. 10cm-15cm of the pileus diameter (major axis, mm), 5cm~7.5cm of pileus diameter (minor axis, mm) and 15-20mm of thickness was measured after 2 year cultivation. And 50~100 ㎟ of strain took the most the area (major axis x minor axis). The 5-10mm of stipe length, 2 times of harvesting and volume was 200g~400g took the most. we are selected a middle model through this result. phylogenetic relationships was drawn based on the ITS sequence analysis in Ganoderma lucidum.

        Mushroom is cultivated as one of the major economical crops in many areas in Korea. The total production has steadily increased from approximately 186,400 M/T in 2007 to 198,563 M/T in 2009. Several bacteria are known as the causal agents of diseases of the cultivated button mushroom (Agaricus bisporus) and oyster mushroom (Pleurotus ostreatus). Pseudomonas tolaasii is the causal agent of brown blotch disease of commercial mushrooms. Colonization of mushroom caps by the bacterium results in development of brown or cream lesions on pileus and stipe. These lesions are slightly concave spots and can be round or spreading. Antagonists against P. tolaasii, HC1 were selected and their control efficacy of brown blotch disease was investigated in this study. The HC1 isolate was selected as an inhibitor of tolaasin activity by bioassay on potato and it was identified as Pseudomonas sp. by the cultural, physiological and biochemical properties and analysis of the 16S rRNA. Control efficacy of brown blotch disease by HC1 treatment was 69% on Agaricus bisporus, 68% on Flammulina velutipes and 55% on pleurotus ostreatus respectively.

        The total production has steadily increased from approximately 186,400 M/T in 2007 to 198,563 M/T in 2009. Winter mushroom, Flammulina velutipes, with 61,057913 M/T in 2009, showed the highest production. Several bacteria are known as the causal agents of diseases of the cultivated button mushroom (Agaricus bisporus) and oyster mushroom (Pleurotus ostreatus). Pseudomonas tolaasii is the causal agent of brown blotch disease of commercial mushrooms. Antagonists against P. tolaasii, HC5 were selected and their control efficacy of brown blotch disease was investigated in this study. After proceeding antagonistic test, HC5 was selected as a strong antagonist against P. tolaasii and the HC5 strain was identified as P. azotoformans with the cultural, physiological and biochemical properties and analysis of the 16S rRNA. Control efficacy of brown blotch disease by HC5 treatment was 73% on Agaricus bisporus, 78% on Flammulina velutipes and 71% on pleurotus ostreatus respectively.

        Cobweb disease symptoms were observed in a mushroom farm in Buye, Korea during a disease survey in 2008-2011. Five isolates of Cladobotryum sp. were obtained from the infected caps and stipes. These isolates of Cladobotryum sp. were identified as C. mycophilum based on their morphological, cultural characteristics and analysis of the ITS sequences. Early symptoms were noticed as round, fleshy, yellowish brown lesions on mushroom caps. Late symptoms progressed when the parasitic fungus formed white cobweb circular colonies on dead or damaged pinheads, spread on the surface of the casing, and covered entirely fruiting bodies. Optimal temperature and pH for mycelial growth on MEA is 23℃ and 6.0. Microscopically the spores of the fungus are large and most 2~3 celled produced on vertically branched conidiophores. Mushroom caps turned dark brown and shrunk due to soft rot. Testing of sensitivity to selected fungicides showed that isolate was highly resistance to Mancozeb and Thiophanate-methyl, moderately sensitivity to Iprodione, and highly sensitivity to Benomyl, Prochloraz-Mn and Carbendazim.

        품종 저항성 조사방법별로병원균과 버섯품종을 다르게 하여 품종간의 차이점이 있는지를 검토하였다. 푸른곰팡이 균과 품종별 대치배양시험에서 대치라인 형성, over growth, Lysis 증상이 나타났으며, 공시품종 중에 ASI 2183, ASI2504, ASI 2477 균주가 병저항성이 있을 것으로 판단되었다. 배양여액을 이용한 방법에서는 ASI 2240, ASI 2479, ASI 2181들이 저항성을 나타냈으며, 대치배양에서는 다른 경향을 보이는 경우도 있었으나, ASI 2479, ASI 2240 균주는 대치배양, 배양여액 시험결과에서 모두 저항성을 보였다. 배양후 접종법에서는 저항성을 보이는 버섯균주는 ASI 2479, ASI 2333, ASI 2181 등이었으며, 가장 감수성 품종은 큰느타리버섯인 ASI 2302 균주로 나타났다. 푸른곰팡이균주 같은 균주에서도 품종에 따라 각기 다른 특성을 나타냈으며, 동일품종에서 푸른곰팡이균주에 따라 각기 다른 병원성을 나타냈다. 느타리버섯과 푸른곰팡이균의 상호관계에서 다양한 증상이 발생하며 느타리버섯 품종에 대한 저항성 유무를 판단할 수 있다고 판단된다. 하지만 처리량이 많은 경우 동일 처리 내의 반복처리에서도 병징의 차이가 발생하는 경우가 있으며, 이는 시험에 사용한 버섯균과 병원균의 접종원의 활력의 차이에 의해 발생하는 것으로 추정되므로 주의해야하며, 저항성 균주로는 붉은느타리종, 감수성균으로 ASI 2302 균을 대조 균으로 적용하면 효과적으로 저항성 검정을 수행할 수 있을 것으로 판단된다.

        본 연구는 느타리버섯 자실체가 생육온도에 따른 수량성과 자실체의 형태적 특성을 확인하기 위하여 느타리버섯 품종 중에서 저온, 중온, 고온성 품종을 구분하여 12개 품종에 대하여 실험을 실시하였다. 수량성에 있어서는 품종의 온도 반응적 특성에 맞는 온도에서 수량성이 가장 높게 나타났다. 형태적 특성에 있어서는 갓크기는 온도처리에 따른 일정한 경향을 보이지 않았으며, 갓두께에 있어서는 저온 처리인 10℃에서 품종에 관계없이 가장 두껍게 나타났다. 대길이는 삼복 외 4품종은 온도가 증가하면서 같이 증가하였으며, 그 외의 품종은 품종에 따라 약간의 차이를 보였다. 대굵기에 있어서는 대체적으로 온도 증가에 따라 가늘어지는 경향이나 품종간 약간의 차이가 나타났다. 버섯수확소요일수에서는 10℃처리에서 가장 기간이 길었으며, 품종별 가장 짧은 소요일수를 보인 것은 품종의 온도 특성에 따라 다르게 나타났다. 품종에 따라서 온도에 반응하여 기형적 형태를 보이는 경우가 있었는데 이런 특이적 현상은 수한1호의 경우 저온 처리인 10℃에서 버섯이 발이후 어린 버섯 갓 표면에 백색 인피가 형성하며 성장 후에는 갓의 일부분에만 발생하거나 없어지는 현상이 있었으며, 춘추2호에서는 고온인 20℃ 이상에서는 대가 뒤틀리며, 대기 휘어지는 특이적 현상이 발생하였다.

        최근 수요가 많은 식용버섯 중 팽이버섯을 대상으로 저장기간 중에 발생하는 자실체 내의 당 종류 및 함량변화를 검토하고자 시험을 수행하였다. 팽이버섯 자실체에는 단당류로는 xylose 및 glucose, ribose, 이당류의 당은 lactose 및 trehalose, 당알코홀류 는 glycerol 및 mannitol, myoinositol, sorbitol 등이 확인되었다. 팽이버섯에 가장 많은 것은 단당류인 xylose로 백색계열은 47.68㎎/g, 갈색계열은 63.28㎎/g로 갈색계열의 함량이 높으며, 품종에 따라 약간의 차이를 보였다. 저장기간에 따른 당 함량은 단당류 및 이당류, 당알코올 모두 저장 시간이 증가하면서 같이 증가하는 추세를 보이고 있으나, trehalose는 감소하였다. 저장온도에 따라서는 큰 차이는 없었으나, lactose는 4℃ 저장에서 저장기간이 길어질수록 크게 증가하는 것으로 나타났으나, -1℃ 저장에서는 검출되지 않았다.

        느타리버섯 푸른곰팡이병에 대한 품종 저항성은 포장에서 효과적인 검토가 불가능하여 한천 배지상에서 푸른곰팡이균이 균을 저해하는 특징 fungistatic, lysis, toxin enzyme 등에 대한 각각의 특성을 실내에 검정할 수있는 방법을 검토하기 위하여 균사생장과 특이 현상을 조사하였다. 대치배양에서는 느타리버섯 균주에 따라 대치선 형성위치, 푸른곰팡이균의 느타리균사 생장부분 위로 생장(overgrowth), 대치선 이후의 버섯균의 용균(lysis)현상이 발생하여 효과적인 저항성 검정이 가능하였다. 배양여액을 여지에 적용하는 방법은 버섯균과 병원균의 종류에 따른 균사생장 및 특이적 현상이 타나나지 않았으나, well plate를 이용한 배양여액 희석방법으로 균사생장의 억제정도의 확인이 가능하였다. 분활 petri dish를 활용하여 동시배양의 경우 약간의 버섯균이 억제되는 현상은 보였으나, 푸른곰팡이균이 버섯균 생장 부분 위로 덮어 효과검정이 불가능하였다. 버섯균을 petri dish 전체에 배양한 후 그 위에 병원균의 균총을 접종하는 방법은 overgrowth, lysis등의 현상이 발생하여 병원성의 검정이 용이하였다. 실내에서의 느타리버섯균의 푸른곰팡이병에 대한 저항성 검정은 대치배양, 배양여액법, 생장후 접종법에 의한 방법으로 가능한 것으로 판단되었다.

        There are an estimated about 20,000 species of mushrooms in the world. Of these, approx. 2,0000 species is edible and around 1,500 species of them grow naturally in Korea. About 300 species of Korean natural mushroom could be classified as edible and medical purpose, but actual use is restricted to only 20 species. Mushrooms has been used for food ingredients since early times. Recently it is being used as a good pharmaceutical material or functional bio-material. Especially Ganoderma lucidum, Phellinus and Inonotus are used for medicinal purposes. In this meaning, the study about chemical component separation and active component effect is necessary. For the study, the systematic establishment of mushroom should be done. In this study, we are trying to build a library by using extracts from the edible, medicinal and wild mushrooms. The physiological activities(antioxidant, anticancer and anti-inflammatory effects) were performed to investigate the effectiveness of mushrooms.

        This study was carried out to investigate neuronal protective activity of fruiting body of Hericium erinaceum. In order to search the effective active compound against amyloid beta peptide-induced oxidative stress on neuronal cells, rat pheochromocytoma cells (PC12), Extracts of Hericium erinaceum were screened and evaluated using both the 2’,7’-dichlorofluorescin diacetate assay (DCF-DA), and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. According to above assays, Solvent partitions of extracts were selected for further purification and isolation of anti-Alzheimer’s disease compound as it exerted the highest protective effects against hydrogen peroxide (H2O2)-induced oxidative stress.

        The standard does much to improve merchantable quality, distribution efficiency and fair dealings by shipping of the standard agricultural products. Mushrooms notified as the standard are five items; Pleurotus ostreatus, Pleurotus eryngii, Flammulina velutipes, Agaricus bisporus and Ganoderma lucidum. But many farmers are suffering from a strict standards. So these standard is required modification to fit farmhouse situations. This study was carried out to investigate mushroom quality of farm shipping and quality change during preservation at various treatment. Flammulina velutipes and Agaricus bisporus preserved at different temperature( 4℃, 10℃, 20℃) to investigate quality changes. Pileus diameters of Flammulina velutipes was 67% of first grade and 33% of second grade at the early stage. After two weeks, pileus diameters was not signigicant changes; 50% of first grade, 50% of second grade at 4℃ and 50% of second grade, 50% of third grade at 10℃. Although there were no significant changes of diameter at 20℃, most of the fruit bodies were decayed. In case of Agaricus bisporus, pileus diameter was 15% of first grade, 81% of second grade and 4% of third grade at the early stage. The more storage period longed, the more the color of the fruit body was browned. But pileus diameters was not signigicant changes. Hardness and chromaticity of these mushroom was declined as time passed. Now we are carrying out more research on some mushroom’s standards for improve consumer confidence.

        We investigated 18 amino acid contents in the fruiting bodies of Agaricus strains to understand their variation in different growth temperature. Although total content of amino acid was more or less different depending on the isolates, it increased until the temperature was 22℃ and decreased above 22℃. Commercial strains of A. bisporus, such as Yangsongi-505, Yangsongi-705 and commercial strain A collected in the market exhibited these general trend in total amino acid content. However, another commercial strain B did not follow these trend. A strain of A. bitorquis did not produce mushrooms at 10 and 13℃, but mushrooms harvested in the other treated temperatures exhibited with the similar patten. Generally, the most amino acid contained in Agaricus spp. was cysteine and followed by phenylalanine, glutamic acid, lysine, proline and histidine. We detected two trends for the contents of each amino acid in the fruiting bodies of Agaricus spp. grown at the different growing temperature. Some amino acid contents were largely correlated with the growth temperature, but the others showed the trend of increase until 22℃ and decrease above 22℃.

        We investigated 18 amino acid contents in the fruiting bodies of Agaricus strains to understand their variation in the harvesting periods. Total content of amino acid was different depending on the isolates. In the case of A. bitorquis and A. brunnescens, the total amino acid content increased and correlated with the harvesting periods. For commercial strains of A. bisporus, such as Yangsongi-505, Yangsongi-705 and commercial strain A collected in the market, it increased until the second harvesting period, but decreased in the third harvesting period indicating that these three isolates exhibit the exceptions for the general trend. Among isolates we used in our experiments, the isolate of A. brunnescens contained the highest and two isolates of A. bisporus were the lowest in total amino acids. We found two trends for the contents of each amino acid in the fruiting bodies of Agaricus spp.. One is the same with the trend of the total amino acid content and the amino acid content were largely correlated with the harvesting period. The other is the trend of increase of amino acid until the second harvesting period and decrease of amino acid in the third harvesting period, which was also shown in the total amino acid content of A. bisporus. Generally, the most amino acid contained in Agaricus was cysteine and followed by phenylalanine, glutamic acid, lysine, proline and histidine.

        To understand the saccharide contents and quantity of winter mushroom (Flammulina velutipes) according to its growth temperature, we measured the saccharide contents at different growth temperature. In our results, the saccharide of its fruiting body turned out to be mainly composed of xylose, trehalose and mannitol in all treatments. In the other hand, Ribose, myo-inositol and sucrose were detected in some treatments. The quantity of trehalose decreased as the growth temperature increased with a variation of its quantity depending on the isolates used in the experiments. In the case of xylose and mannitol, detected in all treatments, the pattern of their quantities was not possible to be profiled and the pattern might be largely depending on the isolates. However, the quantities of xylose and mannitol were largely in a direct proportion and the fluctuation of their quantities was congruent with the exception of ASI 4103, ASI 4166 and ASI 4065. The xylose quantity of ASI 4103 and ASI 4166 increased until the temperature was up to 10℃ and decreased when the temperature was above 10℃. That of ASI 4065 decreased as the temperature rose and increased when above 13℃. The mannitol quantity of ASI 4065 and ASI 4166 decreased until the temperature was up to 10℃ and increased when the temperature was above 10℃. That of ASI 4103 increased as the temperature rose and decreased when above 13℃.

        This study was carried out to decrease the royalties through the dissemination of domestic mushroom varieties by following UPOV (Union Internationale Pour la Protaection des Obtentions Vegetablue: the international union for the protection of new varieties of plants). Although RDA has bred various mushrooms of 25 species of 78 varieties since 1990, most of these were not cultivated in mushroom farms due to two reasons. One is the weaknees of spawn supply system caused by increasing own spawn production and the other is the difficulty in replacing a new variety due to the year-round production in bottle cultivation. Therefore, we made a program to disseminate domestic mushroom varieties. In 2010, five species of 14 varieties were provided as a form of spawn for 43 farms that do not have their own cultivation facility and 11 species of 18 varieties were provided for 45 farms that were equipped with their own cultivation facility. In addition, the breeders explained the cultural characteristics of their own varieties and consulted cultural farms. Since these activities were helpful to mushroom farmers, we plan to open the presentation for mushroom instructor in February 2011 with the consideration of the opinion of mushroom farmers to fully disseminate the spawn of domestic mushroom varieties for the bottle cultivation farms.

        This study was carried out investigate to availability of commercial microbial pesticide and antibacterial activity of isolates isolated from different mushroom media. Ten commercial microbial pesticide and EM liquid were collected from different company. EM of these one has been used for control of mushroom disease and growth promotion at the mushroom farms. The density of bacteria and yeast in EM cultural liquid was higher than those of the crude liquid. The pH values of EM showed the low acid levels(pH 3.5~3.9) by organic acid secreted from microorganism. The kinds of organic acid was acetic acid, lactic acid and succinic acid. The dominant bacteria isolated from EM liquid was Lactobactillus sp.(21 strains), Acetobacter sp.(9 strains), PaeniBacillus sp.(9 strains) and others(12 strains). The organic acid bacteria isolated from fermentation foods( was 92 strains and the dominant genus was Weissella sp.(41 strains), Leuconostoc sp.(21 strains), Enterococcus sp.(9 strains), Lactococcus sp.(9 strains) and others(12 strains). And we isolated 2,500 bacteria from oyster mushroom and button mushroom cultural media for selection of antagonistic bacteria. Thirty five strains of these isolates showed very strong antagonistic activity. These strains were identified Bacillus amyloliquefaciens, Bacillus subtilis, Brevibacterium halotolerans, Pseudomonas libanensis, Stenotrophomonas maltophilia and Alcaligenes faecalis by 16S rDNA analysis.
