        The present study was designed to investigate the antiproliferative activity and molecular mechanisms of Bibimbap in HT-29 human colorectal adenocarcinoma cells. Bibimbap extract inhibited the proliferation of HT-29 cells by 50% at a concentration of 10.1±0.17 mg/mL for 48 h. The population of live cells decreased slightly, and the morphology changed with a reduction in cell volume (pyknosis) with Bibimbap. Treatment with 5 mg/mL of Bibimbap resulted in slight cell shrinkage. Furthermore, as the Bibimbap dose increased to 10 mg/mL, these characteristics were more evident, and HT-29 cells exhibited partial detachment by staining with the DNA-binding dye Hoechst 33342. Flow cytometric analysis by Annexin V and PI double staining showed that Bibimbap increased the levels of apoptosis. Analysis of the mechanism of these events showed that Bibimbap-treated cells exhibited a mitochondria-dependent apoptotic pathway through the modulation of caspase-3, caspase-8, caspase-9, and poly-ADP ribose polymerase, as well as Bax and Bcl-2 expression in dose- and time-dependent manners. Consequently, Bibimbap exerts a significant antiproliferative effect on HT-29 human colorectal adenocarcinoma cells.

        The purpose of this study was to investigate the biological activity of fucoidan, a sulfur-containing polysaccharide, on cytotoxicity and apoptosis in the human HT-29 colorectal cancer cell line using cell viability, Flow cytometry, Western blot, and RT-PCR analyses. Fucoidan inhibited the proliferation of HT-29 cells by 39.6% at a concentration of 100 μg/mL for 72 h. The inhibition was dose-dependent and accompanied by apoptosis. Flow cytometric analysis showed that fucoidan increased early apoptosis and late apoptosis by 65.84% and 72.09% at concentrations of 25 and 100 μg/mL, respectively. Analysis of the mechanism of these events indicated that fucoidan-treated cells exhibited increases in the activation of caspase-3, caspase-8, and PARP in a dose-dependent manner. These results suggest that fucoidan may inhibit the growth of human colorectal cancer cells by various apoptosis-promoting effects, as well as by apoptosis itself.

        The objective of the present study was to investigate the molecular mechanisms involved in the activity of [10]-gingerol using A2058 human melanoma cells. [10]-Gingerol inhibited the proliferation of A2058 cells by 50% at a concentration of 52 μM. Such inhibition was dose-dependent accompanied by morphological change indicative of apoptosis. Furthermore, flow cytometric analysis by Annexin V and PI double staining showed that [10]-gingerol increased the extent of apoptosis. Analysis of the mechanism of these events indicated that [10]-gingerol increased the ratio of Bax to Bcl-2, resulting in the activation of caspase-9, caspase-3, and poly-ADP-ribose polymerase in a dose-dependent manner.

        The aim of study to investigate the phytochemicals and biological activities the bark of Betula schmidtii. The studies consisted of the solvent extraction, followed by the isolation of phenolic components 1~3 from ethyl acetate-soluble fraction of Betula schmidtii Bark. Their chemical structures were identified as arbutin (1), ρ-coumaric acid (2) and ferulic acid (3) using Ultraviolet-Visible (UV-Vis) Spectrophotometer, Electrospray Ionization Mass Spectrometry (ESI-MS) (negative ion mode), 1H-Nuclear Magnetic Resonance (NMR), 13C-NMR, 1H-1H Correlation Spectroscopy (COSY) and 1H-13C Hetero Nuclear Multiple Quantum Correlation (HMQC) spectral data. Compounds 1~3 shows the anti-oxidant effect with IC50 values of 29.74±1.52, 21.32±1.07 and 34.41±1.24 in 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity, respectively. Also, compounds 1~3 exhibited mushroom tyrosinase inhibitory activity with IC50 values of 31.14±1.07, 42.54±1.46 and 69.22±1.43 μM, respectively.

        Red raspberry (Rubus crataegifolius) is a well-known fruit with excellent antioxidant, anti-inflammatory, anti-bacterial and anti-cancer resources containing bioactive phenolic acids, terpenoids, glucosinolates, and glycosides. The aim of our study is to investigate the effect of red raspberry cookies on quality characteristics and antioxidant activities. Cookies were prepared adding different concentrations of red raspberry powder (0, 5, 10, 20, 30, and 50% to the flour weight). The bulk density, pH, spread ratio, loss rate, leavening rate, color, hardness of dough, and cookies were investigated. As a result, the pH of the dough, leavening and loss rate, and L- and b-value of the cookies are significantly decreased with increasing levels of red raspberry powder (p < 0.05). On the other hand, spread ratio, a-value, and brix of the cookies are significantly increased with increasing levels of red raspberry powder (p < 0.05). Total phenol compounds and FRAP assay are significantly increased with increasing levels of red raspberry powder (p < 0.05). Based on the overall observation, red raspberry cookies with 10-20% Rubus crataegifolius powder is recommended to take advantage of its functional properties with consumer acceptability.

        본 연구는 인체 유래 유방암세포인 MDA-MB-231 세포 증식을 억제하고 세포사멸을 유도하는 천연소재 발굴을 목적으로, 북한산 두릅 추출물을 MDA-MB-231 세포에 처리하여 세포사멸 및 작용기전을 규명하였다. 실험 결과, 두릅 추출물 처리 농도가 증가할수록 세포증식이 감소하였고, 세포질의 응축과 핵이 분절되는 등 apoptotic bodies를 형성하였다. 또한 유세포 분석기를 사용하여 MDA-MB-231의 세포주기가 억제되었고, 세포사멸의 특징인 sub-G1 수치가 증가되는 것이 관찰되었고, 두릅 추출물 농도가 증가함에 따라 apoptotic 세포가 유의적으로 증가하였으며, 특히 early apoptosis 보다 late apoptosis가 더 많이 진행됨을 확인할 수 있었다. 이를 바탕으로 MDA-MB-231 세포사멸과 관련된 유전자를 확인하고자 RT-PCR과 western blotting을 수행한 결과, 세포사멸의 주요한 조절인자인 anti-apoptotic인 bcl-2 발현이 두릅 추출물 처리 시 농도 의존적으로 감소되었고, 반대로 Bax의 발현은 증가됨을 확인하였다. 이를 통해 불활성화 형태로 존재한 pro-caspase-9과 pro-caspase-3의 발현이 시료 농도가 증가할수록 감소되었고, 활성화된 형태인 cleaved caspase-9과 cleaved caspase-3의 발현은 두릅 추출물 처리 농도에 의존적으로 증가하였다. 뿐만 아니라, 세포 사멸 과정의 주요 인자인 PARP 또한 시료 농도가 증가할수록 절단 현상이 비례적으로 증가하였다. 이상의 실험 결과를 근거로, 북한산 두릅 지상부의 70% ethyl alcohol 추출물이 MDA-MB-231 인체 유방암 세포의 증식을 억제하는 효과가 있음을 확인하였고, 세포사멸과 관련된 활성기전을 규명하였다. 추후 암 예방/치료제로서 두릅을 활용하기 위해서는 활성 본체와 안정성 규명 및 임상실험 등 추가 연구가 필요할 것으로 사료된다.

        Epimedium koreanum Nakai has been used in traditional medicine as an aphrodisiac, hypotensive, and neurasthenia;however, the cellular mechanism of E. koreanum Nakai cultivated in North Korea on HCT116 colon cancer cellactivity has not been investigated. This study is to investigate the effect of E. koreanum Nakai on apoptosis of thehuman colon cancer cell line HCT116. The anti-proliferative activity of E. koreanum Nakai on HCT116 was iden-tified through MTT, Western Blot, and RT-PCR analyses. The results show that 70% ethyl alcohol extracts of E.koreanum Nakai inhibited the growth of HCT116 colon cancer cells (IC50: 1.2mg/mL on 24 hr). Concomitant acti-vation of the mitochondria-dependent apoptotic pathway occurred via modulation of Bcl-2 and Bax expressions,resulting in activation of cleaved caspase-3 and cleaved caspase-9.

        본 연구에서는 영양과 건강, 그리고 편의성까지 고려한 즉석식품 형태의 쌀미음을 제조하였다. 제조된 쌀미음 분말의 영양성분 구성비를 일반성분 분석을 통하여 분석 하였고, 과립공정에 따른 재수화능의 향상을 연구하였다. 과립분말은 혼합분말보다 지방함량과 단백질 함량이 각각 0.9, 1.9% 높게 나타난 반면, 탄수화물 함량은 상대적으로 3.2% 낮게 나타났다. 과립분말의 단백질 조성은 71.1-118.5 g으로 1일 권장 단백질 섭취량 45-55 g보다 많았는데, 이는 일반적인 환자식이 지니는 공통적인 특성이다. 분말 입자의 침강 속도를 보여주는 분산성의 경우 과립분말(93.7%)은 혼합분말(77.0%) 보다 높게 나타났고, 재수화에 걸리는 시간도 과립분말(122.3 초)이 혼합분말(305.3 초)보다 빨랐으며 이들 결과는 과립공정이 분말의 재수화능을 향상시켜 주었음을 보여준다. 호화쌀가루를 이용한 쌀미음은 기존의 우유 단백질 위주의 환자식을 곡류로 대체하며 영양강화 쌀 가공제품의 개발에 의미가 있다.

        In this study, soy-powder yogurt (SPY) with enhanced levels of γ-aminobutyric acid (GABA) and isoflavone aglycone was produced from sprouting high-protein soybeans (HPSs). The fermented steam-HPS sprouts (0 to 4 cm) were fermented (72 h) with Lactobacillus brevis, and the total free amino acids (FAAs) of the formed mixtures were determined to be 79.53, 489.93, 877.55, 780.53, and 979.97 mg/100 mL in the fermented HPS (FHPS), and the fermented steam-HPS with 0 cm (FSHPS-0), 1 cm (FSHPS-1), 2 cm (FSHPS-2), and 4 cm sprouting lengths (FSHPS-4), respectively. The levels of glutamic acid (GA) and GABA were observed to be the highest, 100.31 and 101.60 mg/100 mL, respectively, in the unfermented HPS (UFSHPS-1, 1 cm) and FSHPS-1 sprouts, respectively. Moreover, the total contents of the isoflavone glycoside form decreased proportionally to the increasing total levels of isoflavone aglycones after fermentation in FSHPS-0, FSHPS-1, FSHPS-2, and FSHPS-4. The levels of isoflavone aglycones were detected as 350.34, 289.15, 361.61, 445.05, and 491.25 μg/g in FHPS, FSHPS-0, FSHPS-1, FSHPS-2, and FSHPS-4, respectively. While FSHPS-1 exhibited the highest DPPH (63.28%) and ABTS (73.28%) radical scavenging activities, FSHPS-4 contained the highest isoflavone aglycone ratio (81.63%). All in all, the FSHPS-1 mixture prepared in this study exhibited high GABA content and functional prosperity, thereby making it suitable for potential applications in the soy-dairy industry.
