본 연구는 관상가치가 뛰어난 산꼬리풀의 분화 재배법을 확립하고자 차광, 기비, 추비, 왜화제 및 적심처리를 각각 달리하여 수행하였다. 연구의 결과 차광 시 산꼬리풀의 생육이 감소 하였으며, 차광률이 증가할수록 크게 억제되었다. 기비 처리는 초장, 절간장, 경직경 및 엽수를 유의적으로 증가하였으나, Hyponex를 이용한 추비 처리는 무처리와 비교했을 때 생육증가에 큰 영향을 미치지 않았다. 왜화제 처리시 diniconazole은 농도에 관계없이 왜화효과가 나타났으며, 농도가 증가할수록 크게 억제되었다. Daminozide는 저농도 처리시 무처리와 비교하여 큰 차이가 없었으나, 2000mg·L-1 농도로 처리하였을 때 왜화효과가 나타났으며, 개화율은 diniconazole 저농도에서 가장 낮은 결과를 보였다. 한편 적심 처리에 따른 유의적인 생육 차이는 확인되지 않았다.
This study aims to reveal how EA affects BAX and NF-kB involved in cell deaths from global ischemia, and to do this, observes the changes of BAX and NF-kB caused by EA application after transient global ischemia. The experimental method is to give rise to global ischemia and apply EA to 27 SD rats with the particulars of being six-week-old, male, around-300 gram-weighing, and adapted to laboratory environment for more than a week, and divide them into three groups, that is, GV20 EA group(n=9), L14 EA group(n=9), no-treatment GI group(n=9), and then observe their changes of BAX and NF-kB at the time lapse of 6 hours, 9 hours and 12 hours after ischemia, using western blotting. The numerical decrease of BAX expression at the time lapse of 9 hours after EA application, though not statistically significant, was observed in GV20 EA group and L14 EA group, and the NF-kB expression appeared statistically significant decrease in GV20 EA group and L14 EA group, but the expression was higher in the group with EA application. Therefore, EA application at the early phase of global ischemia is considered to affect BAX and NF-kB and play a positive role in decreasing apoptosis and cell deaths by inflammation.
This study investigated a suitable method that could be applied for Asian chain fern [Woodwardia japonica (L. f.) Sm.] to propagate gametophytes and promote sporophyte formation. The gametophytes used in all experiments were obtained from germinated spores in vitro and were subcultured at 8-week intervals. The most appropriate media for gametophyte propagation was identified by culturing 300 ㎎ of gametophyte in Murashige and Skoog (MS) basal medium (1/8, 1/4, 1/2, 1, 2), and Knop medium for 8 weeks. As a result, fresh weight of the gametophyte was increased by 56.7-fold on MS medium. Moreover, antheridium formation as well as gametophyte growth was improved on MS medium, especially. To improve the sporophyte formation ex vitro, 1.0 g of gametophyte was ground with distilled water and spread on eight combinations onto four different culture mediums, such as bed soil, peat moss, perlite and decomposed granite. Then generation and growth of sporophytes were investigated after cultivation for 10 weeks. As a result of this experiment, peat moss had a promotive effect of sporophyte formation at single-use and mixed culture soils. In particular, a mixture of bed soil, peat moss and perlite in a 1:1:1 ratio (v/v/v) led to the accelerated formation (782.5 ea/pot) and the frond growth of sporophytes. This included increases in length and width of fronds. However, promotive effect of gametophyte growth and sporophyte formation was not found at single-use and treatment with high ratio of bed soil.
The study was conducted to understand the relationship among university students’ interest in horticulture, psychological well-being and social development and the influence of measured variables. In order to collect data, the surveys were executed by convenience sampling on university students attending four-year universities in Daegu and Gyeongbuk regions of South Korea during the period from July 3 to July 17, 2016. A total of 307 survey results which measured the university students’ interest in horticulture, psychological well-being and social development were analyzed. The results showed that there are differences in the interest in horticulture, psychological well-being, and social development depending on the students’ general characteristics. According to the results analyzed by t-test and one-way ANOVA, the interest in horticulture depending on gender, religion, academic marks, family life satisfaction and school life satisfaction had significant differences showing high interest for the students with religion, good academic marks, high family life satisfaction and school life satisfaction as well as female students. Psychological well-being showed significant differences in academic marks, family life satisfaction and school life satisfaction, while social development displayed significant differences in gender, grade, academic marks, family life satisfaction and school life satisfaction. According to the results analyzed by correlation analysis, there was a significant correlation among university students’ interest in horticulture, psychological well-being and sociality development. Furthermore, regression analysis verified that university students’ interest in horticulture has a positive influence on psychological well-being and social development. The results of this study implies that university students with a higher interest in horticulture have higher levels of psychological well-being and social development.
Background : Bitter gourd (Momordica charantia) is a vegetable with pantropical distribution and contains the active ingredient, such as momordicine and charantin. Moreover, bitter gourd has been reported to exhibit antidiabetic, anticancer, cardiovascular-protective and antioxidant effects. But, the distinctive bitter taste of bitter gourd was not suitable for food preference. This study was conducted to evaluate the improvement effects of bitter taste of bitter gourd with natural fermentation and roasting process. Methods and Results : Bitter gourd fruits were obtained from Headeulnyeokae Co., Ltd.(Gang-Jin 59253, Repubilc Korea). Fruits of bitter gourd were prepared by natural fermentation process for 4h at 25℃. After natural fermentation, the fruits were dried in hot-air dryer for 15h at 49-55℃. The dried fruits were roasted in 500 g batches at 120℃ for 10 min in the electric rotisserie oven. After roasting, fruit pieces were extracted with hot water and cold water. One of them was cool-type, and the other was hot-type bitter gourd tea. In the sensory evaluation, hot tea and cold tea showed the high scores on color, flavor and overall acceptability. Conclusion : These mean that roasted bitter gourd had less bitter, and it could be utilized widely as drink and food material.
This study was carried out to develop an effective seed propagation method for Thalictrum rochebrunianum var. grandisepalum (H. Lev.) Nakai by analyzing seed dormancy types and germination characteristics. Seeds were collected between September to October at Gangwon province, and well-selected seeds were used while being dry-stored at 4±1℃. The seed size ranged 4.52 × 1.58 ㎜ and the weight of thousand seeds were 1,603.5 ± 0.02 ㎎. The moisture content was 7.2%. Seeds were achene type, and morphology characters showed an elliptical shape and rough texture, and light brown in color. Moist-chilling treatment was conducted for dormancy breaking because the seeds had an undeveloped embryo of liner type. The embryo had developed during a moist-chilling period, constantly, and fully developed in 10 weeks. Consequently, it seemed to be non-deep complex or intermediate complex type of morphophysiological dormancy, and embryo dormancy was broken by wet-chilling for 10 weeks. After 10 weeks of wet-chilling treatment, seed germination began. Germination percentage was higher in dark condition raher than light condition and recorded the maximum at 25℃ in the dark (16.3%). A pre-soaking treatment with a combined plant growth hormones promoted germination and shortened T50. Specifically, seed germination of 84.5% was achieved by pre-soaking of seeds with a combined solution of 500 ㎎/L GA3 and 10 ㎎/L kinetin for 24 h after a wet-chilling treatment for 10 weeks. Thus the effect of plant growth hormones coupled with chilling temperature on seed breaking dormancy provide asubsequent growth of seedlings for successful plantation.
In a mercury leaching test for waste using the Korean Standard Method (ES 06404.1), the pre-treatment process of an eluate is very complicated with a high possibility of contamination and low mercury recovery rate. It is also difficult to analyze multiple samples in a short time and the process generates experimental wastes. Accordingly, a direct mercury analyzer (DMA) applying thermal decomposition gold-amalgamation analysis has been recently used. The method shows a relatively high recovery rate for solid samples without complicated pre-treatment and it can be applied to both liquid and solid samples as the EPA method 7473 does. In order to use the auto-sampler in DMA for analyzing many elution samples from waste, this study checked recovery rates depending on acid solutions and additives during continuous analysis. The result showed a significant drop in recovery and precision except for an L-cysteine added sample. Considering commonly used acid-treatment of wastes, three types of acid solutions (nitrate, hydrochloric acid and sulfate) were chosen for analysis, and precision and accuracy were relatively high in nitric acid solution. It has been determined that accuracy and precision improved when 0.01% L-cysteine was added as an additive and this reduced the impact of continuous measurement. Therefore, during analysis of liquid samples or eluted samples using DMA continuously, introducing suitable additives is necessary depending on pre-treatment method in order to improve accuracy and precision in the analysis of mercury.
One-dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis (1D SDS-PAGE) was used to determine whether it would provide improved resolving power of hordein proteins concomitant with improved identification of Korean barley cultivars and germplams. This system gave rapid and reproducible separations of hordein polypeptides. Total fourteen of clear and easily scorable subunits were identified in Korean barley cultivars and germplasms and their polymorphic constitutions could provide biochemical genetic information in progeny analysis and endosperm quality improvement in barley breeding programs. Each hordein polypeptides residing in B, C, and D hordein pattern designations were scored to prepare a cultivar catalogue of protein patterns. On the basis of this character, 7 hordein polypeptide patterns were constructed from 108 barley cultivars and experimental lines. The molecular weight of hordein subunits in Korean barley cultivars and experimental lines varied in the range of 98 to 48 kDa. In contrast, less polymorphic hordein polypeptides were found in the low protein barley lines including malting barleys than those found in Korean barley cultivars and experimental lines
