        Immuno-MemBlot is a technique for detecting, analyzing, and identifying proteins, similar to the Western blot technique but differing in that protein samples are not separated electrophoretically but are spotted through circular or slot templates directly onto the membrane. Recently we developed a new Immuno-MemBlot (IMB) method applying immunoreactions and coloring procedures directly in the wells of MemBlot apparatus, which were connected by canals to perform drainage for reagent application and buffer irrigation. This IMB method was designed to get theimmunoblot results more rapidly and clearly than the previous immunoblot ones. This study is aimed to evaluate the analytical accuracy of IMB using different biological assay. In the sensitivity test of IMB the monoclonal antibody can clearly detect the 30 ng (about 12 pM) of Mucocidin peptide (35 mer), and is also available to detect at least 10 ng (about 4 pM) of Mucocidin peptide (35 mer). The IMB was effective in the quantitative analysis of methothrexate (MTX) assay for cellular apoptosis. And more, this IMB is useful to screen large number of specific samples with ease and accuracy in a short time. In the screenings for the presence of Mucocidin in saliva the quantitative comparison is conspicuous among 48 persons depend on the different conditions ofgender, drinking and smoking habits, and oral diseases. Therefore, it is presumed that, even though the target proteins were partly degraded, a specific epitope can be detected if a monoclonal antibody was still reactive. Conclusively, these data suggest that the IMB can be useful in the primary qualitativeand quantitative analysis of proteins in various fluids, i.e., blood, saliva, tear, urine, etc.

        This study is conducted to investigate the effects of growth characteristics (GC) and DPPH radical scavenging activity (DRSA) of the heading lettuce ‘Fidel’ depending on four concentrations of activated mineral groups (AMG: 1.6% active minerals and 0.03% CaO) added to basic nutrient solutions (diluted by 0, 0.05, 0.1, and 0.2%) and on four different light-emitting diodes (LED; B:R:W ratio = 0:1:12, 0:1:9, 0:5:5, and 2:3:5). Both 0.1 and 0.2% AMG showed better GC of lettuce in plant width, plant length, leaf width, leaf length, and the number of leaves than those of other AMG, while leaf thickness and chlorophyll value did not show significant difference among all AMG. Moreover, 0.1 and 0.2% AMG showed heavier shoot fresh weights than those of other AMG. As for the combinations of AMG and LED, B0:R5:W5 showed the best lettuce GC regardless of AMG compared to other AMG and LED combinations. As shown in the above results of LED, although there was a difference in lettuce growth by LED, the differences of lettuce growth by AMG were statistically significant. The DRSA was the highest at 82.8% in B2:R3:W5, followed by B0:R5:W5 at 77.8%. LED showed differences but AMG did not affect DRSA. Therefore, the optimal conditions in plant factory for GC and DRSA of the lettuce were 0.1% AMG and B0:R5:W5.

        Fruit trees are a key part of agriculture in rural areas and have recently been a part of ecotourism or agrotourism. This study analyzes user segmentation based on user motivation to determine characteristics of potential customers in fruit tree farms, and thereby develop fruit tree experience and educational programs. We conducted a survey of 253 potential customers of fruit tree experience programs in September 2017. Data were evaluated using factor and cluster analyses. The results of the cluster analysis identified four distinct segments based on potential customers' motivations, that is, activity-oriented, learning-oriented, leisure-oriented, and purchase-oriented. These clusters showed that significant differences in the preference of potential customers exist. Different markets were segmented based on the benefits sought by users. The segments’ characteristics were identified and activities relevant to each segment were proposed for rural tourism. Lastly, this study suggests directions for development of fruit tree farm experience and educational programs.

        Polyploidy is occurred by the process of endomitosis or cell fusion and usually represent terminally differentiated stage. Their effects on the developmental process were mainly investigated in the amphibian and fishes, and only observed in some rodents as mammalian model. Recently, we have established tetraploidy somatic cell nuclear transfer-derived human embryonic stem cells (SCNT-hESCs) and examined whether it could be available as a research model for the polyploidy cells existed in the human tissues. Two tetraploid hESC lines were artificially acquired by reintroduction of remained 1st polar body during the establishment of SCNT-hESC using MII oocytes obtained from female donors and dermal fibroblasts (DFB) from a 35-year-old adult male. These tetraploid SCNT-hESC lines (CHA-NT1 and CHA-NT3) were identified by the cytogenetic genotyping (91, XXXY,-6, t[2:6] / 92,XXXY,-12,+20) and have shown of indefinite proliferation, but slow speed when compared to euploid SCNT-hESCs. Using the eight Short Tendem Repeat (STR) markers, it was confirmed that both CHA-NT1 and CHA-NT3 lines contain both nuclear and oocyte donor genotypes. These hESCs expressed pluripotency markers and their embryoid bodies (EB) also expressed markers of the three embryonic germ layers and formed teratoma after transplantation into immune deficient mice. This study showed that tetraploidy does not affect the activities of proliferation and differentiation in SCNT-hESC. Therefore, tetraploid hESC lines established after SCNT procedure could be differentiated into various types of cells and could be an useful model for the study of the polyploidy cells in the tissues.

        Background : Cudrania tricuspidata Bureau is a widely used medicinal perennial woody plant. Obtaining information about the genetic diversity of plant populations is highly important for conservation and germplasm utilization. In this study, we developed single nucleotide polymorphism (SNP) markers derived from chloroplast genomic sequences to identify distinct Korean-specific ecotypes of C. tricuspidata via amplification refractory mutation system (ARMS)-PCR analyses. We performed molecular authentication of twelve C. tricuspidata ecotypes from different regions using DNA sequences in the chloroplast TrnL-F intergenic region. Methods and Results : SNPs were identified based on the results of nucleotide sequence for the intergenic region of TrnL-TrnF gene (chloroplast). Molecular markers were designed for those SNPs with additional mutations on the second base from SNPs for amplification refractory mutation system-polymerase chain reaction (ARMS-PCR). HRM pattern analyses were performed using the Mx3005P QPCR System (Agilent Technologies, CA, USA). Conclusion : We collected 12 individual lines of C. tricuspidata from various region in South Korea and China. Based on the nucleotide sequence in the trnL-trnF intergenic region of these lines, six SNPs and a deletion of 12 bps were identified and 12 individual lines were able to be grouped in one Korean ecotype and two different ecotypes of chinese lines, chinese line 1 and 2. The SNP markers developed in this study are useful for rapidly identifying these specific C. tricuspidata ecotypes collected from different regions.

        Background : Although ginseng has various bioactive compounds in it, there is lack of study on the variations of bioactive compounds in ginseng according to the cultivation soil and the applied fertilizer types (or amount). Therefore, this study aims to examine the variations of 37 fatty acids (FA) and 8 vitamin E (Vit-E) vitamers in 6-year-old ginseng root cultivated in different soil types with different fertilizers regimes. Methods and Results : The profiling of 37 FAs and 8 Vit-E vitamers in 6-year-old ginseng roots was measured by gas chromatography coupled with a flame ionization detector, and then these results were statistically analyzed with chemometrics. The FA and Vit-E content in ginseng roots varied significantly with respect to soil cultivation conditions due to organic fertilizer types and amounts used. Unsaturated FA in ginseng is approximately 2.7 fold higher than the saturated FA. Linoleic, palmitic, and oleic acids were the most abundant FAs found in the ginseng roots. Also, the major Vit-E vitamer found in ginseng root is α-tocopherol. In particular, the application of rice straw compost or food waste fertilizer was increased to create nutritionally desirable FAs and bioactive Vit-E in ginseng root. In addition, phytonutrient profiling coupled with chemometrics can be used to discriminate the cultivation conditions of ginseng. Conclusion : This study extends our understanding about the variations of FA and Vit-E in ginseng root depending on cultivation conditions. Hence, these results can be useful as basic information for reliable ginseng production containing high amounts of phytonutrients in a paddy-converted field.

        Background : The geographical origin of Panax ginseng Meyer, a valuable medicinal plant, is important to both ginseng producers and consumers in the context of economic profit and human health benefits. We therefore aimed to discriminate between the cultivation regions of ginseng using the stable isotope ratios of C, N, O, and S, which are abundant bio-elements in living organisms. Methods and Results : The C, N, O, and S stable isotope ratios were measured by isotope ratio mass spectrometer, and then these isotope ratios profiling was statistically analyzed with chemometrics. The various isotope ratios found in Panax ginseng roots were significantly influenced by region, cultivar, and the interactions between these two factors (P ≤ 0.0002). In particular, δ18O was lower in ginseng roots grown at high altitudes (r = −0.47), while δ34S was higher in ginseng roots grown close to coastal areas (r = −0.48). Chemometric results provided discrimination between the majorities of different cultivation regions. Conclusion : Our case study extends the understanding about the variation of C, N, O, and S stable isotope ratios in ginseng root depending on cultivation region. Hence, the analysis of stable isotope ratios is a suitable tool for discrimination between the regional origins of ginseng samples from Korea, with potential application to other countries.

        Background : Corrosion is one of the most devastating problems faced by most industries. Mild steel has played a vital role in various fields due to the excellent mechanical properties of mild steel such as low density, high strength-to-weight ratios, good environmental stability, high thermal conductivity, and corrosion resistance. Methods and Results : The total phenolic contents (TPC) and total flavonoid contents (TFC) of the methanolic extract of C. grandiflora and R. verniciflua leaf have been examined, and its corrosion inhibition performance was investigated by weight loss and electrochemical impedance spectroscopy (EIS) and polarization measurements. The surface morphology of mild steel was analyzed by scanning electron microscopy (SEM) equipped with energy dispersive X-ray spectroscopy (EDX) and by atomic force microscopy (AFM). The percentage composition of polyphenolic compounds was found to be higher in C. grandiflora and R. verniciflua plant extracts, and it was proved to be a superior, eco-friendly, and anti-corrosive inhibitor for mild steel in 1M of H2SO4. The Tafel polarization studies indicate that the plant extract is a mixed-type inhibitor. Scanning electron microscopy/energy -dispersive X-ray spectroscopy (SEM-EDS), and atomic force microscopy (AFM) studies confirmed the formation of a protective film on the metal surface. The corrosion inhibition of the C. grandiflora and R. verniciflua plant extracts was characterized by Fourier transform infrared (FT-IR), UV-visible spectra, and wide-angle X-ray diffraction (XRD) studies; these show the strong interaction between the metal surface and the inhibitor. Conclusion : The methanolic extract was prepared the two different plants like C. grandiflora, and R. verniciflua was studied the corrosion inhibition on the mild steel specimen in acidic medium through various methods involving weight loss measurements, EIS, and potentiodynamic polarization. The results shows that the C. grandiflora, and R. verniciflua plant extracts illustrate an effective corrosion inhibitor for mild steel with good anticorrosion properties in acidic environmen

        As a result of inquiring into the analysis consequent on a symptom by research subject, it was found that there were 1 paper (1.5%) which did research for the purpose of rehabilitation of the general public, 26 papers (38.8%) targeting diagnosis-related groups (DRG), and 40 papers (59.7%) targeting social consideration subject. Also, as a result of the inquiry about the frequency & number of times of horticultural therapy program implementation, it was found that 49 papers (73.1%) implemented one time a week, and 45 papers (67.2%) were surveyed as the highest by conducting a total of 11~20 sessions of horticultural therapy programs, In the analysis of horticultural therapy activities by type, plant cultivation activity was found to be the most 506 times, accounting for 41.5%, followed by 297 time crafts activity (24.4%), 213 time floral decoration activity (17.5%), and 203time others activities (16.7%). In cultivation activity, soil-using cultivation activity (25.1%) was found to be lower than the proportion (74.9%) of soiless cultivation (16.4%), crafts activity (24.4%), floral decoration activity (17.5%) and other activities (16.7%). The most used plants in a restricted place like a hospital were found to be in the order of Hedera helix, Chamaedorea elegans, Succulent plant, Syngonium podophyllum, Neofinetia falcata HU, Hoya carnosa (L.f.) R.Br.), Rosmarinus officinalis, and Spathiphyllum spp.

        The study was conducted to understand the relationship among university students’ interest in horticulture, psychological well-being and social development and the influence of measured variables. In order to collect data, the surveys were executed by convenience sampling on university students attending four-year universities in Daegu and Gyeongbuk regions of South Korea during the period from July 3 to July 17, 2016. A total of 307 survey results which measured the university students’ interest in horticulture, psychological well-being and social development were analyzed. The results showed that there are differences in the interest in horticulture, psychological well-being, and social development depending on the students’ general characteristics. According to the results analyzed by t-test and one-way ANOVA, the interest in horticulture depending on gender, religion, academic marks, family life satisfaction and school life satisfaction had significant differences showing high interest for the students with religion, good academic marks, high family life satisfaction and school life satisfaction as well as female students. Psychological well-being showed significant differences in academic marks, family life satisfaction and school life satisfaction, while social development displayed significant differences in gender, grade, academic marks, family life satisfaction and school life satisfaction. According to the results analyzed by correlation analysis, there was a significant correlation among university students’ interest in horticulture, psychological well-being and sociality development. Furthermore, regression analysis verified that university students’ interest in horticulture has a positive influence on psychological well-being and social development. The results of this study implies that university students with a higher interest in horticulture have higher levels of psychological well-being and social development.

        This study started with the issue that the criteria for the general image scale used in outdoor color planning are vague and the vocabulary and expressive area of the scale are not accurate. As an exterior color planning guideline, it is required to suggest a sensible image scale, which is an appropriate assessment tool for outdoor environment. We identified psychological attributes of colors and set the color image scale axis as “Soft–Hard” in a vertical and “Warm–Cool” in a horizontal. 26 sensibility languages were finally selected for the image scale through literature review, pre-analysis, and expert evaluation. All types of colors extracted from analyzing over 600 photo images were categorized according to 26 sensibility languages. 3 colors by each sensibility language were finally selected and arranged in the color image scale in accord with the outdoor environment of the apartment houses. The significance of this study is that wide range of color position in the color image scale could be utilized as an effective guideline for actual color planning by upgrading the existing color image scale to a higher level with the color reflecting sensibility language. The result also implies that the development of diversified color palette is required especially for the exterior color planning of apartment houses, and the systematical color design process should be established in order to improve the quality of color environment.

        Background : The natural stable isotope ratio of common bio-elements like carbon (C), nitrogen (N), oxygen (O), or sulfur (S) varies with diverse isotope fractionation processes in nature. Therefore, measuring the variation of these stable isotope ratios in ginseng roots can be a feasible tool to discriminate the geographical origins of ginseng in Korea. Methods and Results : The 3-year-old six Korean ginseng cultivars were cultivated at the five regions in Korea, and then used for measuring the stable isotope ratios of C, N, O, and S by isotope ratio mass spectrometry (IRMS). The mean C, N, O, and S stable isotope ratio values in the ginseng roots significantly differed according to the cultivation regions (p < 0.05). However, these isotope ratios in ginseng roots had relatively weak discriminative power against to the ginseng cultivars at each cultivation region. The interaction of the cultivation region and ginseng cultivar type also significantly affected to the C, N, O, and S stable isotope ratio in ginseng roots (p < 0.0001). The two-dimensional plots associated with the N stable isotope ratio can effectively separate the ginseng roots in Jinan compared to those in the other regions. The partial least squares-discriminant analysis showed more significant separation between ginseng geographical origins compared to the principal component analysis. Conclusion : Our findings improve our understanding of how the isotope composition of ginseng roots varies with respect to cultivation regions and cultivars, and suggest that the analysis of the stable isotope ratios combined with chemometrics can be used as a feasible tool to discriminate geographical origin of ginseng in Korea.

        Background : ROS produced by oxidative stress damaged endothelial cells, and cause a variety of vascular complications. In diabetic hyperglycemia state, ROS increase. The polyol pathway occur in diabetic complications, the excess glucose is absorbed into the polyol pathway when aldose reductase increased, NADPH changes it to sorbitol. Glutathione (GSH) removes ROS. GSH level is reduced by glutathione reductase, using NADPH as an electron donor. Activation of the polyol pathway decrease NADPH, and GSH also reduced. As a result, ROS is increased. In diabetic hyperglycemia state, Glycolysis increases. Effects of increased glycolysis, protein kinase C (PKC) is increased. NAD(P)H oxidase, stimulated by PKC-dependent pathway, increases ROS in the cell. In this study, we measured the ROS scavenging activity of 5 natural products (Lycii fructus, Astragalus membranaceus, Cassia Tora, Polygonatum odoratum, Rubus Coreanus), to confirm the efficacy as diabetic antioxidants. Methods and Results : We extracted 5 natural product by distilled water and ethanol. DPPH radical scavenging activity was significantly higher in Lycii fructus, Rubus coreanus. ABTS radical scavenging activity was better Rubus coreanus, Lycii fructus, Cassia Tora. In addition to, Rubus coreanus, Cassia Tora, Lycii fructus was comparatively higher reducing power activity than other natural products. And total phenolic and flavonoid contents were much higher in Rubus coreanus compared with other extracts. Conclusion : These results suggest that Lycii fructus, Rubus coreanus can be applied as diabetic antioxidant that prevent vascular complications caused by ROS.

        In this study, genetic diversity of wild Codonopsis lanceolata collected in Korea were analysed using SSR makers. Wild C. lanceolata roots were collected in Jeollanam-do Jangheung-gun Choentae Mountain as in roots. The wild C. lanceolata plants were cultivated in Chungbuk National University greenhouse and the leaves were sampled from 36 plants. The genomic DNA of C. lanceolata was extracted using CTAB. PCR was performed using a program of 35 cycles at 94℃ for 30 sec, 60℃ for 30 sec, and 72℃ for 30 sec with an pre-denaturation of 94℃ for 5 min and a final extension of 72℃ for 30 min. The PCR reaction mixture contains 5 pmole of primers and 20 ng of DNA template in a 20 μL reaction volume. The genotype of the analyzed samples were very different. Therefore, the wild C. lanceolata collected in Korea look genetically diverse.

        Codonopsis lanceolata is a perennial climber. The roots are used as medicinal materials or vegetables. Recently, demand for C. lanceolata is increasing as a healthy food. C. lanceolata is distributed in India and East Asia such as China, Japan as well as Korea. In South Korea, this plant is widely cultivated in Gangwon-do province. No C. lanceolata varieties were developed in Korea. The objective of this study is to analyze genetic diversity of C. lanceolata cultivated in Korea using SSR makers. C. lanceolata roots were collected in each region were cultivated in Chungbuk National University greenhouse. Samples were obtained from fresh leaves of 5 plants from each collection region. The genomic DNA was extracted using CTAB. Genetic diversity was analysed using 4 sets of C. lanceolata SSR makers. PCR was performed in total 20 μL reaction volume containing 20 ng of DNA template, 5 pmole of primers. The genotypes of the analyzed samples were very similar. That means that the genetic diversity of C. lanceolata cultivated in Korea is very low.
