Fetal Bovine Serum (FBS) plays a crucial role in animal cell culture; however, the increasing number of bovine fetuses used and sacrificed solely for FBS collection has raised ethical concerns globally. The welfare of fetuses during FBS blood collection has become a key focus of debate among animal welfare and ethics organizations worldwide. Previous studies indicate that heat-inactivated coelomic fluid (HI-CF) from the earthworm Perionyx excavatus may serve as a viable FBS alternative in adherent cell cultures. This study evaluates the potential of HI-CF as an FBS substitute during the in vitro maturation (IVM) stage of bovine embryo culture, with a focus on improving developmental rate through antioxidation effects. In this study, 2% HI-CF was incorporated into IVM media, assessing its impact on cell growth, differentiation, and the expression of genes related to antioxidation. The group of 2% of HI-CF exhibited a trend toward increased cleavage and blastocyst development rates compared to the control group. Although antioxidant genes such as NRF2 and GSR showed no statistically significant differences between the control and treatment groups, a trend toward increased expression was observed. Conversely, GPX1 displayed a trend of decreased expression. Notably, IGF1 and NQO1 were significant upregulated (p < 0.05) in the 2% HI-CF group. Additionally, oocytes stained with H2DCFDA showed a significantly reduced ROS levels (p < 0.05) in the 2% HI-CF group compared with controls. These findings suggest that HI-CF's antioxidative effects support enhanced cell growth and blastocyst development rate, surpassing those observed with FBS. Consequently, HI-CF shows promise as an effective alternative to FBS in vitro maturation of bovine oocytes.
This study was performed to investigate how changing the period of light and dark influences the vegetative growth and the photosynthesis of Doritaenopsis Queen Beer ‘Mantefon’. Clones of Dtps. Queen Beer ‘Mantefon’ at 4-month-old stage were grown in a closed-plant factory system with four different light/dark cycles; 06/06 h, 08/08 h, 10/10 h, and 12/12 h. Temperature and relative humidity were set at 28oC and 80%, respectively, with a photosynthetic photon flux density of 160 ± 10 μmol·m-2·s-1. Repetitive measurements showed that the leaf length and the leaf width were the longest under 12/12 h closely followed by 10/10 h. The fresh weight and the dry weight of leaves and roots were the heaviest at 10/10 h treated samples. Different CO2 uptake patterns were observed from different light/dark cycles. Under 10/10 h and 12/12 h treatments, the CO2 uptake started at early dark period. When the light/dark cycles were shortened to 06/06 h and 08/08 h, the CO2 uptake started at the middle of dark period. Total CO2 uptake amounts were the highest under 12/12 h treatment followed by 10/10 h, 06/06 h, and 08/08 h treatments. Quantitative measurements showed that the vegetative growths under 10/10 h treatment were comparable with that of 12/12 h treatment. These studies indicated that manipulating light/dark can modify the photosynthesis patterns and vegetative growth of Dtps. Queen Beer ‘Mantefon’, resulting in the reduction of the production period.
To determine the medicinal properties of Pleurotus cornucopiae and Safflower Oil (PS), the osteoblast effect was investigated. PS can prevent estrogen deficiency-induced bone loss. It was taken the measurements of biochemical factors such as serum osteocalcin, alkaline phosphatase, IGF-1, and bone status. Forty adult female Sprague-Dawley rats were randomly assigned to four equal groups for 12 weeks of treatment: (1) sham operation (SHAM) + vehicle; (2) ovariectomy (OVX) + vehicle; (3) OVX + 17β-estradiol (25 μg/kg); (4) OVX + PS (300 mg/kg/d, PO). At the end of the experiment, bone turnover, and trabecular microarchitecture were assessed by micro-computed tomography (micro-CT), and histopathology, respectively. Ovariectomized rats experienced remarkable increases in global volumetric bone density, and trabecular microarchitecture deterioration. These OVX-induced pathological changes are reversible in that most of them could be mostly corrected upon 17β-estradiol treatment. PNS treatment significantly prevented bone mass loss and microarchitecture deterioration. Moreover, PS enhanced osteoblast activity but suppressed osteoclast turnover, as evidenced by decreased levels of serum C-terminal telopeptides of type I collagen and elevated levels of alkaline phosphatase. PS mitigates estrogen deficiency-induced deterioration of trabecular microarchitecture.
본 연구는 6조 보리를 이용하여 양조한 맥주의 발효 중여러 특성의 변화를 측정하여 6조 보리 맥주의 제품 가능성을 확인해 보았다. 실험은 2조 보리 1종과 6조 보리4종을 이용하여 제맥, 당화, 발효 공정을 통해 맥주를 제조하였으며, 제조 공정 중 발효 특성의 변화를 확인하였다. 발효 특성 항목으로 효모의 생육, 비중, 색도, 거품 안정성,free amino acid, 13개의 관능 속성 변화를 측정하였고, 최종 제품인 맥주의 알코올 함량, 쓴맛, 탁도, diacetyl을 측정하였다. 결과적으로 6조 보리인 다향이 2조 보리인 진양과 발효 kinetics 특성이 가장 유사하게 나타나 맥주 제조에 적합한 것으로 나타났다.
The aim of this is to examine the effects of SSP therapy on the internal carotid arteries blood flow of 24 tension type headache patients(study group) and non tension type headache patients(control group), and on the reduction of their headaches, when applied to the acupuncture points. It stimulated the acupuncture point of headache 6 place with the SSP. It measured VAS and the blood flow of the internal carotid arteries with TCD. When the internal carotid arteries blood flow of study group and control group were compared by period, a significant difference was found in the 4th period(p<.05). When the Visual Analog Scale of study group and control group were compared by period, a significant difference was found in the 4th period(p<.05). The comparison of each measurement result of the internal carotid arteries blood flow of the study group found significantly increased. The comparison of each measurement result of the VAS of study group found significantly decreased. With regard to the control group, the VAS significantly decreased. The silver spike point low frequency electrical stimulation treatment, when applied to the acupuncture point, can significantly influence the internal carotid arteries blood flow of headache patients and can significantly reduce their headaches.
The purpose of this study is to investigate the effects of closed and open kinetic chain exercise for increasing knee joint function on the knee position sense in the normal adults. Thirty normal adults(male 15, female 15; mean age: 22.13±2.58 years) were participated in this study into two groups, each with 15 people. The group I was trained that closed kinetic chain exercise on the knee joint and the group II was trained that open kinetic chain exercise on the knee joint. Exercise programs performed for 4 weeks, 3 times a week were using Shuttle 2000-1 closed kinetic chain exercise and Knee Extensor open kinetic chain exercise(HUR, Filand). The results of this study were as follows: 1) There were statistically significant decreasing of measuring error degree in 0-20。were found between before and after training in closed kinetic chain exercise(p<.05). 2) There were statistically significant decreasing of measuring error degree in 21-40。were found between before and after training in closed kinetic chain exercise(p<.05). 3) There were statistically significant decreasing of measuring error degree in 41-60。were found between before and after training in closed kinetic chain exercise(p<.05). 4) There were statistically significant decreasing of measuring error degree in 0- 20。were found between before and after training in open kinetic chain exercise(p<.05). 5) There were statistically significant decreasing of measuring error degree in 21-40。were found between before and after training in open kinetic chain exercise(p<.05). 6) There were statistically significant decreasing of measuring error degree in 41- 60。were found between before and after training in open kinetic chain exercise(p<.05). In conclusion, these results suggest that closed and open kinetic chain exercise has increased in the knee joint proprioception between before and after training. Especially, closed kinetic chain exercise could be more useful intervention than open kinetic chain exercise for increasing proprioceptive sense.
Background: The Rosa multiflora, a well-known plant belonging to Rosacea, is widely used in orthodox medicine in worldwide. However, its biological activity and cosmetic preservative efficacy have not yet been studied. Thus, this species is yet to be defined as a functional cosmetic material. Accordingly, an investigation of the above mentioned atrributes was performed on a 50% ethanol extract of Rosa multiflora.
Methods and Results: The antioxidant activity was assessed through free radical scavenging assays with 2,2-diphenyl-1-picrylhydrazyl (DPPH). Additionally, the contents of total phenols and flavonoids were analyzed. The phenolic compounds were detected using HPLC. The antimicrobial activity against Staphylococcus aureus, Escherichia coli, and Candida albicans was assessed using the disc diffusion assay. The preservative effect (challenge test) on a formulation of soothing gel was performed for 28days. The DPPH radical scavenging ability, denoted by the SC50 (half maximal inhibitory concentration for DPPH radical scavenging) value was found to be 131.63 ㎍/㎖. The content of total polyphenol and flavonoid content were 202 ㎎/g and 86.77 ㎎/g, respectively. In additon, astragalin and gallic acid were identified in the extract. The antimicrobial activity of the extract against S. aureus and E. coli was observed to be 5 - 0.5%, and no significant activity was noted against C. albicans. The ethanol extracts (5% and 10%) met the preservation standards of the Cosmetics, Toiletry, and Fragrance Association (CTFA).
Conclusions: Thus the ethanol extract of R. multiflora can be used in cosmetics as a natural preservative and antioxidant.
Background : Cynanchum wilfordi is a typical myocardial medicinal crop. Because it penetrates more than 1m, it requires a lot of labor and harvesting costs. In addition, we needed 4 - 5 times of weeding until July, and in the soil with poor drainage, root rot disease occurs and yield is greatly reduced. Therefore, this study was conducted to develop new cultivation technology that can minimize the effort cost and increase the yield. Methods and Results : As a test material, container standards were used plume tube of diameter 30 ㎝ × height 50 ㎝, 80 ㎝, 100 ㎝. The culture soil manufactured by mixing cokofit 60%, peatmoss 10%, pearlite 6%, vermiculite 10%, zeolite 5%, and charcoal 5%. The compost was made by mixing fowl droppings 45% and cattle manure 5%. The mixing ratio of the culture soil and the compost was 100 : 0, 90 : 10, 80 : 20, 70 : 30, respectively, and the mixture was filled in the container. Cynanchum wilfordi seedling sow raising seedlings during one month seeds become dormancy breaking. Culture soil and container of Cynanchum wilfordi was effective 87% increase (384 ㎏/10 a → 720) in quantity contrast conventional culture in height 50 ㎝ container + culture soil 100 : 0. 80 and 100 ㎝ containers were found to be unsuitable for production throughout the year because of thin thickness of subterranean part and low commercial yield due to the long time required for roots to reach the hardpan layer. The yields of compost mixture decreased from 0% (720 ㎏/10 a) > 10% (642) > 20% (560) > 30% (475) in 50 ㎝ containers. When the compost content was more than 20%, the yield decreased due to physiological damage during the early growth. Conclusion : The culture soil and container cultivate for harvesting ease of Cynanchum wilfordi was effective saving more than 15 times more than the harvest time contrast manpower harvest and the yield was increased by 87%. Also, container cultivation was effective the cost cutting of installation dismantlement and redres since no need to move the cultivation area.
Background : Schisandra chinensis is being weighted difficulties in stable production, there is increasing drought damage caused by climate change as shallow rooted crops. Therefore, the study was performed for water supply capacity and growth characteristics analysis by setting the irrigation method for the drought damage reduction. Methods and Results : Test material was used sophomore V-shaped planting Schisandra chinensis. Irrigation method were surface watering, underground watering, sprinkler and untreated. Underground irrigation was irrigation that buried hose and then dug up the 15㎝. Soil moisture tension was the irrigation after fixed at -30 ㎪(23%). Irrigation timing was performed in June-July that high drought damage and made the most fruit enlargement. The main investigating items were investigated fruit growth, normal fault rate, soil moisture and EC content according to the irrigation method. Normal fruit rate according to irrigation method were appeared in sprinkler(81, 74 %)>underground irrigation(76, 69 %)>surface irrigation(76, 67%)>untreated(66, 52 %). Cluster length of yield component was determined to effective irrigation method in fruit growth the highest in sprinkler. Soil moisture contents was maintained at appropriate level with significant -30㎪(23 %) in the sprinkler. EC content low with a downward trend in underground irrigation and sprinkler. Water supply capacity according to Irrigation Method were sprinkler 40 tons, underground irrigation 85 tons, surface irrigation 138 tons. Conclusion : Appropriate watering methods for drought damage reduction of Schisandra chinensis caused by climate change was determined in the most efficient irrigation method in sprinkler that high fruit growth and normal fruit rate, lower the required of water supply capacity.
Background : Aralia cordata and Polygonum multiflorum GAP cultivation requires a stable drying and storage settings after harvesting. therefore, this experiment was performed in order to effectively manage the physical, chemical and biological hazards. Methods and Results : Test materials were used biennial Aralia cordata, Polygonum multiflorum harvested from the medicinal testing ground. The drying temperatures were treated with 40, 50, 6 0℃ and natural drying. Storage containers were stored in plastic boxes, styrofoam boxes and kraft paper containers, examined the color and quality changes for eight months. Aralia cordata and Polygonum multiflorum drying temperature is dry it took natural drying 720 hours, 40℃ hot air drying 180 hours, 50℃ hot air drying 168 hours and 60℃ hot air drying 108 hours. However, the difference chromaticity of the Lab value corresponding to the temperature does not appear, it was good to dry in a short time at 60℃. Aralia cordata and Polygonum multiflorum stored in a styrofoam box storage method but can be stored at room temperature for up to four months, began to decay caused by moisture content it continues to increase. In plastic box in case of Aralia cordata and kraft vessel in case of Polygonum multiflorum can be stored for eight months in room temperature without decay. Styrofoam boxes stored at 5℃ cold storage were higher water absorption such as room temperature, but decay did not occur. Plastic box and styrofoam box were a tendency such as room temperature. Conclusion : Aralia cordata and Polygonum multiflorum are thought that the color change is not large depending on the drying temperature the lower the water content. Styrofoam storage box, the air permeability is higher than plastic boxes and containers Kraft vessel, decay occurs expected increase.
