        Pyrifluquinazon, as a quinazinalone chemical group, based on a new mode of biological activity. It is reported that mode of action is modifies insect behavior, rapidly stopping feeding such that insects starve to death. Time-release feature and mortality effect on M. persicae using different pyrifluquinazon nano type and non-nano type were compared. Pyrifluquinazon nano type was formulated with different molecular weight and density of used chitosan (CS 30000 0.1% and CS 3000 0.3%). In the CS 30,000 0.1%, the mortality was weakly occurred at early time, but steadily increased after 4days. Finally, we confirmed more than 70% mortality as a peak at 16days. In CS 3000 0.3%, the mortality showed about 70% until 18days as a effective controlled release. Also, We examine time-release feature and mortality effect on M. persicae according to the different pyrifluquinazon nano type(CS 30000 0.1% and CS 3000 0.3%) of concentrations. The CS 30000 0.1% bioassay results of different concentration were showed that the highest concentration(100ppm) was measured better mortality than other concentration at 0 day, but cannot confirm different effect about dissimilar concentration. However, increasing rates of M. persicae were low as treatment concentrate was high. In CS 3000 0.3% 100ppm concentration bioassay result, aphid mortality reached peak at 24 days and increasing rate also low. Additionally, for the comparing of bioassay and feeding behavior of M. persicae against pyrifluquinazon nano types and non-nano type, EPG technique was carried out. In case of non nano type, feeding inhibition efficacy was showed during 4 days after treatment, but appeared similar level with control after 10days. In CS 3000 0.3% 50ppm, residual efficacy was specially showed until 28days after treatment whereas treatments with CS 30000 0.1% were similar to the control after 22days. These result show that the change of feedinng behavior and motrality of M. persicae is correlated with the change of nano type or non nano type of pyrifluquinazon.

        Several species of the genus Aphidius are used in biological control programs against aphid pests throughout the world and their behavior and physiology are well studied. But despite knowing the importance of sensory organs in their behavior, their antennal structure is largely unknown. In this study, the external morphology and distribution of the antennal sensilla on the antennal of both female and male adults of A. colemani were described using scanning electron microscopy (SEM). Generally, the filaform antennae of males (1,515.20±116.48 ㎛) are longer than females (1,275.06±116.42㎛). Antennae of this species is made up of scape, pedicel and flagellomeres. Male and female antennae differed in the total number of flagellomeres as 15 in males and 13 in females. Female and male antennae of A. colemani has samely seven types of sensilla. We classified sensilla placodea, Bohm bristles, 2 types of sensilla coeloconica, , 2 types of sensilla basiconica as with a tip pore and with wall pores, sensilla trichodea. In addition, the possible functions of the above sensilla types are discussed in light of previously published literature; mechanoreception(Bohm bristles, sensilla coeloconicaⅡ and sensilla trichodea) and chemoreception(sensilla coeloconicaⅠ, sensilla basiconicaⅠ,Ⅱ and sensilla placodea). Future studies on the functional morphology of the antennal sensilla of A. colemani using transmission electron microscopy (TEM) coupled with electrophysiological recordings will likely confirm the functions of the different sensilla identified in this study.

        This study was conducted in order to investigate repeated-dose toxicities of Magnolia ovobata ethanol extract (MEE). MEE was administered orally to male and female Sprague Dawley rats at dose levels of 0, 500, 1,000, or 2,000 mg/kg for four weeks. Repeated administration of MEE did not induce abnormalities in general signs, body weight gain, feed and water consumption, necropsy findings, or organ weights. In addition, no abnormality was observed in hematological analyses; red blood cells and their indices, white blood cells, platelets, and coagulation times. In male rats, BUN and creatinine showed an increase at doses of 2,000 mg/kg and 500-1,000 mg/kg, respectively, while in female rats, lactate dehydrogenase and creatine phosphokinase showed a decrease at 2,000 mg/kg, the upper-limit dose of repeated-dose toxicity studies. However, there were no dose-dependent increases or gender-relationship. In addition, other parameters of the hepatic and muscular toxicities as well as energy and lipid metabolism were not affected. In microscopic examination, no considerable pathological findings were observed. The results indicate the safety of oral administration of MEE to the upper-limit dose.

        Apolipophorin-III (apoLp-III) is a hemolymph protein whose function is to facilitate lipid transport in an aqueous medium in insects. Recently, apolipophorin-III in Galleria mellonella and Hyphantria cunea was shown to play an unexpected role in insect immune activation. We show here a novel possible function/role of the apoLp-III in insects. To investigate the genes which have a relationship with apoLp-III in fall webworm larvae, we reduced endogenous Hc apoLp-III mRNA levels in larvae via RNA interference (RNAi). The RNAi-mediated Hc apoLp-III reduction resulted in the reduction of antioxidants, like MnSOD, catalase, and glutathione S transferase as well as immune proteins. In particular, expression of MnSOD commonly decreased in fat body, midgut, and hemocytes following the knockdown of Hc apoLp-III, which induced an elevated level of superoxide anion in Hyphantria cunea larvae. The observed effect of Hc apoLp-III RNAi suggests that Hc apoLp-III is related to the action/expression of antioxidants, especially MnSOD.

        A new insect member of the STAT family of transcription factors (HcSTAT) has been cloned from the lepidopteran, Hyphantria cunea. The domain involved in DNA interaction and the SH2 domain are well conserved. The gene is transcribed at a low level during all stages of development, and transcribed in hemocyte, fat body, midgut, epidermis, and Malpighian tubule. Especially, hemocyte and Malpighian tubule showed transcriptional activation of HcSTAT upon Gram-negative and -positive bacteria challenge. Gram-negative and -positive bacteria challenge specifically results in nuclear translocation of HcSTAT protein and induction of DNA-binding activity that recognizes a STAT target site in H. cunea hemocyte. In vivo treatment with sodium orthovanadatetranslocates HcSTAT to the nucleus in hemocyte cells.

        Effects of Vegemil® containing soybean proteins and isoflavones on the growth and bone density of broiler chickens were investigated. One-week-old male and female Arbor Acres broiler chickens were fed on Vegemil® A containing 3% soybean proteins and 162 ppm isoflavones, instead of water, for 30 days and their growth indices (body weight, leg weight and femur length) and bone density were analyzed. The body weight gains in male and female chickens were increased by 15.6% and 31.7%, respectively, following feeding Vegemil® A compared to normal water. Vegemil® A increased leg weight as well as femur length of females by 22.9% and 15.0%, respectively. In addition, Vegemil® A feeding enhanced femoral bone density by 21.3% in comparison with water feeding. Therefore, it is suggested that Vegemil® A not only facilitates body growth, but also strengthens bone density of normal chickens, and that it could be a promising candidate for the improvement of infant growth and for the prevention of menopausal osteoporosis.

        The present study was carried out to establish an animal model, displaying long-term learning and memory dysfunction, since single intracerebroventricular (icv) injection of amyloid β peptide (Aβ) causes a short-term memory impairment. Male ICR mice were fed a high-cholesterol diet (HCD) containing 3% cholesterol, 1% corn oil and 0.5% cholic acid, and 1 week later, icv injected with Aβ1-42 (5 μg/head). Learning/ memory function was assessed via passive avoidance performances 1 day and 2, 4, and 6 weeks after Aβ1-42 injection, in addition to blood biochemical analyses for lipid profiles and hepatic function. Total cholesterol, lowdensity lipoproteins and hepatic dysfunction parameters markedly increased, while high-density lipoproteins were reduced following HCD feeding. Whereas single injection of Aβ induced temporary memory loss 1 day after administration, exhibiting full recovery after 2 weeks, Aβ treatment in combination with HCD feeding lasted the learning/memory impairment up to 6 weeks. Therefore, it is suggested that hypercholesterolemia augments Aβ-induced memory loss, and that Aβ injection plus HCD feeding could be a long-term memorydeficit model suitable for long-term treatment with drugs or stem cells.

        A novel recombinant baculovirus, NeuroBactrus, was constructed to develop an improved baculovirus insecticide with additional beneficial properties such as higher insecticidal activity and recovery to wild-type baculovirus. For this, Bacillus thuringiensis crystal protein gene (cry1-5) was introduced into Autographa californica nucleopolyhedrovirus (AcMNPV) genome by fusion of polyhedrincry1- 5-polyhedrin under the control of poyhedrin gene promoter. In the opposite direction of this fusion gene, an insect-specific neurotoxin gene (AaIT) under the control of early promoter from Cotesia plutellae bracovirus was introduced by fusion of orf603 partial fragment. Western hybridization and confocal microscopy revealed that AaIT neurotoxin and Polyhedrin-Cry1-5-Polyhedrin fusion protein expressed by the NeuroBactrus and that the fusion protein occluded into the polyhedra. In addition, the fusion protein was activated as about 65 kDa of crystal protein when treated with trypsin. The NeuroBactrus showed high level of insecticidal activity against Plutella xylostella larvae and significant reduction in median lethal time (LT50) against Spodoptera exigua larvae compared to those of wild-type AcMNPV. Re-recombinants derived from the NeuroBactrus, NBt-Del5 (deleted cry1-5), NBt-DelA (deleted AaIT) and NBt-Del5A (deleted cry1-5 and AaIT; wild-type baculovirus) were generated in serial passages in vitro and in vivo. These results suggested that the NeuroBactrus could be transferred to wild-type baculovirus along with serial passages by the homologous recombination between two polyhedrin genes and two partial orf603 fragments.

        Reactive oxygen species (ROS) is toxic to living organisms, because its high reactivity causes oxidative damage to proteins, nucleic acids, and lipids. Superoxide dismutase (SOD) is an enzyme facilitating the removal of superoxide anions from living organisms. This study focused on the cloning of MnSOD cDNA from Hyphantria cunea and its induction upon bacterial infection and various stresses. The open reading frame of MnSOD is composed of 645 bp, encoding 215 amino acid residues. The theoretical molecular mass and pI of putative MnSOD was evaluated to be 24276 Da and 9.14, respectively. The MnSOD from H. cunea is highly similar to human MnSOD (59.5%) as well as Bombyx mori MnSOD (76.2%). MnSOD showed no big induction upon bacterial infection and stresses, compared to that of Cu/ZnSOD.

        Innate immunity responses are triggered by the immune challenge and therefore involve signaling processes. The cellular response is initiated by hemocytes and mainly involves phagocytosis and encapsulation of intruders by these cells. To address whether Hc-STAT is activated upon bacterial challenge, we examined the subcellular location of STAT protein in hemocyte by immunostaining. A new insect member of the STAT family of transcription factors (Hc-STAT) has been cloned from the lepidopteran, Hyphantria cunea. Especially, hemocytes and Mt showed transcriptional activation of Hc-STAT upon Gram (-) bacteria and fungal challenge. Gram (-) bacteria and fungal challenge specifically results in nuclear translocation of Hc-STAT protein and induction of DNA-binding activity that recognizes a STAT target site in H. cunea hemocyte. In vitro treatment with pervanadate translocates Hc-STAT to the nucleus in hemocyte cells. Here we report the first evidence for the involvement hemocyte JAK/STAT pathway upon microbial infection in lepidopteran insect.

        In our present study, total methanol extracts prepared from B. platyphylla var. japonica showed a significant increase in cell proliferation upon the induction of oxidative stress by hydrogen peroxide or y-ray irradiation. Total methanol extracts were fractionated into five separate preparations i.e. n-hexane, dichloromethane, ethylacetate, n-butanol and water fractions. Among these, the ethylacetate and butanol fractions of B. platyphylla var. japonica showed the highest protective effects against oxidative stress induced by hydrogen peroxide. These fractions also showed strong protective effects against y-ray irradiation. When we evaluated the cytotoxicity of these fractions, the butanol fraction showed no effects in a colony formation assay. In addition, the butanol fraction showed a cell proliferation activation effect evidenced by significant increase in the colony formation of y-ray irradiated cells. Both a radical scavenging activity and clonogenic activity assay suggested that the mechanism behind this protective effect against reactive oxygen species may be due to the radical scavenging and cell proliferation activity of B. platyphylla var. japonica extracts.

        The principal objective of this study was to assess the antioxidative activities of Petasites japonicus against oxidative stress in bovine brain tissue . Petasites japonicus is found with a relatively widespread distribution, and is cultivated as a culinary vegetable in Korea. Petasites japonicus samples were dried either by freeze-drying or by hot air-convection drying (80℃), then evaluated for their antioxidative activity by measuring 1-dipheny-1,2-picrylhydrazyl (DPPH) radical scavenging, and by measuring thiobarbituric acid-reactive substances (TBARS) in brain homogenates subjected to Fe2+ -mediated lipids with or without the addition of botanical extract. Hot air convection-drying resulted in a slight increase in the extraction yie1d as compared with freeze-drying. However, total phenol and flavonoid contents in freeze-dried Petasites japonicas were significantly higher than those of hot air convection-drying. Freeze-drying increased the free radical scavenging activity of Petasites japonicas, leaves, and stems by 52.6, 28.6, and 248.0%, as compared with hot air convection-drying. Additionally, the IC50 values measured by TBARS in hot air convection-dried Petasites japonicas, leaves, and stems were increased by 36.0, 31.6, and 15.9%, as compared to those of freeze-drying. Although antioxidative activity was reduced slightly by heat processing in Petasites japonicas, freeze-drying for each portion of Petasites japonicus was the most appropriate for use as a functional food and pharmaceutical material.

        As indigenous aphid parasitoid, Aphelinus varipes kill aphids for feeding in addition to parasitization. Because of this characteristic of A. varipes, this parasitoid may have the possibility of biological control agent against aphids. So we have evaluated traits such as daily paratization, total parasization, number of aphids killed by host feeding, sex ratio, development time, pupal mortality of A. varipes parasitizing green peach aphid, Myzus persicae. At 25°C and 16L:8D, longevity, total paratization and host feeding of A. varipes female was 11.0, 25.3, and 63.3 days, respectively. And development time of male and female, sex ratio (M:F), pupa mortality of offspring of A. varipes were 12.0 days, 12.5 days, 0.88, and 11.6%, respectively. However, because these results are not enough to estimate potential of A. varipes as biological control agents/factors, other factors such as host suitability (Macrosiphum euphorbiae, Aulacorthum solani), effect of temperature, and host seeking behavior of A. varipes continually will be investigated.

        To develop an advanced baculovirus insecticide with additional advantages, such as higher toxicity and recovering to wild-type baculovirus, a novel recombinant baculovirus, NeuroBactrus was constructed. Bacillus thuringiensis crystal protein gene (cry1-5) and an insect-specific neurotoxin gene (AaIT) were introduced into Autographa californica nucleopolyhedrovirus genome by fusion of polyhedrin-cry1-5-polyhedrin under the control of poyhedrin gene promoter, and by fusion of orf603 partial genes and AaIT under the control of early promoter of ORF3006 from Cotesia plutellae bracovirus. About 150 kDa of Polyhedrin-Cry1-5-Polyhedrin fusion protein expressed by NeuroBactrus was occluded into the polyhedra, and activated as about 65 kDa of crystal protein when treated with trypsin. RT-PCR analysis indicated that transcription of AaIT gene occurs by 2 h postinfection (p.i.) and increased at 16 h p.i.. NeuroBactrus showed high toxicity against Plutella xylostella larvae and significant reduction in median lethal time (LT50) against Spodoptera exigua larvae compared to those of wild-type AcNPV. Re-recombinants derived from NeuroBactrus, NBt-Del5 (deleted cry1-5), NBt-DelA (deleted AaIT) and NBt-Del5A (deleted cry1-5 and AaIT; wild-type baculovirus) were generated in serial passages in vitro. This result showed that the NeuroBactrus could be transferred to wild-type baculovirus along with serial passages by the homologous recombination between two polyhedrin genes and two partial orf603 genes.

        Propylea japonica is small ladybug(approximately 4-5mm) and is met with everywhere in Korea. When there was no aphid, the survival rate of P. japonica was 0% among another instar bugs and 26.7~32.8% among same instar bugs after 48 hours. When there were sufficient aphids, the cannibalism of P. japonica was low, so the survival rate of P. japonica was 78.6~95.8% among another instar bugs and 80.6~100% among same instar bugs after 48 hours. In the comparison of the number of eggs at different oviposition site, P. japonica preferred the corrugated cardboard. In experiment using T-tube, the rate of decoy was 81% at cotton aphid compared with pepper leaf and 62% at cotton aphid compared with artificial diet. In the artificial diet using shrimp and chicken liver, the developmental period from egg to adult was 17days and the survival rate from egg to adult was 66.7% but the vitality was not good. These data suggest that P. japonica can be used effective natural enemy for control of aphids and is needed to research about artificial diet, mass rearing system and control effect in field.

        In order to establish a year round indoor-rearing system for Copper butterflies; large copper butterfly, Lycaena dispar, the effect of temperature on larval development and diapause was investigated. Temperature has been suggested as an important factor regulating the developmental rate, length of life, and survival rate from insect. As temperature increased, the developmental period was gradually shorten. The developmental periods of large copper larvae had a range of 11.0 days to 28.5 days at 30℃ and 17.5℃ respectively. The highest emergence rate was 94.2% recorded at 20℃. And the low emergence rate was 72.7% under 17.5℃. We investgated the sensitive stages to diapause induction in the larger copper effect of temperature and photoperiod. The experiment involves transfer of individuals from diapause averting (LD 16:8h, 25℃) to diapause inducing condition (LD 8:16h, 20℃) at various stages. Diapause was induced in 95.2% insect transferred at hatching larvae, in 15.6% of insects transferred at 2nd stadium molt, in 0% of insects transferred at after 3rd stadium molt. Percentage diapause induction increase with the length of short days and low temperature experienced. The main stage sensitive to photoperiod and temperature induction of diapause determination is the early first larval instar. The diapause induction began 14 days and ended 20 days after hatching larvae (LD 8:16h, 20℃).

        수평 배향 액정 모드는 양의 유전율 이방성과 음의 유전융 이방성 액정을 사용한 프린지 필드 스위칭과 양의 유전율 이방성 액정을 사용한 인플래인 스위칭 모드가 대표적이다 . 이 대표적인 세 구동 방식의 화질 특성을 비교하기 위하여 각각의 최척화된 위상지연 값 조건하에서 밝기, 명암대비율과 색 특성을 조사하였다. 그 결과 양액정과 음액정을 사용한 프린지 필드 스위칭 모드가 밝기와 명암대비율 면에 있어서 인플래인 스위칭 모드보다 우수한 특성을 보인다. 또한 양액정을 사용한 프린지 필드 스위 칭 모드는 시야각 방향에서 적은 색 변이 특성을 보인다.

About 150 kDa of Polyhedrin-Cry1-5-Polyhedrin fusion protein expressed by AcB5B-AaIT was occluded into the polyhedra produced by the recombinant virus, and activated as about 65 kDa of crystal protein when treated with gut-juice of Bombyx mori. The AcB5B-AaIT showed about 50% reduced LT50 value compared to that of the recombinant virus, Ap1Ac, expressing Cry1Ac against Plutella xylostella larvae. In addition, Spodoptera exigua larvae fed the recombinant polyhedra of AcB5B-AaIT showed about 4 fold higher refusing diet effect compared S. exigua larvae fed the recombinant polyhedra of the recombinant virus, Ap1C, expressing Cry1C. AcB5B-AaIT could be transferred to wild-type baculovirus along with serial passage by the homologous recombination between two polyhedrin genes contained in polh-cry1-5-polh fusion protein gene. These results suggested that the novel recombinant baculovirus, AcB5B-AaIT, could be applied as advanced viral insecticide.

        The multicolored Asian ladybird beetle, Harmonia axyridis (Pallas) (Coleoptera: Coccinellidae), is common to a wide range of natural and agricultural habitats. Applications of gamma irradiation minimized the losses of stored food and the death or failure of emergence in larval and pupal stages. On the other hand, degrade toxin waste as one of alternative to chemical pesticide for both quarantine and sprout control purposes of storage crop pest. So, we have investigated whether gamma irradiation exposed to eggs, lava, pupa and adults of H. axyridis. It may be affected the emergence, fertility, fecundity, development period and sex ratio of H. axyridis. Some changes of physiological characteristics may be applied to more efficient agents as biological control of several aphids. Insects were exposed to gamma irradiation from 0 to 500 Gy of 60Co depended on their developmental stages. The results showed that the first instar, eggs, third instar, pupae, and adults were more sensitive in order of irradiation dose. And fecundity and fertility of female adults were significantly decreased with increasing gamma irradiation dose at all tested individuals.
