Natural uranium-contaminated soil in Korea Atomic Energy Research Institute (KAERI) was generated by decommissioning of the natural uranium conversion facility in 2010. Some of the contaminated soil was expected to be clearance level, however the disposal cost burden is increasing because it is not classified in advance. In this study, pre-classification method is presented according to the ratio of naturally occurring radioactive material (NORM) and contaminated uranium in the soil. To verify the validity of the method, the verification of the uranium radioactivity concentration estimation method through γ-ray analysis results corrected by self-absorption using MCNP6.2, and the validity of the pre-classification method according to the net peak area ratio were evaluated. Estimating concentration for 238U and 235U with γ-ray analysis using HPGe (GC3018) and MCNP6.2 was verified by -spectrometry. The analysis results of different methods were within the deviation range. Clearance screening factors (CSFs) were derived through MCNP6.2, and net peak area ratio were calculated at 295.21 keV, 351.92 keV(214Pb), 609.31 keV, 1120.28 keV, 1764.49 keV(214Bi) of to the 92.59 keV. CSFs for contaminated soil and natural soil were compared with U/Pb ratio. CSFs and radioactivity concentrations were measured, and the deviation from the 60 minute measurement results was compared in natural soil. Pre-classification is possible using by CSFs measured for more than 5 minutes to the average concentration of 214Pb or 214Bi in contaminated soil. In this study, the pre-classification method of clearance determination in contaminated soil was evaluated, and it was relatively accurate in a shorter measurement time than the method using the concentrations. This method is expected to be used as a simple pre-classification method through additional research.
Molten Salt Reactor (MSR) is one of Generation-IV nuclear reactors that uses molten salts as a fuel and coolant in liquid forms at high temperatures. The advantages of MSR, such as safety, economic feasibility, and scalability, are attributed from the fact that the molten salt fuel in a liquid state is chemically stable and has excellent thermo-physical properties. MSR combines the fuel and coolant by dissolving the actinides (U, Th, TRU, etc.) in the molten salt coolant, eliminating the possibility of a core meltdown accident due to loss of coolant (LOCA). Even if the molten salt fuel leaks, the radioactive fission products dissolved in the molten salt will solidify with the fuel salt at room temperature, preventing potential leakage to the outside. MSR was first demonstrated at ORNL starting with the Aircraft Reactor Experiment (ARE) in 1954 and was extended to the 7.4 MWth MSRE developed in 1964 and operated for 5 years. Recently, various start-ups, including TerraPower, Terrestrial Energy, Moltex Energy, and Seaborg, have been conducting research and development on various types of MSR, particularly focusing on its inherent safety and simplicity. While in the past, fluoride-based molten salt fuels were used for thermal neutron reactors, recently, a chlorine-based molten salt fuel with a relatively high solubility for actinides and advantageous for the transmutation of spent nuclear fuel and online reprocessing has been developing for fast neutron spectrum MSRs. This paper describes the development status of the process and equipment for producing highpurity UCl3, a fuel material for the chlorine-based molten salt fuel, and the development status of the gas fission product capturing technologies to remove the gaseous fission products generated during MSR operation. In addition, the results of the corrosion property evaluation of structural materials using a natural circulation molten salt loop will also be included.
Subunit vaccines are being developed as a potential therapy for preventing microbial pathogen infection. In this study, the immunogenicity of recombinant Brucella (B.) abortus Fe/Mn superoxide dismutase (rFe/Mn SOD) protein as a subunit vaccine against B. abortus was investigated in BALB/c mice model. Brucella Fe/Mn SOD gene was cloned into a pcold-TF DNA vector. The bacterial recombinant protein was expressed using the Escherichia coli DH5α strain with a size of 82.50 kDa. The western blotting assay showed that rFe/Mn SOD reacted with Brucella-positive serum, indicating the potential immunoreactivity of this recombinant protein. After the second and third vaccinations, the peripheral CD4+ T cell population was increased significantly in the rFe/Mn SOD-immunized mice group compared to the PBS control group. Moreover, immunization of this recombinant protein increased the CD4+ T cell population from the first vaccination to the third vaccination. Meanwhile, the CD8+ T cells were slightly enhanced after the second vaccination compared to the first vaccination and compared to control groups. Fourteen days after the bacterial infection, the splenomegaly and the number of bacteria in the spleen were evaluated. The result showed that both rFe/Mn SOD and positive control RB51 decreased the bacterial replication in the spleen and the splenomegaly compared to control groups. Altogether, these results suggested that rFe/Mn SOD could induce host immunity against B. abortus infection.
This study aims to investigate the effects of exogenous succinic acid (SCA) on Brucella (B.) abortus infection in macrophage RAW 264.7 cells and ICR mice. Firstly, the in vitro experiment was conducted by MTT cytotoxicity and bacterial internalization assay to evaluate the uptake of B. abortus into macrophage cells. Two non-cytotoxic concentrations of SCA demonstrated attenuated invasion of Brucella into macrophages at 30 and 45 min post- infection (pi). Secondly, ICR mice were treated with SCA and infected with B. abortus. On day-14 pi, spleen and blood serum were collected to evaluate the bacterial burden and total spleen weight as well as the production of cytokine/chemokine, respectively. The results showed that SCA treatment promoted bacterial growth and reduced the total spleen weight in mice. Furthermore, SCA treatment increased the level of IL-10 cytokine in the sera, while dampening the production of MCP-1 chemokine compared to the control. The results of bacterial load in spleen and spleen weight together with cytokine/chemokine production profile in the sera indicated that SCA induced the host anti-inflammatory response which is beneficial for the survival of Brucella. Therefore, these findings suggest that SCA contributed to host immunity against Brucella infection and the emerging potential topic-immunometabolism should be invested for further investigations.
This study investigated the efficacy of four Brucella (B.) abortus recombinant proteins, namely adenylate kinase (Adk), nucleoside diphosphate kinase (Ndk), 50S ribosomal protein (L7/L12) and preprotein translocase subunit (SecB), as a combined subunit vaccine (CSV) against B. abortus infection in BALB/c mice. Immunoblotting assay showed that these four recombinant proteins as well as pcold-TF vector reacted individually with Brucella-positive serum, but not with Brucella-negative serum. The peripheral blood CD4+ T cell population was increased in CSV-immunized mice compared to PBS and pcold-TF vector groups. In addition, CSV and pcold-TF groups displayed induced IgG1 and IgG2a antibodies production compared to PBS and RB51 group, whereas IgG2a titer was higher than IgG1 titer in CSV group. The secretion profiles of IgG1 and IgG2a production together with an enhancement of CD4+ T cell population suggested that CSV did not only induce T helper 1 (Th1) T cell immunity but also humoral immunity. Therein, Th1 T cell immunity is more predominant in eliminating intracellular bacteria B. abortus. Furthermore, CSV immunization significantly reduced the bacterial burden in the spleen as well as the spleen weight in comparison to PBS and pcold-TF groups. Altogether, combination of these antigens could be potential to induce protective immunity against B. abortus infection in animals.
This study was conducted to investigate effect of feeding beet pulp on reproductive performance, colostrum composition and microbiological characteristics in sows. A total of 12 sows(Landrace×Yorkshire) were randomly allotted three dietary treatments at gestating 90 days. The diets were prepared by adding 0, 5 and 10% of the beet pulp in basal diets. The pigs fed 5% beet pulp showed higher litter size than the 0 and 10% beet pulp treatments, although there was not significant among the treatments. Similarly, birth and 21days weight in piglet were not different among the treatments. In colostrum fat, protein, lactose, citric acid, solid, ffa, true protein were ranged to reference values, but not difference among the treatments. E.coli of feces was lower(P<0.01) in the 5% beet pulp than the 0 and 10% beet pulp treatments. But no effect was observed on Lactobacilli of feces among dietary treatments. These our results indicate the beet pulp has no significant effect of reproductive performance and colostrum composition in sows. But, 5% level beet pulp is effective for reduction of intestinal pathogenic microorganisms.
The objective of this study was to evaluate the effect of the Allomyrina dichotoma larva (AD) on allergy and inflammation. We examined inhibitory effect of AD on allergic reactions in mast cells (RBL-2H3) activated by Compound 48 / 80and inflammatory response in macrophages (Raw 264.7) activated by LPS. Anti-allergy and anti-inflammatory actions ofAD water extract had no cytotoxicity. At these concentrations AD inhibited ẞ-hexosaminidase, tumor necrosis factor- α(TNF- α), interleukin-4 (IL-4), interleukin-6(IL-6) and cyclooxygenase-2(COX-2). AD also inhibited the expression of inducibleNO synthase (iNOS). AD reduced the release of inflammatory cytokines including IL-4, IL-6, TNF-α, and ẞ-hexosaminidase.These results suggest that AD may be potential anti-allergy and anti-inflammatory therapeutic agent.
The occurrence pattern of lepidopteran adults in a paddy-upland rotation field and an organic upland field for 8 maize and 7 sorghum varieties was surveyed from May 8 to September 27, 2016. In addition, their damage ratio and the invasive pores by Ostrinia furnacalis larvae as well as harvest yields were monitored. The density of the lepidopteran adults in two fields showed similar pattern, although a little different pattern in Heliothis assulta. The damage ratio of maize and sorghum varieties by O. furnacalis at harvest periods was depended on both varieties and cultivated environments. A middle negative linear correlation was observed in damage ratios vs yields, the number of invasive pores vs yields, and the number of larvae in corn ears vs yields, but there was not a specific correlation between the damage ratio vs the number of invasive pores. These results may be useful for the establishment of a management strategy to control oriental corn borer in paddy-upland rotation or organic upland fields for maize and sorghum.
The purpose of this study was conducted in order to analyze the effects of the manual intervention and self-corrective exercise models of general coordinative manipulation(GCM) on the balance restoration of spine & extremities joints with distortions and mal-alignment areas. The subjects were the members who visited GCM Musculoskeletal Prevent Exercise Center from March 1 2012 to December 31 2013 because of spine & extremities joints distortion and mal-alignments, poor posture, and body type correction. All subjects were diagnosed with the four types of the GBT diagnosis. And according to the standards of the mobility vs stability types of the upper & lower body, they were classified into Group 1(40 persons) and Group 2(24 persons). For every other day for three times a week, GCM intervention models were applied to all subjects for four weeks, adding up to 12 times in total. Then the balance restoration effects were re-evaluated with the same methods. The results are as follows. 1) Balance restoration effects of VASdp(Visual analysis scale pain & discomfort) and ER(Equilibrium reaction: ER) came out higher in GCM body type(GBT) Ⅱ·Ⅲ·Ⅳ of Group 1. 2) In case of balance restoration effects in Moire and postural evaluation areas, Group 1 was higher and cervical and scapular girdle were higher in Group 2. The balance restoration of the four GBT types was significant in all regions(p<.05), and the scapular girdle came out as high in the order of GBTⅡ·Ⅳ·Ⅰ. 3) In case of thoracic-lumbar scoliosis and head rotation·facial asymmetric· cervical scoliosis·ribcage forward, the balance restoration effects of the upper body postural evaluation areas came out the highest in Group 1 and Group 2, respectively. The balance restoration effects of the four GBT types were significant in all regions(p<.05), and came out the highest in lumbar scoliosis GBTⅢ·Ⅰ, ribcage forward and thoracic scoliosis GBTⅡ·Ⅳ. 4) The balance restoration effects of the lower body postural evaluation areas came out higher in Group 1 and Group 2 for pelvis girdle deviation·patella high·umbilicus tilt and hallux valgus·foot longitudinal arch: FLA·patella direction, respectively. The balance restoration effects of the four GBT types were significant in all regions(p<.05), and came out the highest in pelvis girdle deviation GBTⅢ·Ⅰand patella high-direction GBTⅣ·Ⅱ·Ⅰ. 5) The balance restoration effects between the same GBT came out significant (p<.05) in all evaluation areas and items. The conclusions of this study was the manual intervention and self-corrective exercise models of the GCM about the mal-alignment of the spine & extremities joints across the whole body indicated high balance restoration effects(p<.05) in spine & extremities joints in all evaluation areas.
Experiments were conducted in order to assess the healing effect of bee venom (BV) cream on full-thickness skin wounds in rabbits. BV cream was compared with silver sulfadiazine (SS) as a topical medicament against a control on experimentally created full-thickness wounds. Two wounds measuring 2 × 2 cm were created bilaterally (four wounds/rabbit) on the dorsolateral aspect of the trunk of seven New Zealand white rabbits. Wound treatments were evenly distributed on four sites, using a Latin square design. The contact layer of wounds was treated with physiological saline (control), SS cream, and BV cream over a period of 28 days. Assessment of wound healing was based on scab hardness, wound exudates, wound area, unepithelialized granulation tissue, and histopathological findings. Topical application of BV and SS creams to wounds resulted in reduced inflammation, debridement of necrotic tissue, and promoted granulation and epithelialization. Wound healing was faster, with statistical significance in BV and SS treatments, compared to the control (P<0.05). Treatment with BV evoked an anti-inflammation effect in a rabbit model. BV cream produced a wound healing effect similar to that of commercially available SS cream. Anti-inflammation effect as a topical treatment with BV cream appears to be better than that with SS cream. These results suggest that topical application of BV cream may be an alternative treatment for full-thickness skin wounds.
The objective of this study was to determine the effects of E. coli isolated from porcine semen on sperm viability, motility, and semen pH. Semen samples were prepared using commercial extender, SeminarkPro (Noahbio Tech, Korea) that did not contain antibiotics. And 4 different levels of E. coli were artificially innoculated to semen with following concentrations; 4,000 of sperms with 1 of E. coli (T1), 400 with 1 (T2), 40 with 1 (T3), and 4 with 1 (T4). Semen samples were preserved at 17℃ for 5 days in semen storage box until analyzed by flowcytometer. Aliquots were subjected to measure the sperm viability (Live/Dead® stain), motility (mitochondrial function), and semen acidity (pH) from day 0 (day of semen collection) to day 5. Sperm motility and viability were significantly decreased (p<0.05) on day 0 (4 hrs after preservation at 17℃) in T3 and T4 compared to control groups and were significantly decreased (p<0.05) in all groups from day 3. Sample pH was acidic in T3 (6.90~6.86) and T4 (6.86~6.65) from day 3 to day 5 (p<0.05). On the other hand, sample pH was maintained 7.0~7.1 in control, T1, and T2 during the experimental period. Sperm motility and viability were significantly decreased from day 0 to day 5 compared to control in samples contaminated with E. coli above a value of 40:1 (20×106 sperm cells/ml : 5×105 cfu/ml). Even on day 1 in T4 and on day 3 in T3, semen pH was acidic probably due to the acidification of dead spermatozoa. These results suggest that E. coli contamination has a concentration-dependent detrimental effect on extended porcine semen quality.
