        치자(Gardenia jasminoides Ellis fructus, GJE) 씨에서의 70% 에탄올, CM (chloroform: methanol, 부피 비 2:1(CM)) 및 노르말 부탄올 추출 용매별 flavonoid 함량 및 항산화 능, 금속 킬레이 트 능력 측정을 통하여 치자 씨의 기능성 식품 소재로서의 가치를 검토한 결과, 치자 씨의 proanthocyanidin 함량은 건조 무게(g) 중 카테킨(catechin) 당량으로 계산하여 70.035±0.772 mg으로 나타났으며, 추출 수율은 CM (36.39%), 70% 에탄올(27.32%), 노르말 부탄올(26.23%) 순으로 나타났 다. 추출 용매별 항산화 능은 농도(0.2, 0.4, 0.6 mg/mL)가 증가할수록 유의적으로 증가하였으며 (p<0.05), 양성대조군으로 사용된 ascorbic acid, butylated hydroxyanisole (BHA), ethylenediamnetetraacetic acid (EDTA) 보다는 낮은 활성이 확인되었다. Flavonoid 함량(mg)은 시료 g 당 케르세틴(quercetin) 당량으로 계산하여 70% 에탄올(0.830), CM (0.752), 노르말 부탄올(0.105) 순 으로 확인되었으며, 항산화 능 분석 실험과 금속 킬레이트 능력 실험에서도 이와 유사한 양상으로 동정 되어 모든 분석에서 70% 에탄올과 CM 추출물이 강한 생리활성을 나타내었다. 따라서 치자 씨의 용매 별 flavonoid 함량에 따라 항산화 능 및 금속 킬레이트 능력이 증가하는 것으로 관찰되었다. 이에, 치자 씨는 proanthocyanidin과 flavonoid 화합물을 다량 함유하고 있으며, 높은 항산화 능 과 생리활성을 가 지고 있어 기능성 식품 및 천연 항산화제로서의 가치가 있을 것으로 추정된다.

        치자 껍질의 70% methanol, ethyl acetate (EA) 및 distilled water (DW)의 용매 별 추출물 의 total phenol 함량 및 질소 산화물 소거능, 환원력, β-carotene 탈색을 이용한 항산화력 및 지질과 산화 저해능 측정을 통하여 치자의 기능성 식품 소재로서의 가치를 검토한 결과, anthocyanin 함량은 3.519±0.635 mg/100 g DW로 나타났으며, 용매 별 추출 수율은 DW (39.87%), 70% methanol (36.26%), EA (2.88%) 로 관찰되었다. 추출 용매 별 항산화 활성은 농도(0.2, 0.4, 0.6 mg/mL)가 증가 할수록 유의적으로 증가하였으며 control로 사용된 ascorbic acid, BHA, trolox 보다는 낮은 활성이 확 인되었다. 치자 껍질의 total phenol 함량은 EA, 70% methanol, DW 추출물 순으로 EA 추출물에서 26.59±0.20 CAE (caffeic acid equivalents) mg/g으로 가장 높았으며, Nitric oxide (NO) radical 소거 능에서는 70% methanol (70.32~76.15%), DW (52.66~59.31%), EA (34.65~46.98%) 추출물 순으로 나타났다. Nitrite (NO2) 소거능은 70% methanol (34.57~39.33%), DW (32.53~38.47%), EA (27.59~32.62%) 순으로 관찰되었다. β-carotene 탈색 저해능은 DW (41.55~50.97%), 70% methanol (23.37~44.80%), EA (13.37~25.24%) 순으로 동정되었다. Reducing power (optical density)는 70% methanol (0.044~0.127), DW (0.033~0.099), EA (0.026~0.097) 순으로 확인되었다. 지질과산화 저해 능은 껍질 추출물 중 70% methanol (56.51~76.21%), EA (54.59~63.34%), DW (47.92~61.11%) 순 으로 관찰되었다(p<0.05). 이에, 치자 껍질은 기능성 식품 및 천연 항산화제로서의 가치가 매우 높을 것 으로 판단된다.

        Total phenol contents, total flavonoid contents and antioxidant activity of 70% methanol, 70% ethanol and chloroform-methanol (CM, 2:1, v/v) extracts from onion (Allium cepa L.) peels were studied. The IC50 values of DPPH (2,2-diphenyl-1-picrylhydrazyl) radical and ABTS [2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity in 70% ethanol extract were remained to be lowest followed by 70% methanol extract and CM extract. And the total phenol content (113.56±0.86 mg CAE/g), total flavonoid content (49.63 mg QE/g) and ferric reducing antioxidant power value were also found to be the highest. In contrast, 70% methanol extract possessed the strongest antioxidant activity by β-carotene bleaching assay. CM extract displayed the lowest antioxidant activity compared with other extracts. Onion peels exhibited strong antioxidant activity and abundant phytochemicals, which could be used in a various food products to add phytochemicals and promote good health.

        This study was performed to evaluate the possibility of application of lactic acid bacteria fermentation to increase the anti-allergic activity of the extracts from Houttuynia cordata Thunb. H. cordata Thunb was fermented on 25, 30, 35 and 40℃ for 5 days by two species of lactic acid bacteria, Leuconostoc mesenteroides 4395 and Lactobacillus sakei 383. The anti-allergic activity of water extracts of H. cordata Thunb was then analyzed both before and after fermentation. Anti-allergic activity was determined in vitro assays by using 5-lipoxygenase (5-LO), cyclooxygenase-2 (COX-2) and β-hexoseaminidase release of RBL-2H3 cells (degranulation marker). The extracts fermented at 35℃ by both bacteria had the highest inhibitory activities against 5-LO, and also higher than the control, and the one fermented at 30℃ by both bacteria had the highest inhibitory activity against COX-2. The degranulation of RBL-2H3 cells induced by IgE-antigen complex was estimated as β-hexoseaminidase release rate as reference of 100%, the release rates were inhibited in 25 μg/ml of the extracts fermented at 30, 35 and 40℃ only by L. mesenteroides 4395. These results indicate that anti-allergic activity of H. cordata Thunb is increased by lactic acid bacteria fermentation.

        This study was conducted to investigate the effects of drying methods for Citrus junos peels on quality characteristics of the hot-water leachate from teabags containing those peels. Fresh peels were hot-air (50℃), cold-air (30℃), or freeze-dried (-45℃), powdered to a size of 40 mesh, packaged with a paper sachet, and then the packaged teabags were leached for 10 min with hot-water (70℃). L*value (lightness) and -a* value (greenness) of the peel powder were the highest in the freeze-dried samples. Soluble solids and titratable acidity of the teabag leachate were in the following order; cold-air, freeze, and hot-air dried samples. Among free sugar contents in all samples, fructose content was the highest, followed by glucose and sucrose. Fructose and glucose contents were not affected by drying methods. There was no significant difference in the flavonoid content among the peels dried using three drying methods. DPPH radical-scavenging activity of the leachate was the highest in the cold-air dried sample. These results suggest that cold-air drying would be an effective method to enhance the quality of hot-water leachate of teabags prepared from C. junos peels.

        This study was conducted to develop rice cookies added with young persimmon fruits. Effects of varying amounts (0-12%) of the fruit powder from a young astringent persimmon fruit (picked at July) on the quality characteristics of rice cookies were studied. Ingredients [rice (Oryza sativa subsp. japonica ‘Ilpum’) flours, persimmon (Diospyros kaki Thunb. ‘Cheongdobansi’) powder, sugar, butter, salt, baking powder, egg] were mixed, cut (thickness 3 mm, diameter 35 mm), baked at 170-180℃ for 9 min, cooled, and packaged in polyethylene/nylon bags. The loss rate and spread factor after baking of cookies increased and then decreased with an increase in the amount of fruit powder added. The moisture content and color values (L* and a*) of cookies decreased with an increase in the amount of the persimmon powder added. Phenolic compounds content and DPPH radicals scavenging activity increased with an increase in persimmon powder content; in particular, the DPPH activity of the cookies sharply increased after the addition of 3% persimmon powder. These results suggest that the addition of the young persimmon fruit powder affected the quality characteristics of rice cookies and this fruit powder (approximately 3%) can be utilized as an additive during rice cookie processing.

        The purpose of this study was to measure the bioactivity and antioxidant activity of peel from Gardenia jasminoides fructus Ellis (GJE) in Namhae, Korea, following some established methods. CM (Chloroform:Methanol, 2:1, v/v), 70% ethanol, and n-butanol extracts were collected. Flavonoid content and value as a functional food ingredient of GJE peel was investigated through assessing antioxidant [DPPH (1,1'-diphenyl-2-picrylhydrazyl), ABTS (2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid)], and hydroxyl radical scavenging activities; superoxide dismutase like ability; ferrous ion-chelating capacity; and tannin content by solvent extraction. Solvent extract antioxidant activities significantly increased (p<0.05) at increasing concentrations (0.2, 0.4, 0.6 mg/mL). GJE peel extracts were less active than the positive control [ascorbic acid, BHA (butylated hydroxyanisole), and EDTA (ethylenediaminetetraacetic acid disodium salt dihydrate)]. Based on the results of this study, GJE peel could be used as a natural antioxidant source due to its high antioxidant activity and bioactive compound content.

        The effects of ultrasound-assisted extraction (UAE) on the physicochemical and sensory characteristics of coffee beverage were investigated. Coffee powder (4 g) was soaked into 80 mL distilled water and extracted for 15, 30, 60, 120, 180, 240, or 300 sec at 60℃ in an ultrasonic bath (40 kHz, 300 W). Hot-water extraction (HE) at 80℃ for 300 sec was used as a control. Content of soluble solids in coffee beverage prepared by UAE (60~300 sec) was same or higher than those of HE, however, no significant differences were observed among the four beverages extracted for more than 120 sec. Clarity and browning index of beverage prepared by UAE (15 sec) and by HE were significantly higher and lower, respectively, compared to those of the other. Content of phenolic compounds was lower and higher respectively, in beverage prepared by UAE (15 sec, 30 sec) and UAE (60~300 sec) than by HE. DPPH radical scavenging activity was highest in beverage prepared by UAE (15 sec) and HE. Sensory evaluation showed that color, aroma, taste, and overall acceptability were rated with the best score for beverage prepared by UAE (15 sec) and the worst score for beverage prepared UAE (120~300 sec). Therefore, ultrasound-assisted extraction can be used for improving quality of coffee beverage.

        부추를 활용한 가용성 분말 제조에 적합한 공정 개발의 일환으로, 부추원료의 증숙처리 유무와 착즙액의 분무건조 부형제 종류에 따른 분말제품의 품질특성을 조사하였다. 생부추를 세척, 탈수, 절단 후 증숙처리(100℃, 3분)하거나 무처리한 다음 착즙하여 액을 얻고 여기에 부형제로 dextrin(DE=10)이나 β-cyclodextrin를 5% 첨가하고 분무건조하여 분말을 각각 제조하였다. 부추의 증숙처리는 착즙액 분무건조 분말의 L*값은 높게 하였지만 -a*값, b*값, C*값, ho값은 낮게 하는 효과를 보였다. 분말의 수분함량과 수용성지수는 증숙처리와 부형제의 영향을 받지 않았지만, 입자크기는 증숙처리와 dextrin 첨가구에서 가장 적은 것으로 나타났다. 분말의 클로로필, 총페놀, 비타민 C 함량은 증숙처리구보다 무처리구에서 유의적으로 높은 수준을 보였으나 부형제 종류별로는 유의적인 차이를 보이지 않았다. DPPH 유리기 소거능은 증숙처리에 의해 낮아지며 무처리구에서는 β-cyclodextrin 첨가구가 다소 높은 경향을 보였다. 증숙처리구의 관능적 색, 냄새, 종합 기호도는 무증숙처리구보다 유의적으로 높게 평가되었으며 부형제의 영향은 크게 보이지 않았다. 이로써 부추 착즙액의 분무건조 분말의 품질은 착즙 전 증숙처리의 영향을 크게 받는 것으로 확인되었고, 부추 고유의 이화학적 품질 특성유지에는 무처리가, 관능적 기호도를 고려할 경우에는 증숙처리가 유효한 방법으로 판단되었다.

        대추를 첨가한 드레싱의 제조와 대추퓨레의 첨가량이 드레싱의 이화학적 및 관능적 품질특성과 항산화능에 미치는 영향을 조사하였다. 드레싱은 생대추를 박피, 제심하고 스팀 블랜칭, 마쇄하여 퓨레를 제조하고 이의 첨가량이 0, 10, 20, 30, 40, 50%가 되게 간장, 식초, 올리고당, 올리브유, 물 등을 혼합하여 제조하였다. 드레싱의 색도 중 L*값은 대추퓨레 10% 첨가구를 제외한 나머지 대추퓨레 첨가구들이 무첨가구 보다 높은 수치를 보였으며, a*값은 대추퓨레 첨가구가 무첨가구의 수치보다 낮게 나타났으나 대추퓨레 첨가구 간에는 첨가량에 따라 수치가 비례적으로 증가함을 보였고, b*값은 대추퓨레 첨가구가 무첨가구 보다 높은 수치를 보였다. 총산도는 대추퓨레의 첨가량이 증가할수록 유의적으로 높아짐을 보였다. 가용성 고형분 함량은 대추퓨레의 첨가량이 증가함에 따라 높아져 50% 첨가구에서 가장 높은 수준을 보였다. 점도는 대추퓨레 첨가구가 무첨가구 보다 높은 수준을 보였으며 첨가구에서는 첨가량에 비례하여 점도가 증가하였다. 총페놀 함량은 대추퓨레의 첨가량이 높아짐에 따라 증가함을 보였다. 항산화능의 척도인 DPPH radical 소거능은 대추퓨레의 첨가량에 비례하여 증가하였다. 드레싱의 색, 냄새, 맛, 질감 및 종합적 기호도에 대한 관능검사 결과, 전반적으로 대추퓨레 첨가구가 무첨가가 보다 좋게 평가되었고, 모든 평가 항목에서 대추 퓨레 30% 첨가구가 유의적으로 가장 좋게 평가되었다. 이로써 대추퓨레를 첨가한 드레싱의 제조가 가능하였고, 드레싱의 항산화능과 관능적 품질특성을 고려한 대추퓨레의 첨가량은 30% 정도가 적합한 것으로 판단되었다.

        Direct Sky Blue-5B is an Azo dye known as general for staining of textile and leather, etc., and as materials which are difficult to be biodegraded in nature. The bacterium strain which could degrade direct Sky Blue-5B was isolated from activated sludge of dyeing factory and identified as Proteus sp. by experiment on morphological, cultural and biochemical characteristics, and so named Proteus sp. ST-1. The optimum condition of the strain for degradation of Sky Blue-5B were at about 35℃ and pH 7∼8. The strain had been capable of degradation with organic nitrogen effectively and had completely degraded 200㎎/ℓ of the dye within 12hrs at 37℃. The enzyme system related to degradation of Azo dye may be intracellular, and so degraded the dye after absorption into cell. The degradation products of Sky Blue-5B by Proteus sp. ST-1 were analyzed by Gas Chromatography /Mass Spectrometry and Spectrophotomer, from this observation, it may be infered that the strain degraded the dye directly without any mediate.
