Abstract In this study, the antioxidant activity of water and ethanol extracts from Hibiscus esculentus, Cirsium japonicum, Zizania latifolia and Kalopanax pictus for functional food source were examined. The optimal conditions for phenolic compounds extraction from medicinal plants were at 50% ethanol with Hibiscus esculentus and Cirsium japonicum var. ussuriense, at 40% ethanol with Kalopanax pictus and at 60% ethanol with Zizania latifolia. The total phenolic contents from the extracts of medical plants were determined to be 2.72~34.15 mg/g in the water extracts and 2.83~34.23 mg/g in the ethanol extracts. The electron-donating abilities (EDA) of the water and ethanol extracts were both above 74% at the low concentration of 50 μg/mL. The ABTS radical-cation decolorization was above 88% at 100 μg/mL concentration in all the extracts of various medicinal plants. The antioxidant protection factor (PF) in the water and ethanol extracts of the Cirsium japonicum var. ussuriense extracts was 1.73 ± 0.02 PF and 1.76 ± 0.01 PF at 50 μg/mL concentration respectively, and was higher than those of the other medicinal-plant extracts. The TBARs inhibition rates of all the medicinal-plant extracts, were above 80% at the 50 μg/mL concentration except Hibiscus esculentus. These results confirmed that the various oriental medicinal plants (Hibiscus esculentus, Cirsium japonicum var. ussuriense, Kalopanax pictus and Zizania latifolia) that were included in this study are useful anti-oxidant and functional-food resources.
The aim of this research was to investigate the anti-oxidantive activities of extracts from Chionanthus retusus leaves, fruits and flower. The content of phenolics were 20.8 mg/g in water extracts and 32.2 mg/g in 90% ethanol extracts from flower. The DPPH free radical scavenging activity of extracts from Chionanthus retusus was above 70% at phenolic concentration 100 μg/mL. The ABTS radical decolorization activities of water and ethanol extracts were both above 80% at 100 μg/mL phenolic concentration, respectively. The antioxidant protection factor(PF) of water and 80% ethanol extracts from leaves was the highest as 2.27 PF and 1.70 PF at 50 μg/mL phenolic concentration. The TBARs inhibition rate of the Chionanthus retusus extracts, was above 70% at 200 μg/mL phenolic concentration. The anti-oxidantive activities of extracts from leaves were shown more active than BHT as a positive control except TBARs. These results confirmed that the extracts from Chionanthus retusus leaves, fruits and flower was shown the high anti-oxidant activity. The results can be expected isolated phenolic compounds from Chionanthus retusus to use as functional beauty food resource.
In this study, we selected some material to have potential bioactivity from natural plants, confirmed as basic data for industrializing and tried to develope the food materials using them. DPPH, ABTS, antioxidant protection factor, TBARs and antimicrobial activity of extracts from Rosa multiflora Thunberg fruits were determined. The total phenolics extracted from Rosa multiflora were 12.08, 11.82, 11.1 and 12.6 mg/g when using water, 70% ethanol, 70% methanol and 70% acetone as the solvent, respectively. The optimum conditions for extracting the phenolic compounds were 70% ethanol over for 12 hrs(11.82 mg/g). The electron donating ability and inhibition rate on ABTS of the 70% ethanol extracts were 97% and 92.2%, respectively while the antioxidant protection factor(PF) of the water extracts and 70% ethanol extracts were 1.79 and 1.34 PF, respectively. The TBAR (thiobarbituric acid reactive substance) value were 1.3 μM for the control and 0.15 μM for the 70% ethanol extracts. The inhibitory activity against α-amylase was 26% for the 70% ethanol extracts. The 70% ethanol extracts from Rosa multiflora Thunberg fruits exhibited antimicrobial activity against H. pylori, S. epidermidis, S. aureusand and E. coli with clear zone diameters of 14, 25, 14 and 13 mm, respectively when using 200 μg/mL of the phenolic compounds. An HPLC analysis identified 6 major phenolic metabolites in the Rosa multiflora Thunberg fruits extracts: rosmarinic acid, caffeic acid, chlorogenic acid, courmaric acid, protocatechuic acid and quercetin. In particular, the content of rosmarinic acid was the highest in the 70% ethanol extracts. Therefore these results indicate that 70% ethanol extracts from Rosa multiflora Thunberg fruits can be useful as a natural antioxidant and in functional foods.
The extracellular polysaccharide production from suspension culture of Aloe vera L. was analyzed, and the optimization for suspension cultivation was investigated. From specific polysaccharide assay of glucomannan, FT-IR and 1H NMR spectrum, the extracellular polysacchride from suspension culture was found to be similar to β-1,4-glucomannan originated from Aloe vera fresh leaf. The polysaccharide existed in an acetylated form and its molecular weight by gel permeation chromatography was estimated to be 490 kD (Mn = 440 kD; polydispersity = 1.115). From medium optimization by simplex-cemtroid design using MINITAB®, the callus growth on the 2,4-D of 11.82 μM, kinetin of 13.84 μM and α-naphthalene acetic acid of 4.34 μM was turned out to be significantly superior than the growth on the other culture media combinations. Under this optimal culture media, other conditions such as the suspension culture's temperature, pH and the growth terms were determined. These results showed that the callus growth was optimum at 25oC, pH 5.5 and in 2 weeks. The maximum levels of growth and extracellular polysaccharide production of callus by optimal conditions were about 20.4 and 2.5 g/L, respectively, showing the profile of extracellular polysaccharide production was closely related to the callus growth.
The objective of this study was to identify the anti-oxidation, astringent, and inhibition effects of wild Ligularia fischeri on hyaluronidase and angiotensin conerting enzyme (ACE). In order to identify the total phenolic compound (TPC), various solvents were used for extraction showing hot water extract with the highest value of 14.42 GAE mg/g. In addition, ABTS radical scavenging activity measurements revealed an anti-oxdiation effect of 98.64-99.84% a hot water extract concentration of 50-200 μg/mL and a radical scavenging activity of 95.14-98.96% at a 60% ethanol extract content. If expressed in antioxidant protection factors (PF), the hot water extract showed 0.59-1.02 PF and the 60% ethanol sample displayed 0.30-0.74 PF. To identify the bio-activity effect, the hyaluronidase inhibition effect was determined as 4.66-35.00% in a 50-200 μg/mL hot water extract. Considering ACE inhibition effect, the hot water extract and 60% ethanol sample showed 0-64.24% and 46.12-69.64% inhibition effect, respectively. Lastly, when taking into account the astringent effect, the hot water extract with 50-200 μg/mL TPC concentration showed 15.68-26.92% and the 60% ethanol sample with an equal concentration exhibited 49.48-86.84%, which indicates the possibility to apply this product as a cosmetic source for pore contraction. Therefore, wild Ligularia fischeri extract can be used for anti-inflammation, high-blood pressure prevention, and as a source for health functional food with anti-oxidative properties.
This study was designed to determine the biological activities of Chionanthus retusus flower extracts. Water and 90% ethanol extracts of C. retusus flower were prepared. The inhibitory activities of water and ethanol extracts with a phenolic content of 200 μg/mL against xanthine oxidase were 25.60% and 15.92%, respectively. Further, the water extract did not show any inhibitory activity against α-glucosidase whereas the ethanol extract showed 100.00% inhibition of α-glucosidase. The inhibitory activities of the extracts against tyrosinase were 17.27% (water extract) and 36.13% (ethanol extract), which suggest that the extracts may have a whitening effect. The water extract did not inhibit elastase activity but showed a collagenase-inhibitory activity of 20.21%. On the contrary, the ethanol extract showed 96.26% and 35.93% inhibition of collagenase and elastase, respectively. These findings suggest that the extracts may have an anti-wrinkle effect. Lastly, the extracts showed a hyaluronidase inhibitory activity of 36.96% (water extract) and 88.70% (ethanol extract), suggesting that they may have an anti-inflammatory effect. The results indicate that C. retusus flower extracts containing phenolic compounds can be used as functional resources because they have anti-gout, carbohydrate degradation-inhibitory, whitening, anti-wrinkle, and anti-inflammatory effects.
본 연구는 2015년과 2016년에 ‘상주둥시’ 감의 수확시기 를 달리하여 수확 후 PE필름, 1-MCP 및 AVG를 처리가 저온저장동안 과실의 품질에 미치는 영향을 조사하였다. 2016년 과실 수확시기는 2015년에 비하여 10일 빠른 조기 수확을 실시하였다. 과실의 에틸렌 발생 정도는 조기수확 한 과실들에서 현저히 낮았고, 과실의 경도는 저온저장동안 1-MCP 처리구들이 다른 처리구의 과실에 비하여 높게 유지되었다. 과실의 감모율은 수확기를 달리한 모든 과실 들에서 PE 필름 처리 과실들에서 현저히 억제되었다. 성숙기에 수확한 과실은 저온저장 중 과실의 꽃받침 부위과 경와부의 과피색(L* 및 b*) 변화를 보면 무처리와 PE필름 처리구에 비하여 1-MCP 처리구들이 그 변화정도가 낮았다. 과실의 부패와 연화정도는 PE필름 처리 과실들에서 높게 나타났지만 AVG 처리구들에서는 부패과 발생이 없었다. 그리고 조기수확한 과일은 성숙기에 수확한 과실에 비하여 경도가 높게 유지되었고, 과피색의 변화가 낮은 경향을 보였고, 성숙기에 수확한 과실들은 당도, 과피의 적색 (Hunter a 값) 발현 및 호흡율이 높게 나타났다. 따라서 감 과실의 저장성 향상을 위해서는 조기수확이 중요한 요인으로 작용하고 또한 수확 후 1-MCP 처리가 과일의 경도를 높게 유지하고 과피색의 변화를 억제하였으며, 과실의 감 모율은 PE 필름 처리에 의해 현저하게 억제됨을 확인하였다.
본 연구에서 마삭줄(Trachelospermum asiaticum var. intermedium Nakai)을 용매별과 ethanol 농도별로 추출한 결과, 용매별 추출에서 ethanol 추출물이 20.8 mg/g으로 가장 높은 phenolic compounds 용출량을 보였고, 농도별 추출에서 70% ethanol 추출물이 38.1 mg/g으로 가장 높았다. 물 추출물은 16.8 mg/g의 phenolic compounds 용출량을 보였다. 마삭줄을 물과 ethanol을 추출용매로 사용하여 항산화 효과와 항통풍 및 항당뇨 효과를 확인하여 건강 기능성 식품으로의 가능성을 증명 하고자 하였다. DPPH radical 소거능 측정 결과, 100 μg/mL phenolic compounds의 농도에서 물 추출물은 80.9%, 70% ethanol 추출물은 83.1%의 활성을 나타내었다. ABTS radical 소거능 측정 결과 200 μg/mL phenolic compounds의 농도에서 물 추출물은 95.0%, 70% ethanol 추출물은 95.8%의 소거능력을 나타내었다. PF 측정결과, 100 μg/mL phenolic compounds의 농도에서 물 추출물은 2.43 PF, 70% ethanol 추출물에서는 2.45 PF를 나타내었다. TBARs 측정 결과, 200 μg/mL phenolic compounds의 농도에서 물과 70% ethanol 추출물에서 89.9%와 89.3%의 활성을 나타내었다. 통풍 억제 효과 측정 결과, 200 μg/mL phenolic compounds의 농도에서 물 추출물은 33.3%, 70% ethanol 추출물은 50.5%의 저해 효과를 나타내었다. α-glucosidase 저해 효과 측정 결과, 물 추출물에는 저해 효과가 나타나지 않았고, 70% ethanol 추출물에서는 200 μg/mL phenolic compounds의 농도에서 92.6%의 저해 효과를 나타내었다. 이러한 결과들로 미루어 보았을 때 마삭줄 추출물은 항산화 작용과 건강 기능성 식품으로 활용 가능성을 확인할 수 있었다.
본 연구에서는 긴병꽃풀의 항산화 효과와 기능성을 활용한 양갱을 제조하여 기계적 특성을 측정하고, 관능적 기호도 평가를 실시하였다. 생리활성 물질의 양을 측정하기 위하여 페놀성 화합물 정량을 실시한 결과 긴병꽃풀 열수추출물에서 12.99±0.30 mg/g, 70% ethanol 추출물에서 3.14±0.07 mg/g으로 나타내었다. 50-200 μg/mL의 농도에서 항산화 효과를 측정한 결과, 전자공여능은 열수추출물과 70% ethanol 추출물에서 각각 77.16-78.24%, 73.04-77.00%로 나타내었으며, ABTS는 84.35-99.75%, 83.74-99.55%로 나타내었다. Antioxidant protection factor(PF)는 1.54-1.62, 1.62-2.09 PF를 나타내었으며, TBARs는 42.93-94.09%, 91.05-95.19%로 나타내었다. 긴병꽃풀 추출물의 열수추출물과 70% ethanol 추출물은 거의 비슷한 항산화 효과를 나타내었으나, PF와 TBARs에서는 70% ethanol 추출물에서 더 우수한 항산화 효과를 나타내었다. Tyrosinase 저해활성은 열수추출물에서는 효과가 나타나지 않았으며, 70% ethanol 추출물에서는 30.32-40.56%의 저해활성을 나타내었다. Hyaluronidase 저해활성은 14.23-40.93%, 57.45-72.88%로 70% ethanol 추출물이 더 우수한 저해활성을 나타내었으며, 50-100 μg/mL의 저농도에서는 positive control로 사용한 PDTC보다 우수한 저해활성을 나타내었다. 색도는 긴병꽃풀 분말의 첨가량이 증가할수록 L 값과 a 값은 감소하는 경향을 나타내었고 b 값은 일정하게 나타내었다. 기계적 조직감은 경도와 탄력성에서 긴병꽃풀 분말의 첨가량이 증가할수록 감소하는 경향을 나타내었고, 응집성은 첨가량과 관계없이 일정한 값을 나타내었다. 씹힘성은 2.0%에서 가장 높은 수치를 나타내었으나 대조군과 첨가군 간의 유의적 차이는 나타나지 않았고, 긴병꽃풀 분말이 양갱의 기계적 조직감에 영향을 준다는 것을 확인할 수 있었다. 긴병꽃풀 분말 첨가 양갱의 관능적 기호도는 점도, 경도, 씹힘성, 탄력성, 맛, 향, 전반적인 기호도 항목에서 대조군을 제외하고 1.0% 이하 첨가군에서 기호도가 높게 나타내었으며 2.0% 첨가군은 모든 항목에서 가장 낮은 기호도를 나타내었다. 위 결과에 따라 긴병꽃풀 분말을 이용한 양갱 제조에는 1.0% 이하의 농도를 사용하여 제조하는 것이 가장 적합하다고 판단되었다.
In this study, beauty food activities were determined for extracts of Saurus chinensis as functional beauty food. Tyrosinase inhibitory effect was increased as more 4 folds by ultra-fine grind technology than normal grind technology in ethanol extracts from Saurus chinensis. A ultra-fine ground technology was used to increase the extraction yield for bioactive compounds from medicinal plants. Tyrosinase inhibitory activity, which is related to skin-whitening, was confirmed to be 100% for ethanol extracts at 1,000 ppm of phenolics. Anti-wrinkle activity was determined showing 30% of inhibition effect at above 1,000 ppm phenolic concentration. The elastase inhibitory activity of water and ethanol extracts from ultra-fine ground Saurus chinensis showed higher inhibition activity than compared to extracts from normal ground Saurus chinensis. Astringent activity in ethanol extracts was determined showing over 70% at 250 ppm of phenolics. Based on these results, Saurus chinensis extracts can be used as a functional beauty food and cosmetic with whitening, anti-wrinkle and pore contraction effects.
참취로부터 phenolic compounds를 추출 후 항산화 효과, 건강 기능성 식품 및 미용식품 활성을 검정하여 기능성 소재로 활용 가능성을 살펴보았다. 참취에 함유되어 있는 페놀성 물질은 물과 60% ethanol을 용매로 하여 추출하였을 때 각각 11.1 mg/g, 4.18 mg/g 함량을 나타내었다. 추출물의 phenolic 농도를 50~200 μg/mL로 조절하여 DPPH를 측정한 결과, 물 추출물과 ethanol 추출물 50 μg/mL phenolics 농도 에서 각각 87%, 91%였고, ABTS radical decolorization도 50 μg/mL phenolics 농도에서 각각 99%, 99%였다. Antioxidant protection factor는 200 μg/mL phenolics 농도에 서 물 추출물과 ethanol 추출물에서 각각 1.87 PF와 2.22 PF로 측정되었으며, TBARs값은 50 μg/mL phenolics농도에 서 물 추출물이 95%, ethanol 추출물에서 97%를 나타내었 다. Angiotensin converting enzyme(ACE) 저해능은 물 추출 물에서 50.8%, ethanol 추출물에서 69.4%의 저해율을 나타 내었으며, xanthine oxidase 저해능은 물 추출물은 50 μg/mL phenolics농도에서 91%, ethanol 추출물은 200 μg/mL phenolics농도에서 80%의 높은 효능을 보였다. α-Amylase 저해능은 물 추출물과 ethanol 추출물에서 각각 24%, 89% 로 ethanol 추출물에서 높은 효능을 보였다. Tyrosinase 저해 능을 측정한 결과 물 추출물과 ethanol 추출물에서 각각 47%, 25%의 미백 효과를 나타내었다. Elastase 저해능 측정 결과 물 추출물과 ethanol 추출물에서 각각 46%, 14%로 물 추출물에서 주름 개선 효과가 상대적으로 높게 나타내었 다. 이러한 결과로 보아 참취 추출물은 항산화 고혈압억제, 통풍억제, 당분해억제, 미백효과, 주름개선효과 등의 건강 기능성 식품 및 미용식품 활성 소재로서 활용 가능성이 큰 것으로 판단되었다.
In this study, the antioxidative activity and functional food activities of water and ethanol extracts from Pinus densiflora root were examined. It was more effective to use ethanol than water when extracting phenolic compounds. The extracted phenolic compounds from Pinus densiflora root for biological activities were examined. The phenolic compounds extracted with water and 80% EtOH were 1.86±0.04 mg/g and 6.85±0.16 mg/g, respectively. DPPH free radical scavenging activity of water and EtOH were each 86% and 85% at 100 μg/mL phenolics, respectively. ABTS radical decolorization activity was 48% in water and 68% in EtOH at 200 μg/mL. Antioxidant Protection Factor (PF) were 1.74 PF in water and 1.96 PF in EtOH at 50 μg/mL. TBARs of water and EtOH were 93% and 98%, respectively at 100 μg/mL. The inhibition activity on xanthine oxidase was 83.7% in water extracts and 79.6% in ethanol extracts. Inhibition on xanthine oxidase of water and ethanol extracts showed a higher inhibition effect than allopurinol. The inhibition activity on α-glucosidase was 14.8% in water extracts and 91.6% in ethanol extracts. The result suggests that P. densiflora root extracts may be useful as as functional food material.
In this study, the anti-inflammatory effect of hyaluronidase (HAase) inhibition was determined from 92 species of oriental herbal medicine extracted with water and ethanol solvents because of their non-toxicity in the human body. The water extracts of Evodia officinalis (86.8%), Thuja orientalis (80.8%), Carthami semen (66.5%), Melia azedarach (74.7%), Siegesbeckia pubescens (61.3%), Saururus chinensis (49.15%) showed a relatively greater anti-inflammatory activity. The ethanol extracts of Ailanthus altissima and Saururus chinensis demonstrated the highest anti-inflammatory effect at above 90%. Saururus chinensis was selected for its high anti-inflammatory effect in both water and ethanol extract. Ethanol was more effective than water and optimal extraction conditions for phenolic compounds was determined to be extraction with 50% ethanol for 12 hours. The extracts from Saururus chinensis in optimal condition showed 70~80% anti-inflammatory effect when 100~250 μg/mL phenolic concentration was treated. Concentration of above 500 μg/mL decreased the inhibitory effect. The anti-inflammatory effect and extraction yield were increased by ultra-fine grind technology, indicating that this method can be used to increase the extraction yield of phenolic compounds from medicinal plants.
본 연구에서는 감마선 조사가 떫은감 과육의 세포벽변화, 수렴성 및 경도변화에 미치는 영향을 살펴보았다. 감마선 조사 결과 떫은감 과육 중의 가용성 탄닌 함량의 변화는 대조구의 181.04±1.72 μg/g에 비해 처리 방사선의 양이 5~20 kGy로 증가할수록 146.31 μg/g에서 128.09 μg/g으로 과육내의 가용성 탄닌의 양이 감소하는 것이 확인되었다. 감 과육의 경도는 방사선을 조사하지 않은 대조구에 비해 방사선 조사한 감과육의 경도가 감소하였다. 세포벽 성분의 변화는 알콜불용성 물질의 경우 대조구의 39.3 mg/g에 비해 방사선 처리의 경우 37.2 mg/g로 낮아졌다. 수용성 물질의 함량은 대조구의 경우 11.4 mg/g이었으나, 방사선 조사의 경우 13.9 mg/g로 높아졌으며, 세포벽 성분의 함량은 대조구의 경우 26.6 mg/g였으나, 방사선 조사의 경우 23.1 mg/g로 낮아져, 떫은감 과육이 방사선조사에 의해 숙성이 일어나 세포벽성분의 함량이 감소하고 수용성물질의 함량이 증가한 것으로 나타났다. 세포벽 구성다당류 중 lignin의 함량과 pectin질의 함량은 대조구의 0.82 mg/g, 3.56 mg/g에 비교해 방사선 조사의 경우 0.77 mg/g, 3.14 mg/g로 감소하였다. 산가용성 hemicellulose의 함량은 lignin과 pectin처럼 방사선 조사에 의해 감소하는 경향을 나타내었으나, 알칼리 가용성 hemicellulose와 cellulose는 방사선조사에 의해 증가하는 양상을 나타내었다. 감과육에 존재하는 세포벽 분해 및 연화효소인 polygalacturonase, pectinesterase와 β-galactosidase의 활성의 경우 방사선 조사에 의해 모든 효소활성이 증가하였다. 방사선 처리감의 관능검사 결과 수렴성은 대조구의 경우 매우 떫은 것으로 나타났으나 방사선처리 후에는 거의 떫지 않게 느껴졌고, 과육의 경도에 대한 관능검사 결과 대조구는 딱딱한 것으로 나타났으나 방사선처리 후에는 약간 물렁해 지는 것으로 나타나 방사선처리에 의해 탄닌의 중합이 일어나며 과육의 숙성도 동시에 발생하였다. 따라서 감마선 처리는 떫은 감의 수렴성 제거와 숙성 기간을 단축시킬 수 있는 새로운 방법으로 활용이 가능할 것으로 판단되었다.
본 연구에서는 여주의 기능성을 활용한 양갱을 제조하여 물성을 측정하고, 소비자 기호도 조사를 실시하였다. 쓴맛을 느끼지 않게 하기 위하여 건조한 여주를 180℃에서 roasting하였다. 생리활성 물질인 phenolic compound의 함량은 raosting한 여주 물추출물에서 6.30±0.17 mg/g으로 가장 높은 추출률을 보였다. Phenolics 농도를 50~200 μg/mL으로 조절하여, non-roasting한 생여주와 roasting한 볶은여주의 항산화 활성을 측정하였다. 전자공여능은 non-roasting한 여주와 roasting한 여주가 각각 74.06~92.71%, 86.06~94.07%이었으며, ABTS는 36.26~98.03%와 67.02~99.60%이었다. Antioxidant protection factor(PF)는 2.19~2.25 PF와 2.20~2.36 PF이었으며, TBARS는 13.81~40.97%와 23.32~82.47%이었다. 여주 알콜 추출물이 물 추출물보다 상대적으로 50 μg/mL의 저 농도에서 항산화 효과가 더 우수하였다. 또한 roasting한 여주가 non-roasting한 여주보다 항산화 효과가 더 우수하였다. α-Glucosidase 활성저해 효과는 non-roasting한 여주의 경우 알콜 추출물에서는 물추출물에 비해 높은 효소저해 활성을 나타내었으며, 첨가하는 여주 phenolics에 대한 농도 의존적으로 저해 효과가 높아지는 양상을 나타내었다. 여주 분말 첨가량을 달리하여 제조한 양갱은 경도, 탄력성, 응집성, 씹힘성 등의 기계적 조직감과 색깔 등이 첨가되는 여주분말의 농도에 비례하여 품질이 나빠졌다. 또한, non-roasting 여주 분말을 사용했을 때는 첨가농도에 비례하여 색, 향, 맛, 조직감, 전체적 기호도 등의 관능적 특성이 나빠졌으나, roasting한 여주분말을 사용했을 때는 1% 이하의 저 농도에서는 관능적 특성이 오히려 좋아지는 것으로 나타났다. 따라서 여주분말을 부재료로 사용하여 양갱을 제조할 때, 기능적 특성과 관능적 특성을 고려한다면 roasting하여 첨가하는 것이 바람직할 것으로 생각되며, 사용농도는 1% 이하의 농도가 적합할 것으로 판단되었다.
초미세 분쇄를 하였을 때 입자가 작아질수록 추출수율이 높아져 약 2.5배 높은 추출수율을 나타내었다. 일반 분쇄한 시료 추출물에서 69.8%의 전자공여능 억제효과가 관찰되었고, 미세분쇄와 초미세분쇄 추출물에서는 각각 70.7과 83.8%의 억제효과를 나타내었다. 일반분쇄 추출물과 미세 분쇄 및 초미세분쇄 후 추출물 모두 97% 이상의 높은 ABTS 억제효과를 나타내어 분쇄 방법에 따른 항산화력의 차이는 거의 없었다. 일반 분쇄한 시료 추출물 보다 미세분쇄와 초미세분쇄 추출물에서 더 높은 PF값을 확인하였으며, 50% ethanol 초미세분쇄 추출물에서 1.8 PF로 가장 높은 항산화력을 나타내었다. 미세분쇄와 초미세분쇄 추출물에서는 일반분쇄 추출물에 비해 입자크기가 작아질수록 TBARS 억제율이 높아지며, 물 추출물보다 ethanol 추출물의 효과가 더 우수한 것으로 확인되었다. Xanthin oxidase 저해의 경우 초미세분쇄 후 효소억제 증대 효과를 확인할 수 있었다. Angiotensin converting enzyme 억제활성은 일반 분쇄 추출의 경우 물 추출물에서는 억제활성이 나타나지 않았고, 50% ethanol 추출물에서 24%의 억제율이 확인되었다. 또한, ethanol 추출물의 억제효과가 물 추출물에 비해 상대적으로 우수하였다. 50% ethanol 초미세분쇄 추출물에서 Staphylococcus aureus, Escherichia coli에 대해서 아주 약한 항균효과를 나타내었을 뿐 나머지 추출물에서의 항균 효과는 거의 관찰되지 않았다.
The phenolic compounds of water extracts from Prunella vulgaris were highest at 9.25 mg/g, respectively, when various extraction solvents were used. The optimum condition for extracting phenolic compounds from Prunella vulgaris was extraction in water for 18hr. The DPPH-scavenging activities of Prunella vulgaris were highest at the water extracts. The ABTS radical cation decolorization was higher than 40% in the range of 0~100% ethanol extract section. The antioxidant protection factor on the lipophilic phenolic metabolites was shown to be 1.1 PF in the water extracts from Prunella vulgaris. The TBARS was lower than the control (0.53 μM) in all the sections. The tyrosinase inhibitory effect, which is related to skin whitening, was above 40%, and for the anti-wrinkle effect, the elastase inhibition activity was above 40% at 0.2 mg/mL. The astringent effect of the Prunella vulgaris 40%ethanol extracts was 98.1% at 1 mg/mL. As a result, it can be concluded that Prunella vulgaris has the potential to be used as a cosmetic material.
The phenolic compounds which were extracted with 70% ethanol from Ulmus pumila for 12 hr were the highest as 17.9±1.0 mg/g. DPPH scavenging activity of 70% ethanol extracts was also the highest as 89.5±1.9% and it was confirmed to be high as 80% over in both of water and 70% ethanol extracts containing 50 μg/mL over phenolic concentration. ABTS radical cation decolorization activities of water and 70% ethanol extracts were higher as 96.8±2.9%, antioxidant protection factor (PF) was 2.0 PF in 70% ethanol and showed higher activities in both of water and 70% ethanol extracts containing 200 μg/mL phenolic concentration as 2.5 PF than BHA. TBARs of 70% ethanol extracts was 86.5±4.6%, it showed high anti-oxidative activity in 50∼200 μg/mL phenolic concentrations of water and 70% ethanol extracts as 80% over. The angiotensin converting enzyme (ACE) inhibitory activity of Ulmus pumila extracts against hypertension was 77.4% and 90.6% in water and 70% ethanol extracts of 200 μg/mL phenolic concentration. Xanthine oxidase inhibitory activity of Ulmus pumila extracts for anti-gout effect was not observed in water extracts, but it showed 30% inhibitory activity in 70% ethanol extracts, and 48.1% at 200 μg/mL phenolics concentration.
In this study, the optimal conditions for phenolic-compound extraction from medicinal plants were found to be 24 h and about 50% ethanol. The electron-donating scavenging activities (DPPH), ABTS radical-cation decolorization (ABTS), antioxidant protection factor (PF), and thiobarbituric acid reaction substance (TBAR) were measured to determine the antioxidant activities of the extracts of Sanguisorba officinalis Linn., Citrus unshiu Markovich, Melia azedarach L., Asparagus cochinchinensis Merr., Citrus unshiu S., Polygonum aviculare L., and Leonurus sibiricus L. The total phenolic contents of the extracts of medical plants were determined to be 0.45-3.00 mg/g in the water extracts and 0.33-3.15 mg/g in the 50% ethanol extracts. The electron-donating abilities (EDA) of the water and ethanol extracts were both above 85% at the 50 μg/ml concentration. The ABTS radical-cation decolorization was above 80% at the 100 μg/ml concentration in all the extracts of various medicinal plants. The antioxidant protection factor (PF) of the Melia azedarach L. extracts was found to be 1.65±0.40 PF in the water extracts at the 100 μg/ml concentration, and was higher than those of the other medicinal-plant extracts. The TBAR inhibition rates of all the medicinal-plant extracts, except Asparagus cochinchinensis Merr., were above 85% at the 100 μg/ml concentration. These results confirmed that the various oriental medicinal plants (Sanguisorba officinalis Linn., Citrus unshiu Markovich, Melia azedarach L., Asparagus cochinchinensis Merr., Citrus unshiu S., Polygonum aviculare L., and Leonurus sibiricus L.) that were included in this study are useful functional-food sources.
This study was carried out to determine the biological activity of Acanthopanax sessiliflorum fruit extracts. The phenolic compound contents of the extracts were 21.4 and 15.8 mg/g in hot water and 60% ethanol extracts. The total anti-oxidant activities of the water and the 60% ethanol extracts at a 200 μg/mL phenolic concent ration were at 92.4±0.8 and 89.2±1.1% in terms of the DPPH radical scavenging activity, 98.3±1.1 and 96.5±3.5% in terms of the ABTS radical decolorization, 2.0±0.6 and 1.2±2.8 PF in terms of the anti-oxidant protection factor, and 66.3±0.8 and 61.4±2.3% in terms of the TBARs inhibitory activity. The activities that inhibited the angiotensin-converting enzyme and xanthin oxidase were at 85.1±3.2 and 0% in the water extracts and 59.3±1.5 and 9.5±0.8% in the 60% ethanol extracts at the 200 μg/mL phenolic concentration. The tyrosinase and elastase inhibitory activities were at 56.6±1.8 and 53.1±1.1% in the water extracts and 33.7±2.2 and 22.4±3.1% in the 60% ethanol extracts. The astringent effect of the water and the 60% ethanol extracts were at 50.5±0.9 and 11.5±4.1%.
