본 연구는 조제유류 중 지방산에 대해 최신 분석법을 마련하고자 수행하였다. 조제유류 중 지방산 함량 분석을 위해 GC를 이용한 분석법을 확립하고 시중에 유통 중인 제품을 대상으로 적용성을 검토하였다. 분석법 검증은 특이성, 직선성, 검출한계 및 정량한계, 정확성, 정밀성에 대해 수행되었다. Linoleic acid 및 α-linolenic acid의 0.1-5 mg/ mL 농도범위에서 R2=0.999 이상의 우수한 직선성을 확인할 수 있었다. Linoleic acid 및 α-linolenic acid의 LOD는 각각 0.06 mg/mL, 0.01 mg/mL, LOQ는 각각 0.16 mg/mL, 0.03 mg/mL였다. 표준인증물질 분석을 통해 정확성을 검토 하였으며, linoleic acid 및 α-linolenic acid의 회수율은 각각 100.8%와 101.1%로 확인하였다. 정밀성을 검토한 결과 시료 채취량에 따른 반복성은 linoleic acid 1.4-2.9%, α- linolenic acid 1.1-2.7%이었고, 실험실간 재현성은 각각 2.8%, 1.5%임을 확인하였다. 본 연구에서 확립된 분석법을 적용하여 국내 유통 중인 조제유류 및 조제식 제품 12건에 대해 적용성 검토를 실시한 결과 전체 시료에서 분석이 용이 하였으며, 모두 기준·규격에 적합함을 확인하였다. 본 결과로부터 확립된 GC를 이용한 분석법은 조제유류 중 지방산 함량을 확인하기에 적합함을 확인하였으며 국내 식품 영양 성분의 관리 기반을 강화하는데 기여할 것으로 사료된다.
두 종류의 Cronobacter 선택배지(DFI agar, R&F agar) 의 분유 및 건조호박 내 Cronobacter의 선택 분리능을 realtime PCR법과 함께 비교하였다. 분유에서의 Cronobacter 검출률은 세 가지 방법에서 유의적인 차이를 나타내지 않았으나(p < 0.05), 건조호박의 경우 R&F배지와 real-time PCR법이 DFI에서보다 유의적으로 높은 검출률을 보였다 (p < 0.05). 배지 간 선택성에 있어서도, R&F 선택배지는 건조호박에서 DFI에 비해 유의적으로 높은 선택성을 나타냈다(p < 0.05). Real-time PCR 및 R&F배지의 사용은 분유뿐만 아니라, 건조 호박 등의 높은 경쟁세균총을 갖는 영유아식의 원료로 사용될 수 있는 식품군에서도 Cronobacter를 효과적으로 검출할 수 있는 방법으로 사료된다.
Choline is an amine essential to maintenance of good health. Here, we modified the ion chromatographic method for measuring choline described by Laikhtman & Rohrer and validated by the ICH guideline and applied it to monitoring of choline content in infant formula in the Republic of Korea.
The Certified Reference Materials (CRM), NIST 1849a, were used for validation of the modified method. The parameters of validation were linearity, range, specificity, accuracy, precision, LOD, LOQ and recovery. The linearity, R2, was above 0.999 and the analytical range was 2.5 to 50 μg/mL. The specificity was confirmed by the retention time (RT) and positive tests with calibration standard and CRM. The accuracy, precision and recovery values were 99.6 ± 2.41 %, 2.42 % and 90.8 - 92.8 %. The LOD and LOQ values were 0.03 mg/kg and 0.17 mg/kg, respectively. Five livestock products inspection agencies voluntarily participated in the collaborative study. The result, HORRAT value was 0.8 which was within the reference value 0.5 – 2.0. The validated method was applied to the analysis of choline in infant formula in the Republic of Korea. The results also conformed to 'The Standard for Livestock Products Labeling'.
Vitamin K1 is a fat-soluble vitamin essential for human health. We used the Association of Official Analytical Chemists(AOAC) method 999.15, validated by the International Council for Harmonisation of Technical Requirements for Pharmaceuticals for Human Use(ICH) Guideline, to monitor vitamin K1 content in infant formula in the Republic of Korea. The Certified Reference Materials (CRM), NIST 1849a, were used for validation of the test method. The parameters of validation were linearity, range, specificity, accuracy, precision, LOD, LOQ and recovery. The linearity, R2 was above 0.999 and the analytical range was 0.001 to 0.22 μg/mL. The specificity was confirmed by the retention time (RT) and positive/negative sample test. The accuracy, precision and recovery values were 101.13 ± 0.13 %, 3.15 % and 81.60 % - 99.78 %. The LOD and LOQ values were 0.582 ng/mL and 1.76 ng/mL, respectively. Five livestock products inspection agencies voluntarily participated in the collaborative study. The HORRAT value was 0.6, which was within the reference value 0.5 – 2.0. The validated test method was applied for the monitoring of vitamin K1 in infant formula in the Republic of Korea. The results also conformed to 'The Standard for Livestock Products of Processing and Ingredient Specifications' and 'The Standard for Livestock Products Labeling'.
Cronobacter muytjensii는 유아용 조제분유(IFP)의 잠재적 위험요인으로 중요한 식품 기인성 병원균이다. 이 연구에 서는 C. muytjensii 검지를 위한 특이적 면역글로불린 G(IgG)를 개발하고, 이 anti-C. muytjensii IgG를 이용하여 간접 비경쟁 효소면역측정법(INC-ELISA)을 개발하였다. 그 결과, 새롭게 개발한 INC-ELISA 방법은 C. muytjensii에 매우 민감하고, 순수배양 시 6.5×103 CFU/㎖의 검출한계와 유아용 조제분유에서 1 cell/25 g의 검출한계를 나타내었다. INC-ELISA 방법은 또한 C. muytjensii에 탁월한 특이성을 보이고, Cronobacter 속 외 11종의 다른 식품 기인성 병원균 계통과의 교차반응을 보이지 않았다. 이러한 결과는, 개발된 INC-ELISA 방법이 C. muytjensii에 매우 민감하고 효율적 이며, 신속하고 용이한 검출을 위한 진단 키트 개발에 적용할 수 있음을 시사한다.
Three-hundred samples of powdered infant formula milk and related products from four different manufacturers in 2010 were collected and surveyed their contaminations for aerobic bacteria, coliform, Enterobacter (Cronobacter) sakazakii, and food-borne pathogens. Fifteen samples of sterilized infant formula milk were all negative on these microorganisms. In all collected products of unsterilized infant formulas and follow-on infant formulas,aerobic bacteria were detected at 239 (83.9%) among 285 samples, and they all were found below 10³ cfu/g. Coliform bacteria were also detected at four among 285 samples. Salmonella spp. and Ent. sakazakii, weren't detected at the all samples. Bacillus cereus was detected at 24 (8.4%) among 285 samples. The level of B. cereus was below 100 cfu/g but it was suitable for the range of specification of B. cereus in infant formulas. Clostridium perfringens, Escherichia coli O157:H7, Staphylococcus aureus and Listeria monocytogenes weren't also detected. In consequence, it was suitable for total viable count, coliform and potential pathogen to the specification of infant formulas and related products.
In recent years, the national surveillance network reported the isolation of Enterobacter sakazakii and coliform from powdered infant formula (PIF). PIF means a breast-milk substitute specially manufactured to satisfy the nutritional requirements of infants during the first month. PIF is not sterile infant food and may be intrinsically contaminated with pathogens, such as E. sakazakii and Salmonella enterica that can cause serious illness in infants. Intrinsic contamination of PIF with E. sakazakii and S. enterica has been a cause of infection and illness in infants. E. sakazakii is an opportunistic pathogens and more commonly found than S. enterica in manufacturing environment. Even low levels of contamination of E. sakazakii and S. enterica in PIF were considered to be a risk factor. Enterobacteriaceae showed the high prevalence and some Enterobacteriaceae causing disease were detected. The infant food industry should be encouraged to reduce the concentration and prevalence of E. sakazakii in both the manufacturing environment and PIF. The presence of these microorganisms in PIF may occur as a results of contamination through ingredients during PIF manufacturing process and from the processing environment during packaging. In order to ensure that ingredients and packaging materials are microbiologically suitable, PIF manufacturer need to be selection of supplier according to stringent criteria (e.g. HACCP, good hygienic practices) and testing of ingredients especially colostrum. In conclusion, it is continually necessary to monitor the potential pathogens and reduce them to improve the microbial quality of non-sterilized PIF.
열수(Hot water)를 이용 조제분유 및 선식 준비 시 살균효과를 실험한 결과 60℃ 이상에서 살균 효과를 나타내었으나 조제분유와 선식에 열수를 가하였을 때 발생하는 온도 저하와 조제분유 및 선식 등의 분말 식품에서 biofilm을 생산하여 내열성이 증가되는 E. sakazakii의 특성을 고려하여 70℃ 이상의 열수로 조제분유 및 선식 등을 용해하여야 할 것으로 판단된다. 마이크로웨이브를 이용 E. sakazakii의 살균 효과를 실험한 결과 E. sakazakii를 제어하기 위하여 100 mL의 경우 90초 이상 가열하거나 온도측정이 곤란한 경우에는 최초 끓는 시점까지 가열한 후 냉각하여 섭취하여야할 것으로 조사되었다. 용해된 조제분유 및 선식의 저장 안정성을 평가하기 위해 5~35℃에 48시간 동안 보관하며 E. sakazakii의 성장곡선을 실험한 결과 5℃, 10℃ 냉장온도에서는 48시간까지 증식이 미약하였다. 그러나 냉장 온도에서 사멸하지 않고 미약하게 증식하는 E. sakazakii의 특성을 고려하여 조제분유 및 선식의 냉장 보관은 24시간 이내로 제한하여 섭취하여야 할 것으로 판단된다.
Purpose- This paper aims to examines the various factors Influencing Sales of Infant Formula Milk in China under the Covid-19 Pandemic, and base on this analysis, to present some measures for the sellers and consumers. Design/methodology- The 846 number of comments for 84 bands form e-commerce platform was taken as the index reflecting consumers’ purchasing behavior, and the properties of various types of infant formula milk (commodity name, gross weight and milk source) were taken as the independent variables to analyze the influencing factors of purchasing behavior of infant formula milk by multiple regression method. Findings- The important factors influencing the purchase behavior of infant formula milk are that product name, source of milk, domestic or imported products, applicable age, and packing unit. And then Consumers tend to buy 200-500yuan barrels of imported infant formula milk. Originality/value – The originality of this study can be found in its context, that is, under the Covid-19 Pandemic. At present, China has achieved the best epidemic control effects, which creates a great opportunity for Chinese brand infant formula milk to go abroad and realize internationalization in the future.
