        Mycoplasma hyorhinis (M. hyorhinis) is considered an etiological agent of arthritis in suckling pigs. Recently, some M. hyorhinis strains were shown to produce pneumonia that is indistinguishable from the mycoplasmosis caused by M. hyopneumoniae. In this study, we developed a sensitive and specific PCR assay to detect M. hyorhinis and applied the developed PCR assay for detection of Mycoplasma infection in clinical piglets infected with M. hyorhinis. We developed a new PCR assay using a M. hyorhinis-specific primer pair, Mrhin-F and Mrhin-R, designed from the Mycoplasma 16S-23S rRNA internal transcribed spacer (ITS) region. The primers and probe for the assay were designed from regions in the Mycoplasma 16S-23S rRNA ITS unique to M. hyorhinis. The developed PCR assay was very specific and sensitive for the detection of M. hyorhinis. The assay could detect the equivalent of 1 pg of target template DNA, which indicates that the assay was very sensitive. In addition, M. hyorhinis PCR assay detected only M. hyorhinis and not any other Mycoplasma or bacterial spp. of other genera. The new developed PCR assay effectively detected M. hyorhinis infection in pigs. We suggest that this PCR assay using a M. hyorhinis-specific primer pair, Mrhin-F and Mrhin-R, could be useful and effective for monitoring M. hyorhinis infection in pigs.

        This study aims to examine the effects of services marketing mix elements on customer equity through the customer experience in order to determine an optimal marketing mix strategy in the service industry. This paper utilizes confirmatory factor analysis and structural equation modeling (SEM) to analyze and confirm the proposed conceptual model. This study finds that the process, people, and physical evidence of the “7Ps” of the service marketing mix impart a positive and significant effect on the customer experience in the post-office setting. This result would help practitioners to create a service marketing strategy to differentiate their service from competitors in order to retain existing customers and attract new ones. It was also found that the customer experience is positively linked to customer equity. From the customer equity perspective, this study provides theoretical support that the customer experience, as one of the service operations management measurements, can be employed to measure service performance, which is also closely related to customer equity.

        Curcumin (diferuloylmethane), a constituent of turmeric powder derived from the rhizome of Curcuma longa, has been shown to inhibit the growth of various types of cancer cells by regulating cell proliferation and apoptosis. However, a need exists to design more effective analogs because of curcumin's poor intestinal absorption. EF-24 (diphenyl difluoroketone), the monoketone analog of curcumin, has shown good efficacy in anticancer screens. However, the effects of curcumin and EF-24 on salivary gland epidermoid carcinoma cells are not clearly established. The main goal of this study was to investigate the effects of curcumin and EF-24 on cell growth and induction of apoptosis in human salivary gland epidermoid carcinoma cells. Our studies showed that curcumin and EF-24 inhibited the growth of HTB-41 cells in a dose- and time-dependent manner, and the potency of EF-24 was > 34-fold that of curcumin. Treatment with curcumin or EF-24 resulted in nuclear condensation and fragmentation in HTB-41 cells, whereas the control HTB-41 cell nuclei retained their normal regular and oval shape. Curcumin and EF-24 promoted proteolytic cleavages of procaspase-3/-7/-9, resulting in an increase in the amount of cleaved caspase-3/-7/-9 in the HTB-41 cells. Caspase-3 and -7 activities were detected in viable HTB-41 cells treated with curcumin or EF-24. These results suggest that the curcumin and EF-24 inhibit cell proliferation and induce apoptosis in HTB-41 human salivary gland epidermoid carcinoma cells, and that they may have potential properties as an anti-cancer drug therapy.

        Proper oral hygiene is required to maintain oral health and prevent oral disease. Toothbrushing is central to proper oral hygiene. Mechanical tooth cleaning is the mainstay of plaque control. A variety of toothbrushing techniques have been developed and evaluated for their efficacy. However, these evaluations are subjective. To adequately evaluate toothbrushing objectively, a novel toothbrushing monitoring system was developed. The system involves user-monitored brushing patterns including toothbrush motions using a 3-axis gyroscope, 3-axis accelerometer, 3-axis magnetic sensor, one load-cell and Bluetooth devices. To confirm the efficacy of this toothbrushing monitoring system, eight periodontist performed tooth brushing in a dental model, and their brushing motion was monitored and recorded, and evaluated by statistical means. The proposed monitoring system can be used to aid dental care personnel in toothbrushing instruction.

        In this paper, the forward and inverse kinematics equations for demolition fire vehicle with 6-DOF were developed. This was done through the development of an interface that allows co-simulation using MATHEMATICA and ADAMS. The implementation of this project was done entirely in MATHEMATICA by calculating the kinematics equations to demonstrate the usefulness of this product. The forward and inverse kinematics of the demolition fire vehicle represents the position and orientation of the end-effector as any desired location. This research was particularly conducted using the simulator, and it showed good approximation results in terms of solving a reaching task problem.

        Entomopathogenic Beauveria bassiana and Metarhizium anisopliae are well-known biological control agents worldwide and have high potential in industrialization. However their thermo-susceptibility limits long-term storage under high temperature conditions and high insecticidal activity after application to target pests. Herein we isolated highly virulent isolates, B. bassiana JEF006 and JEF007 and M. anisopliae JEF003 and JEF004, and produced in three grains, such as sorghum, millet and Italian millet as substrates for solid cultures, followed by thermotolerance assays to compare the potential of the three substrates for thermotolerance. The JEF isolates were exposed to dry and wet heat at 50°C and overall conidia were more stable under dry heat condition rather than wet heat. Of the three grains, Italian millet was superior to the other grains in the production of thermotolerant conidia. Additionally Italian millet did not severely aggregated, which enabled air to penetrate into the substrate well compared to the sorghum and millet. JEF isolates were more thermotolerant when they were kept in oil conditions as carriers of an oil-based formulation. This work suggests that Italian millet can be used as an effective substrate to produce more thermotolerant conidia, thus maintaining viability for long times under unfavorable environment and biological activity against target pests.

        Bean bug, Riptortus pedestris is an agriculturally serious pest in East Asian countries, reducing the value of crop quality and loss of income in agribusiness. Chemical pesticides have contributed to the management of the pest, but nowadays insect resistance limits the use of chemical pesticides, thus alternatively new pesticides with different mode of actions such as entomopathogenic fungi are considered. Beauveria bassiana and Metarhizium anisopliae JEF isolates were collected, identified and assayed against bean bugs in laboratory conditions. Some isolates showed >80% virulence by spray and contact-exposure methods. The isolates produced high levels of pathogenesis-related enzymes, such as chitinase, Pr1 protease and lipase. The Agrobacterium tumefaciens-mediated transformation generated random transformants and some mutants had reduced virulence against bean bugs, which provided some materials to figure out pathogenicity-related genes in the fungi. Now characterization of flanking region of the integrated fragment is underway and this work may reveal some important genes in the pathogenesis. This work can be a strong platform for the functional genetics of bean bug-pathogenic B. bassiana.

        The entomopathogenic fungus, Beauveria bassiana is widely used in integrated pest management (IPM), however its successful application is often limited by the little effort to explore its functions of unknown genes. In this work, egfp-expression cassette was randomly integrated into B. bassiana using Agrobacterium tumefaciensmediated transformation, and the general features of the mutants with unusual characteristics and the localization of the integrated genes were explored. To construct a transformation vector, egfp-expression cassette including gpdA promoter and trpC terminator was cut from pBARKS1-egfp using SacI and HindIII and integrated into pCAMBIA containing hygromycin B resistant hygR gene, designated as pCAMBIA-egfp. Transformed B. bassiana isolates were grown on quarter strength-Sabouraud dextrose agar containing 150 μg hygromycinB ml-1. Expression of egfp was investigated by RT-PCR and a fluorescent microscope (400×). Through the genome walking of the transformants using adaptor primers and gene specific primers, unique bands were detected on the egfp-expressing transformants, which were sequenced to figure out the flanking regions. This work provides a platform of methodology to figure out unknown functional genes of B .bassiana and possibly suggest an improved strategy to use the entomopathogen in IPM.

        Recently an outbreaked pest belongs to Hemiptera: Recaniidae, Ricanula sp. is greatly concerned about the outspreading throughout the South of Korea by wide range of host, including Cornelian cherry, Jujube, and peach trees. In Chungbuk province, this pest was first occurred at Jincheon and Okcheon in 2012, Cheongju in 2013 and now found out at Eumseong and Goesan in 2015. Ricanula sp. was oviposited directly into one-year twig, did damages on fruit-bearing formation and finally withered the host. This study was performed to understand the ovipositional characteristics and to develop the standardized forecasting method. Oviposition by Ricanula sp. was abundant in tree than in bush, adult laid eggs on new inner twigs and then covered with wax compound. Total no. of oviposited egg-mass was 10 to 318, and that of on new twig was 5 to 185 per tree, with different to host trees. Thickness of oviposited twigs were done within 2 ~ 5.5 mm and the height was mostly founded with range of 1 to 2 m, founded with highest height over 3 m. Oviposited no. of egg-mass within 30 cm twig was appeared differently from 2 to 7 every host. From based on this investigated result, we provide this for standardized forecasting method. This pest will need to control when egg-mass will occur over 2 at new twig, within 30 cm from the tip, set as total 25 point/ 5 plants (5 point per plant).

        An oxidative fumigant is potent to kill insect pests infesting stored grains. Its oxidative activity generates reactive oxygen species (ROS), which has been considered to be a main insecticidal factor. Furthermore, the oxidative fumigant has cytotoxic effect to insect cell lines, but the cytotoxicity is abrogated by antioxidant treatment. This study aimed to extend the usefulness of the oxidative fumigant in terms of medical purpose against cancer cells. Five cancer cell lines HCT 116 (human colorectal), Lovo (human colorectal), SW480 (human colorectal), MDA-MB-231 (human breast), and MCF-7 (human breast) were tested to determine their susceptibility to the oxidative fumigant with reference to two insect cell lines (Sf9 and Hi-Five). All cancer cell lines were highly susceptible to the oxidative fumigant, compared to the insect cell lines. Interestingly, basal ROS levels of the cancer cell lines were much higher than the insect cell lines. Furthermore, the oxidative fumigant significantly increased the ROS levels in the cancer cells. Treatment of vitamin E as an antioxidant mitigated the cytotoxicity of the oxidative fumigant. Thus, the high susceptibility of cancer cells to the oxidative fumigant may be induced by their high inducible ROS production. This study also investigated the antiviral activity of the oxidative fumigant against insect and plant viruses. The oxidative fumigant significantly inactivated a baculovirus (dsDNA virus) by inhibiting polyhedral production in Sf9 cells. It also inactivated tobacco mosaic virus (ssRNA virus) by suppressing phytopathogenicity. These results support a broad effect of the oxidative fumigant, which can be applied to agricultural and medical purposes.

        Cabbage is one of the major vegetable crops especially in Korea. Various insect pests intimidate the cabbage production. Moreover, an excessive spray of chemical insecticides leads to development of insecticide resistance as well adverse effects on environment and humans. The brassica leaf beetle, Phaedon brassicae, is a coleopteran pest. Both larval and adult stages feed cabbage. This study aimed to develop an effective microbial biopesticide against P. brassicae by adding immunosuppressive agent to Bacillus thurigiensis (Bt). To determine an optimal immunosuppressive agent, two bacterial species of Photorhabdus temperata subsp. temperata (Ptt) and Xenorhabdus nematophila were compared in their insecticidal activity against P. brassicae, and Ptt was selected. Among four subspecies of Bt, B. thuringiensis subsp. tenebrionis (Btt) was selected. Ptt bacterial culture broth possessed immunosuppressive agent(s) because the bacterial culture broth significantly increased other bacterial growth in P. brassicae hemolymph. When the Ptt broth was added to Btt, it significantly increased the insecticidal activity of Btt. Bt-Plus was prepared by mixing Btt and Ptt bacterial culture and sprayed the cabbage infested by P. brassicae. The Bt-Plus was much more effective than Btt treatment. This study reports a novel Bt insecticide to control P. brassicae.

        The purpose of the present study is to provide exercises, together with music, that are helpful for elderly people’s balancing ability, and to determine their effects in order to provide data for the promotion of elderly persons’health. Thirty elderly persons were randomly assigned to a balance training group(BTG) of 15 subjects, or a music and balance training group (music therapy + balance training [MTBTG]) of 15 subjects; intervention was implemented three times per week for six weeks. To measure the changes in their balancing ability before and after the experiment, the limit of stability, the“Timed Up and Go”(TUG) test, and the Berg Balance Scale (BBS) were measured. Changes in the limit of stability before and after the experiment were shown to be significant in both the BTG and the MTBTG. Changes in the limit of stability of the right side before and after the experiment showed statistically significant differences between the BTG and the MTBTG. Changes in the TUG test and the BBS before and after the experiment were shown to be statistically significant in both the BTG and the MTBTG. The application of music during balance training thus is considered to have a positive effect on elderly persons’balancing ability.

        Canine respiratory coronavirus (CRCoV) is commonly associated with canine kennel cough worldwide. Clinically infected dogs present coughing, sneezing, and nasal discharge. Severe infections may progress to pneumonia. Through serological surveys, CRCoV has been identified as a worldwide pathogen found in the respiratory tracts of dogs suffering from mild or severe respiratory disease. In this study, three dogs were obtained from a dog kennel. Over the previous 5 days, the dogs showed coughing, sneezing, and nasal discharge. To detect the etiologic pathogen, we performed multiplex RT-PCR (mRT-PCR) to amplify the genes encoding canine influenza virus matrix protein, canine distemper virus nucleocapsid protein, and CRCoV spike protein. Dot blotting was achieved with a CRCoV-specific probe. Nasal-secreting CRCoV was detected by the 442 bp CRCoV-positive PCR reaction in the nasal swabbing samples from dogs. Further, CRCoV-positive reactions by dot blot hybridization were detected in the nasal swabbing samples from dogs. In conclusion, we detected CRCoV in kenneled dogs with respiratory disease in Korea. Multiplex RT-PCR was able to detect successfully CRCoV infection in dogs. We suggest that mRT-PCR would be useful and effective for monitoring CRCoV infection in various kinds of dogs.
