        The JK-1 isolate which was the best producer of indole-3-acetic acid and carotenoid among the 388 strains isolated from 28 wetlands in Jeju, was identified to be Rhodopseudomonas palustirs belongs to a typical group of non sulfur purple bacteria based on 16S sRNA sequencing. This study investigated the effect of different cultural conditions of pH, temperature, agitation, light and aeration on growth, IAA and carotenoid production of photosynthetic bacterium JK-1 for optimization of IAA and carotenoid production. It was found that growth, IAA, carotenoid, and bacteriochlorophyll production with light (3,000~3,500 Lux) and agitation (100 rpm) showed better results than those with dark/static or dark/ agitation (100 rpm) in anaerobic conditions. The optimal pH, temperature and agitation speed for cell growth were 7, 30℃, 150 rpm, for IAA production were 9, 30℃, 150rpm and for carotenoid production were 6, 25℃, 50 rpm, cultured for 72 h under anaerobic light, respectively. The growth and IAA production were high in aerobic culture compared with anaerocic culture, whereas carotenoid and bacteriochlorophyll content were decreased extremely in aerobic condition (0.5~1 vvm). Subsequently, the optimal culture conditions for JK-1 were selected with pH 7, 30℃ and 100 rpm under anaerobic light and the effect on plant growth was tested by pot assay. Inoculation of JK-1 with 3% (v/v) level caused increase in shoot and root dry weigh that varied from 20%~58% to 40%~28% in young radish in camparison to uninoculated treatment at 50 days of growth. The study suggests that the JK-1 isolate may serve as efficient biofertilizer inoculants to promote plant growth.

        본 연구는 제주지역 화산회토 감귤원에서 두과피복작물인 헤어리베치(Vicia villosa Roth) 의 이용 가능성을 진단하고 효율적인 양분관리 체계를 정립하기 위하여 수행하였다. 시험 은 감귤원 재배지 고도별 5개소(해발고도:신촌 37 m, 호근 116 m, 와산 123 m, 상예 161 m, 그리고 용강 232 m)에서 수행하였다. 헤어리베치 품종은 국내에서 개발된 ‘청풍보라’로 하였으며, 파종량은 ha당 30, 60 및 90 kg을 파종하였다. 시험결과 헤어리베치 입모율은 53.0~57.1%로 파종량과 고도별 포장에 관계없이 유사하였다. 파종량에 따른 잡초 발생비율 은 5개소 평균 11.8, 3.8 및 5.1%로 처리간 유의성이 없었으나 30 kg 파종구에서 다소 높은 경향을 보였으며 60 kg 및 90 kg 파종구는 각각 96.2% 및 94.9% 정도의 잡초 경감효과를 나타냈다. 헤어리베치 파종량에 따른 ha당 건물 수량은 각각 7.37, 8.43 및 8.47톤으로 30 kg 대비 60 및 90 kg 파종 시 각각 14% 및 15% 높았으나 파종량별 유의성은 없었다. 질소 수 량은 ha당 최소 175 kg (와산 30 kg ․ ha-1 파종구)에서 최대 467 kg (용강 30 kg ․ ha-1 파종구) 값을 얻었으며 파종량에 따른 5개소 평균 질소 수량은 각각 254, 316 및 315 kg으로 60과 90 kg 파종구간 차이는 없었으며, 30 kg 파종구에 비하여 24% 높았다. 감귤원에서 기상과 토양 화학성 등 재배환경 요인들이 헤어리베치 수량에 미치는 영향을 구명하기 위하여 요 인들 간 상관관계를 분석한 결과 일평균기온(R=0.2746) 및 적산온도(R=0.2714) 등 기상요인 은 헤어리베치 건물 수량에 다소 영향을 줄 수는 있으나 그 영향이 크지 않은 것으로 판단 되었다. 반면에 토양 EC (R=0.4520**), 치환성칼륨(R=0.4078**) 및 유효인산(R=0.2737) 함량 등이 높을수록 건물 수량이 증가하는 비교적 강한 양의 상관관계를 보여 기상요인 보다는 토양 화학성이 헤어리베치 수량에 미치는 영향이 큰 것으로 판단되었다. 따라서 헤어리베 치 재배시 질소 수량은 포장별 여러 가지 재배적 환경요인들의 영향을 받을 수 있으므로 포장별 일정면적의 생초 수량(X)을 측정한 후 이에 따른 질소 수량(Y)을 산출하여 활용하 는 것이 바람직할 것으로 보이며 그 산출식은 「Y=4.4097X+33.594 (R=0.9547)」와 같다.

        A new Gentiana spp. cultivar “Skyblueball” was bred by crossing between “Jinbu-65 line” with violet blue (VB 95B) colored petals as female and “Jinbu-1 line” with violet blue (VB 93B) petals as male in 2000 year. The superior line of 00-168-3 was selected in Daegwallyeong from 2001 to 2003 year, and its growth and flower characterization were conducted from 2004 to 2007 year in field of Daegwallyeong and Jinbu, Pyeongchang, Gangwon-do, Namwon, Jeollabuk-do and Jeju Special Self-Governing-do. The flower showed violet blue (VB 96C) color at opening, the petal of flower had not spot and flower stem color was purple. The flowers were bloomed at the same time from bottom to upside (Table 1). Plant height was 62.9 ㎝ , number of flower stem per plant is 8.3, number of flower node per stem was 5.3, and number of flower per stem was 21.7 (Table 2). The flowering of ‘Skyblueball’ was bloomed from July 26 in Daegwallyeong, August 3 in Jinbu and Namwon, and June 28 in Jeju Special Self-Governing-do. ‘Skyblueball’ cultivar can be used for cut flower.

        This study was carried out to investigate change of ginsenoside contents in red and fresh ginseng according toroot part and age by hydrolysis. Neutral total ginsenoside contents by hydrolysis in 6-year main root and lateralroot were significantly increased than those by non-hydrolysis, as 41.6 and 32.8%, respectively. However, there wasno significant difference in red ginseng. In fresh ginseng, ginsenoside contents of the protopanaxatriol group such as Re, Rf,Rg₁, Rg₂, and Rh₁ were not significantly different, but Rb₁, Rb₂, Rb₃, Rc, and Rd showed significant difference. The increaserate of neutral total ginsenoside content by hydrolysis was higher in epidermis-cortex than stele. Also, the neutraltotal ginsenoside content was fine root>rhizome>lateral root>main root, respectively. While there was no tendencytowards the increase of ginsenoside by hydrolysis with the increase of root age in fine root and rhizome, there was significantdecrease in main root and lateral root.

        This study was carried out to investigate change of ginsenoside contents according to tissue ratio in ginseng root by age and diameter. The epidermis-cortex and xylem-pith extent, fresh weight, dry weight of ginseng increased with the root age increase. They increased higher in xylem-pith than in epidermis-cortex. The ratio of epidermis-cortex decreased and xylem-pith increased as the main root diameter increased. In case of same diameter, the xylem-pith ratio increased by the increase of root age. The epidermis-cortex ratio was 4 > 5 > 6 years, respectively. The total 10 ginsenosides of epidermis- cortex increased with the root age increase. However, the total ginsenoside of xylem-pith decreased and it was 2~5 times lower than epidermis-cortex. The most of ginsenoside contents existed in epidermis-cortex. The diameter decrease in main root is related to the increase of epidermis-cortex ratio. It leads to increase of ginsenoside contents. In order to select high level of ginsenoside cultivar, it suggested that it should be selected main root having narrow diameter and lower epidermis- cortex ratio.

        This study was carried out to investigate ginsenoside content in different root parts and the correlation between root diameter and ginsenoside composition of Panax ginseng C. A. Meyer cultivated by direct seeding. The unit contents of ginsenoside were 29.65, 28.76, 26.34 mg/g, respectively in 4, 5, 6 years old. However, the total contents of ginsenoside were 431.97, 606.56, 657.80 mg/root, respectively. Total ginsenoside content of fine root was higher than that of main root and lateral root. These tendencies were related to decrease by the increase of root diameter. When diameter of main root and lateral root were the same in different ages, the total ginsenoside content was higher in the order of 4 > 5 > 6 years old roots. Except for ginsenoside-Rg1, other ginsenosides components (PD/PT and total ginsenosides) had highly negative correlation with the root diameter within whole root, main root, lateral root and fine root, which indicated that ginsenoside content is correlated to root diameter. As results, it is suggested that ginsenoside content can be predicted.

        This study was conducted to select plant growth regulators effective at ginseng berry set inhibition to help root growth in Korean ginseng (Panax ginseng C. A. Meyer). PGRs (ethephon, gibberellic acid, maleic hydrazide, coumarin) were applied to field grown 5-year-old Korean ginseng between one and two times, before and during bloom in 2009, 2010. The number of treatment was more effective in ginseng berry set inhibition when used two times compared with one time in GA 1,000 ppm, MH (5,000, 10,000 ppm), coumarin (5,000, 10,000 ppm) treatment. According to treatment period of plant growth regulator, ginseng berry set inhibition rate from 20days before flowering date to 5days after blooming was the highest in MH 5000 ppm showing 99.9% and the lowest in GA 100 ppm showing 32.8%. The spray treatments of Ethephon (50, 150 ppm) and MH (5,000, 10,000 ppm) from 20 days before the flowering bloom up to 5 days before, and coumarin (5,000, 10,000 ppm) from 20 days to 6 days and before blooming that induced the inhibitory effect more than 90% after 12 weeks. Considering ginseng berry set inhibition characteristics and treatment period ethephon and coumarin was important about applied period but, MH treatment appeared to effective ginseng berry set inhibition regardless of treatment period.

        This research was conducted to investigate the influence of various organic substrates on growth and yield of ginseng seedling grown organically in the closed plastic house. The pH and EC of substrates used for organically ginseng cultivation ranged 5.93~6.78 and 0.03~0.15 dS/m respectively. The concentrations NH4-N and NO3-N respectively was 14.01~68.63 mg/L, 5.60~58.83 mg/L. The average quantum of the closed plastic house was range from 10 to 16% of natural light. In July and August, the maximum temperature of the closed plastic house did not exceed 30 and the average temperature was maintained within 25 lower than the field because air conditioning ran. The PPV-1 and PPV-2 bed soil substrates produced higher stem length, stem diameter, shoot fresh weight and leaf area than those of conventional culture. In PPV-2 bed soil substrates, root fresh weight and root diameter was the highest. The root fresh weight of PPV-2 bed soil substrates in closed plastic house was maximum 25% heavier than the conventional cultivation. The results of this experiment will be utilized for making new substrate application for organic ginseng culture in the plastic house.
