        Gold kiwifruit was fermented with Lactobacillus plantarum CK10 derived from kimchi and the fermented products were extracted with ethanol at various fermentation time-points The bacterial cellular density, total titratable acidity, total polyphenol content (TPC), and total flavonoid content (TFC) increased during fermentation, while pH values and total soluble solids decreased. Levels of TPC and TFC were highest after five days, at 1.21±0.13 mg GAE/g dry weight and 0.36±0.04 mg RE/g dry weight, respectively. The antioxidant activities of the fermented gold kiwifruit were analyzed using Fe2+ chelating activity, 2,2-diphenyl-1-picrylhydrazyl (DPPH)-, and 2,2'-azino-bis(3- ethylbenzothiazoline-6-sulphonic acid) (ABTS)- radical scavenging activities, and superoxide dismutase (SOD)-like activity. The Fe2+ chelating activity of gold kiwifruit (125-500 μg/mL) peaked after five days of fermentation at 38.40-78.47%. The DPPH radical- scavenging activity and SOD-like activity were somewhat higher after seven days of fermentation (36.01-86.81% and 54.79-93.83% at 2.5-10.0 mg/mL concentration of samples, respectively). On the other hand, the ABTS radical- scavenging activity of fermented gold kiwifruit was similar to that of the non-fermented form. The polyphenol and flavonoid contents were significantly correlated with the antioxidant activity. In conclusion, our results suggest that TPC, TFC, and antioxidant activity were increased after five and seven days of fermentation, respectively. Therefore, fermented gold kiwifruit with its increased antioxidant activity could be useful in the development of functional foods.

        Fermented halla gold kiwifruit (FHK) was prepared with Lactobacillus plantarum CK10, a bacterium derived from kimchi. We investigated the quality characteristics and antioxidative activity of madeleine added with FHK. The madeleine dough was prepared by mixing flour, sugar, baking powder, and then followed by adding salt, rum, different amount of the FHK (0, 1, and 3%) and butter. The total titratable acidity of madeleine increased significantly with the amounts of added FHK (p<0.05), while the pH value and total soluble solids showed the reverse trend. The color of madeleine became substantially redder with increasing amounts of FHK (p<0.05), and it appeared darker and less yellow at the same time. The total polyphenol contents of madeleines increased significantly with increasing amounts of FHK (p<0.05), but there was little difference in the total flavonoid content. When the antioxidant activities were measured in terms of 2,2-diphenyl-1-picrylhydrazyl (DPPH)- and 2,2’-azino-bis-3-ethylbenzothiazoline- 6-sulfonic acid-diammonium salt (ABTS)- radical scavenging, both measured activities of madeleines increased dramatically with added FHK in a dose-dependent manner. Our results suggested that the acidity, color, polyphenol content, and antioxidant activities of madeleines can be improved by adding the fermented gold kiwifruit.

        Platycodon grandiflorum (Bell flower) is an important plant that has traditionally been used as herbal medicine for the treatment of cough, phlegm, sore throats, lung abscesses, chest pains, dysuria, and dysentery. The present study was initiated to investigate the feasibility of inducing shoot and root organogenesis in cultured explants of P. grandiflorum in a range of culture media and through use of various plant growth regulators (PGRs). The plantlets (Stem containing one node) were isolated and cultured on different concentrations of Murashige and Skoog (MS) medium supplemented with PGRs. We found that proliferation and elongation of shoots and roots could be achieved on ¼ MS for P. grandiflorum with wild and green petals and on ⅛ MS for P. grandiflorum with double petals. The highest levels of development and elongation of adventitious shoots and roots were observed when petal explants were cultured on ¼ MS (pH 3.8) supplemented with 5% sucrose. Increasing the agar concentration reduced shoot growth and rooting potential; nevertheless, the highest number of shoots and roots was observed on 0.6% agar. In the case of growth regulators, ¼ MS supplemented with 1 mg L-1 6-benzylaminopurine (BA) was found to be best for shooting, although higher concentrations of BA tended to reduce shoot and root elongation. The highest number of shoots was achieved on 0.5 mg ․ L-1 thidiazuron (TDZ) from double petal explants grown on ⅛ MS. However, root and shoot elongation were found to decrease when TDZ concentrations were increased. Low concentrations of kinetin, naphthalene acetic acid, indole acetic acid, and 3-indole butyric acid induced shoot and root proliferation and elongation. Taken together, our study showed that low concentrations of PGRs induced the greatest root formation and elongation, showing that the optimal concentration of PGRs for shoot proliferation was species-dependent.

        This study analyzed the placement shape and correlation of green space focusing on anion that is one of curing substances. As its result, it was found out that the meteorological element becomes different and the generated amount of anion also gets different along such different meteorological element in case the shape of placing green space. As the result of analyzing the correlation with meteorological element , it was analyzed that it was the positive correlation in case of time, temperature and relative humidity and it was also analyzed that it was negative correlation in case of wind velocity and amount of light. But, in case of wind velocity, it was considered that the further complement research is required as the sampling distribution was shown as so biased by the windproof effect of the trees planted at the outskirts of green space in case of wind velocity. Based on th analyzed results as above, it suggested the model of generating anion by implementing multiple regression analysis such as y = -2462.383 + 0.304x1 x 72.746x2 x 7.315x3 x - 0.138x4 (x1; time, x2; air temperature, x3; relative humidity, x4; luminous intensity, R2; 0.691).

        Background : Chrysanthemum indicum L. is used in medicine, for bee, ornamental as multivoltinism plant resources. The purpose of this study was to evaluate methods of rooting promotion to make high quality medicinal plants, thereby increasing farm income. This study investigated the effects of different cuttage period, extraction site, shading degree, nursery tray for promoting cutting slips rooting in Chrysanthemum indicum L. Methods and Results : It is used New variety "Gamkuk 1" that were forstered in Gyeongsangnam-do Agricultural Research & Extension Services. Method of application with cuttage period were conducted under five condition; Around the middle of April, Toward the end of April, Early in May, Around the middle of May, Toward the end of May. Early in May plant length was longer than other cuttage period as 41.2%, 83.7% and are significant at significance level 0.05. Method of application with extraction site were conducted under three condition; 1 - 3 node, 4 - 6 node, 7 - 9 node. Leaf number in 1 - 3 node was more than other extration site as 19.4%, 33.6%. Also root length in 1 - 3 was more than other extration site as 10.5%, 23.2%. Method of application with shading degree were conducted under three condition; 50%, 70%, 90%. Plant length in 50 - 70% was longer than 90% shading degree as 23.8%, 24.2%. Also shading degree 50 - 70% had many root length and root number. Method of application with nursery tray were conducted under five condition; 72, 105, 128, 162, 200 tray. Plant length in 105, 128 tray was longer as 8.5 - 35.3% than other nursery tray and are significant at significance level 0.05. Conclusion : According to the results, Early in May, extraction site 1 - 3 node, shading degree 50 - 70%, nusery tray 105, 128 showed the highest growth on cutting slips root promotion in Chrysanthemum indicum L.

        Background: Chrysanthemum indicum L. is used in medicine, for bee, ornamental as multivoltinism plant resources. Since ancient times, Chrysanthemum indicum L. had been widely used as drug and tea. From 2000, breeding began and the three varieties "Manhyang", "Geumhyang", Gamro" were developed so far. As demand has increased, growth traits of "Wonhyang" has been tested and fostered to report growth characteristics of "Wonhyang". Methods and Results: It is used New variety "Wonhyang" that were forstered in Gyeongsangnam-do Agricultural Research & Extension Services. After going through selection, proliferation and characteristics of CI0901, production performance test carried out in 2012~2013. As a result, it had named as "Gamkuk 4" in high quality, quantity and of strong in the disease. Local adaptability trial test carried out in 2014~2015. Conclusion: "Wonhyang" was a good growth in plant height, lengths of branches. Leaf color is light green, flower color is light yellow. Disease and pest resistance of "Wonhyang" is relatively better than "Manhyang". Since plant height is small and plant width is big, "Wonhyang" is available for medicine, food and landscaping. As a result of local adaptability trial test, Yield(131.1㎏/10a) of three region Hamyang,Hapcheon,Changnyeongis 10% higher than "Manhyang". Based on the results of this study, "Wonhyang" was to be registered for a new breed through the breed fostering council.

        Background : This study was conducted to developed the propagation method by cutting for mass cultivation of Vitex roundifolia. We were pitched the cutting two times and treated plant growth regulators to enhance the rooting percentage. Vitex roundifolia is live in beach sandy soil south of Hwanghae-Do and Gangwon-Do. Vitex roundifolia have been used to bath foam. It is good for aromatic plant. It has 0.8% essential oil content in leaf and flower. Major components of essential oil were alpha-Pinene, Sabinene, beta-Pinen, 1,8-cineole, d-Limone. Despite the superior usability, it had not yet been made by the artificial cultivation Methods and Results : We were pitched the cutting of a first-year branch on June 5, which was greenwood cutting and July 17, which was semi-hardwood cutting at Kwangseung-ri beach, Gochanggun, Jeonbuk. The length of cutting was 10cm. It had 3~4 nodes, we stuck a cutting remain 2 nodes above ground on ordinary raise seedling soil. Rooting percentage was measured at 60 days after stuck a cutting. Rooting percentage was higher greenwoody cutting(95%) than semi-hardwood cutting(57.6%). In green wood cutting, there was no significance with plant growth, but chemical injury was occurred in IBA 5,000ppm. In semi-hardwood cutting, there was significance with plant growth regulators. The rooting percentages of all the treat were higher than control(no treatment). Rooting percentage was the highest in NAA 5,000ppm treated. Conclusion : Greenwood cutting method was more proper to propagation for Vitex roundifolia than semi-hardwood cutting. The optimum time to cutting for Vitex roundifolia propagation was the early in June. If miss a time to propagation Rooting percent was elevated by plant growth regulator.

        Seed color is an important factor affecting physiological and developmental process in wheat. One of the plant pigments, anthocyanins are a group of flavonoid compounds well known as pigments responsible for blue, purple, red, or yellow coloration of plant tissues. In this study, we investigated the pigmentation of purple and yellow color seed according to wheat grain developmental stages. The contents of anthocyanin and chlorophyll in the purple and yellow seeds were measured. Chlorophyll contents were changed similarly in both purple and yellow color seed, and no significant difference was observed between them. In purple color seed, the content of anthocyanin was significantly induced compared with yellow color seed. The individual anthocyanin components were investigated by ultra performance liquid chromatography (UPLC). Cyanidine-3-glucoside (C3G) and peonidine-3-glucoside (P3G) were detected as predominant anthocyanin in purple color wheat. To investigate whether structural genes in anthocyanin biosynthesis were involved in the trait differences between purple and yellow color seed, we examined the expression of anthocyanin biosynthesis-related genes (CHS, CHI, F3H, DFR, ANS, UFGT) and MYB transcription factor in developing wheat grains by using qRT-PCR. This study indicates that the expression of anthocyanin biosynthesis-related genes and MYB transcription factors correlate with anthocyanin levels of grain.

        Citrus canker caused by Xanthomonas citri is a notorious disease affecting a decrease in fruit productivity and quality. Citrus export to USA is also prohibited by the disease. Therefore, development of citrus canker resistant variety is essential and exploitation of markers for molecular breeding is urgent. To develop DNA molecular markers, we performed whole genome resequencing for 8 varieties: 4 citrus canker resistant varieties including C. hybrid ‘Kioymi’ and 4 citrus canker susceptible varieties including C. iyo ‘Miyauchiiyokan’. In total, 642 polymorphic SNPs were detected between resistant and susceptible varieties. Of the 642 SNPs, 50 SNPs were preferably selected based on integrative genomics viewer. To apply the markers in a broad range of citrus variety, we performed genotyping with 6 other varieties very well known as citrus canker resistant and susceptible varieties in addition to previous mentioned 8 varieties. Three of the 50 SNPs were identified as a marker to distinguish citrus canker resistant varieties from susceptible varieties. Secondly, we developed molecular markers to apply for F1 lines crossed by ‘Kiyomi’ and ‘Miyauchiiyokan’. Of the 50 SNPs, we identified 2 SNP markers to distinguish between F1 resistant and susceptible lines. One of them is a resistance gene that plays a role in plant defense mechanism. In this study, we developed 5 molecular marker candidates possible to apply for molecular breeding to develop citrus canker resistant variety. We are working on development of candidate markers related to citrus canker.

        The goal of marker-assisted backcrossing is to reduce the number of generations significantly by using genome-based molecular markers. Among other types of molecular markers, SNP (single nucleotide polymorphism) is mostly used in genetic diversity analysis due to its abundance. To develop high-throughput SNP marker for MAB system, we selected 20 Chinese cabbage lines each represent traits as inner leaf color, disease resistance, head type and maturity etc. Then, we sequenced the transcriptomes of 20 lines by using Illumina Hiseq2000. The average transcriptome size was 1.37 Gbase, and the average of short reads mapping rate was about 62.15% (30xcoverage). We identified 13,976 SSR markers and 380,198 SNPs by aligning contigs of 20 Chinese cabbage lines. To develop SNP marker set, we chose 409 SNPs that covers the whole Brassica rapa transcriptome. The filtering criteria were depth, polymorphism, segregation ratio, lack of adjacent SNP and copy number. We positioned the selected SNP markers to the Chinese cabbage linkage map. Clustering dendrogram was produced using SNP marker and three different clusters were generated. The result showed that the genotyping data is partially linked to the phenotyping data. We assume that the developed SNP marker set can be applied in the Chinese cabbage MAB system soon.

        This study was performed to investigate the growth characteristics and inorganic components of Codonopsis lanceolata regarding regional differences. The plant height of Japanese Codonopsis lanceolata was 373.6 cm, so it’s revealed that it has more vigorous growth than Korean won. The flowering time of Korean Codonopsis lanceolata was 2 weeks faster than Japanese one. Total fresh weight of root was 41.0 g and 39.0 g for Korean and Japanese respectively, thus, no significance difference was found. However, regarding fresh weight, Korean one had a more fresh weight (35.4 g) of main root parts, but Japanese one had a more fresh weight (9.6 g) of the lateral root part. Each inorganic component was found more in the aboveground parts, regardless of the region and the content of K was the largest. Regarding the content of macroelements for each part of Codonopsis lanceolata, the content of Na, Mg, P, S, and Ca in Korean Codonopsis lanceolata was found the highest on the leaf, followed by stem and root. In the case of Japanese Codonopsis lanceolata, same result was found on the content of Mg and Ca, however, the highest content of Na and P was found in the stem.

        This study was conducted to compare the growth, inorganic components, and proximate components of Codonopsis lanceolata grown in 10 regions of Korea for selecting superior species and breeding by crossing. Among the all tested lines, the shortest plant height (217.12 cm) was observed from the Ulleungdo region line (No. 4) while the longest (273.9 cm) was observed from Hwasun region line (No. 9). In addition, the lines of central and northern region (No. 1~No. 7) tend to have shorter plant height than those of southern region (No. 8~No. 9) except Jejudo region line (No. 10). Flowering tends to be late towards southern region, and lines in central and northern regions were started flowering about 2 weeks earlier than those in southern regions. However, the heaviest root weight was 13.1 g, found in only Jejudo line (No. 10) whereas there was no significant difference found in the other regions which have a range of 8.3~11.0 g. The inorganic components were varied in each line, however, proportion of macroelements, such as K, Ca, and P, was the largest for every line. Especially for Heongseong region line (No. 2), had larger proportion of macroelements than the others. There was a difference of proximate compositions of Codonopsis lanceolata, except the moisture content, among all regions, however, it was generally shown that the content of crude protein (1.31~3.76%) and crude fiber (2.18~3.12%) was the highest.

        Gamma irradiation has been used as a tool for plant mutation breeding to select new cultivar with improved characteristics. Generally, the irradiation of seeds with high doses of gamma rays disturbs the synthesis of protein, hormone balance, and enzyme activity. And also, high dose of gamma rays to reduce plant height, number of tiller, and root length, although the effect of gamma-irradiated plants may depend on the species and cultivar or stress conditions. Biological effects of radiations can be divided into two types according to dose range and periods of exposure. Acute irradiation represents exposure to high-dose of irradiation over short period time, whereas the chronic irradiation is comprised of exposure to low doses of radiation over extended period of time. To compare the effects of acute and chronic exposure to ionizing radiation on two wheat cultivars (K4191 and Geumgangmil), we measured their germination rate, seedling height, and root length. In order to understand the influence of antioxidant-related genes and DNA repair-related genes, we used qRT-PCR methods to identify their expression levels. To study the behavior of a radiation-induced free radical, gamma-irradiated seeds were used for ESR spectroscopy. Plant growth pattern was showed positive correlation with ESR results. This study indicates that low level chronic radiation exposure is even more serious effects than short doses of high level radiation according to different wheat cultivar.

        Citrus canker caused by Xanthomonas citri pv. citri is one of economically important diseases in the citrus industry. The devastating bacterial disease results in unattractive quality and a significant reduction in fruit production. Citrus growers and industry in Korea has been struggling with the serious disease causing the prohibition of export market. Korea also became the top import market for oranges. The development of markers linked to citrus canker resistance is strongly needed. In this study, we investigated molecular markers between ‘Kiyomi’ (Citrus unshiu x C. sinensis), a resistant cultivar, and Natsudaidai (C. natsudaidai), a susceptible cultivar. To develop markers, we focused on structural variation (copy number variation, CNV, and presence/absence variation, PAV). It has been well documented that CNV and PAV of defense-related genes are associated with resistant cultivars. Using a read depth approach following next-generation sequencing, we performed genome-wide analysis of CNV and PAV in two varieties. As a result, 633 genes showing at least two times difference between the mapping reads from two varieties and 61 genes showing presence of the mapping reads in only either one of them were screened. Visual inspection using the Integrative Genomics Viewer (IGV) was performed and experimental validation is being investigated. Interestingly, one of PAV candidates showed polymorphism in ‘Kiyomi’ and ‘Natsudaidai’ as well as other resistant and susceptible cultivars. Our results suggest a necessity for the detection of structural variation and indicate that the candidates may be useful for molecular breeding for citrus canker resistance and understanding disease resistance mechanism.

        Watermelon (Citrullus lanatus) is one of the most economically important cucurbitaceous crop over the world. Screening of proper reference genes was needed to reverse transcription quantitative real-time PCR (qRT-PCR), and it is bausic step of many researches including gene expression analysis. However, the reference genes on watermelon has not yet been reported systematically. Therefore, eight candidates of reference genes were selected with reference to Arabidopsis or cucumber papers. They are β-Actin, elongation factor 1-α, glyceraldehy-3-phosphate-dehydrogenase, NADP-isocitrate dehydrogenase, leunig, polypyrimidine tract-binding protein1, ubiquitin-conjugating eznyme E2, and 18S ribosomal RNA. The expression levels of genes were evaluated by qRT-PCR under biotic stress (Colletotrichum orbiculare treatment), plant hormone treatment (100 μM ABA), and abiotic stresses such as drought, cold (4℃), salt (250 mM NaCl) stresses. We founded appropriate reference genes which did not induce or reduce gene expression levels under broad spectrum of stresses by qRT-PCR analysis. These results may provide proper information for the use of appropriate reference genes for gene expression studies in watermelon qRT-PCR analysis.

        Chronic gamma irradiation can be used an alternative mutation breeding methods for induction of many useful mutants. Seedlings of purple-colored wheat plants were irradiated with wide range doses of chronic gamma-rays (20, 25, 30, 40, 50, 70, 100, 125, 150, 200, 250, 300 Gy) during 6 weeks at gamma-phytotron in the Korea Atomic Energy Research Institute, respectively. To identify the biological responses purple-colored wheat, we examined the plant height, chlorophyll, carotenoid and total anthocyanin contents in leaf. Plant growth, chlorophyll and carotenoid contents in leaf were decreased when the dose rate increased. Anthocyanin contents were increased with the increase of the radiation dose until 50 Gy treatment. To confirm the real contents of anthocyanin, we also investigated cyanidin-3-glucoside in purple-colored wheat leaf by using UPLC analysis. These results indicate that anthocyanin accumuation was observed under chronic gamma irradiation.

        Citrus is one of the major fruits produced in Korea. There are about 20 species mainly grown in Jeju Island, Korea. Four representative species, which are quite different in the shape of leaf and the taste of fruit, were selected and were used to profile the transcriptomes. These species are ‘Miyagawa Wase’ (C. unshiu Marcov.) satsuma mandarin, ‘Kiyomi’ (C. unshiu Marcov. × C. sinensis) mandarin hybrid, ‘Dangyuja’ (C. grandis) and ‘Natsudaidai’ (C. natsudaidai). Classification of the up-regulated and down-regulated genes using the Cluster of Orthologous Groups of proteins (COG) database reveals that the number of genes included in each group differed significantly among the four species. Several genes that showed significant differences in expression on the microarray were selected and their expression patterns were examined by reverse transcription- ploymerase chain reaction. Metabolic genes such as tyrosine decarboxylase and β-glucosidase ligase were found to be highly expressed in Miyagawa Wase, relative to other species. On the other hand, the expression level of mannose phosphate isomerase was lower in Miyagawa Wase. An efflux pump gene was found to be up-regulated in Kiyomi, whereas cinnamyl-alcohol dehydrogenase was down-regulated. β-carotene 15,15’-dioxygenase, which is involved in the vitamin metabolism, was up-regulated in Natsudaidai. Interspecific differentiations of gene expression are analyzed in terms of the metabolic pathways and their possible roles in citrus species.
