        고무화합물 형태로 구성된 조영제의 병에 Syringe Connector의 Spike를 연결 시 고무의 찢김 정도를 알아보고 찢김 및 분쇄로 인한 합성고무의 혼입 유무와 분쇄된 합성고무가 검출 시 분쇄물의 크기를 실험을 통해 알아보고자 하였다. 그 결과 찢김 정도의 경우 Syringe Connector의 끝과 최초 접촉하는 앞면이 약 3.14±0.04 ㎜로 뒷면 보다 많이 찢겼으며, 실험 대상인 10 병의 조영제에서 평균 7 개에서 15 개로 모두 분쇄물이 검출되었다. 검출된 분쇄물을 이용하여 크기를 측정한 결과 평균크기는 약 7.89±0.31 ㎛이었다. 향 후 다양한 실험 및 분석방법을 통한 추가실험과 더불어 흡인된 분쇄물 차단을 위한 미세 필터타입 자동주입장치의 개발이 필요하며, 분쇄물 유입 시 치명적 사고를 대비하여 관련기관의 관심 또한 필요할 것으로 사료된다.

        본 연구는 완주에서 재배된 세계 주요 11개 적포도 품종으로 제조된 적포도주의 향기 성분을 headspace-solid phase microextraction 분석법으로 확인하였다. 향기성분은 총 75종이 확인되었다. 아로마화합물은 그들의 OAV 값에 의해 5 그룹으로 나뉘었다. 알콜, 알데하이드, 에스테르, C6 화합물이 11개 적포도주의 주요한 향기성분이었다. Isoamy alcohol 알콜과 phenylethyl 알콜은 11개 포도주에서 공통적으로 꽃향기, 달콤한 향을 나타내는데 중요한 물질이었다. Octanoic acid, ethyl ester, hexanoic acid ethylester은 모든 레드와인에서 과실향과 꽃향, 달콤한 향을 내는 중요한 성분이었다. 1-Hexanol은 모든 포도주에서 분석되었으나 풀향을 나타내는 향으로 나타났다. Chanceller, Malbec, marchel, Nsrsha, Pinot Meunier, Sangiovetto 포도주의 주요 향기성분은 과실 향인 것으로 나타났으며 Cabernet Franc, Cabernet Sauvignon, Sauvignon Vert 포도주의 주요 향기성분은 풀향인 것으로 조사되었다. 또한, MBA와 Narsha 포도주의 경우 꽃향이 주요 향인 것으로 조사되었다. 본 연구 결과를 바탕으로 적포도주용 품종을 육성할 때 선발기준으로 향기성분 분석을 활용할 수 있을 것으로 판단되었다.

        The study compared the juice and wine odorants of the Cheongsoo grape cultivar with those of Chardonnay and Reisling wines. The volatile compounds of the three grape varieties were analyzed using headspace solid-phase microextraction coupled with gas chromatography-mass spectrometry. The most common volatile compounds in the juices from the three cultivars were terpenes, C13-norisoprenoids, etones, alcohols, and aldehydes. Terpenes were established as the most abundant group of volatiles in Cheongsoo grape juice, where as aldehydes predominated in Chardonnay and Riesling juices. Forty-two volatile compounds (acids, alcohols, esters, and others) were detected in the three white wines. The concentration of esters was about four times higher in Cheongsoo wine than in Chardonnay and Riesling wines. Five esters found in the Cheongsoo wine, namely, isoamyl acetate, ethyl butanoate, ethyl hexanoate, ethyl octanoate, and ethyl decanoate, exhibited high odor activity values (OAV) of >1. Furthermore, only Cheongsoo wine had a high OAV for isoamyl acetate odorant, which is associated with banana and sweet aromas. Therefore, the abundant and varied esters are believed to be key volatile fruity/sweet odorants in Cheongsoo wine.

        4-Nonylphenol (NP) is a surfactant that is a well-known and widespread estrogenic endocrine disrupting chemical (EDC). Although it has been known that the affinity of NP to ERs is low, it has been suggested that low-dose NP has toxicity. In the present study, the endocrine disrupting effects on reproduction, and the weight of gonads, epididymis, and uterus were evaluated with the chronic lower-dose NP exposing. This study was designed by following the OECD test guideline 443 and subjected to a complete necropsy. In male, NP had an effect on the weight of the testis and epididymis in both F0 and F1. In females, NP decreased the weight of ovary and uterus in F0 but not in pre-pubertal F1 pubs. Fertility of male and female in F0 or F1 was no related with NP administration. The number of caudal-epididymal sperm by body weight (BW) was not different between groups in both F0 and F1. Besides, the difference of the sperm number between generations was not detected. The number of ovulated oocytes was similar between groups in F0, but significantly decreased in NP 50 group of F1. The litter size and sex ratios of offspring in F1 and F2 were not different. The accumulated mating rate and gestation period were not affected by the NP administration. Those results shows that chronic lower-dose NP administration has an effect of endocrine disruptor on the weight of gonads and epididymis of F0 and F1 but not in reproduction. Based on the results, it is suggested that chronic lower-dose NP exposing causes endocrine disruption in the weight of gonad and epididymis but not in the reproductive ability of next generations.

        Background : Undulatum Rhubarb, commonly produced in domestic, is rhizome of Rheum undulatum L. that belongs to the family Polygonaceae. It also can be used as a substitute of R. palmatum L., R. tanguticum Maximowicz ex Balf., and R. officinale Baillon which completely depend on import system. However, there should be clear clarification among Undulatum Rhubarb and Rhubarb, because Undulatum Rhubarb contains rhaponticin as marker compound that is not indicated at Rhubarb. Some of the recently imported Undulatum Rhubarbs have been found to be Rhubarb. Also, it is known that only Undulatum Rhubarb is cultivated at domestic environment. But some plant origins of Rhubarb are grown in Korea, too. Further study are needed to clarify clear origin between Undulatum Rhubarb and Rhubarb. Thus, we collected some domestically cultivated samples and identified them. Methods and Reseults : Rheum undulatum L., Rhubarb, Rheum tanguticum Maximowicz ex Balf. which were cultivated in Gangwondo Agricultural Research and Extension Services in Cheorwon were collected and anayzed the DNA sequences. We also compared DNA sequences in Rhubarb collected from England and R. rhabarbarum L., R. undulatum L., and R. franzenbachii on NCBI. As a result, two kinds of rhubarb cultivated in the test plantation were identified as R. rhabarbarum and R. officinale. In addition, R. undulatum (plant origin of Undulatum Rhubarb) was identified as Rhubarb (Rheum rhabarbarum) in England with 99 - 100% identical in nuclear ITS gene region. Conclusion : R. undulatum, plant origin of Undulatum Rhubarb, is reported as synonym of R. rhabarbarum, R. franzenbachii. Rheum speices which are cultivated as tester in Gangwondo Agricultural Research and Extension Services in Cheorwon are estimated as R. undulatum and R. officinale. Therefore, not only Undulatum Rhubarb but Rhubarb could be grown in Korea.

        Background : Five medicinal herbs have been selected from the preliminary screening for in vitro anti-allergy activity (in RBL-2H3 cells). The present study is conducted to investigate the inhibitory effects of the medicinal herbs on allergic inflammation in other kind of cells. Methods and Results : Cells treated with five extracts prepared from Betula costata Trautv. (aerial part), Camellia sinensis L. (aerial part), Polygonatum stenophyllum Maxim. (root), Pyrus pyrifolia var. culta (Makino) Nakai (leaf), and Rehmannia glutinosa (Gaertn.) Libosch. ex Steud. (root) were measured for mRNA levels of TNF-α on HaCaT keratinocytes stimulated by TNF-α /INF-γ and for mRNA levels of IL-2 in Jurkat T cells mediated by PMA/A23187. Pre-treatment with the five extracts reduced the mRNA levels of TNF-α in HaCaT cells and mRNA levels of IL-2 in Jurkat T cells. In particular, the leaf extract of Pyrus pyrifolia var. culta (Makino) Nakai significantly and dose-dependently decreased the mRNA levels of TNF-α and IL-2. To determine the toxicity of the extracts from the selected medicinal herbs to HaCaT cells and Jurkat T cells, the viabilities of the cells treated with several concentrations of the five extracts were measured by MTT assay. Extracts of Polygonatum stenophyllum Maxim. (root), Pyrus pyrifolia var. culta (Makino) Nakai (leaf), and Rehmannia glutinosa (Gaertn.) Libosch. ex Steud. (root) (up to 250 ㎍/㎖) did not show cytotoxic effects on HaCaT cells and Jurkat T cells. On the other hand, 250 ㎍/㎖ of extracts of Betula costata Trautv. (aerial part) and Camellia sinensis L. (aerial part) reduced cell viability in both cells. Conclusion : These results demonstrate that the leaf extract of Pyrus pyrifolia var. culta (Makino) Nakai has an anti-inflammatory effect by inhibiting pro-inflammatory cytokine expression. Therefore, the leaf of Pyrus pyrifolia var. culta (Makino) Nakai can be a useful resource for the development of anti-allergy/anti-inflammatory materials.

        Background : Perilla frutescens L. is valuable as a medicinal plant as well as a natural medicine and functional food. Limonene perilla collected from various places showed 60% limonene compounds. However biological activity of these accession has not been reported before. Therefore, this study was conducted to investigate the biological activity of limonene perilla. Methods and Results : Fractional solvent extracts were obtained by using organic solvents such as n-hexane, chloroform, ethyl acetate, n-BuOH, and aqueous solvent from different parts of limonene perilla extracted initially in 70% EtOH. We investigated the effects of limonene perilla on total phenol and flavonoid contents, FRAP (Ferric Reducing Antioxidant Power), total saponin contents and tyrosinase inhibition activity. Leaves of limonene perilla produced the highest total phenolic contents (29.88 mg·CAE/g), flavonoid (8.39 mg·QE/g) and saponin contents (47.77 mg·GIE/g) than stems and roots of limonene perilla. FRAP of leaves was 823.00±3.58 μM·FeSO4·E/mg. Tyrosinase inhibition activity rate was 40.31% in 70% ethanol extracts from leaves of limonene perilla. Conclusion : This results suggest that leaf of limonene perilla fractions has significant antioxidant activity. Also, limonene perilla could be used as a functional biomaterial in developing cosmetics and functional foods.

        Background : Angelica dahurica Bentham et Hooker, Angelica gigas Nakai, Ostericum koreanum Maximowicz and Peucedanum japonicum Thumberg are a major medicinal plant in north Geungbuk province. Using medicinal plants are impotant it`s ingredient. Dry condition and stroage method are not standard manual. The ingredient variation of dry condition and stroage method were not researched. Methods and Results : Using plant material were cutivated on Gyongsangbukdo Bonghwa area. It were studied ingredient variation after dry and storage condition by HPLC methods. Major ingredient of Angelica gigas Nakai are decurusin, decurusinangelate. Heated air bulk dry get more decursin than natuarl dry and decurusinangelate of natural bulk dry was higher than heated air bulk dry. Major ingredient of Ostericum koreanum Maximowicz are imperatorin and isoimperatorin.. Imperatorin of Ostericum koreanum was highest peak on 50℃ heated-air dry after plastic bag sorage and isoimperatorin was highest peak on 40℃ heated-air dry after mountain cultivation. Imperatorin is a major ingredient Angelica dahurica Bentham et Hooker. Heated air bulk dry get more decursin and decursinangelate than natuarl dry and small heated-air dry. Peucedanol-7o_glucoside is a major ingredient Peucedanum japonicum Thumberg. Natural bulk dry get more peucedanol-7o_glucoside than heated-air bulk dry. Conclusion : Ingredient of Angelica dahurica Bentham et Hooker, Angelica gigas Nakai, Ostericum koreanum Maximowicz are different under various cutivation, drying method, storage. Diffent Ingedients of Angelica gigas Nakai, Ostericum koreanum Maximowicz were not accord it’s optical conditon.

        Background : Plants are the rich source of antioxidants, which plays a very important role in maintaining human health. Their antioxidant property protects cells of different organs of human beings against free radicals and free radical mediated diseases. Even though, there is lack of knowledge on the antioxidant effect of lutein present in plants. In the present study, lutein was isolated from the GreenTea leaves (Camellia sinensis) which is used as a dietary source. Methods and Results : The procedure adopted for the isolation and purification of lutein using acetone extraction and preparative high performance liquid chromatography (HPLC) is simple and less time consuming. Free radicals scavenging activity of isolated lutein from acetone extract of GreenTea was assessed by DPPH radical scavenging assay and reducing power. The isolated lutein scavenged 79% of DPPH radicals at 20 ㎍/㎖ and two fold lower concentration compared to the standard antioxidants (α-tocopherol). No significant differences were found between the reducing power of the lutein and BHT when their concentrations were high. However, significant differences were observed at relatively low concentrations, the reducing power of lutein was isolated from the GreenTea leaves was stronger than those of their acetone extract and standard antioxidants (BHA). Both electron spin resonance (ESR) and in vitro assay confirmed that lutein was isolated from the GreenTea leaves, exhibited a greater capacity for scavenging superoxide (O2 •-) and hydroxyl (OH •) radicals than standard antioxidants β-carotene and α-tocopherol respectively. Conclusion : The results proven that lutein isolated from GreenTea leaves has an efficient antioxidant ability, it could serve as an antioxidant to scavenge reactive oxygen species.

        Background : Lutein, a xanthophyll, consists of chains with 8 conjugated double bounds containing closed rings on each end of the chain. This carotenoid is found in fruits and vegetables, especially dark green leafy vegetables such as green tea. In this study, we investigated the anticancer effects of purified lutein from green tea on human cancer cell lines containing prostate carcinoma cancer cells (LNCaP). Methods and Results : Prostate carcinoma cancer cells (LNCaP) were cultured and evaluated the inhibitory effect of lutein isolated from green tea compared other carotenoids (β-carotene and lycopene) on cell proliferation. Cyclin D1 and PCNA were evaluated as cell differentiation. In results, PCNA/cyclin regulates the initiation of cell proliferation by mediating DNA polymerase. Under cultural conditions, lycopene remarkably suppressed the PCNA expression prostate cancer cell line LNCaP in higher doses (20 μM - 100 μM) statistically. However, β-carotene and lutein presented the less inhibitory effects on PCNA expression. Determination of PCNA expression in control and treated cells demonstrates that lycopene did affect proliferation in LNCaP cancer cells in dose-dependent manner. However, β-carotene and lutein suppressed the cyclin D1 expression in dose-dependent manner but no in lycopene group. These results indicate that differ carotenoids presented the various suppressive ability of PCNA and cyclin D1 expression in cell proliferation. Conclusion : In conclusion, lutein suppressed the carcinogenesis of induced prostate cancer cell line by acting as a suppressor for inhibiting the expression of cyclin D.

        Background : Schizandra chinensis Baillon have five tastes and lately it is using a beverage broadly. Schizandra chinensis is one of the top producing medicinal plant in Korea. Mungyeong of Gyongbuk province produce almost of Schizandra chinensis. Maturity of Schizandra chinensis get 3 years and proliferation of Schizandra chinensis was not a manual. It is needed that a new cultivar has a big fruit and high quality chracteristics using processed food and beverage. Methods and Results : 105 lines of Schizandra chinensis were collected on Mungyeong, Yeongwol, Jinan. It were studied it’ characteristics especially it’s fruit trait. Fruit traits of Schizandra chinensis were researched on fruit length, fruit weight, maturity, number of fruit, male and female ratio, powdery mildew. Fruit length of Schizandra chinensis is relation of fruit weight. It were founded 15 lines of long fruit length. 5 lines were studied high fruit weight and it’s weight were 32 to 41g. Number of fruit has relation with fruit weight and high fruit weight gets many fruits. it’s numer of fruits were 3 to 41. Male and female ratio were very impotant characteristic. High level of female ratio has quantity of fruit. High level of female ratio were founded 2 lines. Finally It was selected 3 good breed lines of Schizandra chinensis. Conclusion : 105 lines of Schizandra chinensis Baillons were collected on Mungyeong, Yeongwol, Jinan. It were founded 15 lines of long fruit length and 5 lines were studied high fruit weight. High level of female ratio were founded 2 lines. 3 good breed lines of Schizandra chinensis were selected.

        Background : Despite the presence of various bioactive compounds in ginseng, there is lack of study about the phenolic metabolites in ginseng especially depending on the cultivation soil and the fertilizer types. Therefore, this study aims to develop an (-)ESI-LC-MS/MS analytical method for the measurement of selected phenolic compounds in the ginseng root. Methods and Results : Total phenol content in ginseng root was measured with the Folin-Ciocalteau method using UV/Vis spectrophotometer. Then, the 56 selected phenolic metabolites in ginseng root were measured with the (-)ESI-LC-MS/MS. The brief LC-MS/MS analytical conditions were as follows; Thermo Scientific Syncronis C18 HPLC Column (250 × 4.6 mm, 5 μm) was used. Optimized instrument settings were as follows: Curtain gas 20 psi, collision gas 2 psi, ion spray voltage –4500 V, nebulizer gas 40 psi, heating gas 70 psi, and its temperature 350℃. Total phenol content was higher in the ginseng cultivated in the paddy-converted field than that in upland. In particular, the total phenol content was about 6% decreased in the ginseng root cultivated with the food waste fertilizer compared to the control (p < 0.05). Six phenolic constituents including caffeic, chlorogenic, p-coumaric, ferulic, gentisic, and salicylic acids were found in the ginseng root by using the LC-MS/MS in MRM (multiple reaction monitoring) Mode. These six phenolic compounds occupied approximately 20% of the total phenol content measured in the corresponding ginseng root. The chlorogenic acid was the most abundant phenolic metabolite found in the ginseng root, accounting for ≥ 95% of the sum of six phenolic compounds, in this study. Conclusion : This preliminary study can be useful for the study on content and composition of phenolic metabolites in ginseng root with the aspect of metabolomics. We plan to further optimize the LC-MS/MS analytical method and then provide the extended understanding on the phenolic metabolism in the ginseng root with respect to the ginseng cultivation conditions.

        Background : Management of air temperature are known to primarily affecting on physiological properties and yield in plant. Methods and Results : The effect of air temperature on characteristics of photosynthesis and chlorophyll fluorescence in Schisandra chinensis Baillon were investigated under controlled temperature using growth chamber. Net photosyntheis rate, transpiration was measured at 1,000 μmol m-2 s-1 of photon flux density and chlorophyll fluorescence was analyzed by OJIP method. Net photosyntheis rate and transpiration rate was higher in treatment of 25℃. As results of chlorophyll fluorescence by OJIP analysis, maximum quantum yield (Fv/Fm) of photosystem II (PSII) and PIabs was higher in treatment of 25℃ which reflects the relative reduction state of PSII. But in treatment of 35℃ the relative activities per reaction center such as ABS/RC, DIo/RC were higher than in treatment of 25℃ which implied that the relative reduction of electron transport at PSI and increasement of photo inhibition at reaction center. Conclusion : This result implies that 25℃ of air temperature may be a adequate temperature to improving the efficiency of photosynthesis through controlling a photosystem in Schisandra chinensis Baillon.

        With the object of examining the effects of horticultural therapy based on structural recall by developmental stage on cognitive, emotional, and hand functions of institutionalized elderly, this study executed a horticultural therapy program once a week and through a total of 14 sessions during the period from May 2 to December 5, 2013. The subjects of this study were 34 elders admitted to S Geriatric hospital for geriatric diseases, and they were divided into a control group and an experimental group. Because the program was designed for institutionalized elders who were able to do only indoor activities, it provided various seasonal horticultural activities that could be done even in a limited space by giving materials and subjects that could be recalled by developmental stage. The results of this study are as follows. As to changes in cognitive functions (MSQ) after the horticultural therapy program, the control group showed a decrease as having more negative ranks, and the experimental group showed an increase as having more positive ranks, but such changes in the control and experimental groups were not significant. As to changes in emotional functions after the horticultural therapy program, the control group showed a decrease but without significance in ego integrity as having more negative ranks, and the experimental group showed an increase with significance as having more positive ranks (P=0.020). As to the experimental group’s changes in the sub factors of ego integrity, positive ranks increased in all the sub-factors and the increase was particularly significant in satisfaction with present life (P=0.038). As to changes in life satisfaction, the control group showed increased positive ranks but without significance, and the experimental group showed significantly increased positive ranks (P=0.022). As to the experimental group’s changes in the sub-factors of life satisfaction, changes in denial of the past (P=0.029) and denial of the future (P=0.016) were particularly significant. As to changes in hand functions, both the control and experimental groups showed some improvement in pinch after the horticultural therapy program but the changes were not significant. Grip strength did not change in the control group, and in the experimental group right-hand grip strength was improved slightly more than left-hand one, the difference was not significant. The results of this study suggest that horticultural therapy based on structural recall by developmental stage is effective in improving institutionalized elders’ emotional functions and, consequently, can contribute to higher quality of life, which is the ultimate goal of elders.

        The GA application on grapevines induces parthenocarpy, fruit set without fertilization, and the inhibition of pollen tube growth. But the molecular mechanism underlying this inhibition is not understood. Similar defective pollen tube growth within the transmitting tract has been reported in the mutant of GABA transaminase (GABA-T), referred to as pollen-pistil-interaction2 (pop2) in Arabidopsis. In spite of the similarity of pollen tube growth inhibition observed in GA-applied grapevines with that of pop2, only the effects of GABA on stress responses in grapevines have been reported. In present study, transcriptional changes of Vitis GABA metabolic genes, together with changes in GABA levels with or without GA application were analyzed to define how GA application restrained the pollen tube growth in grapevines. A GA solution (Dongbu, Seoul, Korea) at 100 ppm was onto inflorescence clusters 14 days before full bloom (DBF) and clusters were harvested at 0, 1, 2, 4, 7, 9, 12, 14, 16, and 19 days after GA application. Harvested inflorescence samples were immediately frozen in LN2 and extracted RNA and amino acid. The GABA contents were analyzed using high-performance liquid chromatography (Agilent 1100 HPLC, Agilent Technologies, Inc., Santa Clara, USA) equipped with a C18 column (4.6 mm×150 mm, 3.5 μm/VDS optilab, Berlin, Germany), according to the manufacturer’s instructions. Without GA application, the simultaneous high expressions of VvGAD1, VvGAD4 and VvGABA-T2 during 10 to 5 days before full bloom (DBF) showing the activation of GABA metabolism. But the contents of GABA were low before 2 DBF, and it peaked only at near full bloom when expression levels of VvGABA-T2 remained low. After GA application, the contents of GABA were constant during 10 to 5 DBF, although transcription levels of both VvGAD1 and VvGABA-T2 rapidly declined less than 30% of the levels observed without GA application. However, the GABA levels increased more than 2-fold only at near full bloom, compared to those without GA application, and at that time, expression levels of VvGAD1 up-regulated more than 3-fold and those of VvGABA-T2 kept low. But other amino acid contents did not show significant changes. In case of VvSSAHDs, their transcriptional changes with or without GA application were not correlated with GABA levels. These results indicates that GABA levels before pollination is tightly regulated, but GA application alters the GABA-shunt to accumulate excess GABA more than needed for proper pollen tube growth at full bloom. Gibberellin application alters the GABA-shunt to accumulate excess GABA resulting in inhibition pollen tube growth in grapevines.

        The patent ductus arteriosus (PDA) is a vascular structure connecting the proximal descending aorta to the roof of the main pulmonary artery, near the origin of the left branch pulmonary artery. Transcatheter closure has become the treatment of choice for most cases of PDA in both children and adults; however, measurement of the exact size and morphology of the shunt in adult cases using only contrast fluoroscopy is difficult. We report on a case of a 49-year-old woman who underwent transcatheter closure of PDA with intravascular ultrasound (IVUS) guideance.         The tight regulators of fruit set initiation, gibberellin (GA) and auxin, have been applied for decades to induce parthenocarpy, fruit set without fertilization. The integration of GA and auxin signaling mediated by either GA or auxin application during parthenocarpy has been actively reported in tomato, and recently we reported that GA application at pre-bloom also activating auxin signaling and down-regulated negative regulators of fruit set initiation in grapevines. However, the activation of auxin signaling upon GA application without up-regulation of auxin biosynthesis is still unclear. In this study, expression patterns of three auxin efflux transporter genes, VvPIN1a, VvPIN2 and VvPIN4, were monitored during inflorescence development in ‘Tamnara’ grapevines with or without GA application. Without GA application, transcription levels of VvPIN1a and VvPIN4 gradually increased from 14 days before full bloom (DBF) to 2 and 5 days after full bloom (DAF), respectively, except down-regulation of VvPIN1a during 5 DBF to full bloom. However, VvPIN2 expression declined steadily after peaking at 10 DBF. With GA application, VvPIN1a did not show significantly different expression patterns when compared to no GA application, with the exception of 4-fold up-regulation at full bloom, but transcription of VvPIN4 was reduced between 5 and 2 DBF. In addition, VvPIN2 was down-regulated between 12 and 10 DBF by more than 50% compared to levels in the absence of GA application. These reductions of both VvPIN2 and VvPIN4 with GA application prior to pollination suggest that GA application might regulate auxin distribution, instead of auxin biosynthesis, to activating auxin signaling during parthenocarpic fruit initiation.

        Gibberellic acid (GA) is a well-characterized plant hormone, which plays a critical role in various plant growth and development. including stem elongation, floral indcution and seed development. GA is known to cause enlargement of ripening fruits and, especially in grapevines, GA shows a unique function: the induction of seedlessness in seeded grape varieties. However, despite extensive previous studies about GA, there has been no clear verification of the mechanism that induces seedlessness in grapes. To understand how GA treatment results in artificial parthenocarpy of seeded grapes at molecular levels, we analyzed transcriptional changes in seeded grapes with and without GA application in various inflorescence developmental stages using RNA-seq. At 14 days before flowering (DBF), seeded grapes were treated with 100 ppm GA and clusters were collected at three developmental stages: 7 DBF, full bloom, and 5 days after flowering (DAF). Of a total of 28,974 genes that were mapped to grape genome reference sequences, 7,013 and 9,064 genes were up- and down-regulated, respectively, in the GA-treated grape as compared to the non-GA-treated control at 7 DBF, full bloom, and 5 DAF. Clustering analysis revealed that these genes could be grouped into 9 clusters with different expression patterns. We also carried out functional annotation based on gene ontology categories. There were significant differences in the expression of the GA and auxin-related gene families. These findings expand our understanding of the complex molecular and cellular mechanisms of GA-induced parthenocarpy of grapes and provide a foundation for future studies on seed development in grapevines.
