The recent development in genetic assisted selection (combining traditional- and genome assisted selection method) and reproduction technologies will allow multiplying elite cow in Hanwoo small farm. This review describes the new context and corresponding needs for genome assisted selection schemes and how reproductive technologies can be incorporated to get more genetic gain for cow genetic improvement in Hanwoo. New improved massive phenotypes and pedigree information are being generated from commercial farm sector and these are allowing to do genetic evaluation using BLUP to get elite cows in Korea. Moreover cattle genome information can now be incorporated into breeding program. In this context, this review will discuss about combining the reproductive techniques (Multiple Ovulation Embryo Transfer; MOET) and genome assisted selection method to get more genetic gain in Hanwoo breeding program. Finally, how these technologies can be used for multiplication of elite cow in small farm was discussed.
This study aims to identify DNA marker related to health index which is derived from fatty acid composition of Hanwoo meat. We investigated a genetic association between two SNPs (-867G>C and 878C>T) of SCD gene and health indexes. Two health index values (index of atherogenicity and index of thrombogenicity) were derived from a combination of fatty acid composition. Phenotypic correlation indicated that oleic acid (C18:1) was negatively correlated to index of atherogenicity (-0.84) and index of thrombogenicity (-0.91), respectively. Statistical analysis revealed that 878T>C SNP was significantly associated with IA (index of atherogenicity, p=0.012) and IT (index of thrombogenicity, p=0.006). There was no association between the regulatory SNPs (-867G>C and -877Gdel) and health indexes. Haplotype analysis detected 4 main haplotype (GdelT; 0.004, GdelC; 0.344, CGT; 0.350 and CGC; 0.261) in Hanwoo. The GdelT haplotype was significant on IA and IT. The effect of GdelT haplotype showed increasing IA and IT values, while GdelC haplotype has a decreasing IA and IT value in Hanwoo. In conclusion, the 878C>T SNP in the SCD gene seems to have an effect on this health index and might be implemented into animal breeding program.
Government and company are unfolding greenhouse gas reduction activity to prevent the effects of global warming. Also, verification business through greenhouse gas inventory construction is spreaded variously. Greenhouse gas verification proceeds by document examination, risk analysis, field survey. Document investigates emission information, calculation standard, emission report, data management system. And through risk assessment result, establish field verification plan. Through study on risk assessment of greenhouse gas inventory verification, wish to reduce risk of verification.
To compare the diversity and abundance of moth fauna between the deciduous and coniferous forest, the light traps have been managed at six survey sites in Hongneung, Korea Forest Research Institute (KFRI) from May to October 2008. Diversity and equitability were estimated by Simpson's index. A total of 1,181 individuals were collected, comprising 678 individuals (18 families, 145 species) and 503 individuals (17 families, 121 species) in the deciduous forest and the coniferous forest, respectively. Noctuidae was the most dominant family, which consist of 42 species, 248 individuals in the deciduous forest and 38 species, 149 individuals in the coniferous forest. The most dominant species was Hydrillodes morosa (Noctuidae) with 95 individuals (deciduous forest), followed by a pyralid moth, Orthopygia glaucinalis and a tortricid moth, Gnorismoneura hoshinoi with 36 individuals (coniferous forest). Interestingly, Arctiidae and Drepanidae, which were collected only in the deciduous and Lecithoceridae were only in the coniferous forest.
We have cloned the Bacillus cellulyticus K-12 avicelase(Avi, E.G.3.2.1.4) gene(ace A) in E. coli. This was accompanied by using the vector pT7T3U19 and Hind Ⅲ -HindⅢ libraries of Bacillus cellulyticus K-12 chromosomal inserts created in E. coli. The libraries were screened for the expression of avicelase by monitoring the immunoreaction of the anti-avicelase(immunoscreening). Positive clones(Ac-3, Ac-5, and Ac-7) contained the identical 3.5kb HindⅢ fragment as determined by restriction mapping and Southern hybridization, and expressed avicelase efficiently and constituvely using its own promoter in the heterologous host. From the immunoblotting analysis, a polypeptide which showed a CMCase activity with an Mr of 54000 was detected.
Seasonal and vertical distribution of water quality were investigated from May 2001 to June 2002 in Kumoh reservoir located nearby Kumi City, Kyungpook. Kumoh reservoir that lost the role of agricultural irrigation is currently of rapid eutrophication. The vertical distribution of DO was observed clinograde with hypolimnetic anoxic zone. T-P concentrations at the surface ranged from 0.008 to 0.152 mgP/L and T-N concentrations ranged from 1.4 to 3.0mgN/L. The vertical and seasonal variation of T-N was smaller than T-P. DOC concentrations, indicator of organic matter pollution, ranged from 2.8 to 5.4 mgC/L. Apportionment of Total-DOC (T-DOC) indicated that 14% of T-DOC was attributed to Labile-DOC(L-DOC) and the rest was due to Refractory- DOC(R-DOC). The values of TSI(Trophic State Index) ranged between 44 and 52 indicating that Kumoh reservoir is under mesotrophic condition. The results of this study indicate that Kumoh reservoir is likely to be under influence of eutrophication and thus water quality will be aggravated. Therefore, the Kumoh reservoir requires further treatment to improve water quality and a plan of the reusing water resource should be developed.
