        To develop mushroom varieties for cultivating at low temperature on oak logs, a strain with a low fruiting body generation temperature was crossed with Di-mon to select for a line with excellent properties. Selection was followed by cultivation testing. From these studies, Heunghwa 1ho was identified. The optimum temperature for cultivating Heunghwa 1ho, was 13.3oC. The fruiting temperature range was 6.4~20.2oC, identical to that of the parent strain. Growth at 25oC for 7 days achieved optimal mycelial growth of 61.9±2.10 mm, superior to growth of the parent strain at this temperature. The cap shape of Heunghwa 1ho was convex, cap diameter was 57.8±8.31 mm, and cap color was brown. Heunghwa 1ho showed similar genetic traits to those of the parental strain. However, dry weight (20.1 kg/m3) and cap diameter and color are superior to those of the parent strain. The 3 year fresh oak mushroom yield was 113.8 kg/m3, superior to the respective yields of the parent strains JMI 10047 and JMI 90021 (92.5 kg/m3 and 66.4 kg/m3).

        In this study, five different Lentinula edodes cultivar (Chamaram, Sanbaekhyang, Sanjo 713ho, Sanjo 715ho, Sanjo 718ho) were evaluated for their ability to decolorize Remazol Brilliant Blue R (RBBR) in MEB medium, respectively. Chamaram and Sanjo 713ho decolorized RBBR rapidly in MEB medium within 3 and 5 days. The activities of manganese peroxidase (MnP) and laccase were determined on the MEB medium with and without lignin. Sanjo 713ho resulted the highest ligninolytic enzyme activities on incubation day 1, indicating of 1,213 U/mg of MnP activity and 1,421 U/mg of laccase activity.

        In order to provide basic data for preparing management standards and to verify the safety of the Chinese oak mushroom-derived moisturizing medium—which is synthesized and imported in large quantities—the presence of 321 residual pesticides, 7 heavy metals, and 3 radioactive materials was analyzed in the moisturizer samples. Examination of residual pesticides in seven moisturizing medium samples prepared using the Chinese oak mushroom and three domestic sawdust samples used for mushroom culture revealed the presence of cypermethrin and iprodione in three moisturizer samples, but the contents of these pesticides were below the standard limits. Zn was detected in ten samples, Cu was detected in nine samples, and Ni was detected in four samples, but their contents were below the standard limits. Pb, Cd, Cr, and Hg were not detected in any sample. No radioactive materials were detected in the samples. In addition, fruiting bodies of the oak mushroom were observed in each medium. Examination did not reveal the presence of any residual pesticides or harmful compounds. In this study, the use of the moisturizing medium prepared using the Chinese oak mushroom was found to be safe. As residual pesticides, heavy metals, and radioactivity—even in trace amounts—remain concentrated in the human body, continuous verification of the safety of hazardous substances and pollutants during the systematic cultivation and management of these mushrooms is required.

        본 연구에서는 원목재배용 표고(Lentinula edodes) 국산 품종에 대한 장흥지역에서 적합한 표고품종 선발을 위해 국내산 총 5개(천백고, 수향고, 백화향, 풍년고, 산조303호) 품종과 일본산 2개(모리290, 모리유지로) 품종을 선택하여 적합품종 선발을 위해 재배시험을 실시하였다. 생산량 조사결과 산조303호가 40.9 kg으로 모리290 39.2 kg, 모리유지로 37.7 kg 보다 우수한 생산량을 보였다. 표고의 품질은 백화향이 갓이 반구형으로 형태가 우수하였으며, 화고형으로 발생하는 특징을 가진 것으로 확인되었다. 기존 재배중인 일본산 품종과 발생특성이 유사하며 생산량이 우수한 산조303호와 고품질의 표고를 생산하는데 유리한 백화향이 장흥지역에서 적합한 국산 표고품종으로 선발하였다.

        본 연구를 통해 국내 주요 한약재인 복령의 유해물질에 대한 안전성 확립과 허용기준치 설정의 기초자료를 제공하고자 한다. 국내 유통 중인 복령 10건(국내산 5건, 중국 산 5건)에 대한 잔류농약(321종), 중금속(7종), 방사능(3종), 회분 함량을 분석하였다. 잔류농약은 국내산 1건에서만 농약 성분인 cypermethrin 0.03mg/kg이 검출되었으나 기준치 이하로 나타났다. cypermethrin은 과수 및 원예농업에서 해충 방제의 살 충제로 사용되는 성분이다. 중금속은 Hg를 제외한 Zn, Pb, Ni, Cu, Hg, Cr가 모든 시료에서 검출됐지만 기준치 이하로 나타났다. Cd는 중국산 1건에서 0.011 mg/kg이 검출되었다. 방사능 농도도 모든 시료에서 MDA값 이하로 나타나 불검출 수준이었으며, 회분 함량도 모든 시료에서 기준치인 1.0% 이하로 나타났다. 이번 조사에서 국내에 유통 중인 복령의 유해물질에 대한 위해성은 안전하게 나타났다. 일반적으로 버섯류는 중금속 함량이 높다고 알려져 있다. 복령은 토양 속에서 생장하는 재배 특성상 유해물질에 항시 노출되어 있으므로 재배, 저장, 유통 등 생산단계 전반에 대한 지속적인 모니터링과 체계적인 관리기준 설정이 필요할 것으로 보인다.

        표고 재배사내 진균류의 발생특성을 분석하기 위해 골목에서 발생한 진균류를 분리·동정하고 재배사내 온·습도 환경을 분석하였다. 표고골목에서 발생한 진균을 분석한 결과 Cladosporium sp., C. cladosporioides, C. anthropophilum, Pleosporales sp., Trichoderma harzianum, Acremonium sp. 6종으로 확인되었다. 재배사내 환경을 조사한 결과는 5월 1일부터 10월 12일 사이에 재배사내 최고·저 온도가 14.0~30.1oC, 평균습도는 82.8%으로 나타났다. 이러한 결과는 표고 재배과정 중 진균류의 발생이 활발한 초여름부터 가을철 사이에 철저한 재배관리가 중요하며, 한번 발병한 진균류는 연중 재배사내 생존이 가능하기 때문에 주기적으로 모니터링하여 사전에 유해 진균류 발생에 유리한 생육환경을 예방해야 한다.

        국내 유통 중인 건조 목이 8건(국내산 3건, 중국산 5건) 을 수집하여 잔류농약 321종, 중금속 7종, 방사능 3종의 유해물질 잔류성분을 조사하였다. 잔류농약 검사결과 국내산 3건에서는 검출되지 않았으나, 중국산 5건 중 4건에서 Chlorpyrifos, Isoprocarb, Mepiquat chloride, Carbendazim이 검출되었다. 농산물 농약 잔류허용기준에 의하면 목이는 Mepiquat chloride (0.5 mg/kg 이하)만 기준치가 설정되어 있다. 또한 중금속 검사결과는 모든 건조 목이에서 미량의 중금속이 검출되었으나, 기준치 이하로 낮게 나타났다. 다만 국내산 시료 1건에서 납 함량이 기준치(0.3 mg/kg)보다 높게 나타나 정확한 원인 규명을 위해서는 톱밥배지, 지 하수, 보관·유통 과정에 대한 추가적인 조사가 필요할 것 으로 보여진다. 방사능 조사결과에서는 모든 건조 목이에서 방사능 농도가 MDA값 이하로 불검출 수준으로 나타났다. 국내산과 중국산의 평균 방사능 농도는 요오드( 131 I)와 세슘 ( 134 Cs)은 비슷한 수준으로 나타났으며, 세슘( 137 Cs)은 국내산보다 중국산에서 다소 높게 나타났다. 방사성 세슘 ( 137 Cs)은 버섯류에 잘 농축되는 것으로 알려져 있는데 이번 조사에서는 국내 유통 중인 건조 목이는 방사능에 안전하게 나타났다.

        본 연구에서는 원목재배용 표고(Lentinula edodes) 품종에 대한 목질섬유소 분해능을 검정하였다. 5개의 국산품종(천백고, 산조 303호, 풍년고, 백화향, 수향고)을 대상으로 malt extract broth(MEB) 배지에 lignin의 첨가에 따른 RBBR(Remazol Brilliant Blue R) 탈색능을 조사한 결과, 산조 303호와 풍년고가 배양 5일째부터 우수한 분해능을 보였다. 섬유소 분해효소인 MnP와 laccase의 활성은 풍년고가 배양 7일째 MnP 활성이 2,809U/mg, laccase 활성이 2,230U/mg으로 나타났고, 산조 303호가 배양 11일째 MnP 활성이 2,673U/mg, laccase 활성이 2,049U/mg으로 최고 활성을 나타났으며, lignin을 첨가하였을 때 효소의 활성이 증가하였다. 산조 303호, 풍년고와 수향고는 배양 5주 만에 filter paper의 분해정도를 육안으로 확인이 가능할 정도로 분해능이 우수하였다.

        본 연구는 표고균사체 발효 적하수오 열수 최적 추출조건 탐색을 목적으로 수행하였으며, 표고균사로 배양된 하수오의 독립변수로는 독립변수(Xi)로서 추출온도(X1), 추출시간(X2) 및 시료에 대한 용매비(X3)에 대한 실험범위를 설정하여 각각을 5단계로 부호화하였고 중심합성계획에 따라 16구로 설정하여 추출하였다. 또한 이들 독립변수에 영향을 받을 종속변수(Yn)로는 고형분 함량(Y1) 및 당도(Y2), TPC (total polyphenol contents, Y3), TFC (total flavonoid contents, Y4), ABTS cation radical scavenging activity (Y5), DPPH radical scavenging activity (Y6)으로 하였다. 고용분 함량 최대값과 최소값은 각각 31.99%와 16.84%로 나타났다. 이를 회귀분석한 결과 고형분 함량에 대한 모델식의 R2은 0.867로 나타났다. 당도는 7번(8 hrs, 85℃, 67 mg/mL)과 15번(6 hrs, 70℃, 100 mg/mL)시험구에서 3.0 brix로 다른 시험구에 비하여 높은 함량을 보였다. 항산화물질인 총 폴리페놀과 플라보노이드 함량은 8번 시험구(8 hrs, 85℃, 40 mg/mL)에서 총 폴리페놀 함량이 30.40 mg GAE/g, 총 플라보노이드 함량이 146.50 mg QE/g으로 가장 높게 나타났으며, 16가지 시험구의 항산화 활성도 총 폴리페놀과 총 플라보노이드 함량의 결과와 같이 8번 시험구(8 hrs, 85℃, 40 mg/mL)에서 가장 높은 활성을 보였다. 이상의 결과를 참조하여 표고균사체 발효 적하수오에 대한 열수 추출조건을 최적화할 목적으로 추출온도를 95℃로 고정하고 용매비와 추출시간에 대한 종속변수들의 contour map을 superimposing하여 추출물의 특성 중 생리활성물질 함량과 생리활성을 만족시켜주는 최적조건은 91.22℃에서 39.71 mg/mL 용매비와 7.72 추출시간으로 각각 분석되었다. 본 연구는 적하수오에 표고균사체를 배양한 후 생리활성이 우수한 원료를 추출 생산하고자 최적 추출조건을 확립하기 위하여 반응표면분석법을 활용하였다. 향후 생리활성부분에 대한 추가적인 연구가 이루어진다면 더욱 완성도가 높은 표고균사체 발효 적하수오 식품개발에 가능할 것으로 사료된다.

        This study was conducted to investigate the proximate composition, free sugars, amino acids, and minerals in five Lentinula edodes cultivars collected in Korea. No significant differences in the content of crude fat and crude fiber were found between the samples; however, there was significant variation in the contents of nitrogen free extract, ash, and crude protein. Three kinds of free sugars (fucose, arabinose, and glucose) were identified by HPLC, with the glucose content ranging from 5.94% to 12.08%. Sixteen kinds of free amino acids were identified: the highest content of amino acids was found in ‘Sanlim5ho’ (13,768.33 mg%), while the highest content of free amino acids was found in ‘Sanlim4ho’ (6,790.57 mg%). The minerals detected in the L. edodes cultivars were potassium, calcium, magnesium, and sodium, the contents of which ranged from 567.16–2,356.09 mg%, 3.85–7.42 mg%, 9.79–20.88 mg%, and 19.60–22.62 mg%, respectively.

        The purpose of this study was to expand the utility of the Polygonum multiflorum. Also, we fermented P. multiflorum by mushroom mycelial, and analyzed for chemical compositions and biological active of fermented P. multiflorum root. The crude protein content did not show a significant difference between control and fermented P. multiflorum root, crude fat, ash and crude fiber content of fermented P. multiflorum root were lower than control. The content of soluble nitrogen free extract of P. multiflorum root was significantly higher than fermented P. multiflorum root. The major amino acids of P. multiflorum root were determined the arginine. The content of arginine and glutamic acid were 586.67 mg%, and 283.78 mg%, respectively. Sixteen kinds of amino acids were detected in fermented P. multiflorum root, and the major amino acids were determined the arginine and threonine. The total amino acid contents of control and fermented P. multiflorum root were 3,469.03 mg%, and 3,630 mg%, respectively. As the results of antioxidant activity test, the antioxidant capacity of fermented P. multiflorum had a higher than the control. As the mushroom fermentation progresses, it is confirmed that the amino acid content and antioxidant capacity were increased, and it is expected to develop the product using the fermented P. multiflorum using mushroom mycelials.

        β-glucan is a safe and highly potent biological response modifier that nutritionally activates the immune response through the Macrophage, Dendritic and additional immune cells to yield various therapeutic effects. Shiitake mushrooms (Lentinula edodes) contanining β-glucans may be beneficial for human health; they have been used in the treatment of cancer, hypertension, and high cholesterol levels. The effect of different substrates and various developmental stages (mycelium growth, primordium appearance, and fruiting-body formation) on β-glucan production in the edible mushroom L. edodes was studied. The cap of the mature mushroom showed the highest β-glucan activity, and β-glucan activity seemed to be influenced somewhat by some well-known inducers or sawdust. In this study, we utilized five strains (JMI 10022, 10036, 10077, 10079, 10080) of L. edodes regardless of origin and growth conditions. This experiment showed that the expression of β-glucan was induced by glucose bond, and increased with the growing of L. edodes. Quantitatively, reverse transcription PCR utilized pairs of primers specific for β-glucan gene expression shows that expressed genes were most commonly indentified during the process of fruiting-body formation. We suggest that the results will provide valuable information to assist L. edodes industry.

        This study was performed to determine the optimal condition for Lentinula edodes JMI-10079 mycelium cultivation on the root of Polygonum multiflorum. We also analyzed the proximate composition, total amino acids, and minerals in the root of P. multiflorum cultivated with L. edodes JMI-10079 mycelia. The optimal temperature and pH for L. edodes JMI-10079 mycelium cultivation on the P. multiflorum root were 25oC and pH 5–6 respectively, whereas the optimal carbon and nitrogen sources were glucose and maltose, respectively. The content of crude protein, crude fat, and ash in the P. multiflorum root cultivated with L. edodes JMI-10079 mycelia was higher than that in the uncultivated P. multiflorum root. The content of crude fiber was the highest in the control. Total amino acid analysis revealed that the contents of total amino acids and total essential amino acids were increased by higher root of P. multiflorum concentration.

        팽이버섯 포장필름의 국산화 및 동남아시아 지역을 대 상으로 한 수출용 필름을 개발하기 위하여, 개발된 포장 필름을 현재 시중에 유통 중인 포장필름 및 수입산 포장 필름과 함께 저장 중 팽이버섯의 저장 온도별 품질변화에 이어 유리당, 유리아미노산, 구성아미노산 등에 대한 영양 학적 특성변화를 검토하였다. 유리당 함량을 분석한 결과, 저장 온도가 높아질수록 유리당 함량이 약간씩 증가하는 경향을 보였으나, 포장 필름의 종류에 따라서는 큰 차이 를 나타내지 않았다. 유리아미노산의 경우 저장온도가 올 라감에 따라 methionine은 증가하는 반면, tyrosine은 감 소하였다. Polyphenol oxidase의 활성 확인결과 본 과제 로 인해 개발된 포장지가 우수함을 확인하였다. 베타글루 칸 함량은 건강 기능성 식품의 표준 지표로 사용되는 성 분이나 버섯 저장성을 판단하는 지표로 사용하기에는 적 당하지 않을 것으로 여겨진다.

        Many wild mushrooms were collected at MT. Sambi areas for 1 day in July 2011. and 1 day in August 2011. They were identified and examined with references. According to the results, Species diversities are 1 division, 2 classes, 2 subclasses, 3 orders, 12 families, 18 genera and 24 species. Dominant families are Tricholomataceae. Resources of Wild Mushrooms were 4 species in edibility, 4 species in culture, 6 species in toxine, 5 species in medicine, 3 species in anticancer, 10 species in ectomycorrizahe and 12 species in rotten wood.

        Schizophyllum commune belongs to Schizophyllum, Schizophyllaceae, Agaricales, Agaricomycetes and Basidiomycotatina. The morphological shape of S. commune was similar with the cap, fan and hair. As for the sensory evaluation, chewness was not good for human's sensory. The major useful component was identified schizophyllan from S. commune extract. Schizophyllan has a great potential for developing medical supplies and functional foods. In our study, we carried out to internal transcribed spacer regions(ITS) for the specimens systematically. Also, we found out the optimal growth conditions of different temperature and media for S. commune cultivation. As the results of these test showed that when we compare with PDA, MCM and ME1 medium, we confirm that optimal temperature is 35℃ in MCM and ME1 medium. And optimal media of growth condition is ME1 and MCM.

        The biological activities of the various solvent extracts of mycelial Schizophyllum commune were investigated. The electron donating ability of mycelial S. commune was showcel 39%, 21%, 35% and 43% in Hot-water extract, water extract, ethanol extract and methanol extract at a 1,000 ㎍/㎖ concentration, respectively. As the results of SOD (superoxide dismutase)-like ability of Hot-water extract, water extract, ethanol extract and methanol extract of mycelial S. commune were showed 39%, 21%, 35% and 43% at a 1,000 ㎍/㎖ concentration, respectively. Measurement of ABTS radical cation scavenging activity of all extracts was showed more than 80% scavenging activity at a 1,000 ㎍/㎖ concentration. The tyrosinase inhibition effects related to skin-whitening by mycelial S. commune in Hot-water and water extract at 1,000 ㎍/㎖ were showed closely 5% activity and ethanol and methanol extracts were showed about 10% at 1000 ㎍/㎖ concentration. Antimicrobial activity of Propionibacterium acnes and Malassezia furfur were ineffective in mycelial S. commune extracts. These results showed that the mycelial S. commune has potential for the development of antioxidative, whitening effests and as a natural and functional additives.

        Many Wild Mushrooms were collected at Mt. Eokbul areas for 2 days in July 2010. and 1 day in August 2010. Identification in the collecting field by taking photos, recording the feature of the surroundings and ecological quality and then observing internal organs through an optical microscope. They were identified and examined with references. According to the result, Species diversities are 2 division, 2 classes, 1 subclass, 5 orders, 17 familys, 30 genera and 37 species. Dominant famillies are Tricholomataceae and Poryporaceae. Dominant species are Trichaptum abietum. Resources of Wild Mushrooms were 5 species in edibility, 6 species in clulture, 8 species in toxine, 6 species in medicne, 4 species in anticancer, 12 species in ectomycorrizahe and 16 species in rotten wood. Therefore the study is aimed at providing fundamental materials required for preserving Mt. Eokbul.

        A Lentinula edodes has abundant nutritious components such as a fiber, minerals and vitamins. It also was known to effective of anticancer and the immune enhancement. However, studies of mushroom-re1ated products were not still enough. Mushroom jerky was examined by using the under grade Lentinula edodes in this study. Content of stipe was most appropriate 24-30% in slices of dried mushroom. A grinder used 2mm sieve, and graund three or four times. A mushroom and spices were boiled to 30min in order to control the microbe occurrence. A mushroom, water, spices were mixed at ratio of 1 : 4: 1.5 and boiled 10 minutes for sterilization. The optimum drying temperature was 60~70℃ but the edge of slice was burned 80~90℃. Dry time was most appropriate for 4~5 hours.

        A Lentinula edodes was known to the excellent foods that abundant in nutritious components such as a fiber, minerals and vitamins. Fermentation conditions for rice wine were examined by using the medicinal herbs and the incuvated-rice with mycelia of Lentinula edodes. The average of ethanol contents was in the range of 3.4~4.1% at initial fermentation, increasing up to 16.9% after 15 days. Total acid, pH, reducing sugar and alcohol content were similar to the control during storage periods. The change of colors was measured by Hunter's color value during fermentation. The L value was the highest on 15th day of fermentation.
